Assessing the pandemic potential of Nipah virus: From molecular pathogenesis to ecological spillover risks.

Nipah virus, a highly lethal zoonotic paramyxovirus with a case‑fatality rate of 40-75%, poses a formidable threat to global health security. This review synthesizes its evolving epidemiology, contrasting the Malaysian and Bangladesh‑India strains, and dissects molecular pathogenesis focusing on G/F glycoprotein recognition of ephrin receptors and the multi‑layered immune evasion mediated by P gene products. Phylogenetic analyses reveal 8-10% nucleotide divergence between lineages, with positive selection in the G protein that may compromise vaccine cross‑protection. Ecological risk amplifiers

Dynamic interactions between Schistosoma haematobium, Schistosoma mattheei and Schistosoma mansoni underscore the complex polyparasitism of intestinal schistosomiasis in southern Malawi.

Schistosomiasis is prevalent among school-aged children (SAC) in Mangochi District, Malawi, where both intestinal and urogenital forms are endemic. In 2024, we identified schistosomiasis cases predominantly associated with excretion of Schistosoma haematobium × Schistosoma mattheei ova in the faeces of two individuals from Samama village, Mangochi District. In this expanded cross-sectional study, we characterize the prevalence and species composition of Schistosoma infections among 247 SAC in Samama, using genus- and species-specific molecular diagnostics. We also present follow-up data from t

Molecular Characterization of DENV-1 Isolates From Pakistan Reveals Immunogenic Variability and Antiviral Potential of Phytochemicals Targeting the CprM Region.

Dengue virus (DENV), a mosquito-borne arbovirus, remains a critical global health concern, causing approximately 390 million infections annually. Dengue fever poses significant challenges to healthcare systems in endemic regions like Pakistan due to the presence of four established serotypes (DENV-1-DENV-4) and the potential emergence of a fifth serotype (DENV-5). The current study aimed to investigate molecular characteristics of DENV-1 isolates from Peshawar, Pakistan, focusing on serotyping, genetic variability in the CprM junction region, and identification of potential antiviral phytochem

Decoding antibiotic resistance in tuberculosis: Role of RRDR and non-RRDR mutations in diagnostic perspectives.

Rifampicin resistance continues to pose a challenge to successful tuberculosis (TB) control, mainly driven by mutations in the rpoB gene of Mycobacterium tuberculosis. Although most diagnostic tools, such as Xpert MTB/RIF, Xpert Ultra, and line probe assays, focus on the standard 81-bp rifampicin resistance-determining region (RRDR), recent findings emphasize the critical role of non-RRDR mutations in the development of rifampicin resistance. Although less frequent, these mutations either directly modify drug-binding sites or drive compensatory evolution that restores bacterial fitness impaire

p53 Pathway-Targeted Therapeutic Strategies in Glioblastoma: A CNS-Specific Framework Integrating Therapeutic Resistance, Molecular Stratification, CNS Pharmacology, and Clinical Translation.

Glioblastoma (GBM) remains one of the most difficult therapeutic problems in adult neuro-oncology. Its resistance to treatment reflects not only malignant cellular biology but also central nervous system-specific constraints, including blood-brain barrier and blood-tumor barrier heterogeneity, diffuse infiltrative growth, uncertain target-site exposure, treatment-induced evolution, and the need to preserve neurological function. The tumor suppressor p53, encoded by TP53, is highly relevant to glioblastoma because it regulates cell-cycle arrest, DNA repair, apoptosis, senescence, genomic stabil

Screening and characterization of DNA aptamers that modulate prime editing.

INTRODUCTION: Precise genome editing is a critical focus in gene therapy, and the CRISPR-Cas9 system has become a powerful and versatile tool for this purpose. However, a significant limitation of the CRISPR-Cas9 system is its low homologous recombination rate, which can impede the restoration of normal gene function. To address some of these challenges, advanced gene-editing technologies, such as base editors and prime editors have been developed. Here, we explored whether Cas9-specific single-stranded DNA (ssDNA) aptamers could enhance the PE2 system's functionality. METHODS: Systematic evo

Systemic therapy for pretreated advanced biliary tract cancer: past developments and recent advances.

Biliary tract cancer (BTC) remains among the most challenging malignancies with a poor prognosis and limited treatment options, particularly in pretreated patients. As most patients experience disease progression after first-line treatment, effective second-line and subsequent treatments are required. Although the addition of modified FOLFOX (fluorouracil, leucovorin, and oxaliplatin) to active symptom control improved the overall survival of patients with progressing advanced BTC despite gemcitabine plus cisplatin treatment, its efficacy was modest. Moreover, most clinical trials demonstrated

Seasonal dynamics and genetic diversity of human adenoviruses in patients with acute respiratory infection in Thailand, 2024.

