Precision exosome engineering for neurological therapeutics: molecular mechanisms and targeted strategies.

Exosomes are nanoscale extracellular vesicles which are essential for intercellular communication. They have shown great promise in the delivery of drugs into the brain. Recent developments in the use of exosomes to treat neurological conditions are reviewed in this article. Further, it also focuses on molecular and genetic changes that improve the therapeutic effectiveness of these exosomes. Receptor-specific targeting, stimulus-responsive exosomes, and hybrid nanovesicle systems are the three main approaches investigated for facilitating exosome transport across the blood-brain barrier (BBB)

Gut microbiota and Parkinson's Disease: a new frontier in understanding neurological health.

Increasingly recognized as a neurodegenerative disease with motor manifestations and progressive cognitive decline, PD has more frequently been linked to the gut microbiome. The gut-brain axis, a bidirectional communication system between the gut and brain, plays a crucial role in PD pathogenesis. Exploration of the intricacies in the interplay between PD and the gut microbiome, together with the important mechanisms involved, will form the basis of this review. Gut microbiome activities as contributors to PD actions include altered intestinal permeability, neuroinflammation, alpha-syn aggrega

[Questionnaire survey about barriers to telemedicine for patients with intractable neurological disorders].

Telemedicine is not widely used in patients with intractable neurological disorders including neurodegenerative diseases even though its application was covered by national insurance in Japan. To clarify what patient factors should be overcome for telemedicine application, we surveyed outpatients with intractable neurological disorders using questionnaires in Tottori University Hospital, Tekeda General Hospital and Fukuoka University Hospitals. The survey showed that more than half of the telemedicine-beginners did not want to use telemedicine in the former two hospitals located in provincial

Subacute intranasal oxytocin improves neurological recovery after ischemic stroke.

Oxytocin (OXT) is a neuropeptide known for its anti-inflammatory and neuroprotective properties; however, its role in post-stroke recovery remains poorly defined. In this study, we investigated whether intranasal OXT administration during the subacute phase of stroke improves neurological outcomes and modulates microglial responses. Male mice underwent permanent middle cerebral artery occlusion and received intranasal OXT (300 ng/g) or saline on days 3 and 5 post-stroke. Neurological function was assessed using the modified neurological severity score; infarct volume was evaluated through hema

Elucidating the mechanistic role of Rip3 in post-thalamic hemorrhage neurological deficits.

Thalamic hemorrhage (TH), a critical subtype of intracerebral hemorrhage, often leads to central post-stroke pain (CPSP) and neurological deficits, yet its molecular mechanisms remain poorly understood. This study investigated the role of Rip3 in TH pathophysiology by constructing Rip3-knockout (KO) mice and integrating behavioral assessments, transcriptomic sequencing, and molecular experiments. Results demonstrated that TH induced motor dysfunction, mechanical pain hypersensitivity, working memory impairment, and anxiety-like behaviors in wild-type (WT) mice, while Rip3 knockout significantl

An Overview of Neuroprotective Effects of Erythropoietin Against Neurological Disorders: Beyond Erythropoiesis.

Brain related disorders are a set of medical ailments that cause motor incoordination, cognitive and memory problems due to neurodegeneration in the brain. Although current therapies alleviate symptoms, they fail to target the fundamental pathological processes driving the disorders. A hematopoietic growth factor, Erythropoietin, stimulates erythroid cell formation and is therapeutically applied for the treatment of anemia. Furthermore, Erythropoietin has shown improved neurological outcomes in preclinical models and is being investigated as a potential therapeutic agent for neurodegenerative

A potential drug target in epilepsy and its role in other neurological disorders.

Epilepsy (EP) has an increased incidence due to several diseases, including neurodegenerative diseases and hemorrhagic stroke. The proteome-based research on developing suitable therapeutic interventions and disease prevention is beneficial. Cis-acting protein quantitative trait loci from 885 plasma and CSF proteins were used to explore potential drug targets for EP. After we got the preliminary association of TMEM106B with EP by drug target Mendelian randomization (MR), co-localization results identified TMEM106B as a potential drug target for EP further. The protein-protein interaction netwo

The space-time continuum in neurological disorders of the autophagosome-lysosome fusion machinery.

Autophagy is a highly conserved cellular pathway for the degradation and recycling of defective intracellular cargo and plays a vital role in the homeostasis of post-mitotic tissues, particularly the nervous system. Autophagosome-lysosome fusion represents the final critical step in macroautophagy with a tightly regulated process mediated by a complex molecular machinery of tethering vesicles for degradation. Since the first reports of human autophagy disorders, the scientific and clinical focus condensed on severe phenotypes with biallelic-truncating genotypes as monogenic models of near-comp

The Recent Applications of Stem Cell-Derived Exosomes and Hydrogels in Neurological Disorders.

Neurological disorders such as Alzheimer's disease, Parkinson's disease, and stroke pose significant challenges for conventional therapy due to the complexities of the blood-brain barrier (BBB) and the restricted delivery of drugs to the central nervous system. Exosomes, a type of small extracellular vesicle secreted by nearly all cell types, hold substantial promise as delivery vehicles for therapeutic agents in treating these conditions. Notably, stem cell-secreted exosomes have emerged as particularly effective due to their regenerative potential and natural ability to cross the BBB. Simila
