Auditory function in patients with systemic lupus erythematosus.

OBJECTIVE: Patients with systemic lupus erythematosus (SLE) may develop hearing and balance disorders as a result of the immune-mediated inner ear damage due to vasculitis or ototoxicity of drugs used in SLE treatment. The aim of the study was evaluation of the hearing organ disorders in patients with SLE with particular regard to their prevalence and relationship to duration and severity of disease. The severity was assessed from involvement of organs that resulted in poorer SLE outcome, i.e. kidneys and central nervous system (CNS), and from the presence of antibodies associated with unfavou

The masking level difference in chinchilla auditory cortex. Effects of inner hair cell loss.

The purpose of the present study was to investigate responses from the unanesthetized chinchilla auditory cortex (AC) to conditions producing a masking level difference (MLD) in perceptual studies, both before and after inner hair cell (IHC) loss caused by carboplatin. Tungsten electrodes were chronically implanted in the right AC (active) and anterior cranium (common) in six adult chinchillas. Following a recovery period, AC responses were obtained from the unanesthetized animal placed in a passive restraint. Toneburst input/output functions were obtained. Tonebursts (500 Hz) ranged from 0 to

Correlations of Clinical and Audio-Vestibulometric Findings with Coexisting Anxiety and Depression in Ménière's Disease Patients.

INTRODUCTION: Ménière's disease (MD) is an inner ear disorder, characterized by vertiginous attacks, fluctuating sensorineural hearing loss, tinnitus, and a feeling of ear fullness. Endolymphatic hydrops has been proven as the underlying pathology. Frequently, psychopathologies accompany the disease. The aim of this study was to investigate the correlation of anxiety and depression with demographic, clinical, and audio-vestibular findings in MD patients. METHODS: The study included 40 consecutive unilateral MD patients. Demographic data (age, sex, education, employment, and marital status), c

Characterizing a Joint Reflection-Distortion OAE Profile in Humans With Endolymphatic Hydrops.

OBJECTIVES: Endolymphatic hydrops (EH), a hallmark of Meniere disease, is an inner-ear disorder where the membranes bounding the scala media are distended outward due to an abnormally increased volume of endolymph. In this study, we characterize the joint-otoacoustic emission (OAE) profile, a results profile including both distortion- and reflection-class emissions from the same ear, in individuals with EH and speculate on its potential utility in clinical assessment and monitoring. DESIGN: Subjects were 16 adults with diagnosed EH and 18 adults with normal hearing (N) matched for age. Both t

Detection of the RNA-Editing Enzyme ADARB1 in Primary Auditory Neurons and Anti-ADARB1 Immunofluorescence as a Means for Automated Counting.

Spiral ganglion neurons (SGNs), the primary auditory afferents in the inner ear, are vulnerable to degeneration from neuropathies, concussion, and aging, among other causes. Accurate quantification of SGNs is critical for assessing degeneration and therapeutic interventions, but manual counting is arduous due to dense cellular crowding and unreliable nuclear markers. Because SGNs receive input from GluA2-containing AMPA receptors, the transcripts for which must be RNA-edited for calcium impermeability, we hypothesized that SGNs express high levels of the RNA-editing enzyme ADARB1. Here, we con

A porcine congenital deafness model with unconditional knockout of GJB2 generated by CRISPR/Cas9 genomic editing.

GJB2, the primary gene responsible for DFNB1, the most prevalent non-syndromic hearing loss (NSHL), has variants that account for over 50% of all prelingual hearing loss (HL). Mice are the main model for congenital hearing loss (CHL) research, but they have delayed auditory maturation postnatally, and unconditional Gjb2 knockout in mice causes embryonic lethality. Pigs have similar inner-ear anatomy to humans and, like humans, have matured auditory function and fully differentiated cochlea at birth. Currently, there is no GJB2 unconditional knockout animal model for GJB2-related CHLs research,

[Advances in research on the molecular mechanisms and gene therapy of hereditary hearing impairment].

Hereditary hearing impairment represents a significant etiology of language and social dysfunction in both children and adults, primarily caused by genetic factors. To date, over 150 genes have been identified in association with this disorder. The pathogenic mechanisms involve multiple molecular levels, including abnormalities in hair cell cytoskeleton and stereociliary structure, dysfunction of intercellular gap junctions (e.g., GJB2, GJB6), dysregulation of ion channels and transporters (e.g., SLC26A4, KCNQ4), alterations in extracellular matrix composition, and disruption of intracellular

Targeting the brain: alternative administration routes and drug delivery systems for antidepressant therapy.

INTRODUCTION: Although numerous antidepressants are clinically available, they are characterized by slow therapeutic onset, systemic effects, and limited brain exposure, which is restricted by the blood-brain barrier (BBB). AREAS COVERED: Alternative administration routes namely intranasal, ocular, and intratympanic delivery, are emerging as promising strategies for a direct drug targeting of the central nervous system. Data were obtained from PubMed, Web of Science, Scopus, and regulatory agencies, 2010-2025. Intranasal route enables rapid brain delivery through the olfactory and trigeminal

Advances in RNA delivery to extrahepatic tissues and cells.

Despite their transformative potential in treating genetic, neurological, and oncological disorders, the clinical application of RNA therapeutics remains largely confined to the liver. Current approved platforms rely on two fundamentally distinct hepatic delivery mechanisms. N-acetylgalactosamine (GalNAc) conjugation achieves liver targeting through designed, high-affinity binding to the asialoglycoprotein receptor on hepatocytes, while lipid nanoparticles (LNPs) accumulate in the liver through intrinsic physicochemical tropism driven by apolipoprotein adsorption and the fenestrated hepatic va

Vogt-Koyanagi-Harada (VKH)-What Do We Know About the Disease, and Can We Recognize It?

