Role of social determinants, lifestyle and clinical factors in the development of diabetic peripheral neuropathy among a multiethnic Asian population in Singapore: a retrospective analysis.

INTRODUCTION: Diabetic peripheral neuropathy (DPN) is a common, underdiagnosed and irreversible complication of diabetes mellitus. However, limited evidence exists on the drivers of DPN in Asian populations, particularly the role of social and lifestyle factors. This study aimed to identify sociodemographic, lifestyle and clinical predictors of incident DPN in a multiethnic Asian cohort in Singapore. METHODS: Data from two ongoing Singaporean cohorts were analysed. Participants included individuals with type 2 diabetes mellitus and no DPN at baseline. DPN was assessed at follow-up using self-

Naturalistic Driving Outcomes and Sensorimotor Function in Cognitively Normal Older Adults.

BACKGROUND: Driving supports independence and quality of life in later life but is vulnerable to age-related and/or disease-related decline. While cognitive impairment is a well-recognized risk for unsafe mobility, the contribution of sensorimotor function is less understood. METHODS: We studied 374 cognitively normal older adults (Clinical Dementia Rating = 0 at baseline) enrolled in the DRIVES Project. A SensoryMotor Impairment Index (SMI) was constructed from six domains (grip strength, gait speed, reaction time, hearing, vision, olfaction) and categorized as 0, 1, 2, or ≥ 3 impairments. N

A systematic review and meta-analysis of the mechanism of action of Tai Chi on cardiovascular disease: evidence map of aerobic and mind-body exercise pathways.

Cardiovascular disease (CVD) remains a leading global health burden, particularly in low- and middle-income countries. Tai Chi is a mind-body exercise with reported cardiovascular benefits, but its mechanistic pathways and comparative effects versus aerobic exercise are not well defined. To synthesize evidence on the effects and proposed mechanisms of Tai Chi on CVD risk factors and related biomarkers, compared with aerobic exercise or standard care. We conducted a systematic review and meta-analysis in accordance with PRISMA. We searched Google Scholar, PubMed, EBSCOhost MEDLINE, Scopus, Scie

Metabolic [¹⁸F]F-AraG PET imaging of T Cell activation: a functional complement to cell-specific immune tracers.

BACKGROUND: Solid tumors remain a clinical challenge due to the complexity of the tumor microenvironment (TME) and high variability of immune responses to treatment. Monitoring immunological activity within the TME has emerged as a critical determinant of tumor progression and therapeutic outcome. With the advent of radiotracers that can non-invasively visualize immunological activity, research interest has grown in their potential to enhance precision oncology and support adaptive clinical decision-making. Among these, metabolic positron emission tomography (PET) imaging with [¹⁸F]F-AraG, a f

Inflammatory Profile of Lipedema: A Comparative Evaluation with Obese and Normal-Weight Women.

Lipedema is a chronic disorder characterized by disproportionate subcutaneous fat accumulation and pain, with an incompletely understood inflammatory component. It remains unclear whether this inflammatory profile is disease-specific or primarily driven by coexisting adiposity. This study aimed to evaluate systemic inflammatory markers in women with lipedema compared with women with obesity and normal-weight controls. This retrospective study included 229 women aged 30-65 years: 78 with lipedema, 76 with obesity without lipedema, and 75 normal-weight controls. Demographic and laboratory data w

An MR-Neuroimaging Study of Structural and Vascular Brain Networks in Elderly Adults with Obesity and Diabetes who Practice Structural Yoga.

BACKGROUND: Obesity and type 2 diabetes mellitus (T2DM) are associated with accelerated brain atrophy, white matter microstructural disruption, and resting-state functional network dysconnectivity in elderly adults, driven by converging vascular, neuroinflammatory, and insulin-resistance mechanisms. Yoga is a recognized mind-body intervention with documented benefits for glycaemic control, vascular health, and neurocognitive function; however, no study has yet employed multimodal MR neuroimaging to systematically characterize these brain alterations in an elderly obese-diabetic population or t

Risk Assessment of Type 2 Diabetes Mellitus among Adults in Western Rajasthan: A Cross-sectional Survey.

BACKGROUND: Diabetes has emerged as a major health challenge, evident by the rapid rise of cases. As per the National Family Health Survey-5 (NFHS-5), high blood sugar levels have been reported among 13.5% of Indian women and 15.6% of Indian men. However, nearly half of the population with diabetes is estimated to be undetected, especially adults. Diabetes-related comorbidities can be prevented by detecting the high risk at an early stage through assessment and screening of this large pool of adults. OBJECTIVES: The objective of this study was to assess the risk of type 2 diabetes mellitus (T

Linking pediatric obesity to youth-onset type 2 diabetes: integrated longitudinal and cross-sectional evidence from population-based studies worldwide.

