[Clinical Frontiers and Applications Progress of ctDNA in Precision Treatment
of Non-small Cell Lung Cancer].

Circulating tumor DNA (ctDNA) enables minimally invasive, repeatable assessment of tumor burden and molecular evolution and is an important candidate biomarker for precision management of non-small cell lung cancer (NSCLC). This review summarizes current evidence on assay strategies, perioperative dynamics, molecular residual disease (MRD) after curative-intent treatment, relapse surveillance, and applications in unresectable stage III disease. The most consistent evidence shows that detectable ctDNA after definitive treatment is associated with a substantially increased risk of recurrence, wh

Hydraulic residence time thresholds and seasonal regimes governing DOM transformation in upper Yangtze River cascading reservoirs.

The molecular characteristics and biogeochemical transformations of riverine dissolved organic matter (DOM) have a significant impact on carbon cycling in freshwater ecosystems. Although individual dams influence DOM dynamics, the cumulative effects of sequential reservoir systems remain unclear. This study investigates five consecutive reservoirs along the Yangtze River through seasonal sampling (2021-2022). We introduce the concept of cumulative hydraulic residence time (cHRT), which represents the sequential water retention in a low-interference cascade reservoir. Our findings demonstrate t

Loss of anthocyanidin synthase gene is associated with white flowers of Salvia miltiorrhiza Bge. f. alba, a natural variant of S. miltiorrhiza.

SmANS deletion leads to white flower mutation in Salvia miltiorrhiza. SmANS deletion leads to white flower mutation in Salvia miltiorrhiza. Abstract Salvia miltiorrhiza is an essential traditional Chinese medicine (TCM) with purple flowers, and S. miltiorrhiza Bge. f. alba is a unique intraspecific variation with white flowers. The molecular mechanism of flower color formation in S. miltiorrhiza will provide vital information for the variation and evolution. Here, we performed HPLC, transcriptomic, and re-sequencing analyses of purple-flowered S. miltiorrhiza line 'Zihua105' (ZH105) and white-

[Current view on non-Hodgkin lymphomas. Origin and evolution in relation to cytogenetic-histologic correlations].

Recent advances in immunology, cytogenetics and molecular genetics has allowed for a better understanding of the origin and evolution of non Hodgkin lymphoma (NHL). Over the last decade a number of recurrent chromosome aberrations has been disclosed and some correlations with well defined histologic subsets of B-cell NHL has been established. Five important cytogenetic-histologic associations has been documented, well defined by combined cytologic, immunologic and genetic investigations: t(14;18) (q32;q21) and NHL of follicle centre cell origin, frequently with follicular histologic pattern; t

Bayesian phylogenetic analysis of the influenza-A virus genomes isolated in Tunisia, and determination of potential recombination events.

The influenza-A virus (IAV) causes seasonal epidemics and presents a pandemic risk with the possibility of genetic re-assortment, allowing the emergence of new strains. The evolution of IAVes is done most often by relatively frequent re-assortment between gene segments, but the hypothesis of their evolution by recombination between RNA segments has not been justified to this date. Here, we examine this hypothesis by Bayesian phylogenetic analysis, to test if recombination events have occurred between genomic RNA segments. Different IAV subtypes are observed in co-circulation in Tunisia, which

Emergence of unusual vanA/vanB2 genotype in a highly mutated vanB2-vancomycin-resistant hospital-associated E. faecium background in Vietnam.

Enterococcus faecium has become a globally disseminated nosocomial pathogen mainly because of acquisition and diffusion of virulence factors and multidrug resistance determinants, including glycopeptides, which are some of the last resort antimicrobials used to treat more serious infections common in high-risk patients. In this study we investigated and characterized hospital-associated (HA) E. faecium isolates collected at Hue Central Hospital, Vietnam. Our results highlighted the spread among hospital wards of a surprisingly heterogeneous multidrug-resistant E. faecium population comprising

Identification of a novel mutation in RIPK4 in a kindred with phenotypic features of Bartsocas-Papas and CHAND syndromes.

Three children from an expanded consanguineous Kuwaiti kindred presented with ankyloblepharon, sparse and curly hair, and hypoplastic nails, suggestive of CHAND syndrome (OMIM 214350) that belongs to the heterogeneous spectrum of ectodermal dysplasias. After exclusion of pathogenic mutations in TP63 we performed homozygosity mapping, followed by exome sequencing of one affected individual. We initially identified three homozygous mutations in the linked region, located in PWP2, MX2 and RIPK4. Recently, mutations in RIPK4 have been reported in Bartsocas-Papas syndrome (OMIM 263650) that shows o

A genome-wide analysis in Anopheles gambiae mosquitoes reveals 46 male accessory gland genes, possible modulators of female behavior.