Background: Vogt-Koyanagi-Harada (VKH) is a multisystem autoimmune disease that ophthalmologists often encounter first. The condition is caused by an immune response against tyrosinase-related proteins in pigment cells (melanocytes) of the uvea, inner ear, meninges, and skin, and the process may be triggered by genetic and environmental factors. Although much is known about the disease, establishing an accurate and timely diagnosis still requires a multidisciplinary team and strong clinical expertise. Treatment demands early and aggressive anti-inflammatory therapy with corticosteroids, often

Bilateral intradiploic dermoid cysts with unilateral rupture causing hearing loss: illustrative case.

BACKGROUND: Dermoid cysts are congenital lesions that typically occur along the midline. They usually remain asymptomatic unless ruptured, causing meningitis, hydrocephalus, or cranial nerve deficits. This case presents an exceptionally rare scenario of bilateral, symmetric intradiploic dermoid cysts of the occipital bone, which initially manifested as sensorineural hearing loss (an unusual symptom). OBSERVATIONS: A 21-year-old woman presented with right-sided hearing loss. Imaging showed bilateral, symmetric intradiploic cystic lesions within the occipital bone. The right-sided lesion had ru

Differential contributions of mt-Tr and Cs variants to developmental cochlear defects and mitochondrial dysfunction in A/J mice.

A/J mice exhibit early-onset hearing loss linked to Cdh23, mitochondrial tRNA-Arg (mt-Tr), and citrate synthase (Cs) variants. Although developmental cochlear defects have been identified in juvenile A/J mice, the hierarchical contributions of mt-Tr versus Cs remain unclear. Using reciprocal intercross-derived strains to decouple mitochondrial haplotypes from nuclear factors, we demonstrate that the mitochondrial background is the primary determinant of auditory dysfunction. Mice with A/J mtDNA (AXB strains) displayed significantly higher ABR thresholds, accelerated hair cell attrition, and se

Circulating Clues in Ménière's Disease: Elevated Cell-Free DNA and a Pro-Inflammatory Signature in Patients' Blood.

Ménière's disease (MD) is thought to involve dysfunction of the blood-labyrinth barrier, but circulating mechanisms of endothelial injury remain poorly understood. The present study investigated whether cell-free DNA (cfDNA) and inflammatory mediators in plasma contribute to vascular stress and barrier disruption in MD. cfDNA levels were significantly elevated in plasma from patients compared with plasma from healthy controls. Exposure of primary human stria vascularis endothelial cell monolayers to plasma from MD patients led to decreased transepithelial electrical resistance and a significan

Longitudinal study of Emotional Intelligence Scale items associated with improvement in Meniere's disease.

BACKGROUND: Meniere's disease is a stress-related inner ear disorder characterized by recurrent vertigo and auditory symptoms. Evidence linking stress-coping abilities to clinical outcomes remains limited. Emotional intelligence (EI), particularly the situation-management domain of the Emotional Intelligence Scale (EQS), may influence symptom improvement through adaptability and behavioral flexibility. OBJECTIVE: To examine whether EI assessed by the EQS is associated with improvement in Meniere's disease. METHODS: This retrospective study included 93 patients aged 23-82 years who had baseli

Timing and Dosage of Notch Inhibition and Wnt Activation Enhanced Hair Cell-Like Cell Generation and Stereocilia Elongation in Otic Organoids.

The modulation of Notch and Wnt signaling was evaluated for its ability to promote the differentiation and maturation of hair cell (HC)-like cells in otic organoids derived from mouse embryonic stem cells (mESCs). Using R1/E mESCs, we generated otic organoids and treated them with the Notch pathway inhibitor (γ-secretase inhibitor, DAPT) and the Wnt signaling agonist (CHIR99021) during two-time windows: days (D) D12-D18 (early treatment) or D28-D34 (late treatment). Among the late treatment conditions, lower concentration (5 μM DAPT and 2.5 μM CHIR99021) yielded greater expression of MyoVIIa a

Extratemporal Cholesteatoma: A Review of 9 Cases.

BACKGROUND: Complete removal of cholesteatoma is critical but challenging when the matrix adheres to or destroys surrounding vital structures, such as the facial nerve, inner ear, internal carotid artery, dura mater, jugular bulb, sigmoid sinus, cavernous sinus, etc. Damage to these structures can lead to severe complications such as cerebrospinal fluid leakage, life-threatening hemorrhage, and nerve deficits. METHODS: We reviewed 9 cases of extensive cholesteatoma extending beyond the temporal bone that were treated in our department between 2010 and 2022, focusing on surgical approaches and

Combined AAV-mediated specific Gjb2 expression restores hearing in DFNB1 mouse models.

Pathogenic mutations in the Gjb2 gene, encoding connexin 26, are the leading cause of autosomal recessive hereditary deafness. Gene therapy holds significant promise for treating this. Adeno-associated virus (AAV)-mediated therapeutic gene delivery has been shown to be safe and effective in restoring hearing in both animal models and human patients. However, Gjb2 gene therapy has been hindered by the limited specificity and efficiency of the available AAV vectors. In this study, we screened AAV serotypes and found that co-administration of AAV1 and AAV-ie could effectively target Gjb2-expressi
