Functional and structural connectivity of thalamic subnuclei in major depressive disorder at 7 Tesla.

AIMS: Major depressive disorder (MDD) is widely considered to be a mood disorder characterized by altered connectivity. The thalamus plays an important role in MDD by connecting large areas of the brain. Here, we explored thalamic connectivity in MDD at the subnuclear level using ultra-high-field MRI. METHODS: We combined ultra-high-field functional and diffusion MRI at 7.0 Tesla to map the connectivity of thalamic subnuclei in MDD patients (n = 47) and healthy controls (n = 13). We segmented thalamic subnuclei and calculated the functional and structural connectivity of thalamic subnuclei an

Eight-year longitudinal changes in functional connectivity of the fatigue network in Parkinson's disease: evidence from four timepoints.

Fatigue is a prevalent non-motor symptom in Parkinson's disease (PD), yet its long-term neural substrates remain poorly understood. The aim of this study was to characterize eight-year, four-timepoint longitudinal changes in the fatigue-related brain functional connectivity (FC) in PD and examine their associations with fatigue severity. Fifty-one PD patients and 37 healthy controls (HC) were recruited. All variables were assessed at four timepoints: baseline (year-1), year-3, year-5, and year-8. Region of interest (ROI)-to-ROI analyses targeted a predefined fatigue-related network. A visual n

Re-experiencing trauma: Occipital lobe resting-state functional connectivity in posttraumatic stress disorder.

Re-experiencing symptoms in posttraumatic stress disorder (PTSD) often involve vivid visual imagery, yet occipital resting-state functional connectivity (rsFC) remains understudied, particularly in women exposed to interpersonal violence. Clarifying these neural mechanisms may advance understanding of sensory and contextual processing disturbances that contribute to intrusive memories. Notably, occipital rsFC has not been evaluated in relation to re-experiencing using the Posttraumatic Stress Diagnostic Scale (PDS), a validated self-report measure that supports fine-grained symptom-level analy

Distinct within- and between-network functional dysconnectivity of the default-mode and frontoparietal networks in young individuals with first-episode bipolar disorder and major depressive disorder.

BACKGROUND: Evidence of whether disruptions in functional connectivity (FC), particularly within the default-mode network (DMN) and frontoparietal network (FPN), differ between first-episode bipolar disorder (BD) and major depressive disorder (MDD) is inconsistent. METHODS: This study included 31 individuals (mean age, 25 years) with first-episode BD, 31 with MDD, and 31 healthy controls. Resting-state FC was assessed using magnetic resonance imaging. FC was analyzed in the DMN and FPN by using Dosenbach's 160 functional regions of interest. RESULTS: FC within the DMN (ventromedial prefronta

Precuneus-to-hippocampus connectivity links LTP-like plasticity to cognitive function in subjective cognitive decline and mild cognitive impairment.

BACKGROUND: Disruptions in synaptic plasticity and alterations in effective connectivity (EC) involving the hippocampus and amygdala are hallmarks of early Alzheimer's disease (AD). However, the interplay between these neurophysiological changes and their relationships with cognitive functions in subjective cognitive decline (SCD) and mild cognitive impairment (MCI) remains poorly understood. METHODS: Transcranial magnetic stimulation (TMS) and resting-state functional magnetic resonance imaging (rs-fMRI) were used to assess long-term potentiation (LTP)-like plasticity and EC involving the am

Value-based cognitive control moderates the relation of inflammation with depression in adolescents.

Depression has been linked to both elevated systemic inflammation and altered brain function related to reward processing and cognitive control. We know little, however, about how these factors jointly confer risk for this disorder, especially during adolescence, when depressive symptoms typically emerge. In this study, we examined brain function during an incentivized go/no-go task in a community sample of 100 adolescents (mean [SD] = 15.8 [1.1] years). We assessed neural activation and task-dependent functional connectivity using a psychophysiological interaction analysis during the contrast

Altered Interictal Bed Nucleus of Stria Terminalis Connectivity in Patients With Temporal Lobe Epilepsy.

BACKGROUND AND OBJECTIVES: Temporal lobe epilepsy (TLE) is a highly prevalent neurologic disorder, with 30%-50% of patients developing drug-resistant epilepsy. Pharmacoresistant seizures remodel critical arousal and respiratory networks, impairing autonomic function and chemoreception and putting patients at increased risk of adverse respiratory events and sudden unexpected death (SUDEP). Given that the bed nucleus of stria terminalis (BNST) serves as a key relay between brainstem respiratory nuclei and cortical arousal networks, we characterized interictal BNST connectivity alterations in pat

Electroencephalography source-space functional connectivity reveals frequency-specific brain network dysfunctions in obsessive-compulsive disorder.

BACKGROUND: Obsessive-compulsive disorder (OCD) is characterized by disruptions in large-scale brain networks. However, the role of high-frequency neural synchrony in these abnormalities remains unclear. Elucidating frequency-specific alterations may offer critical insights into the neurophysiological mechanisms underlying brain network dysfunction in OCD. METHODS: We analyzed resting-state electroencephalography data from 88 OCD patients and 100 healthy controls (HCs), and constructed source-space functional connectivity networks using weighted phase lag index. Graph-theoretical metrics incl

Effects of chronic ethanol exposure on dorsal medial striatal neurons receiving convergent inputs from the orbitofrontal cortex and basolateral amygdala.

