Huntington's disease (HD) is increasingly recognized as a multisystem disorder in which perturbations of the gut microbiota may influence peripheral homeostasis and shape central neurodegeneration. Across human cohorts, HD is marked by reproducible β-diversity shifts, loss of butyrate-producing taxa, and disruptions in bile-acid and tryptophan-derived metabolites, indicating broad remodeling of microbiota-host metabolic interfaces. Integrating metabolomic evidence with mechanistic data from HD models, we delineate three convergent axes through which dysbiosis may modify disease biology: (i) sh

Grapes are a fruit with origins dating back to ancient times. Their first recorded use, as mentioned in the Bible, was in winemaking. The abundance of bioactive compounds in grapes makes them highly valuable. So far, many varieties of cultivated grapes have been developed for table grapes, wine grapes, and raisin production. In addition to these uses, since grapes contain a variety of nutrients, including resveratrol, flavonoids (such as flavonols, anthocyanins, and catechins), melatonin, vitamins, acids, tannins, and other antioxidants, grape extracts have been widely studied for medical appl

Science and other peer-reviewed studies recognize cadmium (Cd), arsenic (As), mercury (Hg), and fluoride (F) as highly hazardous toxic elements that threaten global health. However, understanding their toxic mechanisms and effective mitigation remains a global challenge. Growing evidence suggests that interleukin-17A (IL-17A) not only plays a role in inflammatory and immune diseases but may also be the brightest target in the mechanisms of toxic injury and its mitigation. This review summarizes IL-17A signaling in toxic element poisoning, highlighting distinct effects on apoptosis, autophagy,

Recent studies have shown a link between disrupted circadian rhythms and the development of chronic opioid-induced negative effects. Both animal and human studies show a significant bidirectional relationship between the circadian system and opioid effects. Opioids can perturb circadian rhythms, and perturbation of the circadian rhythms can aggravate opioid-mediated adverse effects. These bidirectional interactions may attenuate the outcomes of long-term opioid therapy when not considered. A better understanding of the potential mechanisms underlying these interactions may be essential for mor

Complex post-traumatic stress disorder (Complex PTSD) in pediatric patients is challenging to manage, particularly when conventional therapeutic approaches are insufficient. We report the case of Miss A, a 15-year-old girl with a history of severe neglect and abuse, adopted at age 5, who presented with frequent reliving of trauma memories, significant emotional dysregulation, dissociative episodes, recurrent self-harm, and aggression. Her treatment was complicated by comorbid mild-grade learning disability and suspected autism spectrum disorder. Initial management with promethazine, melatonin,

Chemotherapy-induced premature ovarian insufficiency (POI) is a major cause of infertility and hormonal imbalance in young female cancer survivors. In this study, developed a biomimetic scaffold is developed that incorporates polydeoxyribonucleotide (PDRN) and melatonin to restore ovarian function. The scaffold is designed to mimic the ovarian extracellular matrix (ECM), enhancing angiogenesis, promoting antioxidant effects, and reducing reactive oxygen species (ROS). Human embryonic stem cell-derived mesenchymal progenitor cells (hESC-MPCs) are also incorporated to further support tissue rege

This paper pilots a method for the assessment of non-visual lighting effects based upon annual full-spectrum lighting calculations termed an ipRGC-influenced/Non-Visual Spectral Occupant Model (iNSOM). iNSOM calculates annualized melanopic irradiance, described in our previous paper, and derives seasonal and time-of-day metrics based on a collection of photobiological models from Postnova et al., Abeysuria et al. and Tekieh et al. to predict circadian dynamics, alertness and melatonin levels due to light exposure. Quantitative outputs of these metrics and novel spatial visualizations are then

This study evaluates the protective effects of Iris germanica (I. germanica) methanolic extract against L-arginine-induced acute pancreatitis in male rats. The extract's antioxidant potential was confirmed by in vitro assays, revealing 30.76 mg/g of phenolics, 15.86 mg/g of flavonoids, and strong antioxidant activity. HPLC analysis identified catechin and chlorogenic acid as major components. In vivo, rats were divided into five groups: control, L-arginine-only, two I. germanica-treated groups (100 and 400 mg/kg), and a melatonin-treated group. L-arginine elevated serum amylase and lipase leve

Insomnia affects 30% of the U.S. population, with 5% to 15% meeting criteria for chronic insomnia. It can negatively impact quality of life, decrease productivity, increase fatigue and drowsiness, and put patients at higher risk of developing other health problems. Initial treatment focuses on nonpharmacologic therapies such as cognitive behavior therapy, which improves negative thought patterns and behaviors through sleep restriction, stimulus control, and relaxation techniques. Other nonpharmacologic treatments include exercise, mindfulness, and acupuncture. If these approaches are ineffecti

