Functional alterations in cerebral GABAA receptor complex associated with formation of alcohol dependence: analysis using GABA-dependent 36Cl- influx into neuronal membrane vesicles.

The effect of alcohol dependence induced by ethanol inhalation on GABA-dependent 36Cl- influx into membrane vesicles prepared from the mouse brain has been examined. Ethanol, flunitrazepam and salsolinol induced a significant facilitation of the GABA-dependent 36Cl- influx into membrane vesicles obtained from the normal mouse brain. Ethanol inhalation induced the facilitation of GABA-dependent 36Cl- influx at 3-12 hr after the initiation of inhalation, but this facilitation returned to a normal level within 12 hr. In membrane vesicles obtained from the brain of an alcohol-dependent mouse at 7 …

Calcium-mediated repression of β-catenin and its transcriptional signaling mediates neural crest cell death in an avian model of fetal alcohol syndrome.

Fetal alcohol syndrome (FAS) is a common birth defect in many societies. Affected individuals have neurodevelopmental disabilities and a distinctive craniofacial dysmorphology. These latter deficits originate during early development from the ethanol-mediated apoptotic depletion of cranial facial progenitors, a population known as the neural crest. We showed previously that this apoptosis is caused because acute ethanol exposure activates G-protein-dependent intracellular calcium within cranial neural crest progenitors, and this calcium transient initiates the cell death. The dysregulated sign…

Alcohol and pregnenolone interaction on cerebral arteries through targeting of vascular smooth muscle Ca2+- and voltage-gated K+ channels of big conductance.

Despite the significant number of people who may be taking pregnenolone supplements while drinking alcohol (ethanol), the widely documented cerebrovascular actions of pregnenolone and ethanol, and the critical dependence of cerebrovascular function on cerebral artery diameter, there are no studies addressing the effect of pregnenolone + ethanol in combination on cerebral artery diameter. We investigated this by evaluating the effect of this combination on middle cerebral artery diameter in male and female C57BL/6J mice, both in vivo and in vitro. The use of de-endothelialized, in vitro pressur…

Alcohol-induced retrograde memory impairment in rats: prevention by caffeine.

RATIONALE: Ethanol and caffeine are two of the most widely consumed drugs in the world, often used in the same setting. Animal models may help to understand the conditions under which incidental memories formed just before ethanol intoxication might be lost or become difficult to retrieve. OBJECTIVES: Ethanol-induced retrograde amnesia was investigated using a new odor-recognition test. MATERIALS AND METHODS: Rats thoroughly explored a wood bead taken from the cage of another rat, and habituated to this novel odor (N1) over three trials. Immediately following habituation, rats received salin…

Acetic acid enhances attraction to and colonization of wood bolts by ambrosia beetles (Coleoptera: Curculionidae: Scolytinae).

Certain invasive ambrosia beetles are attracted to and colonize physiologically stressed trees emitting ethanol. Xylosandrus germanus (Blandford) and Xylosandrus crassiusculus (Motschulsky) are attracted to trees emitting ethanol and are 2 of the most damaging invasive ambrosia beetles in North America. Acetic acid has been detected in stressed trees emitting ethanol, but whether it has a role in ambrosia beetle ecology is unknown. Sections of tree stems (bolts) were soaked in various dilutions of acetic acid alone and mixed with a 5% dilution of ethanol, then deployed in a field near a woodla…

A potential therapeutic role of resveratrol in mitigating hepatotoxicity induced by paracetamol and alcohol.

OBJECTIVES: Drug-induced hepatotoxicity, particularly from ethanol and acetaminophen (APAP), is a pressing global health challenge. This damage arises from oxidative stress and inflammation, manifesting as elevated liver enzymes and structural liver alterations. Resveratrol and silymarin, recognized for their antioxidant and anti-inflammatory properties, offer potential hepatoprotective benefits. METHODS: This study investigated the hepatoprotective efficacy of resveratrol and silymarin, alone and in combination, in a rat model of ethanol- and APAP-induced liver injury. Thirty Wistar rats wer…

Alcohol exposure induces chick craniofacial bone defects by negatively affecting cranial neural crest development.