Alcohol use disorder is associated with altered function of cortical-amygdala-striatal circuits such as the orbitofrontal cortex (OFC), basolateral amygdala (BLA) and their connections to the dorsal medial striatum (DMS) shown to be involved in goal-directed actions. Using retrobead tracing, we previously reported enhanced excitability of DMS-projecting OFC neurons in mice following 3-to-7-day withdrawal from chronic intermittent ethanol (CIE) exposure. In the same animals, spiking of DMS-projecting BLA neurons was decreased at 3-days post-withdrawal followed by an increase in firing at 7- and

Effect of a single dose of lorazepam on resting state functional connectivity in healthy adults.

Lorazepam is a fast-acting benzodiazepine that is widely used to manage anxiety symptoms through modulation of GABAergic activity. Despite being one of the most prescribed benzodiazepines, the effects of a single dose of lorazepam on brain functional connectivity at rest is not known. In this placebo-controlled, crossover study, twenty healthy adult participants (9 women; 26.2 ± 5.2 years) underwent two resting-state fMRI (rs-fMRI) scans following administration of either lorazepam (2.5 mg) or placebo. Imaging data were analyzed using an individual brain network parcellation approach and diffe

Audio-visual crossmodal connectivity in perceiving distorted speech: A comparison between patients with cleft palate and typical listeners.

This study investigates audio-visual crossmodal connectivity during the perception of distorted speech using task-based functional magnetic resonance imaging (fMRI). A randomized block design was employed, involving 20 patients with cleft palate and 20 typical listeners. Participants underwent perceptual tasks involving both cleft-related glottal stop and typical speech while fMRI scans were conducted. Regional interactions between the auditory and visual cortices were analyzed using dynamic causal modeling (DCM). Individual-level effective connectivity analysis revealed that, during the perce

The ventral hippocampus and nucleus accumbens as neural substrates for cocaine contextual memory reconsolidation.

Addictive drugs engage molecular pathways of associative learning and memory. Drug craving triggered by cues that were once associated with drug intoxication contributes to continued drug-seeking behaviors. Reactivated memories are vulnerable to disruption by interference with the process of reconsolidation, hence targeting reconsolidation could be beneficial in reducing cue-induced drug craving and relapse. Here, we sought to identify the neuronal pathways and neuroplasticity involved in cocaine contextual memory reconsolidation. Mice expressing inhibitory DREADDs in either nucleus accumbens

Extinction of Heroin Seeking Does Not Require the Infralimbic Cortex or Its Projections to the Nucleus Accumbens Shell or Amygdala.

Evidence indicates that the activity of the infralimbic cortex (IL), as well as its projections to the nucleus accumbens shell (NAshell) and amygdala, following an unreinforced lever press is critical for cocaine extinction learning and retention. It is unclear whether the same neural circuitry is involved in extinction encoding for other classes of addictive drugs, including opioids. In this study, we used a behaviour-controlled optogenetic approach in female and male Sprague-Dawley rats to examine the role of the IL and its projections in the extinction of heroin seeking. Rats first underwen

Alterations in static and dynamic functional network connectivity in chronic low back pain: a resting-state network functional connectivity and machine learning study.

Low back pain (LBP) is a prevalent pain condition whose persistence can lead to changes in the brain regions responsible for sensory, cognitive, attentional, and emotional processing. Previous neuroimaging studies have identified various structural and functional abnormalities in patients with LBP; however, how the static and dynamic large-scale functional network connectivity (FNC) of the brain is affected in these patients remains unclear. Forty-one patients with chronic low back pain (cLBP) and 42 healthy controls underwent resting-state functional MRI scanning. The independent component an

Structural Cerebellar and Lateral Frontoparietal Networks are altered in CUD: An SBM Analysis.

Repetitive drug use results in enduring structural and functional changes in the brain. Addiction research has consistently revealed significant modifications in key brain networks related to reward, habit, salience, executive function, memory and self-regulation. Techniques like Voxel-based Morphometry have highlighted large-scale structural differences in grey matter across distinct groups. Source-based Morphometry (SBM) takes this a step further by incorporating the Independent Component Analysis to detect shared patterns of grey matter variation, all without requiring prior selection of re

Melanin-Concentrating Hormone Projections to the Nucleus Accumbens Enhance the Reward Value of Food Consumption and Do Not Induce Feeding or REM Sleep.

Regulation of food intake and energy balance is critical to survival. Hunger develops as a response to energy deficit and drives food-seeking and consumption. However, motivations to eat are varied in nature and promoted by factors other than energy deficit. When dysregulated, nonhomeostatic drives to consume can contribute to disorders of food intake, adding to the increasing prevalence of restrictive eating disorders and obesity. Melanin-concentrating hormone (MCH) neurons have been implicated in the regulation of feeding behavior, in addition to a number of other fundamental behaviors inclu
