Reln Haploinsufficiency Enhances Fentanyl-Induced Locomotion and Dorsal Striatal Activity Without Affecting Opioid Reinforcement and Relapse-Like Behavior.

The Reln gene encodes the extracellular glycoprotein Reelin that regulates synaptic plasticity and activity-dependent gene expression with implication in several neuropsychiatric disorders, including substance use disorder. Although reduction of Reln expression has been associated with altered responses to psychostimulants and cannabinoids, its role in opioid-evoked behaviors has not been investigated. Here, we tested whether Reln haploinsufficiency alters behavioral and molecular responses to the synthetic opioid fentanyl. Heterozygous Reeler (Reln+/-) mice and wild-type littermates were asse

ANOS1 Facilitated Tumorigenesis in Gastric Cancer through the Regulation of Oxidative Phosphorylation Pathway.

Gastric cancer (GC) is the fifth leading cause of cancer-related deaths, with poor early detection and prognosis. Biomarkers related to its development are desperately needed. This study explores ANOS1 expression in GC and its mechanism in tumor metabolic reprogramming. Core molecules were identified via co-expression analysis of The Cancer Genome Atlas (TCGA) and Gene Expression Omnibus (GEO) data. RT-PCR and Western blot were used to confirm the levels of ANOS1 in GC and normal tissues. Assays conducted both in vitro and in vivo verified that ANOS1 accelerates the development of GC. Seahorse

Endometrial HOXA-10, HOXA-11, ß-1 integrin, ECM-1, FAK, and CD44 immunohistochemical expressions in endometriosis-related recurrent IVF failure: a retrospective case-control study.

BACKGROUND: Endometriosis is a common cause of infertility and is frequently associated with recurrent implantation failure in assisted reproductive technologies. Impaired endometrial receptivity, mediated by altered transcription factors, adhesion molecules, and extracellular matrix components, has been proposed as a contributing mechanism. This study aimed to evaluate compartment-specific immunohistochemical expression patterns of HOXA-10, HOXA-11, CD44, β1 integrin, ECM-1, and focal adhesion kinase (FAK) in women with endometriosis-related implantation failure. METHODS: This retrospective c

HAPLN3 p.T34A contributes to incomplete penetrance of moyamoya disease in Chinese carrying RNF213 p.R4810K.

BACKGROUND AND PURPOSE: The penetrance of the RNF213 p.R4810K, a founder mutation of moyamoya disease (MMD), is estimated to be only 1/150-1/300. However, the factors affecting its penetrance remain unclear. Therefore, our study aims to identify modifier genes associated with the incomplete penetrance of this founder mutation. METHODS: Whole-exome sequencing (WES) was performed on 36 participants, including 22 MMD patients and 14 non-MMD controls with RNF213 p.R4810K mutation. Fisher's exact test was used to assess the presence of genetic variants that differed significantly between MMD patie

Mispositioned Neurokinin-1 Receptor-Expressing Neurons Underlie Heat Hyperalgesia in Disabled-1 Mutant Mice.

Reelin (Reln) and Disabled-1 (Dab1) participate in the Reln-signaling pathway and when either is deleted, mutant mice have the same spinally mediated behavioral abnormalities, increased sensitivity to noxious heat and a profound loss in mechanical sensitivity. Both Reln and Dab1 are highly expressed in dorsal horn areas that receive and convey nociceptive information, Laminae I-II, lateral Lamina V, and the lateral spinal nucleus (LSN). Lamina I contains both projection neurons and interneurons that express Neurokinin-1 receptors (NK1Rs) and they transmit information about noxious heat both wi

Reelin controls the positioning of brainstem serotonergic raphe neurons.

Serotonin (5-HT) acts as both a morphogenetic factor during early embryonic development and a neuromodulator of circuit plasticity in the mature brain. Dysregulation of serotonin signaling during critical periods is involved in developmental neurological disorders, such as schizophrenia and autism. In this study we focused on the consequences of defect reelin signaling for the development of the brainstem serotonergic raphe system. We observed that reelin signaling components are expressed by serotonergic neurons during the critical period of their lateral migration. Further, we found that ree

Deep hypothermia reverses behavioral and histological alterations in a rat model of perinatal asphyxia.

The consequences of perinatal asphyxia (PA) include alterations which may manifest as schizophrenia. Characteristic features of this disease include a decrease in specific subpopulations of GABAergic cells and deterioration of social interaction. The purpose of this study is to assess if a deep and short-hypothermic treatment can ameliorate this damage in a model of PA. Rats offsprings were exposed to 19 min of asphyxia by immersing the uterus horns in water at 37 °C followed by 30 min in air at 10 °C that resulted in 15 °C body temperature. At postnatal day 36-38, the rats were tested in the

The established and future biomarkers of malignant pleural mesothelioma.

Malignant pleural mesothelioma (MPM) is an asbestos-related cancer with a median survival of 12months. The MPM incidence is 1-6/100,000 and is increasing as a result of historic asbestos exposure in industrialized countries and continued use of asbestos in developing countries. Lack of accurate biomarkers makes diagnosis, prognostication and treatment prediction of MPM challenging. The aim of this review is to identify the front line of MPM biomarkers with current or potential clinical impact. N-acetyl-cysteine prevents pyramidal cell disarray and reelin-immunoreactive neuron deficiency in CA3 after prenatal immune challenge in rats.

