Immunotherapy has revolutionized cancer treatment paradigms due to its high specificity, favorable safety profile, and remarkable clinical efficacy. Immune checkpoint blockade (ICB) therapy has been widely adopted in clinical practice for various malignancies. Unfortunately, its limited response rates restrict broader application. Myeloid-derived suppressor cells (MDSCs) serve as key mediators of ICB resistance. In this study, we demonstrated that fucoidan extracted from Saccharina japonica LMWF modulated MDSCs activity to sensitize tumors to ICB therapy. Mechanistically, LMWF activated the TL

BACKGROUND: Steroid hormone-driven endometrial decidualization involves the differentiation of endometrial stromal cells into decidual cells, which form the decidua for blastocyst implantation. This process is a prerequisite for successful embryo implantation. O-GlcNAcylation is a nutrient-sensitive post-translational modification. Our prior work demonstrated that O-GlcNAcylation promotes embryo recognition by enhancing endometrial epithelial cell proliferation, migration and invasion, thereby facilitating embryo implantation. However, its regulatory role in endometrial stromal cell decidualiz

Cerebral ischemia/reperfusion injury (CIRI) is a major cause of neurological dysfunction after ischemic stroke, and effective therapies remain limited. Gomisin A, a bioactive lignan isolated from Schisandra chinensis, exhibits antioxidant and anti-apoptotic activities, but its role in CIRI remains unclear. Here, the protective effects of Gomisin A were investigated using middle cerebral artery occlusion (MCAO) rats and oxygen-glucose deprivation/reoxygenation (OGD/R) cell models. Gomisin A significantly reduced cerebral infarct volume and improved neurological outcomes in MCAO rats. In OGD/R m

The high mortality rate of prostate cancer remains a major threat to men's health worldwide, highlighting an urgent need for novel therapeutic agents with improved efficacy and safety profiles. In this study, we employed a rational structural modification strategy to synthesize 14 novel 1,2,3-triazole-hybridized derivatives of piperlongumine (PL), a natural alkaloid with proven anticancer activity. The antiproliferative activity of these derivatives was evaluated in vitro against multiple cancer cell lines (PC-3, A549, T47D, and SMMC-7721), with normal HEK293 cells used to assess their selecti

Diazinon (DZN) is an organophosphorus (OP) pesticide implicated in carcinogenesis. Its primary mechanism of action is inhibition of acetylcholinesterase (AChE). We evaluated the impact of low-dose DZN on two gastric cancer (GC) cell lines (AGS, MKN-45), focusing on cholinergic and serotonergic signaling. Proliferation-stimulatory concentrations were identified by MTT assay (0.1 µM for AGS, 0.01 µM for MKN-45). Scratch assays demonstrated enhanced migration at low doses but suppression at high dose. AO/EB staining confirmed reduced viability with increased apoptosis at high dose. Cell cycle ana

Organometallic ruthenium complexes have gained significant attention as promising anticancer agents due to their favorable properties in medicinal chemistry. In this study, four adamantane-N-acylthiourea-based ligands (ADL1-ADL4) and their corresponding Ru(η6-p-cymene) complexes (RuAD1-RuAD4) were synthesized and characterized using 13C{1H} NMR, FT-IR, UV-vis spectroscopy, and ESI (HR)-MS. Additionally, the structures of all complexes were confirmed through single-crystal X-ray diffraction analysis. The in vitro cytotoxicity of the compounds was evaluated against human melanoma (A375), triple-

The Drosophila melanogaster intestine has emerged as a powerful model system for studying the fundamental mechanisms underlying cancer biology, including tumorigenesis, metastasis, and paraneoplastic syndromes such as cachexia. The key signaling pathways implicated in human colorectal cancers play similar roles in the Drosophila intestinal system. Dysregulation of these pathways in the Drosophila intestine leads to overproliferation of intestinal stem cells, loss of cell differentiation, and potential transdifferentiation, mirroring tumorigenic processes in humans. Recent research utilizing Dr

Preeclampsia (PE) is a hypertensive disorder of pregnancy characterized by maternal endothelial dysfunction and end-organ damage. Our previous work demonstrated that PE patient-derived exosomes contained higher levels of soluble FMS-like tyrosine kinase-1 (sFlt-1) and significantly induced endothelial dysfunction and PE development. However, the mechanisms underlying the effect of sFlt-1-enriched exosomes (sFlt-1-Exo) on PE development are poorly characterized. Here, we revealed that trophoblast-derived sFlt-1-Exo treatment induced significant inhibition of human umbilical vein endothelial cel

The second most common cancer among men is prostate cancer, which is also the fifth leading reason for male cancer deaths worldwide. Bone metastases are the main factor affecting the prognosis of prostate cancer. Consequently, antitumor and anti-prostate cancer-induced bone destruction medicines are urgently needed. We previously discovered that aminooxyacetic acid hemihydrochloride (AOAA) suppressed bone resorption and osteoclast growth by decreasing adenosine triphosphate (ATP) production and limiting oxidative phosphorylation (OXPHOS). Here, we evaluated the impacts of AOAA on prostate canc

Hirschsprung disease (HSCR), one of several neurocristopathies in children, is characterized by nerve loss in the large intestine and is mainly treated by surgery, which causes severe complications. Enteric neural crest-derived cell (ENCC) transplantation is a potential therapeutic strategy; however, so far with poor efficacy. Here, we assessed whether and how fecal microbiota transplantation (FMT) could improve ENCC transplantation in a rat model of hypoganglionosis; a condition similar to HSCR, with less intestinal innervation. We found that the hypoganglionosis intestinal microenvironment n