Excess alcohol consumption during pregnancy could lead to fetal alcohol syndrome (FAS). However, the molecular mechanism leading to craniofacial abnormality, a feature of FAS, is still poorly understood. The cranial neural crest cells (NCCs) contribute to the formation of the craniofacial bones. Therefore, NCCs exposed to ethanol was investigated - using chick embryos and in vitro explant culture as experimental models. We demonstrated that exposure to 2% ethanol induced craniofacial defects, which includes parietal defect, in the developing chick fetus. Immunofluorescent staining revealed tha…

HepG2 cells recovered from apoptosis show altered drug responses and invasiveness.

BACKGROUND: Cancer relapse, associated with increased drug resistance and rate of metastasis, often follows completion of chemotherapy but the cancer escape mechanisms are still incompletely understood. Percutaneous ethanol injection (PEI) has been used for treating hepatocellular carcinoma (HCC) for decades, while the recurrence after PEI treatment remains a major limitation. Recent evidence mounted that cancer cells could survive from chemical induced apoptosis, suggesting a potential route through which cancer relapse may occur. This study focuses on the consequence of HepG2 recovery from e…

Lithium prevents long-term neural and behavioral pathology induced by early alcohol exposure.

Fetal alcohol exposure can cause developmental defects in offspring known as fetal alcohol spectrum disorder (FASD). FASD symptoms range from obvious facial deformities to changes in neuroanatomy and neurophysiology that disrupt normal brain function and behavior. Ethanol exposure at postnatal day 7 in C57BL/6 mice induces neuronal cell death and long-lasting neurobehavioral dysfunction. Previous work has demonstrated that early ethanol exposure impairs spatial memory task performance into adulthood and perturbs local and interregional brain circuit integrity in the olfacto-hippocampal pathway…

Chronic administration of alcohol in the developing rat: expression of functional tolerance and alcohol olfactory aversions.

Three experiments were designed to evaluate the acquisition and expression of tolerance to ethanol in preweanling rats. Administrations were begun on Postnatal Day 13 (PD13) and continued, on alternating days, through PD23. Following either three or five administrations of a 2.5 g/kg dose of ethanol, animals expressed tolerance to the motor-impairing consequences of acute ethanol, as measured through attenuated latencies to perform a righting reflex. However, no tolerance was evident when it was assessed through the hypothermic response to ethanol delivery (Experiment 1). The results of subseq…

Cholinergic and Neuroimmune Signaling Interact to Impact Adult Hippocampal Neurogenesis and Alcohol Pathology Across Development.

Alcohol (ethanol) use and misuse is a costly societal issue that can affect an individual across the lifespan. Alcohol use and misuse typically initiates during adolescence and generally continues into adulthood. Not only is alcohol the most widely abused drug by adolescents, but it is also one of the most widely abused drugs in the world. In fact, high rates of maternal drinking make developmental ethanol exposure the most preventable cause of neurological deficits in the Western world. Preclinical studies have determined that one of the most consistent effects of ethanol is its disruption of…

Individual susceptibility and alcohol effects:biochemical and genetic aspects.

The large interethnic and interindividual variability in alcohol-induced toxic effects comes from a combination of genetic and environmental factors, influencing ethanol toxicokinetics. The hepatic enzymatic systems involved in ethanol metabolism are alcohol dehydrogenase (ADH), aldehyde dehydrogenase (ALDH) and microsomal P4502E1 (CYP2E1). ADH oxidizes ethanol to acetaldehyde, which is very efficiently oxidized to acetate by ALDH. About 10% of moderate quantities of ethanol is metabolised by CYP2E1; the percentage increases when ADH is saturated. During ethanol metabolism reactive oxygen spec…

Contrasting effects of anesthetics in tadpole bioassays.