BACKGROUND: Prenatal infection is a major risk factor for the occurrence of neuropsychiatric disorders. These have been associated with hippocampal neuroanatomical and functional abnormalities. In the present study, we evaluated the occurrence of pyramidal cell disarray and reelin neuronal deficit in the hippocampus, and the protective role of N-acetyl-cysteine (NAC) in a rodent experimental model of prenatal immune challenge. METHODS: Sprague-Dawley rats received either 500 μg/kg of endotoxin (lipopolysaccharide, LPS) or 2 ml/kg of isotonic saline by i.p. injection on day 19 of gestation. Af

ErbB/integrin signaling interactions in regulation of myocardial cell-cell and cell-matrix interactions.

Neuregulin (Nrg)/ErbB and integrin signaling pathways are critical for the normal function of the embryonic and adult heart. Both systems activate several downstream signaling pathways, with different physiological outputs: cell survival, fibrosis, excitation-contraction coupling, myofilament structure, cell-cell and cell-matrix interaction. Activation of ErbB2 by Nrg1β in cardiomycytes or its overexpression in cancer cells induces phosphorylation of FAK (Focal Adhesion Kinase) at specific sites with modulation of survival, invasion and cell-cell contacts. FAK is also a critical mediator of in

Modulation of collagen fibrillogenesis by tenascin-X and type VI collagen.

Tenascin-X (TNX) is an extracellular matrix glycoprotein. We previously demonstrated that TNX regulates the expression of type VI collagen. In this study, we investigated the binding of TNX to type I collagen as well as to type VI collagen and the effects of these proteins on fibrillogenesis of type I collagen. Full-length recombinant TNX, which is expressed in and purified from mammalian cell cultures, and type VI collagen purified from bovine placenta were used. Solid-phase assays revealed that TNX or type VI collagen bound to type I collagen, although TNX did not bind to type VI collagen, f

Biophysical and computational characterization identifies FSTL1 as a novel binding target of doxorubicin.

Mapping the protein-binding landscape of doxorubicin (DOX) is pivotal for optimizing therapeutic efficacy and overcoming resistance. In this study, we integrated data from prior protein microarray screenings that identified 363 DOX-binding proteins (with a signal-to-noise ratio (SNR) > 4) along with 224 Homo sapiens expression profiling series obtained from the Gene Expression Omnibus (GEO). Cross-analysis indicated that follistatin-like 1 (FSTL1) consistently demonstrated bidirectional expression across three GEO series (GSE202536, GSE13477, GSE155478), indicating sensitivity to DOX. Network medicine and single-cell mapping identify a collagen-rich, fibrosis-associated hub module in Duchenne muscular dystrophy.

Progressive fibrosis is a hallmark of Duchenne muscular dystrophy (DMD) pathology, driving muscle degeneration and failure. However, the key transcriptomic programs and hub gene networks associated with extracellular matrix remodeling in DMD remain incompletely characterized. We employed weighted gene coexpression network analysis (WGCNA) on transcriptomic data to identify disease-associated modules. Through intersection with GeneCards and topological screening of protein-protein interaction networks, key hub genes were isolated. We further characterized the immune microenvironment via CIBERSO

Cell-type specific contribution of KLF4 to fibrotic nephropathy in tubular epithelium and fibroblast.

Kidney tubulointerstitial disease culminates in renal fibrosis, which is a key determinant of subsequent end-stage renal disease. Krüppel-like factor 4 (KLF4), one of the Yamanaka transcription factors, controls various essential cellular functions, and has been implicated in kidney diseases. However, evidence regarding the role of KLF4 in renal fibrosis specifically within tubular epithelial cells and fibroblasts remains limited. In our study, we observed significant induction of KLF4 protein in tubular and interstitial myofibroblasts from mouse and human fibrotic kidneys. Mice with epitheliu

HRX-215 attenuates cartilage fibrosis in osteoarthritis progression and microfracture repair by targeting MKK4.

Osteoarthritis (OA) is a common degenerative joint disease characterized by progressive cartilage degeneration. Cartilage fibrosis is one of the key pathological events and is also the direct cause of repair failure due to fibrocartilage formation after microfracture surgery. Mitogen-activated protein kinase kinase 4 (MKK4) is a MAP2 kinase that activates c-Jun N-terminal kinase (JNK) and p38 mitogen-activated protein kinase. It is a key kinase in the stress-activated protein kinase (SAPK)/mitogen-activated protein kinase (MAPK) signaling network. The OA-related data set from the GEO database

Changes in the biochemical and gene expression profiles of Palaemon carinicauda during ovarian development.

Palaemon carinicauda is an economically important shrimp species in China. In this study, muscle, hepatopancreas and ovaries were collected to measure their biochemical composition during ovarian development. In addition, dynamic changes in the transcriptome of P. carinicauda ovaries were investigated. The results showed steady increases in glycogen, triglyceride (TG) and protein levels in the ovaries during maturation. The glycogen level increased consistently in the hepatopancreas, while in the muscle, glycogen showed a downward trend after reaching its highest value at Stage III. Similarly,
