An updated look into reactive oxygen species and cyclophosphamide-induced ovarian damage.

It is estimated that 70-75% of cancer survivors are interested in parenthood with an astounding 80% struggling with reduced fertility. Cancer treatments can deteriorate the ovarian primordial follicular pool and reduce the quality of fertilizable eggs through cytotoxicity and induction of premature ovarian insufficiency (POI). Cyclophosphamide is one of the most commonly used chemotherapeutic agents and is also utilized as an immunosuppressant in autoimmune diseases and in bone marrow transplants. Accordingly, those treated with cyclophosphamide face increased risks of POI and infertility, pot

Exploring Anticonvulsant Effects of Pomalidomide by Targeting Oxidative Stress and Nrf2-Ho1 Signaling Pathway in Male Wistar Rats: A New Insight in Seizure Control.

Current medications for seizure symptoms can reduce seizure severity but do not stop or slow their progression. These drugs often have unpleasant side effects and may not work for all patients. The search for new therapeutic targets for seizure progression can be expedited through drug repurposing, which leverages existing approved medications, ultimately reducing clinical trial costs. This study investigates the neuroprotective properties of pomalidomide, an immunomodulatory drug, in a male rat model of pentylenetetrazol-induced seizures. Pomalidomide pretreatment significantly decreased the

Redox Regulation of Endogenous Gasotransmitters in Vascular Health and Disease.

Hydrogen sulfide (H2S), nitric oxide (NO), and carbon monoxide (CO) are now recognized as key gasotranmitters that regulate vascular function, contributing to vasodilation, angiogenesis, inflammation control, and oxidative balance. Initially regarded as toxic gases, they are produced on demand by specific enzymes, including cystathionine γ-lyase (CSE), endothelial nitric oxide synthase (eNOS), and heme oxygenase-1 (HO-1). Their activity is tightly controlled by redox-sensitive pathways. Reactive oxygen species (ROS), particularly superoxide and hydrogen peroxide, modulate gasotransmitter biosy

UHPLC-HRMS-MS analysis, antioxidant and hepatoprotective effects of methanolic extracts from Ficus carica L., leaves and stem barks against acute CCl4-induced liver damage.

Methanolic leaf (LME) and stem bark (SBME) extracts from Ficus carica L., were analysed using UHPLC-HRMS-MS analysis. Fifty-two compounds were identified belonging to flavonoids, phenolic acids and alkaloids. Their hepatoprotective activity was tested in a CCl4 rat model. In the CCl4-treated groups, the investigated extracts improved body weight (LME: 245.7 and SBME: 236.5 g) and relative liver weight (LME: 3.9 and SBME: 3.6 mg/g). They also reduced levels of aspartate aminotransferase (LME: 102.6 and SBME: 205.3.0 (UI/L), alanine aminotransferase (LME: 170.3 and SBME: 260.6 UI/L), lactate deh

Inflammation-Targeting Multienzyme Activity Carbon Dots Loaded with Methotrexate for Synergistic Immunotherapy in Rheumatoid Arthritis.

In rheumatoid arthritis (RA), excessive reactive oxygen species (ROS) and chronic inflammation drive damage to the synovium, cartilage, and bone. Developing precise and synergistic therapy for RA is crucial for improving remission rates. Here, carbon dots (CDs) with multienzyme activity and inflammation-targeting capabilities are designed to deliver methotrexate (MTX) for synergistic RA treatment. Specifically, positively charged CDs with porphyrin iron cores and amino-functionalized surfaces are synthesized to simultaneously scavenge hydrogen peroxide, superoxide anions, and hydroxyl radicals

Assessment of the Behavioral and Neurochemical Characteristics in a Mice Model of the Premotor Stage of Parkinson's Disease Induced by Chronic Administration of a Low Dose of MPTP.

Parkinson's disease is the second most common neurodegenerative movement disorder caused by the death of dopaminergic neurons in the Substantia nigra. The motor symptoms of Parkinson's disease only become apparent in the late stages, whereas non-motor impairments often manifest earlier. Therefore, devising adequate experimental models to study the pathogenesis of Parkinson's disease is of fundamental scientific importance. In this study, we aimed to evaluate the behavioral and neurochemical characteristics in a model of the premotor stage of parkinsonism in mice induced by chronic administrati

Microalgae Parasite Diseases of Mytilus galloprovincialis: Infections, Immunology and Antioxidant Defense.

Coccomyxa parasitica-like algae pose a growing threat to bivalve aquaculture. In this work, for the first time under controlled conditions, the effect of the green parasitic microalgae of genus Coccomyxa sp. in the Sea of Japan on the immune and antioxidant protection of Mytilus galloprovincialis was studied in two ways of infection-through filtration (with feed) and injection (into an adductor). By day 7, mortality in both experimental groups reached 68%. The phagocytic activity of hemocytes significantly decreased in the feed group, which may be due to the masking of the parasite as a food p

SIRT1 and SIRT3 Impact Host Mitochondrial Function and Host Salmonella pH Balance during Infection.

Mitochondria are important organelles that regulate energy homeostasis. Mitochondrial health and dynamics are crucial determinants of the outcome of several bacterial infections. SIRT3, a major mitochondrial sirtuin, along with SIRT1 regulates key mitochondrial functions. This led to considerable interest in understanding the role of SIRT1 and SIRT3 in governing mitochondrial functions during Salmonella infection. Here, we show that loss of SIRT1 and SIRT3 function either by shRNA-mediated knockdown or by inhibitor treatment led to increased mitochondrial dysfunction with alteration in mitocho

Testicular protective effects of hesperidin against chemical and biological toxicants.

