[Effects of heat stress on DA mediated PI signal transduction system in rat striatum].

Objective. To study the effects of heat stress on dopamine (DA) and related phosphatidylinositol (PI) signal transduction system members: PLA2, PI, Ca2+ in rat striatum. Method. Male Wistar rats were randomly divided into control group and heat stressed group. Heat stressed rats were placed in small hot chambers and taken out as soon as their rectal temperatures (Tr) reached the preset temperature. Then the animals were killed and their striatums were taken out. Fluorospectrophotometry, HPLC, Fura-2/AM fluoresence labelled method and acidalkaline titration were used to measure the content of D

Special relationship between mitochondria and hypolemmal nerve terminals in salivary glands of some bats.

BACKGROUND: Hypolemmal nerve terminals, which are abundant in many types of salivary glands, are naked axons that have penetrated the basement membrane to take up a position between adjacent parenchymal cells. Although they may form vesicle-filled varicosities, there usually are no obvious morphological indications as to which cell actually is being innervated. METHODS: Specimens of over 200 species of bats were live-trapped and their major salivary glands extirpated and prepared for electron microscopic examination. RESULTS: In 13 species of bats out of the more than 200, mitochondria-hypol

Synergism between cAMP and ATP in signal transduction in cardiac myocytes.

ATP transiently increases the intracellular Ca2+ concentration in cardiac myocyte suspensions. Pretreatment with norepinephrine (NE) greatly potentiates the ATP response. We performed experiments on adult rat myocyte suspensions loaded with fura-2 to investigate the mechanism of NE potentiation. We found that forskolin (an activator of adenylate cyclase), 3-isobutyl-1-methylxanthine (an inhibitor of phosphodiesterase), and permeative adenosine 3',5'-cyclic monophosphate (cAMP) analogues potentiate the increase in cytosolic Ca2+ concentration induced by ATP. NE, forskolin, and 8-(4-chlorophenyl

Activity-dependent Notch signalling in the hypothalamic-neurohypophysial system of adult mouse brains.

Notch signalling has a key role in cell fate specification in developing brains; however, recent studies have shown that Notch signalling also participates in the regulation of synaptic plasticity in adult brains. In the present study, we examined the expression of Notch3 and Delta-like ligand 4 (DLL4) in the hypothalamic-neurohypophysial system (HNS) of the adult mouse. The expression of DLL4 was higher in the supraoptic nucleus (SON) and paraventricular nucleus (PVN) compared to adjacent hypothalamic regions. Double-labelling immunohistochemistry using vesicular GABA transporter and glutamat

Differential regulation of Notch ligands in dendritic cells upon interaction with T helper cells.

The Notch signalling pathway has recently been linked to T helper 1 (Th1)/T helper 2 (Th2) cell polarization via a mechanism involving differential expression of Notch ligands, Delta-like and Jagged, in antigen-presenting cells. However, whether stimuli other than pathogen-derived factors are involved in the regulation of Notch ligand expression in dendritic cells (DCs) remains unknown. Here, we address the effect of T helper cells (Th1 and Th2) on Delta-like 4 and Jagged 2 expression in bone marrow-derived DCs. We demonstrate that both Th1 and Th2 cells induce Delta-like 4 mRNA expression in

Th2 Cytokines Reshape the Transcriptome: Insights from a Canine Organoid Model of Atopic Dermatitis.

Atopic dermatitis (AD) and canine atopic dermatitis (CAD) are common allergic and pruritic skin diseases characterized by immune dysregulation and epidermal barrier dysfunction. To delineate how Th2 cytokines contribute to CAD pathogenesis, canine primary epidermal organoids (cPEOs) were established from keratinocytes, and exposure to IL-4/IL-13 induced morphologic changes characteristic of CAD. RNA sequencing analysis comparing IL-4/IL-13-treated cPEOs to untreated controls identified 224 differentially expressed genes (DEGs). Further rigorous filtering narrowed this down to 69 key DEGs, with

Comprehensive vasodilatory and antihypertensive effects of 3,7,3',4'-tetrahydroxyflavone (fisetin): Ex vivo and in vivo evidence.

