Flagellin is a potent activator of a broad range of cell types that are involved in innate and adaptive immunity. Therefore, it is a good adjuvant candidate for vaccines, and it might function as a biological protectant against both major acute radiation syndrome during cancer radiotherapy and a mitigator of radiation emergencies. However, accumulating evidence has implicated flagellin in the occurrence of some inflammatory diseases, such as acute lung inflammation, cardiovascular collapse and inflammatory bowel disease. The aim of this study was to elucidate whether only flagellin-TLR5 signal

Infection with high-risk human papillomavirus (HPV) causes cervical intraepithelial neoplasia (CIN) and invasive cervical cancer (ICC). The distribution of HPV types in cervical diseases has been previously described in small studies for Canadian women. The prevalence of 36 HPV genotypes in 873 women with CIN and 252 women with ICC was assessed on cervical exfoliated cells analyzed with the Linear Array (Roche Molecular System). HPV16 was the most common genotype in CIN and ICC. The seven most frequent genotypes in order of decreasing frequency were HPV16, 51, 52, 31, 39, 18, and 56 in women w

This review summarizes clinically important findings from 17 systematic reviews and 2 guidelines on skin cancer indexed between April 2008 and April 2009. Melanoma primary-prevention measures, such as education, are more likely to be successful in younger children than adolescents, and general population screening for melanoma by whole-body examination is not currently supported by the evidence. A large systematic review of melanoma and pregnancy concluded that pregnancy does not affect prognosis. Two systematic reviews imply that sunburn later in life also increases the risk of melanoma, and

The range of medicines and technologies that are essential for sexual and reproductive health care is well established, but access to them is far from universally assured, particularly in less developed countries. This paper shows how the pharmaceutical industry plays a major role in the lack of access to essential medicines for sexual and reproductive health care, by a) investing in products for profit-making reasons despite their negative health impact (e.g. hormone replacement therapy), b) marketing new essential medicines at prices beyond the reach of countries that most need them (e.g. HP

The efficacy of current cancer vaccines is limited by the functional heterogeneity and poor availability and expansion of professional antigen-presenting cells (APCs). Besides their potent innate effector properties, gammadelta T cells have been suggested to be involved in the initiation and maintenance of adaptive immune responses. Here, we investigated the capacity of human gammadelta T cells to induce expansion of virus-specific T cells to Epstein Barr virus (EBV) antigens. Aminobisphosphonate-stimulated human peripheral blood-derived gammadelta T cells (Vgamma2+Vdelta2+) acquired a dual ph

Therapeutic nucleic acids (TNAs) and its precursors are applied to treat several pathologies and infections. TNA-based therapy has different rationales and mechanisms and can be classified into three main groups: 1) Therapeutic nucleotides and nucleosides; 2) Therapeutic oligonucleotides; and 3) Therapeutic polynucleotides. This review will focus in those TNAs that have reached clinical trials with anticancer and antiviral protocols, the two most common applications of TNAs. Although therapeutic nucleotides and nucleosides that interfere with nucleic acid metabolism and DNA polymerization have

Recent developments in recombinant DNA research promise to have a substantial impact on medicine. The technology now allows for DNA pieces to be isolated and ligated to other DNA, such as that from bacteria, and be replicated in foreign hosts. The replicated DNA can then be isolated and used to study its structure, to study gene expression and its regulation, to program microorganisms to synthesize medically, agriculturally or industrally important proteins, to transfer genes back into mammalian cells and to diagnose genetic diseases. The DNA used for cloning can be obtained by copying mRNA, b

BACKGROUND: Many emerging adults (EAs) are prone to making unhealthy choices, which increase their risk of premature cancer morbidity and mortality. In the era of social media, rigorous research on interventions to promote health behaviors for cancer risk reduction among EAs delivered over social media is limited. Cancer prevention information and recommendations may reach EAs more effectively over social media than in settings such as health care, schools, and workplaces, particularly for EAs residing in rural areas.
OBJECTIVE: This pragmatic randomized trial aims to evaluate a multirisk fac

