Spontaneous release of acetylcholine from Torpedo synaptosomes: effect of cetiedil and its analogue MR 16728.

The effects of cetiedil and its analogue MR 16728 were examined on spontaneous acetylcholine release measured with a chemiluminescent assay using choline oxidase in a synaptosomal suspension obtained from Torpedo marmorata electric organ. Evoked acetylcholine release is inhibited by cetiedil, whereas this drug enhances spontaneous extracellular Ca(2+)-independent acetylcholine release (up to 340%). This effect was examined as a function of cetiedil concentration and incubation time. On the other hand, the analogue MR 16728, which enhances A23187-evoked acetylcholine release, also enhances spon

[Effects of the pineal body and melatonin on sensitivity to pain in mice].

Sensitivity to painful stimuli was measured by hot plate, writhing tests and electric caudal stimulation in mice. The mice were kept under light-dark 12/12 cycle with light out at 18:00 for at least 2 wk. Both basal analgesia and meperidine (pethidine)-induced analgesic effect exhibited parallel circadian rhythms, with the marked peak and through occurring at mid-dark and mid-light phases, respectively. The day-night differences in pain threshold 10 d after pinealectomy were not evident, especially in the loss of dark time augmentation of analgesic responses, but persisted in sham operated mic

Left and right vagus nerve stimulation: historical perspectives, clinical efficacy, and future directions.

Neuromodulation has profoundly transformed medical science, offering new treatments for various neurological conditions. Stimulation techniques that target the brain, spinal cord, trigeminal nerve, and vagus nerve (VN) use electrical impulses to modulate neural functions. Among these, vagus nerve stimulation (VNS) is distinguished for its use to stimulate the VN to modulate neural functions. VNS shows promising applications across a wide range of neurological conditions, exemplifying the ongoing evolution of neuromodulation. As VNS continues to prove its efficacy, an important consideration in

Non-invasive Autonomic Neuromodulation Is Opening New Landscapes for Cardiovascular Diseases.

Autonomic imbalance plays a crucial role in the genesis and maintenance of cardiac disorders. Approaches to maintain sympatho-vagal balance in heart diseases have gained great interest in recent years. Emerging therapies However, certain types of emerging therapies including direct electrical stimulation and nerve denervation require invasive implantation of a generator and a bipolar electrode subcutaneously or result in autonomic nervous system (ANS) damage, inevitably increasing the risk of complications. More recently, non-invasive neuromodulation approaches have received great interest in

Transcranial Static Magnetic Field Stimulation over the Primary Motor Cortex Induces Plastic Changes in Cortical Nociceptive Processing.

Transcranial static magnetic field stimulation (tSMS) is a novel and inexpensive, non-invasive brain stimulation (NIBS) technique. Here, we performed non-invasive modulation of intra-epidermal electrical stimulation-evoked potentials (IES-EPs) by applying tSMS or sham stimulation over the primary motor (M1) and somatosensory (S1) cortices in 18 healthy volunteers for 15 min. We recorded EPs after IES before, right after, and 10 min after tSMS. The IES-EP amplitude was significantly reduced immediately after tSMS over M1, whereas tSMS over S1 and sham stimulation did not affect the IES-EP ampli

Hypomania Risk in Noninvasive Brain Stimulation.

Noninvasive brain stimulation, using electromagnetic waves (repetitive transcranial magnetic stimulation (rTMS)) and direct electrical current (transcranial direct current stimulation (tDCS)), is a new frontier in treating psychiatric maladies. While still being developed as viable treatment options, both options have had numerously reported side-effects, with hypomania being a significant concern during investigations. While there has been a relatively rare incidence of hypomania among rTMS/tDCS trials, it still posits an important issue regarding the safety of both treatment modalities. This

Microthalamotomy effect during deep brain stimulation: potential involvement of adenosine and glutamate efflux.

