Antimicrobial peptides (AMPs) are promising alternatives to conventional antibiotics for the treatment of multidrug-resistant (MDR) pathogens; however, their clinical translation is limited by proteolytic degradation. We developed dPA-13, an all-D-enantiomer of the 13-mer AMP PA-13, to improve resistance. dPA-13 exhibited potent bactericidal activity against Pseudomonas aeruginosa, superior protease stability, and a favorable safety profile. Confocal imaging and comparative proteomics revealed distinct mechanistic differences: while PA-13 localized to the membrane, dPA-13 exhibited enhanced cy

Bacterial biofilm-associated infections (BBAIs) and antibiotic resistance constantly threaten human health and life. Herein, we report a series of anionic or mixed-charge copolypeptides that can simultaneously prevent and eradicate Pseudomonas aeruginosa and Staphylococcus aureus biofilms without the development of bacterial resistance. The copolypeptides composed of anionic l-glutamic acid (E), hydrophobic l-phenylalanine (F), and/or cationic l-lysine (K) residues were readily synthesized by ring-opening polymerization and side-chain modifications. Anionic or mixed-charge copolypeptides with

OBJECTIVES: Staphylococcus epidermidis (S. epidermidis) is a common opportunistic pathogen in clinical settings. The widespread misuse of antibiotics has led to a continuous increase in its antimicrobial resistance, greatly complicating treatment. Drug repurposing, characterized by low cost, high safety, and short development cycles, represents an effective strategy for combating drug-resistant bacterial infections. This study aims to investigate the antibacterial activity of the antiparasitic drug resorantel against S. epidermidis and to preliminarily elucidate its antibacterial mechanism.

UNLABELLED: Colistin remains a last-line antibiotic against multidrug-resistant Gram-negative pathogens, but its clinical utility is limited by nephrotoxicity and emerging resistance. This study identifies macelignan, a natural lignan from nutmeg, as a potent synergist that enhances colistin activity against Escherichia coli (E. coli) through a novel metabolic disruption mechanism. In vitro, macelignan reduced the colistin minimum inhibitory concentration (MIC) by approximately 100-fold, with the combination (8 µg/mL macelignan + 0.008 µg/mL colistin) achieving complete bacterial eradication w

The prevalence of macrolide-resistant Mycoplasma pneumoniae in Japan is increasing once again. The therapeutic efficacy of quinolone garenoxacin was compared with minocycline, which is recommended as the first-choice drug in patients with macrolide-resistant M. pneumoniae pneumonia. Defervescence within 48 h after initiation of antibiotics was observed in 90.6% of garenoxacin and 90% of minocycline recipients. Our results indicated that garenoxacin can be considered as the first-choice drug for the treatment of macrolide-resistant M. pneumoniae pneumonia.

The rapid emergence of multidrug-resistant (MDR) bacteria poses a critical global health threat, severely limiting the effectiveness of current antibiotics. Polymyxin B (PMB) remains a last-line treatment for MDR Gram-negative infections; however, its clinical use is restricted by nephrotoxicity, neurotoxicity, and the growing prevalence of resistance. This study reports the development of PMB-loaded silver nanoparticles (AgNPs) stabilized with polyvinylpyrrolidone (PVP) as a novel antimicrobial delivery system. The nanoparticles were synthesized via chemical reduction and optimized for PMB lo

BACKGROUND: We aimed to examine the accuracy of 0.25 μg/mL-interval minimum inhibitory concentration (MIC) measurement of vancomycin (VCM) for Staphylococcus aureus.
METHODS: We collected microbiological data on S. aureus from June 2020 to November 2023 at Okayama Univer-sity Hospital, Japan. The VCM MIC values were determined by the microdilution method using Dry Plate Eiken (Eiken Chemical Co., Ltd, Tokyo, Japan) at ≤ 0.5, 0.75, 1, 1.25, 1.5, 1.75, and 2 µg/mL. We performed day-to-day reproducibility testing for S. aureus standard strain (ATCC 29213) and evaluated the within-run reproducibi

Facial thread lifting is widely employed in minimally invasive rejuvenation for its rapid recovery. However, its clinical acceptance is heavily limited by common complications, particularly local infections and severe inflammatory responses. Herein, a multifunctional silk fibroin (SF) lifting thread incorporating tannic acid (TA) and gentamicin (GEN) was fabricated using a custom-designed mold with a biomimetic fishbone architecture and was denoted as TA/GEN@SFs. TA/GEN@SFs exhibited excellent mechanical properties, with a breaking force of up to 40.00 ± 2.30 N and an elongation at break of 13

