BACKGROUND: Pyroptosis-mediated neuroinflammation represents a critical pathological mechanism in drug-resistant temporal lobe epilepsy (TLE), while Adipose-derived stem cell exosomes (ADSC-Exos) may target this process through NLRP3 inflammasome inhibition. Our study investigated the therapeutic effects of ADSC-Exos by mitigating NLRP3-driven pyroptosis in TLE.
METHODS: We isolated ADSC-Exos, the characteristics of which were confirmed. The Kainic acid-induced mouse TLE model were used to assess the in vivo effect of ADSC-Exos. To evaluate ADSC-Exos penetration, brain tissues were collected

The leucine-rich repeat kinase 2 (LRRK2) G2019S mutation is a major genetic risk factor for Parkinson's disease (PD). Most studies have shown that this mutation alone is insufficient to induce marked motor impairment or notable dopaminergic loss, and that pathological changes have been reported mainly in aging models or when additional stressors are applied. A decrease in progranulin (PGRN) is a major cause of frontotemporal dementia and has also been observed in patients with LRRK2 G2019S PD. In this study, we sought to determine how PGRN is regulated during the prodromal stage of PD. In 12-w

BACKGROUND: Behçet's disease (BD) is a complex systemic vasculitis with a poorly understood etiology that involves genetic, environmental, and immunological factors. Increasing evidence suggests that viral and bacterial infections may trigger or exacerbate BD through immune-mediated pathways. This review aims to clarify how different infectious agents may contribute to BD pathogenesis.
METHODS: For this review, articles addressing microbial involvement in BD were collected from established databases such as PubMed, Scopus, and Web of Science. Priority was given to studies evaluating classical

DNMT3A and TET2 are epigenetic regulator genes commonly mutated in age-related clonal hematopoiesis (CH). Despite having opposed epigenetic functions, these mutations are associated with increased all-cause mortality and a low risk for progression to hematologic neoplasms. Although individual impacts on the epigenome have been described using different model systems, the phenotypic complexity in humans remains to be elucidated. Here, we make use of a natural inflammatory response occurring during coronavirus disease 2019 (COVID-19), to understand the association of these mutations with inflamm

Neurodegenerative diseases, such as Alzheimer's disease, Parkinson's disease, and Huntington's disease, are characterized by progressive loss of neurons in the brain regions, including the hippocampus, cortex, substantia nigra, and striatum. Multiple pathological mechanisms drive this neuronal loss, including oxidative stress, chronic inflammation, mitochondrial dysfunction, protein misfolding, and excitotoxicity. Recent evidence suggests that these processes are intricately linked to the dysregulation of key signalling pathways, such as the IĸB/NFĸB, and KEAP1/Nrf2 pathways, which play centra

Even with reperfusion therapy, ischemia-reperfusion (I/R) injury remains to be a major driver of organ failure associated with myocardial infarction, stroke, and liver transplantation, with effective therapeutic targets still elusive. Using in vitro hypoxia/reoxygenation (H/R) models, we discovered that the pharmacological inhibition of 15-lipoxygenase (ALOX15) by thiolox effectively mitigates myocardial I/R injury. While ALOX15-a well-established promoter of lipid peroxidation and ferroptosis-has been extensively studied in cardiac I/R, its involvement in multi-organ I/R injury and non-ferrop

Accumulating evidence suggests that NLRP3-mediated alveolar macrophage (AM) pyroptosis and subsequent high mobility group box protein 1 (HMGB1) secretion play significant roles in the pathogenesis of acute respiratory distress syndrome (ARDS). Nrf2 has been shown to be individually involved in regulating pyroptosis. In this study, we investigate the ability of CDDO-imidazolide, a potent Nrf2 activator, to regulate AM pyroptosis and HMGB1 secretion in sepsis-associated ARDS, along with its underlying mechanism. The in vitro alveolar macrophage (AM) pyroptosis model, established by stimulating J

The female reproductive system ages before any other physiological system, making it a sensitive indicator of aging. Early reproductive aging is associated with the early onset of infertility and an increased risk of several diseases. During aging, systemic and reproductive oxidative stress and inflammation levels increase through inflammasome activation, leading to ovarian follicle loss. Other markers of reproductive aging include increased fibrosis and shortening of telomeres in ovarian cells. The factors that accelerate reproductive aging are unclear, but likely involve exposure to endocrin

Over 90% of patients with myelodysplastic syndromes/neoplasms (MDS) exhibit anemia at diagnosis, primarily due to ineffective erythropoiesis. This is characterized by abnormal proliferation and differentiation of erythroid cells influenced by signaling pathways including heme synthesis, ferroptosis, senescence and apoptosis. Despite widespread anemia, the specific mechanisms and pathway alterations at different disease stages are not well understood. This study employed the NUP98-HOXD13 (NHD13) transgenic mouse model, which mimicked the erythroid changes observed in MDS patients, to explore th

