Prognostic factors in patients with primary transitional cell carcinoma of the upper urinary tract.

A total of 50 patients with primary transitional cell carcinoma of the upper urinary tract underwent deoxyribonucleic acid ploidy characterization by flow cytometric analysis of paraffin embedded specimens. The primary tumor was diploid in 29 patients (58%) and aneuploid in 21 (42%). Aneuploidy was identified more frequently in grade 3 than in grades 1 and 2 neoplasms (p = 0.001). Additionally, grade 3 neoplasms occurred more often with invasive (stages T2 to T3) compared to superficial (stages TA, TIS and T1) tumors (p = 0.002). However, deoxyribonucleic acid ploidy was not significantly asso

Favorable outcomes in elderly patients undergoing high-dose therapy and autologous stem cell transplantation for non-Hodgkin lymphoma.

High-dose therapy and autologous stem cell transplantation (HDT-ASCT) can offer potential long-term remission or cure in patients with non-Hodgkin lymphoma (NHL). Limited experience is available on the safety and efficacy of HDT-ASCT in elderly patients. This is a single-center, retrospective study examining outcomes of HDT-ASCT for 202 NHL patients, ages 60 years and older, between January 2001 and December 2012. Overall survival (OS) and progression-free survival (PFS) were analyzed according to age at HDT-ASCT, hematopoietic cell transplantation comorbidity index (HCT-CI), NHL histology, an

Integrative bulk and single-cell transcriptomics identifies a HERC5-linked immune-attenuated risk program for pulmonary metastasis/relapse in osteosarcoma.

BACKGROUND: Pulmonary metastasis/relapse (PM/R) is a major cause of osteosarcoma treatment failure. We aimed to develop a compact transcriptomic PM/R risk score and characterize associated immune and single-cell programs. METHODS: TARGET-OS bulk RNA-seq was used for discovery and GSE21257 for external validation. Differential expression, multi-algorithm feature selection and LASSO regression were used to construct the score. Clinical associations were assessed by PM/R classification, survival and calibration analyses. Immune signature scoring, weighted gene co-expression network analysis (WGC

Galloyl-RGD induces oxidative stress-mediated apoptosis in human melanoma cells.

Melanoma is a malignant tumor arising from melanocytes and is characterized by rapid metastasis, making it the most aggressive form of skin … l electrophoresis and peptide mass fingerprinting suggested that Galloyl-RGD may influence melanoma cell survival and potentially trigger apoptosis-related pathways in SK-MEL28 cells. To validate these findings, we performed functional assays to evaluate the effects of Galloyl-RGD on cell viability, cell cycle progression, migration, and apoptosis in SK-MEL28 c …

Enteroblastic and Hepatoid Colorectal Carcinomas are Aggressive Cancers With a Distinctive Immunophenotype, While Clear Cell Carcinoma Appears to Represent Clear Cell Change in Conventional Colorectal Cancer.

Enteroblastic, hepatoid, and clear cell carcinomas are rare subtypes of colorectal carcinoma (CRC) with histologic and immunohistochemical (IHC) overlap. We assessed 37 CRCs that possibly represent these subtypes. Provisional criteria for each subtype, along with case photomicrographs and IHC data, were circulated among the authors to provide diagnostic interpretations. Based on plurality consensus, the carcinomas were classified as enteroblastic (n=13), hepatoid (n=6), and clear cell (n=18). Unanimous agreement was reached in 18% (3 clear cell, 3 hepatoid). Interobserver analysis revealed a m

Long-term outcomes of co-administration of CD19 and CD22 CAR-T cell therapy in pediatric patients with relapsed/refractory Philadelphia chromosome-positive acute lymphoblastic leukemia.

BACKGROUND: Despite initial responses, resistance to tyrosine kinase inhibitors (TKIs) and disease relapse remain major challenges in patients with Philadelphia chromosome-positive acute lymphoblastic leukemia (Ph+ ALL). Given the efficacy of chimeric antigen receptor T (CAR-T) cell therapy in Relapsed/Refractory (R/R) ALL, we aimed to evaluate the long-term outcomes of co-administration of CD19 and CD22 CAR-T cell therapy for pediatric patients with R/R Ph+ ALL. METHODS: We conducted a retrospective subanalysis of patients with R/R Ph+ ALL to assess the long-term outcomes of co-administratio

Incorporating Immunotherapies into the Evolving Standard of Care for Pediatric B-Cell Precursor Acute Lymphoblastic Leukemia.

Survival for pediatric B-cell precursor acute lymphoblastic leukemia (BCP-ALL) exceeds 90% in high-income countries due to risk-adapted chemotherapy and measurable residual disease (MRD)-guided strategies. However, relapse remains a leading cause of death, and chemotherapy-related toxicities highlight the need for equally effective, less toxic approaches. B-cell-directed immunotherapies targeting CD19 and CD22 have emerged as transformative modalities. This review synthesizes clinical evidence for three major immunotherapeutic classes: CD19/CD3 T cell engagers (blinatumomab), CD22 antibody-dru

UAP1 as a prognostic biomarker regulating malignant biological functions in multiple myeloma cells.

BACKGROUND: Multiple myeloma (MM) is a hematological malignancy characterized by proliferation of malignant plasma cells. UDP-N-acetylglucosamine pyrophosphorylase 1 (UAP1) has been shown to promote progression in various cancers. However, its specific role in MM remains unclear. METHODS: We analyzed six datasets comprising 3,117 patients from the Multiple Myeloma Research Foundation and Gene Expression Omnibus databases. The reliability of UAP1 as a prognostic predictor was also systematically evaluated. To further elucidate the specific mechanisms underlying the role of UAP1 in promoting MM

[Expert consensus on the management practice of CD19 CAR-T cell therapy for B-cell lymphoma in China (2025 edition)].

