Unguided web-based brief intervention with genetic risk education to reduce unhealthy alcohol consumption in Japan: Protocol for a randomized controlled trial.

BACKGROUND: Alcohol consumption is a major global contributor to morbidity and mortality and is a well-established risk factor for multiple cancers. Acetaldehyde, a toxic metabolite of ethanol, is a causal factor in alcohol-related esophageal cancer. A substantial proportion of the Japanese population carries the aldehyde dehydrogenase 2*2 (ALDH2*2) allele, which impairs ALDH2 enzymatic activity and increases acetaldehyde exposure, thereby elevating esophageal cancer risk. Among individuals with the ALDH2*2 allele, cancer risk demonstrates a clear dose-response relationship with alcohol consum

Alcohol Abstainer Status and Prazosin Treatment in Association with Changes in Posttraumatic Stress Disorder Symptoms in Veterans with Comorbid Alcohol Use Disorder and Posttraumatic Stress Disorder.

BACKGROUND: The noradrenergic system has been implicated in alcohol use disorder (AUD) and posttraumatic stress disorder (PTSD), with adrenergic agents reducing drinking in individuals with AUD and improving sleep disturbances in individuals with PTSD. In a recent clinical trial, prazosin, an α1-adrenergic antagonist, was not superior to placebo in reducing PTSD symptoms, sleep problems, or alcohol consumption in a comorbid population; however, patients in both treatment conditions improved in all symptom domains over the course of treatment. It remains unknown whether alcohol abstinence is re

Effect of alcohol consumption and psychosocial stressors on preterm and small-for-gestational-age births in HIV-infected women in South Africa: a cohort study.

OBJECTIVES: Psychosocial stressors such as depression and stress, intimate partner violence (IPV) and alcohol use have been linked to preterm and small-for-gestational-age (SGA) births in general populations. The prevalence of psychosocial stressors and alcohol abuse is high in many HIV-infected (HIV+) populations. Our objective was to evaluate the effects of psychosocial stressors and alcohol abuse on birth outcomes in HIV-infected women. METHODS: Antenatal depression and non-specific psychological distress, periconception IPV and alcohol consumption were measured during the second trimester

Binge alcohol consumption 18 h after induction of sepsis in a mouse model causes rapid overgrowth of bacteria, a cytokine storm, and decreased survival.

Alcohol abuse increases vulnerability to infections and infection-related mortality. In previous studies, we found that acute alcohol abuse in a binge-drinking model in mice decreased resistance to bacterial sepsis when alcohol was administered near the time of bacterial challenge. In the present study, we investigated the effects of alcohol administered later in the course of sepsis (18 h after injection of Escherichia coli). Our working hypothesis was that decreased production of cytokines caused by alcohol at this time would actually improve survival, because overproduction of pro-inflammat

Methylation Biomarker of Chronic Heavy Alcohol Consumption (HAC), but Not Acute HAC, Predicts All-Cause Mortality in Prostate, Lung, Colorectal and Ovarian Cancer Screening Trial.

BACKGROUND: Due to variability in patterns of consumption as well as well-known difficulties in obtaining valid self-report from heavy drinkers, quantifying the effects of heavy alcohol consumption on mortality is challenging. Using a DNA methylation biomarker of chronic heavy alcohol consumption (HAC) named the alcohol T-score (ATS), we previously showed that chronic HAC was a strong predictor of mortality. However, whether there is a similar effect when measures of shorter-term heavy alcohol use, i.e., recent "binge" drinking, were used to predict mortality was not examined. This is a critic

Navigating alcohol's impact: A mixed-methods analysis of community perceptions and consequences in Northern Tanzania.

BACKGROUND: Worldwide, alcohol is a leading risk factor for death and disability. Tanzania has particularly high rates of consumption and few resources dedicated to minimizing alcohol-related harm. Ongoing policy efforts are hampered by dynamic sociocultural, economic, and regulatory factors contributing to alcohol consumption. Through the voices of Kilimanjaro Christian Medical Centre (KCMC) patients, this study aimed to investigate community perceptions surrounding alcohol and the impact of its use in this region. METHODS: This mixed-methods study was conducted at KCMC between October 2021

Fetal growth and maternal alcohol consumption during early pregnancy.

OBJECTIVE: The relationship between light maternal alcohol consumption and fetal outcome remains contentious and the professional advice women receive is conflicting. The aim of this large epidemiological study was to examine the relationship between fetal growth and maternal alcohol behaviour before and during early pregnancy. STUDY DESIGN: Clinical and sociodemographic details of women who delivered a baby weighing ≥500 g during the eight years 2010-18 were analysed. Details on lifestyle behaviour before pregnancy and at the time of the first antenatal hospital visit were computerised using

A new model of interactive effects of alcohol and high-fat diet on hepatic fibrosis.

