DNMT3A/TET2-mediated epigenetic alteration in Type-1 diabetes dysregulates the haematopoiesis.

Type 1 diabetes (T1D) is an autoimmune disorder characterized by insulin deficiency and hyperglycemia, driven by environmental insults and epigenetic reprogramming beyond genetic predisposition. Recent evidence suggests that systemic epigenetic dysregulation in T1D influences the differentiation of hematopoietic stem cells (HSCs) towards pro-inflammatory myeloid subsets, such as macrophages and dendritic cells, and compromises lymphoid tolerance, including Treg dysfunction. Mechanistically, perturbations in mitochondrial metabolism in HSCs disrupt the α-ketoglutarate/succinate ratio, which dir

Case Report: Investigation and characterization of a multiple endocrine neoplasia type 1 case and its pedigree.

Multiple Endocrine Neoplasia Type 1 (MEN1) is an autosomal dominant inherited disease characterized by the combined occurrence of tumors in multiple endocrine organs. As a rare disease, the clinical manifestations of MEN1 are currently considered to be associated with the development of combinations of more than 20 different tumors, such as parathyroid adenomas, neuroendocrine tumors, pituitary tumors, as well as lipomas, thymic carcinoids, pheochromocytomas, adrenal adenomas, and ovarian tumors. However, the concurrent occurrence of MEN1 and teratoma is extremely rare in reported cases to dat

Report of Concomitant Intracranial Cysts in Unrelated Patients With Heterozygous Germline NF1 Pathogenic Variants.

Neurofibromatosis 1 (NF1) is one of the most common genetic diseases of the central nervous system. The nature of intracranial lesions that are associated with NF1 are yet to be fully defined. Arachnoid, velum interpositum, and odontogenic cysts are among the more common intracranial cystic lesions that can be congenital. Although odontogenic cysts are well known to be associated with Gorlin-Goltz syndrome, the possibilities of other genetic disorders in patients with odontogenic and other intracranial cystic lesions have continued to stir research interests. Here, we report 2 cases of unrelat

Mitochondrial Quality Control: A Gatekeeper of Spermatogenesis.

Male infertility is rising worldwide. Spermatogenesis is energetically demanding and tightly coupled to mitochondrial quality control (MQC)-including biogenesis, fusion-fission dynamics, and mitophagy. Here we synthesize current evidence across developmental stages of germ cells, highlighting stage-specific shifts from glycolysis-supported ATP production to oxidative phosphorylation (OXPHOS)-dominated metabolism and remodeling of mitochondrial architecture into the midpiece sheath. We summarize genetic and functional data implicating OPA1/MFN1-2 in inner/outer membrane fusion, DRP1/FIS1/MFF in

[Evolution of integrated morphological prognostic indices in breast cancer diagnosis. Total malignancy score and nottingham prognostic index].

Breast cancer (BC) is a complex, clinically and morphologically heterogeneous disease for which integral morphological and molecular genetic parameters are of key importance. This paper describes the evolution of semiquantitative tumor grading systems, beginning with the classical Greenough, Patey-Scarff, and Bloom-Richardson systems and ending with modern mathematical methods based on the grade of BC malignancy. Integral prognostic indices are presented in detail, including the Nottingham Prognostic Index (NPI), its modifications (NPI+, METASTASIS), the Total malignancy score (TMS), as well a

Case report: an underdiagnosed multifocal ectopic Purkinje-related premature contractions caused by SCN5A mutation.

Multifocal Ectopic Purkinje-related Premature Contractions (MEPPC) syndrome is a recently recognized rare channelopathy caused by gain-of-function mutations in the SCN5A gene, leading to a high burden of premature ventricular contractions (PVCs) originating from the Purkinje system and often manifesting as a dilated cardiomyopathy-like phenotype. This case report presents a young woman without structural heart abnormalities or a known family history of similar conditions who was initially diagnosed with idiopathic fascicular ventricular arrhythmia (VAs) and underwent two unsuccessful radiofreq

Different Patterns of Lichen Planus in Three Members of One Family.

The pathogenesis of Lichen Planus (LP) and its variants, despite many investigative efforts, remains incompletely understood. The occurrence of the disease in siblings suggests a potential role for both genetic and shared environmental factors. Here, three Iranian siblings-one male and two females-who presented with various clinical forms of LP were introduced. Case No. 1 exhibited bilateral facial pigmentation and was diagnosed with Lichen Planus Pigmentosus (LPPigm). Case No. 2 presented with facial pigmentation accompanied by facial papules and was diagnosed with LPPigm and Lichen Planopila

CRISPR-based genome editing reveals the roles of efflux pumps in Mycobacterium abscessus.

