Chronic exposure to MK-801 leads to olfactory deficits and reduced neurogenesis in the olfactory bulbs of adult male mice.

BACKGROUND: MK-801 is a drug widely used in preclinical studies to model schizophrenia in animals. Its distinctive feature is the ability to mimic pathological changes in social interactions. Unlike humans, rodents rely heavily on their sense of smell for social interaction. Since, as previously demonstrated, it also impairs neurogenesis, we set out to determine whether olfactory impairment is associated with chronic administration of the drug. METHODS: The mice were divided into two groups, of which one was administered the drug for 3 weeks, and the other only once. Olfaction and social tran

Drug Discovery in the Age of Artificial Intelligence: Transformative Target-Based Approaches.

The complexities inherent in drug development are multi-faceted and often hamper accuracy, speed and efficiency, thereby limiting success. This review explores how recent developments in machine learning (ML) are significantly impacting target-based drug discovery, particularly in small-molecule approaches. The Simplified Molecular Input Line Entry System (SMILES), which translates a chemical compound's three-dimensional structure into a string of symbols, is now widely used in drug design, mining, and repurposing. Utilizing ML and natural language processing techniques, SMILES has revolutioni

Ceritinib-ibuprofen synergistic hepatotoxicity: Insights from real-world data and liver organoid models.

Ceritinib, a next-generation anaplastic lymphoma kinase inhibitor used to treat non-small cell lung cancer, has been increasingly associated with drug-induced liver injury (DILI), especially in the context of drug-drug interactions (DDIs). To elucidate the hepatotoxic risk of ceritinib in combination therapies, we conducted a real-world pharmacovigilance analysis of the FDA Adverse Event Reporting System (FAERS) and experimentally validated the results using human liver organoids. Disproportionality analysis revealed that ceritinib administered in combination therapy was more likely to induce …

In vitro stability of low-concentration ziconotide alone or in admixtures in intrathecal pumps.

OBJECTIVES: Ziconotide is often administered in combination with other analgesics via an intrathecal pump. Studies have established that ziconotide is stable when delivered alone in high concentrations. No stability data are available, however, for ziconotide given in low concentrations and/or with other analgesics as usually occurs in clinical oncology practice. The objective of this study was to assess the in vitro stability of ziconotide alone and combined with other analgesics in intrathecal pumps at 37 °C, as well as in syringes at 5 °C, to evaluate conditions for storing and transporting

Trade-offs between drug toxicity and benefit in the multi-antibiotic resistance system underlie optimal growth of E. coli.

BACKGROUND: Efflux is a widespread mechanism of reversible drug resistance in bacteria that can be triggered by environmental stressors, including many classes of drugs. While such chemicals when used alone are typically toxic to the cell, they can also induce the efflux of a broad range of agents and may therefore prove beneficial to cells in the presence of multiple stressors. The cellular response to a combination of such chemical stressors may be governed by a trade-off between the fitness costs due to drug toxicity and benefits mediated by inducible systems. Unfortunately, disentangling t

Etonogestrel-releasing contraceptive implant in a patient using thalidomide for the treatment of erythema nodosum leprosum: a case report.

INTRODUCTION: Thalidomide is an immunomodulatory drug and first choice in the treatment of erythema nodosum leprosum. Given its teratogenic potential, it is essential that an effective contraceptive method is used, especially a long-acting reversible contraceptive (LARC) method. The subdermal etonogestrel (ENG)-releasing implant is an adequate method due to the high effectiveness and long-term use. However, interaction between thalidomide and ENG has not been well documented. Concern arises because thalidomide interacts with cytochrome P450 (CYP450) enzymes that metabolize sexual steroids. AIM

Nanoparticle Delivery Systems in Rheumatoid Arthritis: More Than Vehicles.

Rheumatoid arthritis (RA) is one of the most prevalent systemic autoimmune inflammatory diseases worldwide, causing chronic, progressively worsening arthritis that may ultimately lead to disability. Despite the availability of numerous therapeutic agents, limitations exhibit, including poor aqueous solubility, suboptimal stability, inadequate permeability, short half-lives, and multi-organ toxicity during long-term or high-dose administration. Nanoparticle-based drug delivery offers a robust strategy to mitigate these deficiencies while maximizing therapeutic efficacy through controlled-releas

Achieving Rapid Remission With Guselkumab in a Moderately Active Ulcerative Colitis Patient With High Infectious Risk and Inadequate Response to Advanced Therapy: A Case Report.