BACKGROUND: Alcoholic steatohepatitis (ASH) and nonalcoholic steatohepatitis (NASH) are the most frequent conditions leading to elevated liver enzymes and liver cirrhosis, respectively, in the Western world. However, despite strong epidemiological evidence for combined effects on the progression of liver injury, the mutual interaction of the pathophysiological mechanisms is incompletely understood. The aim of this study was to establish and analyze an experimental murine model, where we combined chronic alcohol administration with a NASH-inducing high-fat (HF) diet. METHODS: Balb/c mice were

Levels of gamma-aminobutyric acid-benzodiazepine receptors in abstinent, alcohol-dependent women: preliminary findings from an 123I-iomazenil single photon emission tomography study.

BACKGROUND: Although alcohol dependence in women is an increasing problem, little is known about the effects of alcohol on the female brain. Evidence from a few structural and functional neuroimaging studies suggests that the female brain may be more susceptible than the male brain to the harmful effects of alcohol. However, no in vivo studies of the neuropharmacology of alcohol dependence in women have been carried out. The aim of this preliminary study was to test the hypothesis that alcohol dependence in women is associated with greater reduction in gamma-aminobutyric acid (GABA)-benzodiaze

Skill vs. Disposition: Examining Paths of Intervention Effects in an Alcohol and Drug Use Prevention Trial Targeting U.S. Adolescents.

The goal of this study was to evaluate a mediation model of two approaches to deterring the onset of alcohol, cigarette, and marijuana among middle school adolescents. Students completed surveys that included yes/no self-reports about their past 30-day and lifetime alcohol, cigarettes, and marijuana use. Surveys assessed dispositions: perceptions that drug use would interfere with desired lifestyles, perceptions about drug use prevalence and acceptability, and drug use intentions. Surveys also assessed skills: students' ability to achieve goals, make decisions, and refuse drug use offers. Clas

Development of deep learning auto-encoder algorithms for predicting alcohol use in Korean adolescents based on cross-sectional data.

Alcohol is a highly addictive substance, presenting significant global public health concerns, particularly among adolescents. Previous studies have been limited by traditional research methods, making it challenging to encompass diverse risk factors and automate screening or prediction of adolescents' alcohol use. This study aimed to develop prediction algorithms for adolescent alcohol use in South Korea using machine learning (ML) and deep learning (DL) models, and to identify important features. The study utilized a combination of DL (i.e., Auto-encoder) and ML (i.e., Logistic regression, R

Youth cannabis and alcohol use expectancies mediate associations between pre-adolescent cognitive function and subsequent use initiation.

Youth substance use increases risk for developing substance use disorders. Previous work using the longitudinal ABCD Study® found distinct neurocognitive factors contribute to youth tobacco and alcohol initiation. Using data for 7776 ABCD Study® participants, this study expands prior work to examine prospective associations between early neurocognitive factors (general ability, executive function, learning and memory, and visuospatial and mental rotation) at ages 9-10 years (sample enrolled 2016-2018) and cannabis use by ages 13-14 years. We also test whether positive and negative tobacco smok

The relationship between nutrition and Ménière's disease.

Ménière's disease is an inner ear disease with attacks characterized by ear fullness, tinnitus, fluctuant sensorineural hearing loss and vertigo. Although pathophysiology of the disease is not fully known, endolymphatic hydrops are believed to play a role. Although there is no certain treatment procedure for Ménière's disease, some treatments are applied to prevent attacks, to treat the symptoms that occur during the attacks, and to prevent the permanent effects of the symptoms on the hearing and balance system. Lifestyle changes, dietary modifications, diuretics, vasodilator corticosteroids,

Drinking to death: Hyponatraemia induced by synthetic phenethylamines.

Synthetic phenethylamines are widely abused drugs, comprising new psychoactive substances such as synthetic cathinones, but also well-known amphetamines such as methamphetamine and 3,4-methylenedioxymethamphetamine (MDMA, ecstasy). Cathinones and amphetamines share many toxicodynamic mechanisms. One of their potentially life-threatening consequences, particularly of MDMA, is serotonin-mediated hyponatraemia. Herein, we review the state of the art on phenethylamine-induced hyponatremia; discuss the mechanisms involved; and present the preventive and therapeutic measures. Hyponatraemia mediated

Association of excise duty reforms with alcohol per capita consumption and alcohol-attributable life expectancy loss in Malaysia, 2000-2022: An interrupted time-series analysis.

BACKGROUND: Malaysia is a predominantly Muslim, upper-middle-income country where approximately 10% of the population consumes alcohol, yet heavy episodic drinking is common among drinkers. Despite multiple excise duty reforms between 2000 and 2022, their population-level impact on alcohol consumption and alcohol-attributable harm remains unclear. This study evaluates the effects of three major excise duty changes on adult alcohol per capita consumption (APC) and estimates alcohol-attributable life expectancy loss over time. METHODS: A longitudinal ecological design was applied using annual n

Association between alcohol consumption and mortality in Parkinson's disease.

Previous studies on the association between alcohol consumption and risk of Parkinson's disease (PD) have produced controversial results. However, the relationship between alcohol consumption and mortality in PD has scarcely been investigated. Among the nationwide population data from Korea National Health Insurance Service, newly diagnosed PD (ICD-10 code: G20 and a rare intractable disease registration code: V124), between 2009 and 2017, were selected. Alcohol consumption habit was obtained from a self-reported questionnaire on the National Health Screening Program. 32,419 individuals with P