Hypertension is a chronic condition characterised by persistently elevated arterial blood pressure and remains a major global health burden. Although many antihypertensive agents are clinically effective, treatment is often limited by poor adherence and adverse effects, underscoring the need for alternative or adjunctive therapies with favourable tolerability. Fisetin, a naturally occurring plant flavonol found in fruits and vegetables, has been reported to exert antihypertensive activity; however, the vascular mechanisms underlying its effects remain incompletely defined. This study investiga

Self-Powered CaO2/Heme@Zn-MOF Enabled Fluorescence Immunosensing Strategy for Accurate and Selective Isoprocarb Monitoring.

Accurate and sensitive detection of pesticide residues is crucial for environmental safety and human health. Herein, a self-powered metal-organic framework nanozyme-based immunoassay was developed for selective and sensitive detection of isoprocarb by encapsulating calcium peroxide and heme into a Zn-MOF to construct the CaO2/heme@Zn-MOF composite. The prepared nanozyme exhibits intrinsic peroxidase-like activity and enables in situ H2O2 generation under acidic conditions, effectively eliminating the need for external oxidants. By coupling antibody-antigen specific recognition with catalytic s

Calretinin-Positive Neurons in Central Medial Thalamus Drive Nociception and Anxiety via Projections to Anterior Insular Cortex in male mice.

The central medial thalamic nucleus (CM), a key component of the thalamic intralaminar nuclei, receives nociceptive inputs from the lateral parabrachial nucleus and is implicated in pain processing. However, the specific neuronal subtypes and downstream pathways that mediate these functions remain poorly defined. Here, we combined tract tracing, immunofluorescence, in situ hybridization, chemogenetics, fiber photometry, and behavioral tests to investigate the role of calretinin-positive (CR+) neurons in the CM (CMCR+) and their projections to the anterior insular cortex (aIC) in nociceptive tr

Piezo1 Mechanotransduction in Skeletal Muscle: Convergence with Noncoding RNA Regulation in Myogenesis, Regeneration, and Sarcopenia.

Skeletal muscle is a continuously load-bearing tissue whose growth, repair, and age-related decline are governed by mechanical signals; failure of this mechano-regulation underlies disuse atrophy and sarcopenia. Piezo1, a mechanically activated cation channel, has emerged as a tractable transducer of these signals in muscle, contributing to satellite-cell quiescence and senescence, regenerative division, myoblast fusion, and the response to loading and unloading. In parallel, the myogenic noncoding RNA program is among the best defined in any lineage, with myomiRs miR-1/133/206, the long nonco

Cell Therapy as Metabolic Rescue after Ischemic Stroke: Rewiring Bioenergetics, Redox Homeostasis, and Neurovascular Repair.

Ischemic stroke represents a dynamic metabolic disorder of the neurovascular unit (NVU) rather than a static vascular occlusion followed by neuronal demise. Immediate oxygen and glucose deprivation rapidly deplete ATP, disrupt the transmembrane ionic gradients, increase glutamate excitotoxicity, and overload mitochondrial with calcium. These events alter glycolytic, lipid, amino acid, and redox pathways. During the subacute and chronic phases, astrocytes, microglia, macrophages, endothelial cells, pericytes, oligodendrocytes, and surviving neurons continue to remodel substrate utilization. The

Obestatin treatment links mitochondrial homeostasis and skeletal muscle repair in Duchenne muscle dystrophy.

Duchenne muscular dystrophy (DMD) is a genetic, progressive neuromuscular disease caused by mutations in the dystrophin protein which compromise the integrity of the sarcolemma. Current care of DMD involves both supportive and targeted disease modifying medications. Obestatin, a peptide derived from preproghrelin, is a potential candidate to enhance existing treatments for DMD. This study was conducted to analyse the molecular mechanism by which obestatin acts on myofiber metabolism and muscle restructuring in DMD. Through human and animal models of DMD, we identify the calcium-activated prote

Comparing the Representation of a Simple Visual Stimulus across the Cerebellar Network.

