Zinc deficiency impacts billions of people and contributes significantly to the increased incidence of community-acquired pneumonia worldwide. Myeloid cells require the zinc transporter ZIP8 for proper host defense. Previously, we observed that infection with S.pneumoniae in myeloid-specific Zip8 knockout mice (Zip8KO) results in increased bacterial burden and mortality despite increased recruitment of macrophages into the lung. Here, we reveal that the lungs of infected Zip8KO mice generate a unique population of dysfunctional macrophages with defects in phagolysosomal function and cell survi

INTRODUCTION: Allogeneic hematopoietic cell transplantation (HCT) can cure hematologic malignancies, but 30-70% of recipients experience acute graft-versus-host disease (GvHD). GvHD is associated with perturbations in the gut microbiome. Bile acids are host derived compounds that are transformed by gut bacteria and bind to specific host cell receptors, informing our hypothesis that changes in bile acid-metabolizing gut bacteria alter bile acid levels to affect gut physiology and immunity during GvHD.
METHODS: In a longitudinal case-control study of patients with and without acute gut GvHD, we

Duckweed-associated plant growth-promoting bacteria have attracted attention for their potential to enhance duckweed biomass production. However, the mechanisms underlying isolate-specific growth promotion are unclear. This study compared the effects of two duckweed-associated bacterial isolates, Terrimicrobium sp. PS02 and Aeromicrobium sp. PS05, on the growth, biomass composition, colonization behavior, and transcriptomic responses of Spirodela polyrhiza. Biomass composition and transcriptome analyses were performed to characterize the host responses. Both isolates enhanced duckweed biomass;

Pathogens deploy specialized mechanisms for tissue entry, barrier crossing, intracellular rerouting, immune remodeling, and persistence within hostile host environments. Although these mechanisms are most often studied in the context of pathogenesis, they also reveal how biological activities can be positioned, routed, sustained, and constrained under the spatial, temporal, immune, metabolic, and tissue-specific pressures encountered in vivo. This Review develops a pathogen-informed design framework for identifying host-facing pathogen functions that can be mechanistically separated from repli

The practical deployment of multi-component luminescent materials is universally hampered by the photodegradation of organic guests, a fundamental challenge that conventional host-centric designs struggle to overcome. Herein, we propose a paradigm-shifting strategy termed "rational guest selection", which elevates the intrinsic molecular photostability of emissive guests to a primary design criterion, equal in importance to emission color. This principle is rigorously validated using the first metal-organic framework constructed from europium ions and pillar[6]arene (Eu-P6MOF) as a model host.

SARS-CoV-2 can infect and adapt to multiple animal species, yet the host determinants that control interspecies viral adaptation remain poorly defined. Here we investigate whether interferon-induced transmembrane protein 3 (IFITM3), a key antiviral protein deficient in certain human populations, affects interspecies adaptation of SARS-CoV-2. We find that SARS-CoV-2 Beta and Omicron BA.4 variants passaged through IFITM3-deficient versus wild type mice exhibit enhanced replication and pathogenesis in this new host species. These enhancements are associated with amino acid substitutions in the vi

Neutrophil migration to bacterial infection sites is key for host defense. Host ribosomal protein SA (RPSA) has been recently reported to regulate the anti-infection immunity of immune cells; however, its role in neutrophil migration remains unclear. Here, using myeloid-specific Rpsa-deficient mice, we found that RPSA deletion inhibited neutrophil infiltration and markedly exacerbated Streptococcus suis serotype 2 infection. Adoptive cell transfer and neutrophil depletion assays identified RPSA as vital for the anti-infective function of neutrophils. Mechanistically, RPSA deficiency induced th

Non-retroviral endogenous viral elements (nrEVEs) are increasingly recognized as potential regulators of host biology, but their functional roles in Lepidoptera remain largely unexplored. Here, we identified seven nrEVEs derived from four RNA virus families in Bombyx mori. All seven nrEVEs were transcriptionally active and exhibited distinct developmental and tissue-specific expression patterns. Their small-RNA profiling revealed that these elements could generate small interfering RNA (siRNA)-like RNAs, primary PIWI-interacting RNA (piRNA)-like RNAs, or ping-pong-associated piRNAs. Functional

Aging men are highly affected by prostate diseases, such as benign prostatic hyperplasia, prostatitis, and prostate cancer. Together, these conditions contribute significantly to the global health burden and are a leading cause of illness in men. Organoid models have transformed disease modeling by allowing the replication of key features of epithelial structure and function in vitro. However, the closed lumen structure of organoids limits experimental access to the apical surface, hindering studies of host-pathogen interactions. To overcome these challenges, we generated a physiologically rel

Proteins containing Toll/interleukin-1 receptor (TIR) domains with catalytic NADase activity have recently emerged as major regulators of innate immunity in both bacteria and plants. In humans, however, only a single protein-SARM1-exhibits TIR-dependent NADase activity. Initially reported to act as a negative regulator of Toll-like receptor (TLR) signaling, SARM1-mediated NAD+ hydrolysis is now recognized as the central driver of Wallerian degeneration, a regulated form of axonal cell death that occurs following injury. Here, we demonstrate that suppression of TLR signaling by SARM1 requires i