Because hair follicles (HFs) are highly sensitive to ionizing radiation, radiotherapy-induced alopecia (RIA) is a core adverse effect of oncological radiotherapy. Yet, effective RIA-preventive therapy is unavailable because the underlying pathobiology remains underinvestigated. Aiming to revitalize interest in pathomechanism-tailored RIA management, we describe the clinical RIA spectrum (transient, persistent, progressive alopecia) and our current understanding of RIA pathobiology as an excellent model for studying principles of human organ and stem cell repair, regeneration, and loss. We expl

The liver plays pivotal roles in immunologic responses, and correct hepatic adaptations in maternal immunology are required during pregnancy. In this review, we focus on anatomical and immunological maternal hepatic adaptations during pregnancy, including our recent reports in this area. Moreover, we summarize maternal pregnancy-associated liver diseases, including hyperemesis gravidarum; intrahepatic cholestasis of pregnancy; preeclampsia, specifically hemolysis, elevated liver enzymes, and low platelet count syndrome; and acute fatty liver of pregnancy. In addition, the latest information ab

We report the green synthesis of trimethyl chitosan-functionalized poly(2-hydroxyethyl methacrylate) (PHEMA-TMC) nanogels via surfactant-free emulsion photopolymerization. TMC, a quaternized derivative of chitosan, was synthesized through methylation of chitosan, resulting in quaternary and tertiary amine groups as the main substitution products. TMC tertiary amine moiety and riboflavin (RF) acted as a redox photo-initiating system to generate free radicals for the polymerization under light irradiation. The effects of polymerization parameters such as irradiation time, concentrations of TMC a

A neuropsychiatric drug overdose impairs physiological function via central nervous system (CNS) depression. In drug-related deaths, only the drug concentration can currently provide information regarding CNS depression in victims. In this study, using a drug overdose model, we investigated the ability of neurotransmitters in the cerebrospinal fluid (CSF) to serve as biomarkers for CNS depression. Four groups of rats were orally administered diazepam (200 mg/kg) and/or phenobarbital (100 mg/kg) or vehicle. In a hot plate test performed to assess physiological impairment, drug-administered anim

Central serous chorioretinopathy is one of the most frequent causes of vision reduction among middle-aged men. This disease usually has a self-limiting course, but sometimes it lasts more than 4-6 months or a second episode follows a complete resolution of the first one. Nevertheless, to date no consensus exists about the duration threshold and therapy protocols for these non-resolving central serous chorioretinopathy. Treatment as half-dose and half-fluence photodynamic therapy, subthreshold micropulse laser treatment, mineralocorticoid receptor antagonists, intravitreal anti-angiogenic drugs

An adult female spayed dog was evaluated after inadvertently receiving a total dose of 1,750 mg oral cyclophosphamide, equivalent to 2,303 mg/m2, over 21 days (days -21 to 0). Nine days after the last dose of cyclophosphamide (day +9), the dog was evaluated at Perth Veterinary Specialists. Physical examination revealed mucosal pallor, a grade 2/6 systolic heart murmur, and severe hemorrhagic cystitis. Severe nonregenerative pancytopenia was detected on hematology. Broad spectrum antibiotics, two fresh whole blood transfusions, granulocyte colony stimulating factor, and tranexamic acid were adm

In this work, we introduce a number of models for human circadian phase estimation in ambulatory conditions using various sensor modalities. Machine learning techniques have been applied to ambulatory recordings of wrist actigraphy, light exposure, electrocardiograms (ECG), and distal and proximal skin temperature to develop ARMAX models capturing the main signal dependencies on circadian phase and evaluating them versus melatonin onset times. The most accurate models extracted heart rate variability features from an ECG coupled with wrist activity information to produce phase estimations with

Mutual interactions between neurohormones, sleep, and the circadian system have been extensively studied. Hormonal secretion is either influenced by sleep and is independent of circadian timing or is closely coupled with the light-dark cycle, although both processes ultimately interact with each other. Sleep has a strong effect on the levels of some hormones (e.g., growth hormone) but little effect on others that are primarily regulated by the circadian system (e.g., melatonin). The exact mechanisms through which sleep affects circulating hormonal levels are not well understood. Much more is k

Transcriptome profiling of the pineal gland has revealed night/day differences in the expression of a major fraction of the genes active in this tissue, with two-thirds of these being nocturnal increases. A set of over 600 transcripts exhibit two-fold to >100-fold daily differences in abundance. These changes appear to be primarily attributable to adrenergic-cyclic-AMP-dependent mechanisms, which are controlled via a neural pathway that includes the suprachiasmatic nucleus, the master circadian oscillator. In addition to melatonin synthesis, night/day differences in gene expression impact gene