Benign prostatic hyperplasia (BPH) is a prevalent urological disorder in aging men, frequently linked to hormonal fluctuations. Agomelatine (AGO), a melatonergic receptor agonist and serotonin 2C antagonist, has demonstrated anti-inflammatory and antioxidant properties. This study aimed to explore the potential of AGO in mitigating BPH induced by testosterone propionate (TP) in rats. Male rats received TP (5 mg/kg/day, subcutaneously) to induce BPH and were pretreated with AGO (80 mg/kg/day) for 28 days. AGO inhibited the rise in prostate index, serum prostate-specific antigen (PSA), and testo

Traumatic brain injury (TBI) triggers a series of pathophysiological events, contributing significantly to secondary injury and long-term functional deficits. While exosome therapy is beginning to emerge as a promising avenue for various injuries, its efficacy in TBI, using preclinical models that mimic the biomechanics of human acceleration/deceleration TBI, remains largely unexplored. This study investigated the capacity of human Schwann cell-derived exosomes (hSC-Exo) to improve outcomes in a model of moderate fluid percussion injury (FPI). We found that jugular infusion of hSC-Exo 30 min

Obesity, characterized by excessive body fat accumulation due to energy metabolism imbalance, has emerged as a significant risk factor for male infertility. Icariin (ICA), a principal constituent of Epimedium, has demonstrated potential in improving obesity and enhancing testicular spermatogenesis. This study aimed to elucidate the role and underlying mechanisms of ICA in mitigating obesity-induced male infertility through the suppression of inflammation and insulin resistance. Utilizing a high-fat diet (HFD)-induced obese mouse model, we investigated ICA's effects on pyroptosis, insulin resis

Hydrogen sulfide (H2S) is a vital signaling molecule involved in various physiological processes; however, the mechanisms underlying its systemic signaling remain poorly understood. We hypothesized that albumin, the predominant plasma protein and a vital sulfhydryl carrier, mediated systemic H2S signaling, which could potentially treat H2S-deficient diseases. This study aimed to investigate this hypothesis. Our results showed the presence of sulfhydrated proteins in normal mouse serum, with albumin being particularly enriched. The level of sulfhydration was influenced by H2S availability and t

Olfactory dysfunction is one of the earliest non-motor symptoms of Parkinson's disease (PD), accompanied by abnormal α-synuclein aggregation in the olfactory bulb (OB). We previously reported that Cordycepin (Cor), a potential anti-inflammatory and anti-oxidative adenosine derivative, has emerged as an attractive candidate for PD treatment. However, existing investigations have predominantly focused on the amelioration of motor symptoms rather than modulating the disease course from prodromal stage. This study aimed to evaluate if Cor exerts a disease-modifying effect at the prodromal stage pr

Hepatic ischemia reperfusion (HIR) injury is a complication that complicates major liver surgeries and contributes to significant hepatic and remote organs damage. Aprepitant (Ap), a neurokinin-1 receptor (NK-1R) antagonists, is an antiemetic commonly used in preventing chemotherapy-induced nausea and vomiting. This study aimed to evaluate the potential protective effect of Ap against acute lung injury (ALI) associated with HIR, utilizing the Pringle maneuver to induce 30 min of hepatic ischemia followed by 1 h of reperfusion, while targeting the NLRP3/IL-1β signaling pathway. Polycystic ovary syndrome (PCOS), a common endocrine disorder affecting over 10% of women, is characterized by hyperandrogenism and ovarian dysfunction. While linked to chronic inflammation and granulosa cell apoptosis, its molecular mechanisms remain unclear. In this study, we elucidated the novel role of Krüppel-like factor 5 (KLF5) in the pathogenesis of PCOS and its regulatory role with thioredoxin interacting protein (TXNIP). In a mouse model of PCOS induced by dehydroepiandrosterone (DHEA), KLF5 expression was significantly elevated in ovarian tissues (up-regulated 2.62-fold, P < 0.001),

BACKGROUND: Hyperuricemia (HUA) is a chronic disease caused by abnormal purine metabolism with high prevalence. Dihydromyricetin (DMY) is a natural flavonoid that is abundant in plants, such as vine tea, grapes and bayberry. DMY has been shown to possess multiple biological properties, but its anti-HUA effect remains underexplored. In the present study, the regulatory effects of DMY on HUA and its complications and mechanism were investigated.
RESULTS: DMY (10 and 20 μmol L-1) treatment significantly reduced xanthine oxidase (XOD) expression and uric acid (UA) synthesis in normal human liver

INTRODUCTION: Podocytopathies are a uniquely renal disease syndrome, in which direct or indirect podocyte injury leads to proteinuria or nephrotic syndrome. Of the many factors that contribute to podocytopathies, the abnormal regulation of autophagy, such insufficient or excessive autophagy levels, have been proposed to play a significant role in the occurrence and development of podocytopathies. However, there still has been a lack of systematic and comparative research to elucidate exact role of autophagy in podocytopathies and its current research status. This study aims to utilize bibliome