The male accessory glands (MAGs) of many insect species produce and secrete a number of reproductive proteins collectively named Acps. These proteins, many of which are rapidly evolving, are essential for male fertility and represent formidable modulators of female postmating behavior. Upon copulation, the transfer of Acps has been shown in Drosophila and other insects to trigger profound physiological and behavioral changes in females, including enhanced ovulation/oviposition and reduced mating receptivity. In Anopheles gambiae mosquitoes, the principal vectors of human malaria, experimental

Evolutionary relationships of conserved cysteine-rich motifs in adhesive molecules of malaria parasites.

Malaria parasites invade erythrocytes in a process mediated by a series of molecular interactions. Invasion of human erythrocytes by Plasmodium vivax is dependent upon the presence of a single receptor, but P. falciparum, as well as some other species, exhibits the ability to utilize multiple alternative invasion pathways. Conserved cysteine-rich domains play important roles at critical times during this invasion process and at other stages in the life cycle of malaria parasites. Duffy-binding-like (DBL) domains, expressed as a part of the erythrocyte-binding proteins (DBL-EBP), are such essen

High affinity mimotope of the polysaccharide capsule of Cryptococcus neoformans identified from an evolutionary phage peptide library.

Cryptococcus neoformans causes a life-threatening meningoencephalitis in a significant percentage of AIDS patients. Mice immunized with a glycoconjugate vaccine composed of the glucuronoxylomannan (GXM) component of the cryptococcal capsular polysaccharide conjugated to tetanus toxoid (TT) produce Abs that, based on the epitope recognized, can be either protective or nonprotective. Since nonprotective Abs block the efficacy of protective Abs, we are interested in developing a vaccine that would focus the immune response specifically to protective epitopes. Previously, we screened a phage displ

Molecular surveillance of adenovirus and coronavirus in Procellariiformes, Brazil: Detection of new divergent adenoviruses.

Adenoviruses are widespread DNA viruses that infect a broad range of vertebrate hosts and are recognized for their long-term host coevolution, remarkable genetic diversity, and broad pathogenic potential. Coronaviruses circulate among wild birds due to their evolutionary potential and are relevant to animal and public health. However, the diversity and ecological role of these viruses in pelagic seabirds remain poorly understood. This study provides the first molecular survey of adenoviruses and coronaviruses in Procellariiformes from the South Atlantic. Fifty stranded individuals representing

Ultrasound-Guided Axillary Management in Early-Stage Breast Cancer: From Diagnosis to De-Escalation.

Axillary lymph node management in early-stage breast cancer has undergone a fundamental transformation over the past three decades, shifting from routine radical clearance to precision-guided de-escalation. Ultrasound (US) has emerged as the central imaging modality in this paradigm shift, serving as the first-line tool for preoperative nodal assessment, guidance for biopsy and clip placement, monitoring of response to neoadjuvant chemotherapy (NAC), and localization of marked nodes for targeted axillary dissection (TAD). This review systematically examines the evolution of axillary management

Ultrastructural Insights Into the Reproductive Anatomy and Eggs of Cotton Pink Bollworm, Pectinophora gossypiella Saunders (Lepidoptera: Gelechiidae).

The pink bollworm, Pectinophora gossypiella Saunders is a major pest of cotton, notorious for its high reproductive potential and rapid evolution of resistance to Bacillus thuringiensis (Bt) toxins. Despite its economic significance, detailed knowledge of its reproductive anatomy and egg ultrastructure has remained limited, constraining the development of advanced molecular control strategies such as CRISPR/Cas9-based genome editing. The present study provides the first comprehensive characterization of the reproductive system and egg surface morphology of P. gossypiella using stereomicroscopy

Whole-genome characterization and phylogenetic analysis of pigeon circovirus in racing pigeons from Heilongjiang, China.