BACKGROUND: Current consensus recommendations are to not initiate cervical cancer screening for immunocompetent adolescent females before 21 years of age. This is in part because of the very low rate of 0.8 per 100,000 new cervical cancer cases diagnosed among women aged between 20 to 24 years. Timely human papillomavirus vaccination further decreases the incidence of cervical cancer to 4 cases per 100,000 persons by the age of 28 years. Screening before 21 years of age has demonstrated no clear benefit in cancer risk reduction or outcomes. In addition, unindicated screening among adolescents

BACKGROUND: Human papillomaviruses (HPVs) are associated with a wide variety of cutaneous and mucosal infections and with malignancies in humans. More than 100 HPV types have been identified, some of which have affinity for skin and others for mucosal sites.
AIM: The purpose of this study came from our desire to support the "Stop Cervical Cancer" campaign, adopted as "Cervical Cancer Removed as a Public Health Problem", following the World Health Organization (WHO) initiative in 2019-2020. At European level, cervical cancer mortality has fallen by more than 30% over the past 30 years, through

INTRODUCTION: New York City was the epicenter of the first wave of the COVID-19 pandemic, and cancer patients were among the most vulnerable groups. We characterized antibody response in a diverse cohort of cancer patients prior to the availability of COVID-19 therapeutics. We report: (1) antibody response to COVID-19 by cancer type; (2) trend of SARS-CoV-2 IgG spike antibodies over time in cancer patients with COVID-19 diagnosis during 2020-2021.
METHODS: Single-center, prospective, observational study of patients treated for cancer at Memorial Sloan Kettering Cancer Center with symptomatic,

Non-cytotoxic cancer therapies increasingly seek to control malignant systems rather than maximize tumor-cell killing. Educative strategies aim to impose or restore less harmful states such as dormancy, differentiation, tumor microenvironment normalization or reduced invasion. Pressure-dependent evolutionary containment is a related control strategy, not evidence of durable reprogramming by itself. The limitations of size-based assessment are already well established: Choi, PERCIST, mRECIST, Lugano and iRECIST each operationalized a disease-specific alternative. This review's contribution is t

Neuroblastoma is the most common extracranial solid tumor in children. It remains a major therapeutic challenge, particularly in high-risk cases where long-term survival rates remain below 50 % despite intensive multimodal treatment. The immunosuppressive tumor microenvironment (TME), marked by low MHC class I expression and infiltration of regulatory immune cells, has historically limited the efficacy of immune-based therapies. Recent advances in immunotherapy have reshaped the treatment landscape, with anti-GD2 monoclonal antibodies such as dinutuximab and naxitamab demonstrating significant

INTRODUCTION: The combination of phototherapy (PTT/PDT) and immunotherapy holds promise for cancer treatment but is hindered by poor spatiotemporal coordination and inadequate targeted co-delivery. Nanomedicine carriers offer a platform to overcome these barriers by enabling precise control over drug release kinetics and reducing off-target toxicity. Methods This review systematically analyzes the rationale, mechanisms, and recent advances in nanocarrier-mediated phototherapy-immunotherapy combination strategies, with a focus on studies published within the last three years. We examine how nan

Oncology drug development continues to have a high attrition rate despite major advances in therapeutic modalities such as antibody-drug conjugates, bispecific antibodies, cell therapies, and cancer vaccines. Critical development decisions are often made under substantial uncertainty, creating a need for quantitative approaches that integrate diverse sources of evidence. Quantitative medicine (QM) and model-informed drug development (MIDD) provide a framework to support decision-making throughout the oncology drug development lifecycle by leveraging pharmacology, disease biology, clinical data

Messenger RNA (mRNA) is a promising therapeutic for genetic defects, gene editing and vaccines. The success of mRNA therapies largely depends on the optimal delivery vehicles. Although lipid nanoparticles are the leading option, some polymers exhibited attractive properties in mRNA delivery. In this study, branched polyethylene-imine (PEI) with molecular weight (Mw) of 1.8 kDa, 10 kDa and 25 kDa were explored for mRNA delivery. The complexation, dissociation and stability of bPEI/mRNA polyplexes were examined by gel electrophoresis. In vitro cellular uptake and transfection efficiency were eva