Aberrant epigenetic transcriptional regulation is linked to metastasis, a primary cause of cancer-related death. Dissecting the epigenetic mechanisms controlling metastatic progression may uncover important insights to tumor biology and potential therapeutic targets. Here, we investigated the role of the SIN3A histone deacetylase 1 and 2 (SIN3A-HDAC1/2) complex in cancer metastasis. Using a mouse model of melanoma metastasis, we found that the SIN3A-HDAC1/2 transcription repressor complex silences BMP6 expression, causing increased metastatic dissemination and tumor growth via suppression of B

Gallbladder cancer (GBC) is a demanding fatal disease with no ideal treatment for inoperable patients. Recent reports have determined TNF-α associated lymphatic metastasis in GBC, while its resistance to TNF-α-killing remains largely unexplored. In this assay, we first found cellular inhibitor of apoptosis (cIAP1) overexpressed in GBC tissues and the roles in promoting the proliferation and migration of GBC in vitro as its homology cIAP2 does. Then how GBC cell survives TNF-α toxicity and TNF-α-induced apoptosis first prevail as follows. The reduction in cIAP1 does not give rise to apoptosis e

BACKGROUND AND AIM: Tripartite motif-containing 29 (TRIM29) is structurally a member of the tripartite motif family of proteins and is involved in diverse human cancers. However, its role in pancreatic cancer remains unclear.
METHODS: The expression pattern of TRIM29 in pancreatic ductal adenocarcinoma was assessed by immunocytochemistry. Multivariate logistic regression analysis was used to investigate the association between TRIM29 and clinical characteristics. In vitro analyses by scratch wound healing assay and invasion assays were performed using the pancreatic cancer cell lines.

Ischemic stroke causes transient increase of neural stem and progenitor cell (NSPC) proliferation in the subventricular zone (SVZ), and migration of newly formed neuroblasts toward the damaged area where they mature to striatal neurons. The molecular mechanisms regulating this plastic response, probably involved in structural reorganization and functional recovery, are poorly understood. The adaptor protein LNK suppresses hematopoietic stem cell self-renewal, but its presence and role in the brain are poorly understood. Here we demonstrate that LNK is expressed in NSPCs in the adult mouse and

PECAM-1 is a 130-kd member of the Ig superfamily present on endothelial cells, platelets, polymorphonuclear leukocytes, monocytes, and lymphocytes. Its expression begins early in development and persists through adulthood. PECAM-1 functions as an adhesion and signaling molecule between adjacent endothelial cells and between endothelial cells and circulating blood elements. Antibodies directed against PECAM-1 have been shown to affect angiogenesis, endothelial cell migration, and polymorphonuclear leukocyte transmigration. Furthermore, its dimerization is associated with the modulation of integ

Mice immunized with three injections of gamma-irradiated Karp strain of Rickettsia tsutsugamushi were evaluated for the presence of cell-mediated immunity by using delayed-type hypersensitivity, antigen-induced lymphocyte proliferation, and antigen-induced lymphokine production. These animals also were evaluated for levels of circulating antibody after immunization as well as for the presence of rickettsemia after intraperitoneal challenge with viable Karp rickettsiae. After immunization with irradiated Karp rickettsiae, a demonstrable cell-mediated immunity was present as evidenced by delayed

Thirty-four chimeric pyridone alkaloids, including twenty-eight new compounds named aspernigrins F1-F8, G1-G5, H1-H7, and I-P (1-28), were isolated from the mangrove endophytic fungus Aspergillus niger H6a, together with six known ones (29-34). The structures of these compounds were elucidated by HRESIMS, NMR spectroscopic data, single-crystal X-ray diffraction analyses, TDDFT-ECD calculations, and chemical methods, including Marfey's derivatization. The structure of 26 contains two units of 2-aminododecanoic acid, whereas those of 27 and 28 not only contain two moieties of 2-aminododecanoic a

Sphingosine-1-phosphate receptor 1 (S1PR1, also known as EDG1) is a G-protein-coupled receptor. Although there have been conflicting results reported, S1PR1 is generally accepted to be protumorigenic in many cancer types. However, S1PR1 has not been studied in thyroid cancer. This study aimed to investigate the biological activity of S1PR1 in thyroid cancer. S1PR1 protein levels were found to be higher in thyroid cancer tissues than adjacent normal tissues. Using an S1PR1-GFP construct, we showed that S1PR1 is localized in the cell membrane; however, when stimulated by sphingosine-1-phosphate,

Cancer cell resistance and tumor relapse remain major challenges in cancer treatment. Chloroquine, an FDA-approved antimalarial drug currently undergoing clinical trials for various cancers, has emerged as a promising candidate for combination therapy with conventional anticancer agents. In this study, we demonstrate that in patients-derived osteosarcoma cells who had undergone multiple chemotherapy treatments, as well as in murine colorectal cancer cells, administration of standard chemotherapeutic agents induces autophagy, which likely serves as a cytoprotective mechanism promoting therapy r

Protein kinase CK2 is a pleiotropic protein kinase, which phosphorylates a number of cellular and viral proteins. Thereby, this kinase is implicated in the regulation of cellular signaling, controlling of cell proliferation, apoptosis, angiogenesis, immune response, migration and invasion. In general, viruses use host signaling mechanisms for the replication of their genome as well as for cell transformation leading to cancer. Therefore, it is not surprising that CK2 also plays a role in controlling viral infection and the generation of cancer cells. Epstein-Barr virus (EBV) lytically infects