Human adenoviruses (HAdVs) are a significant cause of acute respiratory infections (ARIs), particularly in pediatric populations. Continuous molecular surveillance is essential to understand their epidemiology, genetic diversity, and seasonal dynamics. In 2024, a surveillance study was conducted in Bangkok, Thailand, involving 8,130 nasopharyngeal swabs collected from ARI patients. HAdV was detected in 5.3% (429/8,130) of ARI cases, with peak weekly positivity reaching 17.7% during weeks 10-12 (March). These HAdV-positive samples were subsequently genotyped through partial hexon gene sequencin

Evolutionary dynamics of sex determination in Branchiostoma belcheri driven by repeated transposition of a single novel gene.

Sex determination systems display striking evolutionary flexibility, yet the mechanisms underlying their transitions remain poorly understood. Using newly generated genome assemblies, we investigated the evolving sex-determining system in the amphioxus Branchiostoma belcheri. We identified two female-specific sex-determining regions (SDRs) on chromosome 13, both derived from independent transpositions of the autosomal gene tesD, which shows testis-specific expression in amphioxus species. CRISPR/Cas9 knockout experiments in Branchiostoma floridae confirmed that tesD functions as a male-determi

Stringent selection drives convergence toward omicron-like SARS-CoV-2 receptor-binding motifs.

In vitro protein evolution can provide powerful insights into the amino acid sequences that underlie key biological functions. Here, we use this to explore the evolutionary trajectories of the SARS-CoV-2 spike protein receptor-binding motif (RBM) binding the human angiotensin-converting enzyme 2 (ACE2), an essential first step in viral infection. Applying stringent selection pressures starting from the Wuhan or another non-Omicron variant protein-coding sequence results in rapid convergence towards Omicron characteristic mutations and its sub-lineages. Conversely, under mild selection, only so

A strain of high cell-adapted porcine deltacoronavirus: Isolation, genetic evolution, and pathogenicity analysis.

In 2024, an outbreak of piglet diarrhea occurred in Guangxi Zhuang Autonomous Region, China. RT-PCR assay of clinical samples confirmed that porcine deltacoronavirus (PDCoV) was the primary pathogen. In this study, the PDCoV GX/2024 strain was successfully isolated in PK-15 cells with optimized trypsin treatment conditions. The cytopathic effect (CPE) of this strain gradually intensified during serial passages. After plaque purification, the strain achieved stable proliferation with high titer, reaching a peak titer of 107·2 TCID50/mL, which indicated its potential as an inactivated vaccine ca

Harnessing a germ‑free mouse gut bioreactor for directed evolution of probiotics to combat non-alcoholic fatty liver disease.

Directed evolution has transformed biomolecular engineering but remains largely untapped for probiotic optimization, despite its immense promise for human health maintenance and disease therapy. Here, we present an in vivo, host-mediated directed-evolution platform that harnesses the gut's endogenous selective pressures to drive functional enhancement of probiotics. Using Bifidobacterium animalis subsp. lactis as a model, we expose germ-free male mice to stepwise increases in bile-acid stress via a high-fat, high-cholesterol diet. Compared to in vitro evolution, which fails to produce any func

Reactivation of mammalian regeneration by turning on an evolutionarily disabled genetic switch.

Mammals display prominent diversity in the ability to regenerate damaged ear pinna, but the genetic changes underlying the failure of regeneration remain elusive. We performed comparative single-cell and spatial transcriptomic analyses of rabbits and mice recovering from pinna damage. Insufficient retinoic acid (RA) production, caused by the deficiency of rate-limiting enzyme Aldh1a2 and boosted RA degradation, was responsible for the failure of mouse pinna regeneration. Switching on Aldh1a2 or RA supplementation reactivated regeneration. Evolutionary inactivation of multiple Aldh1a2-linked re

Evolution and characteristics of tetracycline resistance in Mycoplasma pneumoniae.

Macrolide resistance in Mycoplasma pneumoniae is becoming prevalent. Tetracyclines are an important alternative for patients with macrolide-resistant M. pneumoniae infections. Tetracycline-resistant M. pneumoniae has never been reported in clinical isolates or in vitro-induced strains. Here, we aimed to explore tetracycline-resistant M. pneumoniae in vitro and the underlying resistance mechanisms. Clinically isolated parental M. pneumoniae strains were selected for tetracycline resistance by inducing them in vitro with increasing concentrations of tetracycline, minocycline, and tigecycline. Impact of prolonged infection on SARS-CoV-2 evolution.

Immunocompromised patients with prolonged severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infections may serve as reservoirs for viral evolution, with suboptimal immune responses facilitating the accumulation of adaptive mutations. This study aimed to characterize the drivers of SARS-CoV-2 adaptive evolution in such hosts through genomic surveillance. We retrospectively analyzed 24 patients with long-term positive nasopharyngeal reverse transcription-polymerase chain reaction results (symptom onset duration: 7-14 days on average, 1 HIV-positive patient with >20 days of infection).