Deep brain stimulation (DBS) of the thalamus is widely used in humans to treat essential tremor and tremor dominant Parkinson's disease. After DBS lead implantation, tremor is often reduced even without electrical stimulation. Often called "microthalamotomy" effect, the exact mechanism is unknown, although it is presumed to be due to micro lesioning. Here, we tested whether microthalamotomy effect may, in fact, be mediated via release of neurotransmitters adenosine and glutamate, using fast scan cyclic voltammetry (FSCV) and amperometry, respectively. Implantation of microelectrodes into the v

Differences in parasympathetic vasodilator and salivary responses in the cat submandibular gland between lingual and chorda-lingual nerve stimulation.

Experimental activation of parasympathetic vasodilatation and salivation in the submandibular gland by a "pseudoreflex" method has definite advantages over other commonly used methods. However, it is unclear which stimulation sites allow for avoidance of direct activation of the parasympathetic efferents to this gland. We examined this question in heavily anesthetized (alpha-chloralose and urethane), artificially ventilated, cervically vago-sympathectomized cats, using the intersection of the chorda-lingual nerve and the submandibular gland duct ("the intersection") as our reference point. Soleus H-reflex to S1 nerve root stimulation.

H-reflexes in normals were elicited by percutaneous electrical and magnetic stimulation of proximal nerve roots at the cauda equina. H-M interval to S1 nerve root stimulation at the level of the S1 foramen was 6.8 +/- 0.33 ms, with side to side difference of 0.16 +/- 0.13 ms. Compression/ischemia of the sciatic nerve in the mid-thigh abolished the H-reflex to stimulation of the tibial nerve at the popliteal fossa when the H-reflex to S1 nerve root stimulation was preserved. The length of the S1 nerve root in human cadavers was measured to be 17.5 +/- 03 cm, providing an estimated dorsal root c

Diclofenac evaluated in a human experimental model of central pain.

The putative central analgesic activity of diclofenac was investigated in a human experimental pain model using intraneural electrical stimulation in the median nerve. Since pain is induced proximal to the peripheral nociceptors, the model can be used to test central analgesic properties of i.a. pharmacological interventions performed during series of repeated stimulations. A single intravenous dose of 50 mg diclofenac or saline was administered during an ongoing series of painful intraneural stimulations in a double-blind cross-over study in 10 healthy volunteers. Neither diclofenac nor salin

Morphine inhibits substance P release from peripheral sensory nerve endings.

Tissue levels of substance P (SP) were determined by radioimmunoassay in cat dental pulps after unilateral electrical stimulation of the inferior alveolar nerve. Nerve stimulation (10 V, 10 Hz, 5 ms for 3 min) reduced the SP level at the stimulated side by approximately 40%, indicating a release of SP. Stimulation performed after infusion of morphine (0.3 mg/kg/min, i.v.) for 10 min did not reduce the pulpal SP-level. Naloxone (3 mg/kg, i.v.) administered immediately before the morphine infusion in three experiments, did not influence the effect of morphine on pulpal SP levels after nerve stim

Vagally induced gastric antral contractions and gastric emptying of a liquid test meal.

The emptying of a liquid test meal from the stomach was studied during, and in the absence of, electrical stimulation of cut ends of a thoracic branch of the vagus in anaesthetized cats. The test meal (154 mmol.1-1 NaCl and 30 mg.1-1 phenol red) was measured by collecting effluent from a duodenal fistula over a 30 min period. The stomach emptied about 60% of the meal under control conditions compared with over 90% during efferent stimulation of the vagus. The increased volumes emptied during efferent stimulation were not accounted for by secretion of gastric acid. Coincident with the vagally e

Excitation of the Auditory System as a Result of Non-invasive Extra-Cochlear Stimulation in Normal Subjects and Tinnitus Patients.