In recent years, the incidence of B-cell non-Hodgkin lymphoma (B-NHL) in China has shown a steady increase, accounting for approximately 85%-90% of all lymphomas. Although standard immunochemotherapy regimens such as R-CHOP have led to long-term remission in some patients, approximately 30%-40% still experience relapse or refractory disease, with dismal prognosis and a median survival of less than one year. For patients who fail multiple lines of therapy, conventional options such as chemotherapy, radiotherapy, or hematopoietic stem cell transplantation offer limited benefits, highlighting an

Prognostic value and therapeutic targeting of XPO1 in chronic lymphocytic leukemia.

Chronic lymphocytic leukemia (CLL) is a subtype of B-cell malignancy with high heterogeneity. XPO1 is highly expressed in many hematological malignancies, which predicts poor prognosis. In the study, we aimed to explore the prognostic role of XPO1 and the therapeutic effect of Selinexor, a selective inhibitor of nuclear export, which targets XPO1. We collected 200 CLL samples in our center to confirm XPO1 mRNA expression and analyzed the correlation between XPO1 expression and prognosis. Then, we decreased XPO1 expression with Selinexor to explore the effect of proliferation inhibition, cell c

Genetically re-engineered K562 cells significantly expand and functionally activate cord blood natural killer cells: Potential for adoptive cellular immunotherapy.

Natural killer (NK) cells play a significant role in reducing relapse in patients with hematological malignancies after allogeneic stem cell transplantation, but NK cell number and naturally occurring inhibitory signals limit their capability. Interleukin-15 (IL-15) and 4-1BBL are important modulators of NK expansion and functional activation. To overcome these limitations, cord blood mononuclear cells (CB MNCs) were ex vivo expanded for 7 days with genetically modified K562-mbIL15-41BBL (MODK562) or wild-type K562 (WTK562). Natural killer cell (NK) subsets and NK-like T-cell populations in acute myeloid leukemias and myelodysplastic syndromes.

The impact of the immune microenvironment on the behavior and therapeutic strategies for hematopoietic and lymphoid neoplasms is being increasingly recognized. Many functional studies of natural killer (NK) cell cytotoxic responses in myelodysplasia (MDS) and acute myeloid leukemia (AML) exist, but with limited data on these lymphocyte proportions and related T-cell subsets. The proportions of these cells and their prognostic implications were therefore investigated in 89 AML, 51 MDS, and 20 control marrows by flow cytometry. The median proportion of NK cells (relative to the total lymphocytes

Targeted radiotherapy potentiates the cytotoxicity of a novel anti-human DR5 monoclonal antibody and the adenovirus encoding soluble TRAIL in prostate cancer.

TRAIL (tumor necrosis factor-related apoptosis-inducing ligand) induces a death signal following binding to death receptors (DR4, DR5). We have developed a novel anti-human DR-5 monoclonal antibody (TRA-8) and adenoviral encoding TRAIL (Ad/TRAIL). Herein, we are testing the combined effect of radiotherapy and TRA-8 or Ad TRAIL in prostate cancer cells. Human prostate cancer cell lines LnCap, PC-3 and DU145 were used in this study. Cells were treated either with TRA-8 alone or Ad/TRAIL, radiation alone, or a combination of each at different doses and intervals. Cell survival using the MTS assay…

Prognostic implications of micoRNA miR-195 expression in human tongue squamous cell carcinoma.

BACKGROUND: miR-195 is aberrantly expressed in multiple types of disease. But little is known about the dysregulation of miR-195 in tongue squamous cell carcinoma (TSCC). In this study, we investigated the roles of miR-195 in the development and progression of TSCC. METHODS: Using quantitative reverse transcription-polymerase chain reaction (qRT-PCR), we evaluated miR-195 expression in TSCC samples from 81 patients. Overall survival of these patients was examined using Kaplan-Meier curves with log-rank tests and the Cox proportional hazards model. The expression of two known miR-195 target ge

Pathogenesis of mantle-cell lymphoma: all oncogenic roads lead to dysregulation of cell cycle and DNA damage response pathways.

Mantle-cell lymphoma (MCL) is a well-defined subtype of B-cell non-Hodgkin's lymphomas (B-NHL), accounts for approximately 6% of all lymphoid neoplasms, and has a median survival of 3 to 4 years. The genetic hallmark of MCL is the chromosomal translocation t(11;14)(q13;q32) that leads to deregulation and upregulation of Cyclin D1, an important regulator of the G1 phase of the cell cycle. This genetic event is present in virtually all cases of MCL, whereas additional genetic alterations that occur in subsets of MCL have been described. Most of these alterations appear to disturb the cell cycle

Survival of normal and magnesium-deficient erythrocytes in rats: effect of magnesium-deficient diet vs. splenectomy.

Magnesium deficiency in rats causes anemia, the mechanism of which is unknown. The effect of dietary magnesium, splenectomy, and magnesium content of erythrocytes on erythrocyte survival was studied in Fisher rats. Half of the animals were splenectomized, the remainder sham-splenectomized; each group was subdivided, and some were placed on a magnesium-deficient diet, the rest on a control diet. After 6 weeks, each of the four subgroups was divided, half were given 51Cr-labeled red cells from magnesium-deficient rats, the remainder, labeled normal red cells. The survival functions of the labele
