Intracellular Mycobacterium leprae Utilizes Host Glucose as a Carbon Source in Schwann Cells.

New approaches are needed to control leprosy, but understanding of the biology of the causative agent Mycobacterium leprae remains rudimentary, principally because the pathogen cannot be grown in axenic culture. Here, we applied 13C isotopomer analysis to measure carbon metabolism of M. leprae in its primary host cell, the Schwann cell. We compared the results of this analysis with those of a related pathogen, Mycobacterium tuberculosis, growing in its primary host cell, the macrophage. Using 13C isotopomer analysis with glucose as the tracer, we show that whereas M. tuberculosis imports most …

Composition of three essential oils, and their mammalian cell toxicity and antimycobacterial activity against drug resistant-tuberculosis and nontuberculous mycobacteria strains.

Tuberculosis (TB) is the most ancient epidemic disease in the world and a serious opportunistic disease in HIV/AIDS patients. The increase in multidrug resistant Mycobacterium tuberculosis (MDR-TB, XDR-TB) demands the search for novel antimycobacterial drugs. Essential oils (EOs) have been widely used in medicine and some EOs and their major components have been shown to be active against M. tuberculosis. The aim of this work was to evaluate the antimycobacterial and cell toxicity activities of three EOs derived from Salvia aratocensis, Turnera diffusa and Lippia americana, aromatics plants co…

Susceptibility of raccoons (Procyon lotor) to infection with Mycobacterium bovis.

Tuberculosis due to Mycobacterium bovis infection is endemic in white-tailed deer (Odocoileus virginianus) in the northeastern portion of the lower Michigan peninsula (USA). Various wild carnivores and omnivores, including raccoons (Procyon lotor), are infected with M. bovis within the endemic area. To investigate the pathogenesis of tuberculosis in raccoons and the likelihood of M. bovis transmission from infected raccoons to other susceptible hosts, we experimentally inoculated raccoons with single oral doses of M. bovis (ranging from 30 to 1.7 x 10(5) colony forming units [CFU]), five daily

Bone marrow examination for the diagnosis of mycobacterial and fungal infections in the acquired immunodeficiency syndrome.

In a series of 342 bone marrow examinations from 314 patients with human immunodeficiency virus infection, 70 examinations (20%) detected opportunistic mycobacterial or fungal infections. One hundred eleven of the 314 patients had such infections, and, hence, 63% (70/111) were detected by bone marrow examination. Special stains for microorganisms detected 16 (32%) of 50 Mycobacterium avium complex infections, 10 (22%) of 45 Mycobacterium tuberculosis infections, eight (73%) of 11 Histoplasma capsulatum infections, and five (83%) of six Cryptococcus neoformans infections. Bone marrow cultures d…

Modeling the Impact of Case Finding for Tuberculosis: The Role of Infection Dynamics.

BACKGROUND: Many individuals previously infected with Mycobacterium tuberculosis (Mtb) may clear their infections or experience substantially reduced progression risks over time. Such dynamics suggest that recent transmission is more important in driving TB incidence in high-burden settings than previously estimated; thus, the impact of interventions to reduce transmission (eg, community-based active case finding) may also be greater than previously thought. METHODS: We constructed 2 models of Mtb transmission that differed only in the inclusion of a clearance mechanism. We calibrated these m…

Bone tuberculosis: molecular pathogenesis, diagnostic advances, therapeutic challenges, and emerging strategies for effective management.

Bone tuberculosis (BTB) remains a diagnostic and therapeutic challenge despite advances in molecular medicine, accounting for 10–15% of extrapulmonary tuberculosis cases with disproportionate morbidity in immunocompromised populations. This comprehensive review synthesizes current understanding of BTB pathogenesis, diagnostic innovations, and therapeutic strategies while highlighting critical knowledge gaps. At the molecular level, Mycobacterium tuberculosis exploits bone-specific niches through granuloma formation, immune evasion mechanisms involving autophagy inhibition and cytokine dysregul…

Translational bacterial biomarkers and pharmacodynamic models in the treatment of tuberculosis.

Treatment of Mycobacterium tuberculosis is uniquely prolonged and complex among bacterial infections due to its unusual propensity for persistence during drug stress. Persisters are subpopulations of organisms tolerant to bactericidal antibiotics and characterized by a slower rate of elimination in the absence of genetic changes associated with resistance. The biological basis of this response is increasingly well understood and novel translational tools have become available that are capable of identifying and characterizing such organisms in vitro and in vivo. Recent in vivo and clinical obs…

Harnessing miRNAs: A Novel Approach to Diagnosis and Treatment of Tuberculosis.

Mycobacterium tuberculosis (Mtb) complex, responsible for tuberculosis (TB) infection, continues to be a predominant global cause of mortality due to intricate host-pathogen interactions that affect disease progression. MicroRNAs (miRNAs), essential posttranscriptional regulators, have become pivotal modulators of these relationships. Recent findings indicate that miRNAs actively regulate immunological responses to Mtb complex by modulating autophagy, apoptosis, and immune cell activities. This has resulted in increased interest in miRNAs as prospective diagnostic indicators for TB, especially…

Tuberculosis preventive therapy: scientific and ethical considerations for trials of ultra-short regimens.

Preventive therapy remains key to the elimination of tuberculosis and is typically offered to people with presumptive Mycobacterium tuberculosis infection to prevent active disease. Although the duration of tuberculosis preventive therapy has been reduced substantially over time, it remains long in absolute terms, and uptake remains low. Treatment-shortening trials using non-inferiority designs have so far led to the implementation of effective regimens of 1-4 months' duration. Such regimens are a substantial improvement on the previous 6-9 months' duration standard of care but still far too l…

Nano-Enabled Effective Tuberculosis Treatments: A Concise Overview.

Tuberculosis (TB) is a severe respiratory infectious disease caused by Mycobacterium tuberculosis (M.tb), which puts enormous pressure on public health and economic systems worldwide. Therefore, accurate diagnosis and timely intervention of TB are critical for interrupting disease transmission and reducing mortality among TB patients. However, the low bioavailability, inadequate targeting, and significant adverse side effects of conventional antibiotics and the emergence of the multidrug-resistant M.tb strain result in limited TB treatment efficacy or even the development of multidrug-resistan…

Lab-in-a-cartridge for real-time detection of tuberculosis via precise measurement of urinary lipoarabinomannan.

Current methods for detecting Mycobacterium tuberculosis (M.tb) in centralized medical facilities are a bottleneck in TB surveillance, particularly in resource-constrained regions. In response, we present a groundbreaking portable bio-tool, the lab-in-a-cartridge (LIC) system, designed for on-site detection of lipoarabinomannan (LAM) in trace amounts within the urine. The innovative design combines pumpless liquid handling and magnetic force-based enrichment with horseradish peroxidase polymer amplification to precisely quantify low biomarker levels. Employing a tetramethylbenzidine-based colo…