Toxic agents can adversely impact the male reproductive system mainly via activating oxidative stress affecting the seminiferous epithelia, spermatogenesis, sperms, and the testis. Toxic agents lead to the excessive generation of reactive oxygen species (ROS), such as hydroxyl radicals, hydrogen peroxide, and superoxide anions. ROS exert a cytotoxic effect and oxidative damage to nucleic acids, proteins, and membrane lipids. Hesperidin is a pharmacologically active phytoflavone abundantly occurring in citrus fruits, such as oranges and lemons. It has shown various pharmacological properties su

ShSPI Inhibits Thrombosis Formation and Ischemic Stroke In Vivo.

Thrombotic diseases, emerging as a global public health hazard with high mortality and disability rates, pose a significant threat to human health and longevity. Although current antithrombotic therapies are effective in treating these conditions, they often carry a substantial risk of bleeding, highlighting the urgent need for safer therapeutic alternatives. Recent evidence has increasingly pointed to a connection between elastase activity and thrombosis. In the current study, we investigated the antithrombotic effects of ShSPI, an elastase inhibitor peptide derived from the venom of Scolopen

CaO assisted mechanochemical remediation of lindane-contaminated soil.

In this study, the planetary ball milling with CaO addition was used to remediate lindane-contaminated soil. Based on Hertzian theory, a mathematical model was proposed to simulate the trajectory of grinding ball and the local energy transfer during a planetary operation at the disk rotation velocities of 150-250 rpm. Besides, the influence of different parameters on lindane removal in soil was investigated, whose results showed that disk rotation velocity and reagent-to-soil ratio had a positive effect, while soil moisture, initial concentration of lindane, and mass of polluted soil demonstra

mTORC1 - TFEB pathway was involved in sodium arsenite induced lysosomal alteration, oxidative stress and genetic damage in BEAS-2B cells.

The mechanistic target of rapamycin (RAPA) complex 1 (mTORC1) - transcription factor EB (TFEB) pathway plays a crucial role in response to nutritional status, energy and environmental stress for maintaining cellular homeostasis. But there is few reports on its role in the toxic effects of arsenic exposure and the related mechanisms. Here, we show that the exposure of bronchial epithelial cells (BEAS-2B) to sodium arsenite promoted the activation of mTORC1 (p-mTORC1) and the inactivation of TFEB (p-TFEB), the number and activity of lysosomes decreased, the content of reduced glutathione (GSH) a

Antibacterial effect of Cu2O/TiO2 visible-light photocatalytic composite on Xanthomonas campestris.

In this study, a Cu2O/TiO2 (CuTi) visible-light photocatalytic composite was employed for the treatment of Xanthomonas campestris and X. campestris-infected Brassica napus seedlings. The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) values against X. campestris were determined to be 8 and 32 μg ml-1, respectively. Transmission electron microscopy analysis demonstrated a direct correlation between the extent of bacterial cell damage and the concentration of CuTi. Noteworthily, a bactericidal rate of 100% was achieved at a concentration of 150 μg ml-1 over a

Facile synthesis of pyrite FeS2 on carbon spheres for high-efficiency Fenton-like reaction.

Designing iron-based catalysts for Fenton-like reactions with peroxymonosulfate (PMS) as oxidants have attracted growing attentions. Herein, pyrite FeS2 supported on carbon spheres (FeS2@C) is synthesized by a facile low-temperature method. The FeS2@C/PMS system can degrade carbamazepine (CBZ) effectively in a wide pH range. Sulfate radicals (SO4·-), hydroxyl radicals (·OH), superoxide radical (O2·-), and singlet oxygen (1O2) are the responsible reactive oxygen species (ROSs) for CBZ degradation. Moreover, in the simulated fixed-bed reactor, the FeS2@C/PMS system can maintain a high CBZ remova

Ferulago angulata extract alleviates testicular toxicity in male mice exposed to diazinon and lead.

In this study, we investigated the protective effects of Ferulago angulata extract (FAE) against the reproductive toxicants Diazinon (DZN) and Lead (Pb) in mice. These pollutants are known to induce oxidative stress (OS), while FAE acts as a natural antioxidant. Adult male NMRI mice were exposed to DZN, Pb, and DZN+Pb, with or without FAE treatment for six weeks. We evaluated OS markers, testicular histology, and expression of mRNA related to enzymatic antioxidants. Exposure to DZN and Pb led to increased levels of thiobarbituric acid reactive substance (TBARS) and nitric oxide (NO) in the tes

Intestinal microenvironment-mediated allergic dynamic phenotypes and endotypes in the development of gluten allergy.

This study aimed to investigate the dynamic changes in intestinal microenvironment factors in the development of gluten-induced allergy (GA). Our results showed that GA provoked increasingly severe allergic phenotypes such as allergic and diarrheal symptoms with the gluten sensitization frequency, which was accompanied by dynamically rising levels of gluten-specific immunoglobulin (Ig) E, IgG2a and IgA, serum histamine, T cell-related inflammatory cytokines, and intestinal indexes. An increase in luminal pH was more significant in the large intestine versus the small intestine, which was due t