Ulcerative colitis (UC) with prior inadequate response to advanced therapy (AT-IR) that requires hospitalization is particularly challenging to manage, especially in cases where a high risk of infection limits the use of immunosuppressive agents. A 32-year-old male patient with pancolitis-type UC was refractory to glucocorticosteroids, infliximab, and tacrolimus. His clinical course was complicated by septic shock and invasive candidiasis. Antifungal therapy limited the use of upadacitinib due to drug-drug interactions. After achieving infection control, intravenous guselkumab (200 mg) was ini…

Invasive aspergillosis in lung and heart transplant recipients.

Invasive aspergillosis (IA) is a significant cause of morbidity and mortality in lung transplant (LT) and heart transplant (HT) recipients. Most of the risk is in the first year post-transplant. In this review, the epidemiology, diagnosis and treatment of IA are examined in detail. The challenges in diagnosis are highlighted. Whilst culture and histology are the cornerstone, the emerging role of serological and molecular methods is discussed. Treatment is a multipronged approach with drug-drug interactions and toxicity impacting outcomes. New antifungals are in development and have the potenti…

Biopharmaceutics Aspects of Sugar Alcohols: Implementation of Patient-Centricity in Pharmaceutical Development and Clinical Use.

Sugar alcohols such as mannitol and sorbitol are frequently used as pharmaceutical excipients in oral dosage forms. Their versatility stems from extensive evidence of use in humans. However, investigations over many years have clearly indicated their osmolarity-derived biopharmaceutics effects on oral drug absorption. Despite this, these accumulated insights have not been fully integrated into pharmaceutical formulation development or clinical use, leading to suboptimal industrial and clinical practices. This review provides a comprehensive summary of the biopharmaceutics of sugar alcohols, de

Unveiling Dynamic Hotspots in Protein-Ligand Binding: Accelerating Target and Drug Discovery Approaches.

Computational methods have transformed target and drug discovery, significantly accelerating the identification of biological targets and lead compounds. Despite its limitations, in silico molecular docking represents a foundational tool. Molecular Dynamics (MD) simulations, employing accurate force fields, provide near-realistic insights into a compound's behavior within a biological target. However, docking and MD predictions may be unreliable without precise knowledge of the target binding site. Through MD simulations, we investigated 100 co-crystal structures of biological targets complexe

Discovery of Indole-Based PDE5 Inhibitors: Synthesis and Pharmacological Evaluation.

Phosphodiesterase 5 (PDE5) inhibitors have been suggested as new treatments for Alzheimer's disease (AD) and other conditions such as cancer and cardiovascular diseases. Utilizing the widespread presence of the indole moiety in biomolecules and drugs, previously synthesized quinoline and naphthyridine compounds were modified into novel indole-containing PDE5 inhibitors. Replacing the amine with an amide group led to identifying a potent analogue, compound 14a, with an IC50 of 16.11 nM. Molecular docking simulations further highlight the significance of the amide group in drug-target interactio

Design, Synthesis, In Vitro Evaluation of Antitumor Activity, and Molecular Docking Studies of D-ring-opened Indazole-Matrine Derivatives as Potential Anticancer Agents.

To develop matrine derivatives with improved anticancer activity, we designed and synthesized 16 derivatives using matrine as the lead compound through drug splicing principles. Using the MTT assay, we evaluated their antiproliferative effects against three human cancer cell lines: cervical cancer (HeLa), hepatocellular carcinoma (Huh-7), and triple-negative breast cancer (MDA-MB-231). Most compounds demonstrated superior activity to matrine, with W12 showing the strongest effects (IC50 values: 4.8 ± 0.33 µM for Huh-7, 8.03 ± 1.28 µM for MDA-MB-231, and 10.11 ± 0.39 µM for HeLa). Discovery of highly potent α-keto ester-based peptidomimetic inhibitors of the Hip1 protease for the treatment of Mycobacterium tuberculosis.

Mycobacterium tuberculosis (Mtb), the bacterium responsible for tuberculosis, is the leading cause of death due to a single infectious agent. Given the alarming increase in drug-resistant cases, therapeutic agents targeting novel Mtb drug targets are urgently needed. Hip1, a serine protease required for Mtb survival in macrophages and tolerance to various antibiotics, has been identified as an attractive therapeutic target. In the current study, we describe the design and synthesis of highly potent (pM range K i) peptidomimetic α-keto ester inhibitors of Hip1. We also report the first two X-ra
