Colistin methanesulfonate against multidrug-resistant Acinetobacter baumannii in an in vitro pharmacodynamic model.

Using an in vitro pharmacodynamic model, a multidrug-resistant strain of Acinetobacter baumannii was exposed to colistin methanesulfonate alone and in combination with ceftazidime. Pre- and postexposure colistin sulfate MICs were determined. A single daily dose of colistin methanesulfonate combined with continuous-infusion ceftazidime prevented regrowth and postexposure MIC increases.

Quinidine-induced agranulocytosis.

A case of agranulocytosis in an 86-year-old man after 8 weeks treatment with quinidine sulfate is described. Acute phase serum from the patient demonstrated antineutrophil activity by the microgranulocytotoxicity assay. Review of the literature reveals that more than one mechanism could cause this idiosyncratic immune-mediated agranulocytosis.

Bioremediation of complex chlorinated-solvent impacted groundwater.

Enhanced in situ bioremediation is a widely adopted and sustainable means of remediating chlorinated contaminant impacted groundwater utilizing organohalide respiring bacteria. These bacteria utilize chlorinated compounds as terminal electron acceptors to harvest energy, producing partially or fully dechlorinated end products. A barrier to successful remediation utilizing organohalide respiring bacteria is the impact of environmental factors which may be toxic or otherwise inhibitory towards these organisms or their respiration pathways, particularly in sites with complex contaminant profiles

The Metabolomic Signature of Stressful Life Events.

Stressful life events impact individual's functionality and contribute to disease outcomes, yet the biological pathways underlying life stress remain unclear. We characterized the metabolomic profiles of stressful life events using data from 3,264 participants (5,163 observations) of the Dutch NESDA cohort. 98 metabolites were identified, with upregulated metabolites overrepresented in phosphatidylethanolamine and downregulated metabolites overrepresented in fatty acid metabolism. 92 of these metabolites were available in the Dutch NEO cohort (N=599): 11 were significantly replicated including

Sulfur‑Containing Additives for Enhanced Kinetics and Interfacial Stability of Phosphorus Anodes in Li‑Ion Batteries.

Red phosphorus offers ultrahigh theoretical capacity for lithium-ion battery anodes but is fundamentally limited by poor electronic conductivity, sluggish lithium-ion diffusion, and severe volume expansion that destabilizes the solid-electrolyte interphase. Here, we introduce a synergistic strategy that simultaneously regulates reaction kinetics and interfacial stability using sulfur-containing additives, lithium sulfide, and in situ generated lithium sulfate. Lithium sulfide confines lithium polyphosphide intermediates, enabling a direct and highly reversible conversion reaction, while lithiu

Chimeric enediynes as potent antivirals against coronaviruses by targeting the conserved free-fatty-acid binding site on the spike protein.

Conventional targeted protein degradation (TPD) strategies operate by hijacking intracellular proteolytic machinery, limiting their therapeutic scope. Here, we present protein-radical-oxidation targeting enediyne chimera (PROTEC), a TPD platform that can be located exclusively outside of host cells and degrades pathogen proteins via a radical-based oxidative mechanism. The maleimide-fused enediyne core undergoes cycloaromatization under physiological conditions to generate reactive radical species. Strategic incorporation of sulfate-based hydrophilicity-adjusting groups (HAGs) restricts PROTEC

Select microbial metabolites promote tau aggregation in a murine tauopathy model.

The gut microbiome is emerging as a modifier of risk for neurodegenerative diseases, but underlying mechanisms remain poorly understood. Here, we show that the hTau.P301S mouse model for progressive tauopathy develops alterations in the composition and function of the gut microbiome that are not recapitulated in amyloid-based 5xFAD or 3xTg models for Alzheimer's disease. Disrupting the gut microbiome via chronic antibiotic treatment exacerbates cognitive deficits and tau pathology in hTau.P301S mice, demonstrating a causal influence of the microbiome on tau-driven disease progression. This cor

Therapeutic Potential of Vanillic Acid in Ulcerative Colitis Through Microbiota and Macrophage Modulation.

This study investigated the protective effects of the dietary polyphenol vanillic acid (VA) on dextran sulfate sodium-induced acute ulcerative colitis (UC) in mice, focusing on its impact on the gut microbiota and inflammatory responses. VA was supplemented following dextran sulfate sodium administration, and key indicators, including body weight, disease activity index, colon length, spleen index, and inflammatory markers, were assessed. VA supplementation significantly alleviated UC symptoms, preserved intestinal barrier integrity, and reduced pro-inflammatory cytokine levels. Additionally,

Targeting cancer stem-like cells via cholesterol modulation and ferroptosis induction using a multifunctional nanoplatform to overcome drug resistance.

Overcoming therapy resistance in triple-negative breast cancer (TNBC) requires the effective targeting of cancer stem-like cells (CSCs). TNBC is characterized by hyperactivation of the mevalonate pathway, leading to cholesterol accumulation in CSC membranes, which alters membrane properties, enhances stemness, and restricts both drug penetration and lipid peroxidation-a key driver of ferroptosis. Here, we develop Fe/CDP, a nanoparticle with a Fe3O4 core coated with chondroitin sulfate and loaded with pravastatin, a mevalonate pathway inhibitor, and doxorubicin (DOX). In TNBC mouse models, Fe/C

Targeting the Gut-Kidney Axis: Modulation of Gut Microbiota by Traditional Chinese Medicine for Chronic Kidney Disease Management.

The interaction between gut microbiota dysbiosis and CKD progression via the "gut-kidney axis" is increasingly recognized. Gut-derived uremic toxins (e.g., indoxyl sulfate and p-cresyl sulfate) accumulate systemically, while beneficial metabolites like short-chain fatty acids (SCFAs) decrease, contributing to inflammation, oxidative stress, and kidney fibrosis. Traditional Chinese Medicine (TCM), including complex formulae, single herbs, and active ingredients, has long been used to manage CKD. Emerging evidence-primarily from animal studies-highlights its potential to alleviate the disease by

Surfactant-rescued gelation: Stabilizing P123-chitosan hydrogels in blood-isotonic aqueous solutions for controlled drug delivery.

The loss of gelation in Chitosan-Pluronic composite hydrogels under physiological saline conditions restricts their potential for drug delivery applications. To this effect, we present a dynamic and composite hydrogel system based on Pluronic P123 and chitosan, which restores gelation upon the addition of sodium dodecyl sulfate (SDS), even in the presence of NaCl at blood-isotonic concentrations. The entry of SDS induces electrostatic interactions and hydrophobic associations, enabling robust gel formation under physiological conditions. The gelation behavior can be desirably modified by varyi

Nickel and Cobalt Recovery from Spent Lithium-Ion Batteries via Electrodialysis Metathesis.

Recycling of spent lithium-ion batteries is important due to the increasing demand for electric vehicles and efforts to realize a circular economy. There is a need to develop environmentally friendly processes for the refining of nickel, cobalt, and other metals contained in the batteries. Electrodialysis is an appealing method for recycling of battery metals with selective separation and low chemical input. In this study, sodium sulfate was used in an electrodialysis metathesis procedure to sequentially separate EDTA-chelated nickel and cobalt. Replacing hitherto used sulfuric acid with sodiu