Youth-onset type 2 diabetes mellitus (T2DM) is a rapidly growing pediatric metabolic disorder that parallels the global childhood obesity epidemic. Despite multiple population-based registries and a global meta-analysis documenting near-universal obesity at the time of a T2DM diagnosis, no integrated synthesis spanning 25 years and including diverse populations has been performed. Here we conducted a systematic review of PubMed and Scopus (January 2000 to March 2025) using longitudinal data from population-based registries in the USA (SEARCH), Canada (Manitoba), England and Wales (National Pae

Exploring recent insights on intermittent fasting in regulating glucocorticoid levels and diet-induced metabolic disorders with focus on MAFLD and hepatic outcomes.

Consumers' risky eating behaviours aided by the current food environment have led to an increase in diet-related metabolic disorders. Metabolic (dysfunction)-associated fatty liver disease origin represents a major global health burden that is increasing at an alarming rate on an annual basis. Modifying the timing of calorie consumption, dietary composition, or caloric intake offers a promising therapeutic approach for the management of this condition. The aim of this review was to provide a concise analysis of the impact of intermittent fasting on the regulation of glucocorticoid levels and d

Evaluation of novel inflammatory biomarkers in overweight, obese, and morbidly obese children: a cross-sectional study.

BACKGROUND: Chronic low-grade inflammation (metaflammation) is a central feature of obesity and contributes to metabolic complications. Complete blood count (CBC)-derived inflammatory indices have been proposed as accessible markers of obesity-related inflammation, but their biological meaning in pediatric obesity remains unclear. OBJECTIVE: To determine whether CBC-derived composite inflammatory indices primarily reflect adiposity-related inflammation or metabolic impairment in children with different obesity severities. METHODS: This retrospective cross-sectional study included 417 childre

What matters in next-generation implanted neurotechnology design? Perspectives from individuals with treatment-refractory depression.

Objective. Major financial investments in implanted neurotechnology have fueled a wave of innovation in device design, implant procedures and therapy delivery methods. These innovations are often presented as making implanted neurotechnologies more appealing to patients. However, it remains unclear whether these innovations meaningfully impact patients' decision-making about adoption. The present study aims to examine how next-generation neurotechnology innovations shape prospective decision-making about adoption among patients with treatment-refractory depression (TRD), a key projected target

Allostatic load profiles, resilience, and psychosocial adaptation in young adults.

INTRODUCTION: Allostatic load (AL) reflects the cumulative physiological burden on the body, quantified using biomarkers across multiple systems. The current study identified latent AL profiles in young adults and examined whether resilience moderates associations between AL and depressive symptoms, social functioning, and role functioning. METHODS: A total of 165 nonclinical young adults (84 women; aged 19-30 years) provided data on 15 biomarkers spanning the hypothalamic-pituitary-adrenal axis, oxidative stress, inflammatory-immune, lipid/glucose-metabolic, and renal systems, which were z-s

Immunometabolic reprogramming and mitochondrial dysfunction in acute respiratory distress syndrome: mechanisms, metabolic resilience, and therapeutic perspectives- a narrative review.

BACKGROUND: Acute respiratory distress syndrome (ARDS) is a biologically heterogeneous condition in which patients exposed to similar injurious stimuli often develop markedly different clinical trajectories and outcomes. While inflammation is central to ARDS pathogenesis, inflammatory burden alone does not fully explain the variability in disease progression, treatment response, or recovery. Emerging evidence suggests that immunometabolic reprogramming, characterized by increased glycolysis, impaired mitochondrial oxidative phosphorylation, and altered metabolite signalling, plays a critical r

Fast and Accurate Parameter Optimization of Transcranial Temporal Interference Stimulation by Selectively Employing Target-Specific Candidate Electrodes.

Transcranial temporal interference (TI) stimulation offers non-invasive modulation of deep brain regions with limited neocortical activation. Achieving focal TI stimulation requires individualized optimization of the stimulation parameters, including electrode configurations and injection currents. However, conventional exhaustive search (ES) approach is computationally expensive, hindering the clinical applicability of subject-specific parameter optimization. We propose a target-specific restricted exhaustive search (RES) strategy that substantially reduces the computational burden while pres
