FABP3 Aggravates Cerebral Ischemia-Reperfusion Injury by Promoting Mitochondrial Lipid Accumulation and Enhancing BAX-Dependent Apoptosis.

We previously demonstrated that fatty acid-binding protein 3 (FABP3) is significantly upregulated in ischemic neurons, and its inhibition mitigates ischemic brain injury in mice and attenuates mitochondrial damage under rotenone-induced oxidative stress. These findings suggest a potential role for FABP3 in mitochondrial dysfunction in ischemic neurons, although the underlying mechanism remains unclear. In this study, we further investigated the role of FABP3 in mitochondrial injury and apoptosis in ischemic neurons. Our findings indicated that FABP3 deficiency significantly decreased infarct v

Exosomes derived from Bcl-2-engineered iPSC-cardiomyocytes mitigate aortic valve calcification and inhibit cardiomyocyte apoptosis.

Calcific Aortic Valve Disease (CAVD) is a progressive cardiovascular disorder, the pathological processes of which are correlated with cell apoptosis, inflammatory response, and osteogenic remodeling of valve interstitial cells. Presently, there exists no effective pharmacological treatment or intervention to impede or reverse the advancement of this disease. Systematically identifying key molecular biomarkers and their regulatory pathways is crucial for the formulation of novel therapeutic strategies. Exosomes, functioning as a novel drug delivery system, can transport specific functional mol

Ethacrynic Oxadiazole 6u Induces Apoptosis by Targeting mTOR/c-Flip Axis and Noxa in Lymphoma Cells.

BACKGROUND: Evasion of apoptosis is a major cause of therapy failure in lymphoma. We studied a covalent compound 6u with an α, β-saturated carbonyl group derived from the diuretic drug ethacrynic acid to induce apoptosis of lymphoma cells by targeting antiapoptotic proteins. RESULTS: We found that 6u induces apoptosis in 4 lymphoma cell lines, Daudi, Romas, Jeko-1 and Jurkat, accompanied by down-regulation of c-Flip and Mcl-1, as well as upregulation of Noxa. The down-regulation of Mcl-1 relies on Noxa induction and that the down-regulation of c-Flip is mediated by inhibiting protein synthesi

CREG1 restricts ALV-J replication via the mitochondrial dysfunction-driven activation of innate immunity and apoptosis.

BACKGROUND: Host antiviral defense relies on key regulatory genes that coordinate immune signaling and cellular homeostasis, yet their roles in J subgroup avian leukosis virus (ALV-J) infection remain poorly defined. Here, we identify cellular repressor of E1A-stimulated genes 1 (CREG1) as a key regulator of mitochondrial function and a critical immune-related gene involved in ALV-J infection. The objective of this study was to explore the effects and underlying mechanisms of CREG1 in the context of ALV-J infection. METHODS: In this study, transcriptomic analysis and RT-qPCR revealed that the

An Inhalable Apoptosis-Mimicking Functionalized Carbon Dots-Methylprednisolone Nanomedicine for Synergistic Therapy in Acute Lung Injury.

Acute lung injury (ALI) is a life-threatening inflammatory syndrome closely associated with reactive oxygen species (ROS) burst and dysregulation of M1/M2 macrophage polarization. Therefore, immune regulation of pulmonary macrophages represents a potential strategy to alleviate lung damage. However, existing approaches suffer from imprecise cellular targeting, monofunctional therapeutic effects, and suboptimal biocompatibility. To address these issues, an inhalable, multifunctional biomimetic nanoplatform is proposed that mimics apoptotic bodies (CTM@PS-Lip). CTM@PS-Lip is synthesized by fabri

Mitochondria-mediated apoptosis induced by acetogenins from Porcelia macrocarpa (Annonaceae) in K562 chronic myeloid leukemia cells.

BACKGROUND: Acetogenins from Annonaceae species are widely recognized for their potent cytotoxic and antitumor properties. Some acetogenin derivatives, such as bullatacin, have been reported to impair mitochondrial function, including inhibition of mitochondrial respiratory chain complex I. However, the cytotoxic effects of acetogenin derivatives isolated from Porcelia macrocarpa in leukemia cells remain poorly explored, as do the mitochondrial mechanisms underlying their biological activity. PURPOSE: The present study aimed to evaluate the cytotoxic potential of three acetogenins, namely (2S

Coenzyme Q10 exerts anti-cancer effects in bladder cancer by inducing ROS-mediated apoptosis and autophagy.

Bladder cancer (BCa) is one of the most common types of cancer that impacts the urinary system, and its incidence is increasing worldwide. Coenzyme Q10 (CoQ10) is a naturally occurring fat-soluble quinone with anti-oxidant and anti-inflammatory properties, and it plays a significant role in the suppression of tumor growth. Our objective was to elucidate the potential molecular mechanisms by which CoQ10 affects the advancement of BCa. In our study, human BCa cells 5637 and T24 and normal ureteral epithelial cells SV-Huc-1 were treated with different concentrations of CoQ10. Cell viability, reac

Vitamin K2 (MK7) alleviates arsenic-induced cardiac hypertrophy by suppressing the EGR3/BOK mitochondrial apoptosis pathway.