Mycobacterium abscessus, one of the most antimicrobial-resistant bacteria, is increasingly recognized as the cause of infections that are difficult to treat. Novel genetic manipulation tools are required to elucidate the biology, pathogenesis, and antibiotic resistance mechanisms of M. abscessus. In this study, we modified the method used to prepare M. abscessus electrocompetent cells to achieve efficient transformation, and then optimized the CRISPR-Cas9-assisted genome-editing tools to allow efficient genetic manipulation. Using these tools, we constructed 66 efflux pump mutants of M. absces

Genetic regulation of lncRNA expression in whole human brain and their contribution to CNS disorders.

Long non-coding RNAs (lncRNAs) play a key role in the human brain, and genetic variants regulate their expression. Herein, expression quantitative trait loci of lncRNAs encompassing 10 brain regions from 134 individuals were analyzed, and novel variants influencing lncRNA expression (eSNPs) and respective affected lncRNAs (elncRNAs) were identified. The eSNPs showed proximity to their corresponding elncRNAs, enriched in the non-coding genome, and have a high minor allele frequency. The elncRNAs exhibit a high-level and complex pattern of expression. The genetic regulation is more tissue specif

Basal-shift transformation leads to EGFR therapy-resistance in human lung adenocarcinoma.

Although EGFR tyrosine kinase inhibitors (EGFR-TKIs) are effective for EGFR-mutant lung adenocarcinoma (LUAD), resistance inevitably develops through diverse mechanisms, including secondary genetic mutations, amplifications and as-yet undefined processes. To comprehensively unravel the mechanisms of EGFR-TKI resistance, we establish a biobank of patient-derived EGFR-mutant lung cancer organoids, encompassing cases previously treated with EGFR-TKIs. Through comprehensive molecular profiling including single-cell analysis, here we identify a subgroup of EGFR-TKI-resistant LUAD organoids that lac

Genomic analyses reveal new insights into Alzheimer's disease.

Alzheimer's disease (AD) is the most common cause of dementia, with global case numbers projected to reach 153 million in 2050 1 . AD is highly heritable, with twin-based heritability estimates of 60-80% 2 . While 1,200 causal loci are predicted to exist for AD 3 , approximately 80 have been associated with AD in two recent studies 4,5 , suggesting that many loci remain to be discovered 6 . Here, we analyzed data from 183,620 AD cases and 2.6 million controls from diverse ancestries, identifying 118 loci in a multi-ancestry analysis and 9 additional loci in ancestry-specific analyses, 48 of wh

Beyond the brain: The colorectal impact of Parkinson's disease.

Parkinson's disease (PD) is a rapidly developing neurodegenerative disorder characterized by degeneration of dopaminergic neurons in the basal ganglia of the brain. Its prevalence is estimated to exceed 1.2 million cases in the United States by 2030. Emerging evidence suggests that PD may originate in the gut and hence is linked to gastrointestinal (GI) dysfunctions such as inflammatory bowel disease and GI cancers. Colorectal cancer (CRC) is one of the most common cancers seen worldwide. It shares several risk factors with PD, including advancing age, male gender, genetic predispositions, and

Malignant Gastrointestinal Neuroectodermal Tumors: Challenging Tumors With Diverse Morphology and Different Considerations for Differential Diagnosis.

Malignant gastrointestinal neuroectodermal tumors (MGNET) are extremely rare malignant mesenchymal tumors derived from ectodermal neural cells of the gastrointestinal tract that most commonly arise in the small intestine. Preoperative diagnosis is challenging owing to the lack of well-established imaging characteristics. However, MGNET exhibits distinctive histopathological features, immunohistochemical profiles, molecular genetic characteristics, and ultrastructural manifestations under electron microscopy. Due to its aggressive clinical course, early diagnosis and intervention are crucial fo

Insights into ancestral diversity in Parkinson's disease risk: a comparative assessment of polygenic risk scores.

Risk prediction models play a crucial role in advancing healthcare by enabling early detection and supporting personalized medicine. Nonetheless, polygenic risk scores (PRS) for Parkinson's disease (PD) have not been extensively studied across diverse populations, contributing to health disparities. In this study, we constructed 105 PRS using individual-level data from seven ancestries and compared two different models. Model 1 was based on the cumulative effect of 90 known European PD risk variants, weighted by summary statistics from four independent ancestries (European, East Asian, Latino/

Integrative Computational Approaches for Understanding Drug Resistance in HIV-1 Protease Subtype C.

Acquired immunodeficiency syndrome (AIDS) is a chronic disease condition caused by the human immunodeficiency virus (HIV). The widespread availability of highly active antiretroviral therapies has helped to control HIV. There are ten FDA-approved protease inhibitors (PIs) that are used as part of antiretroviral therapies in HIV treatment. Importantly, all these drugs are designed and developed against the protease (PR) from HIV subtype B. On the other hand, HIV-1 PR subtype C, which is the most dominant strain in countries including South Africa and India, has shown resistance to PIs due to it
