BACKGROUND: Non-B subtypes account for at least 50 % of HIV-1 infections diagnosed in Belgium and Luxembourg. They are considered to be acquired through heterosexual contacts and infect primarily individuals of foreign origin. Information on the extent to which non-B subtypes spread to the local population is incomplete.
METHODS: Pol and env gene sequences were collected from 410 non-subtype B infections. Profound subtyping was performed using 5 subtyping tools and sequences of both pol and env. Demographic information, disease markers (viral load, CD4 count) and viral characteristics (co-rec

OBJECTIVE: Current treatment options for epithelial ovarian cancer are limited and therapeutic development for recurrent and drug-resistant ovarian cancer is an urgent agenda. We investigated the potential use of genetically engineered Vesicular Stomatitis Virus (VSV) to treat ovarian cancer patients who fail to respond to available therapies. Specifically, we examined the toxicity to hosts and specificity of targeting ovarian tumors using a Wv ovarian tumor model.
METHODS: We first tested recombinant VSV for oncolytic activity in a panel of human ovarian epithelial cancer, immortalized, and

Cyclo-oxygenase-2 (COX-2) is believed to induce neuronal oxidative stress via production of radicals. While oxygen radicals are not directly involved in COX-2-catalytic cycle, superoxide anion radicals have been repeatedly reported to play a critical role in COX-2-associated oxidative stress. To resolve the controversy, we characterized production of free radicals in PC12 cells in which COX-2 expression was manipulated either genetically or by direct protein transfection and compared them with those generated by a recombinant COX-2 in a cell-free system. Using spin-traps alpha-(4-pyridyl-1-oxi

The toothless (tl) osteopetrotic mutation in the rat affects an osteoblast-derived factor that is required for normal osteoclast differentiation. Although the genetic locus remains unknown, the phenotypic impact of the tl mutation on multiple systems has been well characterized. Some of its actions are similar to tumornecrosis factor superfamily member 11(TNFSF11; also called TRANCE, RANKL, ODF and OPGL) null mice. TNFSF11 is a recently described member of the tumor necrosis factor superfamily which, when expressed by activated T cells, enhances the survival of antigen-presenting dendritic cel

INTRODUCTION: Hypoparathyroidism is a rare endocrine disorder caused by deficient secretion or action of parathyroid hormone (PTH), leading to hypocalcaemia, hyperphosphataemia, and disrupted calcium-phosphate homeostasis. Postsurgical hypoparathyroidism is the most common form, whereas autoimmune, genetic, metabolic, and infiltrative causes are less frequent. Chronic PTH deficiency is associated with renal, skeletal, neuromuscular, ocular, and soft tissue complications that impair quality of life.
AREAS COVERED: Conventional treatment with oral calcium and active vitamin D corrects hypocalca

The strain Pseudomonas putida KT2440 has been genetically modified to synthesize 2,3-butanediol (2,3-BDO). To achieve this goal, we have used as a host the P. putida KT2440 acoB::mini-Tn5 mutant unable to mineralize acetoin and 2,3-BDO. This strain has been further engineered by overexpressing in the pIZ2 plasmid, using the inducible promoter Ptrc, the budABC operon from Klebsiella oxytoca, which encodes the three enzymes required to transform pyruvate into 2,3-BDO. Flask and bioreactor experiments were carried out for the optimization of the 2,3-BDO production using acetate or glucose as subs

Secondary lymphedema is a chronic disease affecting an isolated limb following lymph node resection for cancer treatment. Management options are limited and onerous, leading to near-universal progression to subcutaneous fibroadipose tissue deposition. Here, we identify bone morphogenetic protein (BMP) ligands as mediators of fibroadipose tissue deposition through in vitro experiments with human lymphedema fluid and BMP-specific inhibitor. Systemic in vivo delivery of BMP inhibitor reduces fibroadipose tissue deposition in a mouse model of hindlimb secondary lymphedema. Considering systemic del

Type VII secretion system (T7SS) substrate EsxA contributes to the pathogenesis of several Gram-positive bacteria, but its role in viridans streptococci-induced infective endocarditis (IE) remains unclear. Genomic analysis of the Streptococcus gordonii DL1 strain identified a genetic locus encoding five T7SSb core machinery proteins (EsaA, EssA, EsaB, EssB, and EssC) and one substrate, EsxA. T7SSb mediates EsxA secretion, as evidenced by reduced secretion in the essC-deleted mutant and restoration upon complementation. In a rat model of IE, the esxA-deficient strain shows lower survival in cir

Infantile-onset Pompe disease (IOPD) is a rare genetic disorder associated with a deficiency of a lysosomal enzyme, the acid alpha-glucosidase. It is characterized by the accumulation of lysosomal and non-lysosomal-bound glycogen in various organs, such as the heart, skeletal muscle, and brain tissue, resulting in muscle weakness, hypertrophic cardiomyopathy, respiratory insufficiency, and other complications. Without treatment, IOPD has a very high mortality rate, with patients dying within the first year of life. Over the past few decades, significant therapeutic advancements have been made