One of possible approach that may suppress tinnitus is electrical stimulation of the ear. At first invasive techniques were used (promontory or round window stimulation), nowadays a non-invasive method, namely hydrotransmissive electric stimulation (ES) through external acoustic canal, has been developed. The aim of the study is to investigate the effect of applying ES with positive and negative current polarities on the ears of healthy subjects and on the tinnitus ears of patients with tinnitus. This comparison further clarifies the mechanisms of operation of non-invasive extra-cochlear ear E

Optogenetic Hyperpolarization of Cardiomyocytes Terminates Ventricular Arrhythmia.

Cardiac defibrillation to terminate lethal ventricular arrhythmia (VA) is currently performed by applying high energy electrical shocks. In cardiac tissue, electrical shocks induce simultaneously de- and hyperpolarized areas and only depolarized areas are considered to be responsible for VA termination. Because electrical shocks do not allow proper control over spatial extent and level of membrane potential changes, the effects of hyperpolarization have not been explored in the intact heart. In contrast, optogenetic methods allow cell type-selective induction of de- and hyperpolarization with

Neuromodulation in the Treatment of Alzheimer's Disease: Current and Emerging Approaches.

Neuromodulation as a treatment strategy for psychiatric and neurological diseases has grown in popularity in recent years, with the approval of repetitive transcranial magnetic stimulation (rTMS) for the treatment of depression being one such example. These approaches offer new hope in the treatment of diseases that have proven largely intractable to traditional pharmacological approaches. For this reason, neuromodulation is increasingly being explored for the treatment of Alzheimer's disease. However, such approaches have variable, and, in many cases, very limited evidence for safety and effi

Vago-vagal reflexes to the colon of the anaesthetized ferret.

Electrical stimulation of the central end of the vagal communicating branch in the thorax at frequencies between 2 and 20 Hz elicited, after a latency of 7.2 +/- 0.8 s, large-amplitude colonic contractions. 5 Hz stimulation gave near maximal contractions and, because vomiting was more likely to occur at higher stimulus frequencies, was used as the standard stimulus for subsequent experiments. At this frequency the peak colonic contraction was 6.5 +/- 0.9 kPa. Following atropine the characteristics of the response to central vagal stimulation differed from that seen before atropinization. Augmentation of vagal reflex bradycardia be central alpha2-adrenoceptors in the cat.

Vagal reflex bradycardia was induced in anaesthetized cats with high level spinal axotomy by electrical stimulation of either the carotid sinus nerves or a depressor nerve. In both preparations reflex bradycardia increased with the rate of stimulation. Injection of 1 microgram/kg clonidine into a lateral cerebral ventricle augmented reflex bradycardia in response to carotid sinus nerve stimulation while the same dose of clonidine was ineffective when given intravenously. The antagonistic effect of intracerebroventricular yohimbine (50 micrograms/kg) indicated that the effect of clonidine was d

Evaluation of proximal facial nerve conduction by transcranial magnetic stimulation.

A magnetic stimulator was used for direct transcutaneous stimulation of the intracranial portion of the facial nerve in 15 normal subjects and in patients with Bell's palsy, demyelinating neuropathy, traumatic facial palsy and pontine glioma. Compound muscle action potentials (CMAPs) thus elicited in the orbicularis oris muscle of controls were of similar amplitude but longer latency (1.3 SD 0.15 ms) compared with CMAPs produced by conventional electrical stimulation at the stylomastoid foramen. No response to magnetic stimulation could be recorded from the affected side in 15 of 16 patients w

Increase in intracellular sodium ion activity during stimulation in mammalian cardiac muscle.

Changes in stimulation rate alter the electrical and mechanical characteristics of myocardial cells. We have investigated the possibility that intracellular sodium activity (aiNa) changes with stimulation and correlates with changes in contraction strength. Two kinds of liquid membrane Na+-selective microelectrodes were used to measure aiNa in guinea pig and sheep ventricular muscle and in sheep Purkinje strands. Stimulation produced a rate- and time-dependent elevation of aiNa. Small increases in aiNa were seen at stimulation rates as slow as 0.2 Hz, and faster rates of stimulation elevated a
