Rebound swelling of astroglial cells exposed to hypertonic mannitol.

BACKGROUND: Mannitol is widely used in anesthesia and critical care medicine. Although its clinical effects were originally attributed to osmotic dehydration of brain cells, other mechanisms have also been proposed. Osmotic dehydration of astroglial cells is opposed by powerful volume-regulating mechanisms that involve inward transport of electrolytes. These mechanisms have been studied previously by exposing cells to hypertonic saline gradients. Because of its potential clinical relevance, the volume response of astroglial cells exposed to hypertonic mannitol was investigated. METHODS: Rat C

[Cataplexy in type C Niemann-Pick disease].

Cataplexy usually occurs as a part of the tetrad of clinical phenomena of idiopathic narcolepsy. Symptomatic cases are rare. A 4 years old girl from consangineous parents had recurrent loss of muscle tone and fell to the ground, when she laughed. The EEG was normal. Prolonged neonatal jaundice with cholestasis, hepatosplenomegaly, mental regression, supranuclear ophthalmoplegia, and foam cells led to the diagnosis of Niemann-Pick disease type C with symptomatic cataplexy. Symptomatic forms of the narcolepsy-cataplexy complex should be considered, when there is an early onset before puberty, ca

Studies on the cell tropism of Listeria monocytogenes in ovine fetal brain cell cultures.

The uptake of Listeria monocytogenes by cells in primary dissociated brain cell cultures prepared from ovine fetuses at approximately 50 to 60 days of gestation was studied using a sequential double immunofluorescence technique with antibodies against cell type-specific markers and the bacterial pathogen. Cell cultures were inoculated with bacteria at day 4, 8, and 15 in vitro. Listeria monocytogenes was predominately internalized by CD68-positive macrophages, followed by astrocytes, fibronectin-expressing cells, and neurons. An uptake of the bacterium by galactocerebroside (GC)-positive oligo…

Cellular organisation of the optic nerve and the implications for optic neuritis.

Opportunities for studying growth, degeneration and repair in the central nervous system have altered over the last decade with the development of techniques for culturing neurones and glia and the availability of immunological or molecular markers that identify separate lineages and their progeny. Much pioneering work has been carried out in the rodent optic nerve but the principles that emerge are representative for other parts of the nervous system; development of neurones and glia may differ substantially in rats and man, so that assumptions must be made in extrapolating from properties of

Human eastern equine encephalitis. Electron microscopic study of a brain biopsy.

An electron microscopic study was done on brain biopsy tissue from an eight-month-old female with acute eastern equine encephalitis diagnosed by indirect immunofluorescence. Rare clusters of round virions were found almost exclusively in the extracellular space. All virions observed had spiked envelopes, and their sizes averaged approximately 55 nm. Also found were rare enveloped virions along with degenerate organelles in a membrane-bound structure in the cytoplasm of macrophages. Intracytoplasmic development of virions was not found. Tubuloreticular complexes were seen in the endothelial cel

[The experimental study of the cerebral vasospasm--the micro-circulation on the hypothalamus and the brain stem (author's transl)].

The correlation between the cerebral vasospasm and the micro-circulation in the hypothalamus and brainstem was observed histologically and electronmicroscopically using the colloidal carbon infusion method through the right vertebral artery of dogs. The posterior communicating artery of the dog was ruptured with a fine needle, which resulted in subarachnoid hemorrhage. The colloidall infusion was done at 30 min., 48 hours, 1 week and 4 weeks after the subarachnoid hemorrhage. Results 1) The cerebral vasospasm was confirmed by vertebral angiography on all cases with 30 min., 48 hours, and 1 wee

MANF Clears Mutant Uromodulin in Human Kidney Organoids of Autosomal Dominant Tubulointerstitial Kidney Disease.

Autosomal dominant tubulointerstitial kidney disease due to uromodulin mutations (ADTKD-UMOD) is one of the leading hereditary kidney diseases. Currently there is no targeted treatment. To illuminate human relevance of mesencephalic astrocyte-derived neurotrophic factor (MANF)-based therapy, we have established patient induced pluripotent stem cell (iPSC)-derived kidney organoid model carrying UMOD p.H177-R185del, the leading mutation causing ADTKD. We have discovered that MANF can directly bind and repress ER calcium release channel IP3R1, thus enhancing AMPK-induced autophagy in a TRIB3-depe

Sodium-Based Osmotherapy in Continuous Renal Replacement Therapy: a Mathematical Approach.

Cerebral edema, in a variety of circumstances, may be accompanied by states of hyponatremia. The threat of brain injury from hypotonic stress-induced astrocyte demyelination is more common when vulnerable patients with hyponatremia who have end stage liver disease, traumatic brain injury, heart failure, or other conditions undergo overly rapid correction of hyponatremia. These scenarios, in the context of declining urinary output from CKD and/or AKI, may require controlled elevations of plasma tonicity vis-à-vis increases of the plasma sodium concentration. We offer a strategic solution to thi

A soluble endoplasmic reticulum factor as regenerative therapy for Wolfram syndrome.