Mucormycosis is an emerging, life-threatening human infection caused by Mucorales fungi1-3. Metabolic disorders uniquely predispose an ever-expanding group of patients to mucormycosis through poorly understood mechanisms1,2,4,5, suggesting that uncharacterized host metabolic effectors may confer protective immunity against this infection. Here we uncover a master regulatory role of albumin in host defence against Mucorales through the modulation of fungal pathogenicity. Our initial studies identified severe hypoalb uminaemia as a prominent metabolic abnormality and an independent biomarker of

The role of extracellular vesicle non-coding RNAs in host-parasite interactions remains poorly understood, particularly for human liver flukes. Although tRNA-derived small RNAs (tsRNAs) are emerging as new regulatory molecules in parasite exosomes, they have not yet been characterized for the liver flukes. We performed small RNA sequencing to profile tsRNAs in the exosome-like vesicles derived from the liver fluke Opisthorchis felineus. Transcriptomic data from human cholangiocytes were analyzed to assess the enrichment of the predicted target genes among differentially expressed genes. We ide

Environmental stressors, host condition, and parasitic infections are closely linked, as adverse conditions may compromise individual health and increase susceptibility to parasitism. This study evaluated the influence of intrinsic (body condition) and extrinsic host factors (farm, mining activity, cattle load, and dog abundance) on prevalence and parasite load in the Endangered subpopulation of Chaetophractus vellerosus from eastern Buenos Aires Province, Argentina. Fecal samples from 45 individuals were analyzed using coproparasitological techniques, including modified Ritchie sedimentation

Cetaceans are sentinels of marine ecosystem health and are increasingly exposed to protozoal pathogens. This study investigated the occurrence and molecular identity of Sarcocystidae protozoa in 159 stranded cetaceans representing 21 species along the Brazilian coast. Molecular analysis based on PCR amplification and sequencing of the sarcocystid internal transcribed spacer 1 (ITS1) region revealed infections in 14 individuals, showing a clear habitat-associated distribution. Toxoplasma gondii and Sarcocystis neurona were detected in nine coastal cetaceans, consistent with transmission from te

Ascariasis is a prevalent parasitic infection in both humans and pigs, resulting in significant human morbidity and economic losses in the porcine industry. Experimental infections are often studied by mimicking natural settings, exposing the host to frequent, small doses (trickle infection) or by administering a single (bolus) dose of infective eggs. However, the impact of infection schemes and doses on the host immune response inA. suum infected pigs is still unclear. Using trickle and bolus infections and applying different doses, we found systemic GATA3+CD4+ T cells and eosinophils increas

Despite the remarkable success of antiretroviral therapy (ART) in suppressing human immunodeficiency virus type 1 (HIV-1) replication, viral persistence remains a major barrier to cure. This persistence is sustained by heterogeneous cellular reservoirs in which viral expression is tightly regulated by host-dependent molecular mechanisms. Beyond the canonical cluster of differentiation 4 (CD4+) T-cell reservoirs, HIV-1 establishes long-lived infection in myeloid cells, glial populations within the central nervous system (CNS), and additional non-canonical cellular niches, each characterized by

Considering the therapeutic potential of the Proteus mirabilis PM16 podophage, the interaction between PM16, its host strain, and the mouse immune system was investigated. We evaluated how pre-existing humoral immunity to PM16 influences the immune response against P. mirabilis and the neutralization of the phage itself. Balb/c mice were divided into three groups and immunized two times with (1) 0.9% NaCl, (2) adjuvants, or (3) a mixture of PM16 and an adjuvant. Then, each group was subdivided into three subgroups: mock infection, infection with P. mirabilis, and infection with P. mirabilis fo

Following Mycobacterium tuberculosis (Mtb) infection, the pathogen can establish a metabolically quiescent or replication-arrested dormant state within the host, leading to asymptomatic latent tuberculosis infection (LTBI). Under specific conditions, these dormant bacilli may reactivate and replicate, resulting in symptomatic clinical infection and constituting a major reservoir for active tuberculosis (TB). Dormant Mtb exhibits reduced susceptibility to conventional anti-TB drugs, and current TB vaccines fail to prevent the establishment or reactivation of dormant Mtb, rendering latent infect

The global poultry industry continues to face significant challenges due to Salmonella infections, which pose severe public health concerns and economic losses. Recently, the reemergence of antimicrobial resistance has led to the restriction of antibiotic use in poultry, especially as growth promoters, thus accelerating the search for sustainable alternatives. Among these, probiotics have gained attention as potential candidates for improving poultry health and mitigating Salmonella colonization in the gut. This review summarizes the key mechanisms through which probiotics exert anti-Salmonell

Resistant bacterial infections have become a major global public health challenge, claiming millions of lives annually and imposing enormous economic burdens. The core issue lies in the imbalance of the host immune system, particularly macrophage function. This review elucidates the pivotal role of the host ubiquitin system in macrophage antimicrobial immunity. It systematically examines how this system leverages its unique enzyme-substrate network to orchestrate immune responses with dynamic equilibrium and precision, achieved through two critical pathways: the appropriate modulation of infla