BACKGROUND: When arsenic combines with oxygen, chlorine and sulfur, it forms inorganic arsenic compounds. Studies have found that exposure to inorganic arsenic can cause pathological cardiac hypertrophy. MK7 is a long-chain menaquinone with superior bioavailability and extended half-life compared to other vitamin K2 homologues. BCL2-related ovarian killer (BOK) belongs to the BCL2 family and regulates mitochondrial membrane permeability to determine whether to initiate apoptosis. Early growth response 3 (EGR3) is a type of transcription factor that can specifically recognize and bind to its do

Ginsenoside Rh2 Attenuates Intervertebral Disc Degeneration by Modulating the PI3K/AKT Signaling Pathway to Suppress Apoptosis.

Intervertebral disc degeneration (IVDD) is a major cause of chronic low back pain and imposes a substantial socioeconomic burden. Ginsenoside Rh2 (Gin-Rh2), a bioactive ginseng compound with antioxidant properties, may have therapeutic potential in IVDD, but its effects and underlying mechanisms remain unclear. Here, we evaluated Gin-Rh2 in cellular and rat models of IVDD. In vitro, Gin-Rh2 attenuated interleukin-1β (IL-1β)-induced nucleus pulposus cell apoptosis, reduced matrix metalloproteinase-3 (MMP3) expression, and increased aggrecan and type II collagen production. Genetically Predicted 12,13-DiHOME Mediates the Association between TNF-Related Apoptosis-Inducing Ligand and Immune Thrombocytopenia.

INTRODUCTION: The role of Tumor Necrosis Factor (TNF)-Related Apoptosis-Inducing Ligand (TRAIL) has been highlighted in autoimmune and metabolic disorders. However, there is still an inadequate explanation of its causal relationship with primary immune thrombocytopenia (ITP). This study employed Mendelian Randomization (MR) to identify the potential causal associations among TRAIL, plasma metabolites, and ITP. METHODS: This study was conducted focusing on analyzing data from a Genome-Wide Association Study (GWAS) comprising inflammatory proteins, plasma metabolites, and the FinnGen consortium

MTHFD2 modulates neural stem cell proliferation and apoptosis after intracerebral hemorrhage by regulating mitochondrial NADPH homeostasis.

BACKGROUND: Intracerebral hemorrhage (ICH) represents one of the most severe forms of cerebrovascular injury, characterized by high mortality and lasting neurological dysfunction. Neural stem cells (NSCs), which are indispensable for neurogenesis, are mainly distributed in the hippocampus, a brain region essential for learning and memory. However, alterations in hippocampal NSCs following ICH and the mechanisms that mediate these changes remain poorly defined. The present study investigates the effects of ICH on hippocampal NSCs, focusing on the regulatory role of MTHFD2 in maintaining mitocho

Shear stress-induced Ca2+ influx triggers endoplasmic reticulum stress and cardiomyocyte apoptosis: implications for mitral regulation.

BACKGROUND: Mitral regurgitation is highly prevalent and elevates the risk of heart failure. Regurgitant flow induced shear stress disrupts Ca2+ homeostasis in atrial cardiomyocytes. Overloaded Ca2+ is a crucial regulator of endoplasmic reticulum stress-mediated apoptosis, although the regulatory mechanisms remain unclear. We aimed to explore the relationship between shear stress and Ca2+ homeostasis. METHODS AND RESULTS: Through employing an in vitro model replicating atrial cardiomyocytes during mitral regurgitation, we observed that shear stress increased Ca2+ oscillation and the amplitude

Heme and hemozoin induce platelet cell death through UPR-induced apoptosis and ferroptosis in vivax malaria.

Malaria is a highly prevalent infectious disease caused by Plasmodium parasites. Plasmodium intraerythrocytic replication leads to hemolysis-driven intermittent febrile crises in patients. In addition, the lysis of unparasitized red blood cells contributes to anemia and endotoxemia. Because thrombocytopenia is an important feature of vivax and severe falciparum malaria, we hypothesized that increased hemolysis in malaria contributes to severe thrombocytopenia by releasing endogenous and parasite toxins (ie, heme and hemozoin) capable of inducing programmed cell death in platelets. Using comple

Sea cucumber extract enhances glutathione-related antioxidant defense and reduces apoptosis in Eimeria papillata-infected mice.

Eimeria spp. are globally significant parasites infecting many vertebrate hosts and causing coccidiosis. The widespread use of anticoccidial drugs has led to increased parasite resistance, highlighting the need for alternative therapies. This study aimed to evaluate the anticoccidial, antioxidant, and anti-apoptotic effects of Holothuria polii extract (HpE) compared with the reference drug amprolium in mice experimentally infected with Eimeria papillata. Seven groups of mice were studied: a negative control; a group treated with HpE only (200 mg/kg); a positive control (infected untreated); an

Oxidative Stress-Mediated Mitochondrial Dysfunction Drives Genistein-Induced Apoptosis in TM4 Sertoli and ELT3 Leiomyoma Cells.

Genistein (GEN) exhibits concentration- and cell-type-dependent biological activities; however, the mechanisms underlying its cytotoxicity remain incompletely understood. This study investigated whether oxidative stress-mediated mitochondrial dysfunction contributes to GEN-induced apoptosis in TM4 mouse Sertoli cells and ELT3 rat leiomyoma cells. Cells were exposed to GEN (30 or 100 µM) for 48 h. Cytotoxicity, oxidative stress, mitochondrial function, and apoptosis were assessed by measuring cell viability, lactate dehydrogenase release, reactive oxygen species (ROS), malondialdehyde, glutathi