INTRODUCTION: Pompe disease (PD) is a rare genetic disorder that leads to intralysosomal glycogen accumulation because of a deficiency in the lysosomal enzyme acid α-glucosidase (GAA), which is required to break down glycogen to glucose. Enzyme replacement therapy (ERT) with recombinant human GAA (rhGAA) supplies exogenous GAA to reduce glycogen deposits, thereby improving motor and respiratory functioning.
AREA COVERED: The first approved ERT for PD was the rhGAA alglucosidase alfa. Limitations associated with this treatment led to the development of two other rhGAAs: avalglucosidase alfa an

On 01 August 2025, the United Kingdom became the first country in the world to implement a targeted immunisation programme using a meningococcal vaccine (4CMenB) for protection against gonorrhoea. 4CMenB is a recombinant, protein-based vaccine licensed for prevention of serogroup B meningococcal disease but, because Neisseria meningitidis is genetically closely related to Neisseria gonorrhoeae, observational studies estimate that the vaccine also provides some (30-41%) protection against gonorrhoea. Given the rising incidence of gonorrhoea and increasing antimicrobial resistance, the UK progra

Hemophilia A (HA) is a common X-linked recessive hereditary bleeding disorder. Coagulation factor VIII (FVIII) is insufficient in patients with HA due to the mutations in the F8 gene. The restoration of plasma levels of FVIII via both recombinant B-domain-deleted FVIII (BDD-FVIII) and B-domain-deleted F8 (BDDF8) transgenes was proven to be helpful. FVIII-Padua is a 23.4 kb tandem repeat mutation in the F8 associated with a high F8 gene expression and thrombogenesis. Here we screened a core enhancer element in FVIII-Padua for improving the F8 expression. In detail, we identified a 400 bp effici

Real-time PCR assays are valuable tools for the rapid and accurate diagnosis of infectious diseases by identifying the nucleic acid sequences of pathogens. Here, we aimed to develop a recombinant plasmid-based standard for validating the sensitivity of different molecular diagnostic methods adopting the Jonstrup assay (J assay). Chimeric plasmid DNA (cpDNA) harboring various pathogen genes and the target site of the J assay was constructed. Our findings revealed that the J assay could detect a single copy of the target gene similar to digital droplet PCR. The detection sensitivity of each esta

Pseudoxanthoma elasticum (PXE) is a genetic multisystem ectopic calcification disorder caused by inactivating mutations in the ABCC6 gene encoding ABCC6, a hepatic efflux transporter. ABCC6-mediated ATP secretion by the liver is the main source of a potent endogenous calcification inhibitor, plasma inorganic pyrophosphate (PPi); the deficiency of plasma PPi underpins PXE. Recent studies demonstrated that INZ-701, a recombinant human ENPP1 that generates PPi and is now in clinical trials, restored plasma PPi levels and prevented ectopic calcification in the muzzle skin of Abcc6-/-mice. This stu

Autosomal recessive hypophosphatemic rickets type 2 (ARHR2; MIM #613312) is a very rare disorder caused by biallelic loss-of-function mutations in the ENPP1 (ectonucleotide pyrophosphatase/phosphodiesterase 1) gene. ENPP1 deficiency encompasses a spectrum of phenotypes that includes, in addition to ARHR2, generalized arterial calcification of infancy (GACI), ossification of the posterior longitudinal ligament (OPLL), and pseudoxanthoma elasticum. ARHR2 can be found in GACI survivors, but it may also be the first manifestation of ENPP1 deficiency. Although the precise mechanisms are not fully e

Japanese encephalitis (JE), caused by the Japanese encephalitis virus (JEV), is a highly threatening disease with no specific treatment. Fortunately, the development of vaccines has enabled effective defense against JE. However, re-emerging genotype V (GV) JEV poses a challenge as current vaccines are genotype III (GIII)-based and provide suboptimal protection. Given the isolation of GV JEVs from Malaysia, China, and the Republic of Korea, there is a concern about the potential for a broader outbreak. Under the hypothesis that a GV-based vaccine is necessary for effective defense against GV JE

The receptor type protein tyrosine phosphatase D (PTPRD) is expressed by neurons and implicated in interesting phenotypes that include reward from addictive substances, restless leg syndrome and neurofibrillary tangle densities in Alzheimer's disease (AD-NFTs). However, the brain phosphotyrosine phosphoprotein (PTPP) substrates for PTPRD's phosphatase have not been clearly defined. Although we have identified small molecule inhibitors of PTPRD's phosphatase that are candidates for reducing reward from addictive substances, no positive allosteric modulators of this phosphatase that might be can