The increasing threat of healthcare-associated infections and mosquito-borne diseases demands new materials that combine antimicrobial protection, insect repellency, and biocompatibility. Here, we convert Stellera chamaejasme L. (SC), an ecologically harmful invasive weed, into a multifunctional biocomposite (SCZrB/WPU) by integrating SC root fibers, bamboo waste, zirconium phosphate (ZrP), and waterborne polyurethane (WPU). The resulting material exhibits >99.99% antibacterial efficacy against S. aureus and E. coli, and >89.5% repellency against Aedes albopictus. It releases only 0.14 mg L-1

Rapid antibiotic susceptibility tests (ASTs) are essential for quick selection of effective drugs to treat infections at an early stage. However, the most widely used phenotypic ASTs in clinical practice often require 24 to 48 hours of pre-culture enrichment and 4 to 20 hours of testing. They are too slow to guide therapy, even with the most rapid protocol. Here, we report a lab-on-a-silicon chip (LOSC) system, which integrates arrays of silicon nanowire field-effect transistor (SiNWFET) sensors with high-throughput cell-collection microfluidics for rapid ASTs. The microfluidics concentrate ba

Spinal tuberculosis (TB), previously known as Pott’s disease, is the most common bone infection caused by Mycobacterium tuberculosis. The diagnosis of spinal TB is based on clinical, radiographic, microbiological, and histological findings. Its indolent nature, subacute progression, paucibacillary state, and difficulties in obtaining adequate samples often result in delayed diagnosis. The emergence of multidrug-resistant (MDR) and extensively drug-resistant (XDR) TB has posed significant challenges in the management of TB infections. Here, we present a case of pre-XDR tuberculosis of the spine

UNLABELLED: Clofazimine (CFZ) is a key drug used to treat drug-resistant tuberculosis (DR-TB), while pyrifazimine (TBI-166) is an improved riminophenazine derivative with better pharmacokinetics. However, there is a lack of data on its susceptibility and resistance in regions with a high disease burden, such as southwestern China. We compared the in vitro antimicrobial activities of CFZ and TBI-166 against 249 DR-TB clinical isolates (99 multidrug-resistant TB [MDR-TB] and 150 pre-extensively drug-resistant TB [pre-XDR-TB] isolates) from southwestern China. TBI-166 exhibited a concentration-de

This study evaluated the in vitro efficacy of antimicrobial and antibiofilm of three disinfectant-loaded nanoparticles (AgNPs-H₂O₂, CuNPs-Virkon S, and CuNPs-TH4) against Vibrio alginolyticus and Vibrio fluvialis, isolated from naturally infected gilthead seabream in Egypt. Qualitative Congo red assay showed that V. fluvialis isolates were strong biofilm producers, whereas V. alginolyticus showed limited phenotypic variation. Quantitative assay results, analyzed with a Mann-Whitney U test, showed no statistically significant difference in overall biofilm biomass between V. alginolyticus and V.

AIMS: Candida albicans and Staphylococcus aureus polymicrobial biofilms cause chronic wound infections and tolerate standard antimicrobials poorly. Their endurance is driven by fungal hyphal morphogenesis, interkingdom co-aggregation, and virulence proteins, including SAP5, which create a strong polymicrobial biofilm. Homovanillic acid, a plant-derived phenolic molecule, was tested for its antibiofilm effects on hyphal growth, bacterial-fungal interactions, and SAP5-associated pathogenicity.

BACKGROUND: With the increasing severity of antimicrobial compound resistance, exploring the combined effects of novel antibacterial compounds is of significant research value. Polylysine possesses cationic antibacterial activity, while glutathione is renowned for its antioxidant function. However, systematic research on the ionic complexes formed by their electrostatic self-assembly is insufficient. This study preliminarily evaluated the thermal stability and in vitro antibacterial activity of the polylysine-glutathione ionic complex, aiming to optimize its synthesis process and explore its f

The escalating crisis of antibiotics against Gram negative bacteria resistance demands innovative strategies to combat recalcitrant intracellular pathogens. We reposition RS 17053, an α1A-adrenoceptor antagonist with unexplored antibacterial function, as a mechanistically distinct broad-spectrum antibiotic. We proved RS 17053 has strong binding ability with Class A penicillin binding proteins (PBPs), which overcomes the β-lactam barrier by penetrating host cells and disrupts the biosynthesis of peptidoglycan by uniquely targeting MrcA/MecA. RS 17053 eradicated Gram-negative (Brucella, Salmonel
