Correlation between ultrastructural features and adrenal steroid hormone contents was studied in two human adrenocortical adenomas associated with primary aldosteronism. In two adrenocortical adenoma cells, large numbers of lipid droplets were characteristically observed in the cytoplasm, these lipid droplets were closely related to the endoplasmic reticula or mitochondria. On the other hand, biochemically, in two adrenocortical adenomas, aldosterone and progesterone were higher than those in the normal parts of the same adrenal glands. Using a leucocyte migration inhibition test sensitisation to Mallory bodies (alcoholic hyalin) was found in a statistically significant 41% of 17 patients with alcoholic hepatitis. Patients with alcohol-induced fatty liver and cirrhosis did not demonstrate sensitisation. Mallory bodies are a characteristic feature of alcohol-induced liver damage, and immunological sensitisation to them might lead to liver cell death and cell progression of the hepatitis process.

Ultrastructural localization of thiamine pyrophosphatase (TTP) and acid phosphatase (AcPase) activity was performed on jejunal absorptive cells of rats pretreated with the antimicrotubular agent colchicine and of control animals. Demonstration of TPP activity showed that most of the dislocated Golgi stacks after colchicine application lacked positively staining cisternae of the mature side. The ultrastructural alterations of the peripheral blood mononuclear cells of a patient with Reye's syndrome are reported. The involved cells were lymphocytes which showed marked nuclear changes such as irregular or highly convoluted nuclei, multilobulated nuclei, presence of scanty chromatin, and large nucleoli. Changes of the cytoplasmic organelles involved the mitochondria and dense bodies with well defined surrounding membrane appeared. The possibility that these findings indicate a viral etiology of the disease is discussed.

Acute and recovery biopsies of three patients with Reye's Disease are described. Pleomorphic changes of neuronal mitochondria were identified in all of the acute biopsies, similar in appearance to the characteristic alterations of hepatic mitochondria. Guinea pigs were restrained in a smoking machine and were exposed to the smoke of eight cigarettes a day, five days a week for 12 to 15 weeks. One group of animals inhaled filtered smoke from which most of the tar and nicotine had been removed. Heart weight of the smoking animals was found to be significantly increased in both the filtered and unfiltered groups. Halothane-induced contractures in isolated muscle fibers from swine susceptible to malignant hyperthermia (MHS) were significantly less when fibers were incubated in KRB plus 6.2 x 10(-6) M dantrolene sodium prior to the administration of 4 per cent halothane. The presence of denervation-like changes and abnormal mitochondria in the muscle of carriers of the malignant hyperthermia syndrome suggest a neuropathic basis for the human syndrome. A defect in mitochondrial ATP synthesis resulting from denervation, and potentiated by some general anaesthetics, may be the primary muscle fibre lesion in the human malignant hyperthermic syndrome.