Endoplasmic reticulum (ER) stress-mediated cell death is an emerging target for human chronic disorders, including neurodegeneration and diabetes. However, there is currently no treatment for preventing ER stress-mediated cell death. Here, we show that mesencephalic astrocyte-derived neurotrophic factor (MANF), a neurotrophic factor secreted from ER stressed cells, prevents ER stress-mediated β cell death and enhances β cell proliferation in cell and mouse models of Wolfram syndrome, a prototype of ER disorders. Our results indicate that molecular pathways regulated by MANF are promising thera

Neuromyelitis Optica.

Neuromyelitis optica (NMO), formerly known as Devic disease, is an autoimmune astrocytopathic disease characterized by transverse myelitis and optic neuritis. Most patients demonstrate a relapsing course with incomplete recovery between attacks, resulting in progressive disability. The pathogenesis involves production of aquaporin-4 antibodies (AQP4-IgG) by plasmablasts in peripheral circulation, disruption of the blood-brain barrier, complement-mediated astrocyte injury, and secondary demyelination. The diagnosis relies on characteristic clinical manifestations in the presence of serum AQP4-I

Modulation of spinal glial reactivity by intrathecal PPF is not sufficient to inhibit mechanical allodynia induced by nerve crush.

Spinal glial reactivity has been strongly implicated in pain that follows peripheral nerve injury. Among the many therapeutic agents that have been tested for anti-allodynia through immune modulation is the atypical methylxanthine propentofylline. While propentofylline shows a potent anti-allodynia effect after nerve transection injury, we here demonstrate that, when propentofylline is used intrathecally at the effective immune-modulatory dose, allodynia after rat nerve crush injury is completely preserved. Microglial/macrophage Iba-1 and astrocytic GFAP expression, increased in the dorsal hor

Ganglioglioma in the nasal cavity: a case report.

Ganglioglioma is a tumor containing both astrocytic and neuronal components. It may occur any where in the central nervous system and spinal cord but is only encountered rarely. Nasal glial heterotopia (also known as ''nasal glioma''), is a rare developmental abnormality seen in a wide age group. Gangliogliomas may also manifest as a nasal glial heterotopia, and neurogenic tumors should be considered in the presence of a nasal mass. In this article, we present a case of ganglioglioma located in the right-nasal cavity. The mass was excised totally through an endoscopic approach. The ganglioglio

Genetic susceptibility to heat identifies rare neurological diseases at particular risk from climate change impacts.

BACKGROUND: Climate change is one of the greatest contemporary challenges to human health, undermining human health through multiple mechanisms. Among relatively understudied mechanisms are those related to individual genomic variation. We aimed to examine this possibility. METHODS: Through a defined, agnostic literature review-based approach, we curated human genetic variants with functionally characterised temperature-dependent effects: we call these 'calortypic variants', some of which are linked to temperature-sensitive disease phenotypes. Next, we examined their occurrence in whole-genom

Spatiotemporal histogenesis of the developing human cerebellum reveals dynamic layering of Bergmann glia.

Bergmann glia (BG) are a specialized glial population essential for cerebellar development, yet their developmental timeline and molecular i … iptomic datasets identified an ASCL1+ PTF1A+ ventricular zone progenitor cluster giving rise to BG, astrocytes, and oligodendrocytes. Pseudotime analyses delineated three gliogenic lineages and revealed two temporally and transcriptionally distinct BG populations, emerging at 11-12PCW and 17PCW, suggesting multiphasic BG ontogeny. Together, these multimodal …

Morphological Changes in the Rat Brain in the Early Post-Resuscitation Period: The Relationship Between Hypoxia-Sensitive Immunohistochemical Markers, Hemodynamic and Acid-Base Status.

The relevance of studying the molecular mechanisms of ischemia-reperfusion brain injury is due to the ongoing need to identify therapeutic targets to improve neurologic outcomes following cardiac arrest. This study aims to identify early morphological and molecular changes in the rat brain in the post-resuscitation period after asphyxial cardiac arrest, as well as to assess the relationship of these changes with hemodynamic and acid-base status. Morphological analysis using immunohistochemistry (IHC) revealed an increase in the intensity of the IHC reaction to S-100β protein in astrocytes of t…

Host-cell dependent epigenetic profiles associated with survival outcomes in T. gondii infection.

Toxoplasmosis is a widespread parasitic disease affecting roughly one-third of the global population. In immunocompromised individuals or du … pted to fibroblast culture, we compared parasite survival and epigenetic profiles in neonatal mouse astrocytes (AST) and human foreskin fibroblasts (HFF). Comparative analyses revealed marked divergence in parasite population dynamics, accompanied by reduced H3K4me3 enrichment at promoter regions in parasites grown in HFF. This epigenetic shift correlated wi …

Age-Dependent Activation of Pannexin1 Function Contributes to the Development of Epileptogenesis in Autosomal Dominant Sleep-related Hypermotor Epilepsy Model Rats.

To explore the processes of epileptogenesis/ictogenesis, this study determined the age-dependent development of the functional abnormalities in astroglial transmission associated with pannexin1-hemichannel using a genetic rat model of autosomal dominant sleep-related hypermotor epilepsy (ADSHE) named 'S286L-TG'. Pannexin1 expression in the plasma membrane of primary cultured cortical astrocytes and the orbitofrontal cortex (OFC), which is an ADSHE focus region, were determined using capillary immunoblotting. Astroglial D-serine releases induced by artificial high-frequency oscillation (HFO)-ev…