The cerebellum is a conserved structure of the vertebrate brain involved in the timing and calibration of movements. Its function is supported by the convergence of fibers from granule cells (GCs) and inferior olive neurons (IONs) onto Purkinje cells (PCs). Theories of cerebellar function postulate that IONs convey error signals to PCs that, paired with the contextual information provided by GCs, can instruct motor learning. Here, we use the larval zebrafish to investigate (1) how sensory representations of the same stimulus vary across GCs and IONs and (2) how PC activity reflects these two d

SCUBE3 promotes osteogenic differentiation and mitophagy in human bone marrow mesenchymal stem cells through the BMP2/TGF-β signaling pathway.

In clinical settings, addressing large bone defects remains a significant challenge for orthopedic surgeons. The use of genetically modified bone marrow mesenchymal stem cells (BMSCs) has emerged as a highly promising approach for these treatments. Signal peptide-CUB-EGF domain-containing protein 3 (SCUBE3) is a multifunctional secreted glycoprotein, the role of which remains unclear in human hBMSCs. This study used various experimental methods to elucidate the potential mechanism by which SCUBE3 influences osteogenic differentiation of hBMSCs in vitro. Additionally, the therapeutic efficacy o

High expression of P-selectin induces neutrophil extracellular traps via the PSGL-1/Syk/Ca2+/PAD4 pathway to exacerbate acute pancreatitis.

BACKGROUND: Excessive neutrophil extracellular traps (NETs) is involved in the progression of acute pancreatitis (AP) but the mechanisms controlling NETs formation in AP are not fully understood. Therefore, our study sought to investigate the mechanism of the highly expressed P-selectin stimulating the formation of NETs in AP. METHODS: NETs formation was detected by flow cytometry, immunofluorescence staining, and cf-DNA and MPO-DNA complexes were measured as biomarkers of NETs formation. Neutrophils treated with P-selectin and pharmacological inhibitors were examined by western blot, immunof

Carbon monoxide/heme oxygenase system in plant: Roles in abiotic stress response and crosstalk with other signals molecules.

Carbon monoxide (CO) has been recognized as a crucial gasotransmitter mainly produced by heme oxygenase (HO)-catalyzed heme degradation in plant. Recent studies have shown that CO plays an important role in regulating growth and development of plant, as well as and responding to a variety of abiotic stresses. Meanwhile, many studies have reported on CO working in combination with other signal molecules to mitigate abiotic stress. Here, we presented a comprehensive overview of recent developments in which CO reduces plant damage caused by abiotic stresses. The regulation of antioxidant system,

Stem/Proliferative and Differentiated Cells within Primary Murine Colonic Epithelium Display Distinct Intracellular Free Ca2+ Signal Codes.

The second messenger, intracellular free calcium (Ca2+ ), acts to transduce mitogenic and differentiation signals incoming to the colonic epithelium. A self-renewing monolayer of primary murine colonic epithelial cells is formed over a soft, transparent hydrogel matrix for the scalable analysis of intracellular Ca2+ transients. Cultures that are enriched for stem/proliferative cells exhibit repetitive, high frequency (≈25 peaks h-1 ), and short pulse width (≈25 s) Ca2+ transients. Upon cell differentiation the transient frequency declines by 50% and pulse width widens by 200%. Metabolites and

Patient Endothelial Colony-Forming Cells to Model Coronary Artery Disease Susceptibility and Unravel the Role of Dysregulated Mitochondrial Redox Signalling.

Mechanisms involved in the individual susceptibility to atherosclerotic coronary artery disease (CAD) beyond traditional risk factors are poorly understood. Here, we describe the utility of cultured patient-derived endothelial colony-forming cells (ECFCs) in examining novel mechanisms of CAD susceptibility, particularly the role of dysregulated redox signalling. ECFCs were selectively cultured from peripheral blood mononuclear cells from 828 patients from the BioHEART-CT cohort, each with corresponding demographic, clinical and CT coronary angiographic imaging data. Spontaneous growth occurred
