B-cells play a major role in the immunopathogenesis of autoimmune diseases. Not only do they produce autoantibodies, but they regulate other cell types, secrete cytokines, and present antigens. In genetically susceptible strains of mice, the DA strain of Theiler's virus, a picornavirus, causes a persistent infection of the white matter of the spinal cord associated with chronic demyelination. In resistant strains, on the other hand, the infection is cleared within 1 to 2 weeks. In this study we assessed the clinical utility of measuring all major rheumatoid factor (RF) isotypes (IgG, IgA, and IgM) in the diagnostic immunology laboratory using an enzyme-linked immunoassay (ELISA). New Zealand and New York State Flemish giant rabbits were immunized by intrarectal injections of intact sheep red blood cells (SRC) or sonicated whole sheep blood, with or without prior enemas, and were challenged intravenously with SRC. Since Japanese encephalitis virus (JEV) induces the generation of suppressor T cells in mouse spleen which, through the production of a soluble suppressor factor (SF), suppress IgM plaque-forming cells (IgM-PFC) the present study was undertaken to further characterize these suppressor cells and the SF. The suppressor cells were Ly1-2+ and sensitive to hydrocortisone and a high dose of irradiation. SF was trypsin-sensitive, thermolabile and non-dialysable, and passed through a 450-nm filter. This SF was shown to be a low molecular weight substance, with an approximate MW of 18,000 daltons.