The median anesthetic concentrations (AC 50 values) in the bath solution for loss of righting reflex (RR) or nociceptor response (NR) were determined in larval Rana catesbeiana. The AC50 (NR)/AC50(RR) ratios were between 1.6 and 1.9 for ethanol, ether and diazepam. Comparable ratios for hexobarbital and thiopental were 9.8 and 6.2, which indicated a marked lack of "analgesic" effect. The tadpole AC50 values were consistently higher than AC50 values reported for nonionized and unbound drug in mammalian blood water. The tadpole/mammal ratio for loss of NR during thiopental anesthesia was approxi…

Ethyl Alcohol Versus Botulinum Toxin A: A Comparative Study of the Visual and Histopathological Outcomes in the Rabbit Anterior Auricular Muscle Model.

BACKGROUND: Botulinum toxin has long been known for its paralytic effects at the neuromuscular junction. Although it has been widely used for vascular and nervous tissues, there has been no study of the aesthetic effects of the application of ethanol to muscle tissues to date. OBJECTIVE: The authors aimed to demonstrate the effects of the application of ethanol to muscle tissues after an intramuscular injection and to compare the effects of botulinum toxin A (BTA) and ethanol. METHODS AND MATERIALS: A total of 28 rabbits were divided into 4 groups (n = 7 each). Botulinum toxin A (5 units) an…

Alcohol consumption before pregnancy causes detrimental fetal development and maternal metabolic disorders.

Alcohol consumption before or during pregnancy poses serious health risks to the fetus; however, the underlying mechanisms involved remain obscure. Here, we investigated whether ethanol consumption before pregnancy affects maternal or fetal health and whether pharmacological inhibition of CYP2E1, a major ethanol oxidation enzyme, by 4-methylpyrazole (4-MP) has therapeutic effects. We found that ethanol consumption (5%) 2 weeks before pregnancy resulted in a decrease in the number of viable fetuses and abnormal fetal development, and these effects were accompanied by impaired maternal glucose h…

Animal models of motivation for drinking in rodents with a focus on opioid receptor neuropharmacology.

Ethanol, like other drugs of abuse, has motivating properties that can be developed as animal models of self-administration. A major strength of the operant approach where an animal must work to obtain ethanol is that it reduces confounds due to palatability and controls for nonspecific malaise-inducing effects. In the domain of opioid peptide systems, limited access paradigms have good predictive validity. In addition, animal models of excessive drinking-either environmentally or genetically induced-also appear sensitive to blockade or inactivation of opioid peptide receptors. Ethanol availab…

Optic nerve hypoplasia in an acute exposure model of the fetal alcohol syndrome.

The effects of acute prenatal exposure to ethanol on the postnatal optic nerve have been examined in a C57B1/6J mouse model. Pregnant mice were exposed by intraperitoneal injection to ethanol (25% ethanol, each dose at 0.015 ml/g separated by 4 h), or to saline on the 8th gestational day and the optic nerve examined at P15. There was a significant difference in the cross-sectional areas of optic nerves from ethanol-exposed mice (Mean +/- SD: 32,800 +/- 11,000 microns2) compared to control nerves (Mean +/- SD: 52,100 +/- 8,900 microns2). This difference was mainly the result of a reduction in t…

Differential effects of long-chain alcohols in long- and short-sleep mice.

Observations that the long-sleep (LS) and short-sleep (SS( mouse lines differ in their depressant response to barbiturates, and that the difference between lines becomes greater as lipid solubility increases, prompted this investigation of the effects of alcohols that differ in lipid solubility. Results indicate that LS and SS mice differ significantly in their sleep time responses to propanol, butanol, and 3-methyl butanol, as well as ethanol: their hypothermic responses showed a similar pattern, but only the response to ethanol differed significantly between lines. For both sleep time and hy…

Impact of adolescent alcohol use across the lifespan: Long-lasting tolerance to high-dose alcohol coupled with potentiated spatial memory impairments to moderate-dose alcohol.

Understanding how alcohol exposure during adolescence affects aging is a critical but understudied area. In the present study, male rats were exposed to either alcohol or saline during adolescence, then tested every 4 months following either an ethanol or saline challenge; animals were tested until postnatal day (PD) 532. It was found that long-lasting tolerance to high-dose ethanol exists through the test period, as measured by loss of righting reflex, while tolerance to lower doses of ethanol is not found. In addition, alcohol exposure during adolescence facilitated spatial memory impairment…
