A 47-year-old white male developed massive hepatosplenomegaly, a pleural effusion, leucocytosis, and a left parasternal mass following a relatively symptom-free persistent hypereosinophilia for about 5 years. Bone marrow aspiration and biopsy and peripheral blood differential showed eosinophilia and a shift to the left with immature cells. A high serum B12 vitamin level and low LAP activity were found. Biopsy of the soft tissue mass revealed a granulocytic sarcoma (chloroma) with a hyperdiploid karyotype (49,XY, + 10, + 15, + 19,3q-), whereas the bone marrow cells had a normal male karyotype.

Murine B lymphocytes, adipocytes, and olfactory neurons contain a DNA-binding protein that participates in the regulation of genes encoding tissue-specific components of signal transduction. Purification and cloning of this protein, termed early B-cell factor (EBF), from murine B lymphocytes and independent cloning of a protein, termed Olf-1, from olfactory neuronal cells revealed virtual complete amino acid sequence identity between these proteins. As a first step towards identifying a human genetic disorder or mouse mutation for which EBF could be a candidate gene, we have chromosomally mapp

This chapter describes a lambda Red-based recombineering approach for targeted genetic manipulation in Chlamydia species. The method uses a non-replicative plasmid encoding lambda Red recombination proteins together with locus-specific homology arms to generate precise gene deletions in the Chlamydia chromosome. Detailed protocols are provided for construct design, host cell-based selection, and genetic validation of recombinant strains, and noteworthy considerations for transforming different Chlamydia species and strains are given.

Graft-versus-leukemia (GVL) effect is mainly mediated by alloreactive T and NK cells through human leukocyte antigen (HLA). Although most HLA genes are located on the short arm of chromosome 6, the clinical significance of deletion of the short arm of chromosome 6 (del6p) which could attenuate the GVL effect has not been elucidated. We investigated impact of del6p at the diagnosis of hematological malignancies on allogeneic cell transplantation, using a Japanese registry database. Del6p was highly associated with additional chromosomal abnormalities. In the univariate analysis, the del6p group

Telomere biology is often framed as a uniquely eukaryotic problem, yet the challenges of maintaining DNA ends are shared by all organisms harboring linear DNA, including viral entities. Telomeres enable complete replication of chromosome ends while shielding the termini from DNA repair factors and innate immune surveillance systems that would otherwise recognize them as damaged or foreign DNA. A particular threat is posed by Mre11-Rad50, a nuclease complex conserved from bacteria to humans that specializes in the detection and processing of DNA ends. The solutions to these DNA end-related chal

Chromosome 3q (3q) amplification is a frequent event in non-small cell lung cancer (NSCLC) and other cancer types, yet therapeutic strategies targeting oncogenic drivers within this amplicon have been unsuccessful. Our study revealed that the 3q amplicon encodes multiple Golgi-related proteins, including Phospholipase D1 (PLD1), which catalyzes the production of a potential oncogenic lipid, phosphatidic acid. We found that PLD1 is highly expressed in 3q-amplified NSCLC (3q-NSCLC) and its enzymatic activity is further boosted by coamplified PLD1 activator, ARF-like GTPase 14. High PLD1 activity

Proper segregation of meiotic chromosomes depends on the controlled formation and resolution of recombination intermediates that give rise to crossovers (COs). While the BTRR complex-comprising the BLM helicase, topoisomerase IIIα, and the proteins RMI1 and RMI2-is a well-characterized regulator of recombination in somatic cells, its function during meiosis remains incompletely understood, particularly for RMI2 in mammals. Here, we investigate the role of RMI2 in mouse spermatogenesis using a targeted RMI2 knockout model. Loss of RMI2 results in severe meiotic defects, including reduced testis…

Children with Sex Chromosome Trisomies (SCTs) face a risk for developmental language vulnerabilities. Understanding how the Child-Directed Speech (CDS) adapts to these developmental trajectories is crucial for early intervention. This longitudinal study mapped the evolution of CDS addressed to 16 children with SCTs from 8 to 24 months of age. Results revealed significant changes. Structurally, the input transitioned in line with typical development, showing significant increases in lexical variety and syntactic complexity. Functionally, mothers transitioned from a more affective/attention-gett

Axenfeld-Rieger Syndrome (ARS) is a rare genetic disorder defined by ophthalmological abnormalities, frequently accompanied by craniofacial and dental anomalies. Although mutations in the PITX2 and FOXC1 genes are commonly identified, some patients do not receive molecular confirmation. This report describes a clinically compelling case of ARS with a typical phenotype but negative genetic testing. A 10-year-old male demonstrated physical abnormalities from early childhood and underwent multiple medical evaluations without a definitive diagnosis. He experienced difficulty and discomfort during

Chromosome architecture is essential for faithful chromosome segregation and genome stability. However, the mechanisms underlying chromosome organization are not well-characterized. Here, we identify the ubiquitin ligase Ufd4p as a master regulator of chromosome organization. Ufd4p deficiency causes longer chromosome axes and increased meiotic crossovers due to compromised condensation. Multiomics profiling reveals that Ufd4p promotes transcription of condensin components required for chromosome condensation. Mechanistically, Ufd4p targets Sum1p, a subunit of histone deacetylase complex, for p

Polyploidization in peanut (Arachis hypogaea L.) fixed heterosis but also caused a genetic bottleneck isolating cultivated peanut from its wild diploid relatives. Mechanisms such as homoeologous exchange can partially restore genetic diversity by generating new allelic combinations. Double reduction is a rare polyploid-specific segregation pattern in which a single-dosage locus yields duplex gametes. It requires multivalent formation-associated with homoeologous exchange in allopolyploids-and crossing over between non-sister chromatids. Although peanut mainly exhibits disomic pairing, occasion

Triple-negative breast cancer (TNBC) is characterised by high rates of chromosomal instability (CIN) and rewired intercellular communication driven by both soluble factors and extracellular vesicles (EVs). To assess how CIN might affect EV-mediated signalling in TNBC, we studied the EV landscape of TNBC cell lines with induced CIN. We find that CIN leads to increased secretion of EVs and that these EVs promote cell migration of recipient cells. EVs are enriched for extracellular matrix (ECM) proteins, including EFEMP1. Indeed, modulation of EFEMP1 levels in EVs significantly alters migration b

cGAS is the primary innate immune DNA sensor. On binding DNA, cGAS generates cGAMP, ultimately driving inflammation. Although normally silenced on self-DNA, genotoxic stress can activate cGAS, proposed to be mediated by micronuclei, chromosome bridges and DNA:RNA hybrids. However, mechanistically, this is poorly understood due to a lack of sensitive and selective single-cell cGAS activation assays. Here we solve this with an improved cGAMP reporter for microscopy, flow cytometry and biochemical assays. Strikingly, we find that genotoxic stress-mediated cGAS activation is a rare event that is n