Pigeon circovirus (PiCV), a globally distributed pathogen, is associated with immunosuppression and high mortality in racing pigeons. Despite its endemicity in Chinese pigeon populations, molecular epidemiological data on PiCV in Northeast China remain scarce. This study reports the first complete genome sequence of PiCV isolated from racing pigeons in Heilongjiang Province. Through next-generation sequencing (NGS) for whole-genome assembly and systematic PCR-Sanger sequencing for validation, we obtained the complete 2,035-bp genome (designated HLJ2024). Homology analysis revealed nucleotide i

BRAF Mutations in Myeloid Neoplasms: Prevalence, Co-Mutation Landscape, and Clinical Outcomes-A Comprehensive Review.

Background: BRAF is a core component of the RAS-MAPK signaling pathway and an established oncogenic driver in several solid tumors and selected hematologic malignancies. In myeloid neoplasms, BRAF mutations are rare, and their prevalence, molecular context, and clinical significance remain incompletely defined. Available evidence is scattered across heterogeneous reports involving acute myeloid leukemia, myelodysplastic syndromes, myeloproliferative neoplasms, and overlap myelodysplastic/myeloproliferative neoplasms, with variable descriptions of mutation subtypes, co-mutational profiles, cyto

Achieving Blue-Shifted and Optimal Room Temperature Phosphorescence Emission via Hydrostatic Pressure Effect: A Theoretical Perspective.

Hydrostatic pressure provides a powerful external stimulus to modulate the excited-state properties of organic room-temperature phosphorescent (RTP) materials, yet the microscopic origin of pressure-induced spectral evolution remains insufficiently understood. Herein, we present a comprehensive theoretical investigation into the origin of blue-shifted RTP emission in IH-MPT under hydrostatic pressure. The calculations reveal that increasing pressure induces a pronounced blue shift in the phosphorescence spectrum, which originates from a continuous upward shift of the lowest triplet excited sta

Deep Learning-Guided Discovery of Dual Inhibitors of SARS-CoV-2 Entry and 3CL Protease.

The rapid evolution of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) underscores the need for antivirals that are resilient to resistance. Current Food and Drug Administration (FDA)-approved therapies primarily target single viral mechanisms, leaving gaps in efficacy. Here, we developed a Deep Learning-based Activity Screening Model (DLASM), which integrates graph convolutional network with machine learning to identify SARS-CoV-2 inhibitors, using experimental 3-chymotrypsin-like (3CL) main protease assay data. The optimized DLASMs virtually screened ~170,000 compounds from dive

Wavelength tunable ultralong room temperature phosphorescence from 3D-printable benzoindole based materials.

Organic ultralong room temperature phosphorescence (URTP) materials have attracted growing interest due to their persistent afterglow and potential applications in information security, anti-counterfeiting, and advanced photonic systems. Herein, a series of benzoindole (Bd)-based emitters with progressively extended π-conjugation (Bd-NH2, Bd-NAP, Bd-PHE, and Bd-PYR) were rationally designed and synthesized to elucidate the effect of π-extension on triplet-state regulation and phosphorescence behavior. Photophysical investigations reveal that all Bd derivatives exhibit characteristic π-π* trans

Fishing Net-Gravel Interlocking Mechanism to Investigate Molecular Dynamics of Physical Gel Formation in Oil-Water Emulsions: A Simulation Study for an Oil Field in Eastern China.

The viscosity peak phenomenon at the phase inversion point in crude oil emulsions can be understood through the lens of physical gelation. This study employs coarse-grained molecular dynamics (CG-MD) simulations to investigate the gel-like network structures formed at oil-water interfaces across varying water-to-oil particle-number ratios. We reveal that pure water forms a fully connected hydrogen bond network (500 molecules, 313.15 K, 2.68 H-bonds per molecule) behaving as a flexible physical gel scaffold, while pure oil exhibits a dispersed sol-like structure (35.9 clusters average). Venom gland transcriptomics and bioactivity profiling suggest bifunctional hyaluronidase activity in the venom of Mesobuthus crucittii (Scorpiones: Buthidae).

INTRODUCTION: Scorpion venom is a rich source of bioactive molecules with promising biomedical applications. Hyaluronidases are venom-associated enzymes that facilitate toxin diffusion by degrading extracellular matrix glycosaminoglycans, yet their structural diversity and substrate specificity in scorpion venoms remain insufficiently explored. This study aimed to identify and characterize a novel hyaluronidase from the venom gland of the Iranian endemic scorpion Mesobuthus crucittii and evaluate its biochemical and structural properties. METHODS: Venom gland transcriptome profiling was perfo
