Post-weaning high-fat diet results in growth cartilage lesions in young male rats.

To determine if a high-fat diet (HF) from weaning would result in a pro-inflammatory state and affect joint cartilage, we fed male rats either HF or Chow diet post-weaning, and voluntary wheel exercise (EX) or cage only activity (SED) after 9 weeks of age. At 17 weeks body composition, plasma biomarkers and histomorphology scores of femoro-tibial cartilages of HF-SED, HF-EX, Chow-SED and Chow-EX groups were compared. Food intake and activity were not significantly different between groups. HF diet resulted in significantly higher weight gain, %fat, fat:lean ratio, and plasma leptin, insulin an

Metabolic and anthropometric changes in early breast cancer patients receiving adjuvant therapy.

Weight gain and metabolic changes have been related to survival of early breast cancer patients (EBC). ''However, factors influencing metabolism post-diagnosis are not fully understood. We measured anthropometric [body mass index (BMI), body weight, waist and hip circumferences, and waist-to-hip ratio] and metabolic (levels of insulin, glucose, H1Ac, total, HDL, and LDL cholesterol, triglycerides, and the homeostasis model assessment score [HOMA]) parameters in 433 pre- and post-menopausal women with EBC at diagnosis and 3, 6, 9, 12, and 24 months thereafter. At diagnosis, compared with post-m

The influence of endocannabinoid receptor 1 gene variations on anthropometric and metabolic parameters of women with polycystic ovary syndrome.

INTRODUCTION: Polycystic ovary syndrome (PCOS) is associated with an increasing number of metabolic comorbidities. About 50% of PCOS patients are obese, and insulin resistance affects up to 70% of these women. The endocannabinoid system contributes to human energy homeostasis. CNR1 is a biological candidate for human obesity and related metabolic disorders. The aim of this study was to determine the relationships between CNR1 polymorphisms and anthropometric and metabolic parameters in PCOS women. MATERIAL AND METHODS: 130 women diagnosed with PCOS according to the Rotterdam criteria were rec

Infected Sclerosing Lipogranuloma after Hernioplasty: Ultrasonographic and MRI Findings.

We report the ultrasonographic and MRI findings of an infected sclerosing lipogranuloma after scrotal hernioplasty. Sclerosing lipogranuloma is a rare foreign-body reaction of fat tissue, with most cases being associated with the genital and urinary tracts. To the best of our knowledge, MRI findings in sclerosing lipogranuloma in the scrotal sac have not yet been published and this is possibly the first study to report the case of an infected sclerosing lipogranuloma in the English literature.

A Case of Familial Partial Lipodystrophy: From Clinical Phenotype to Genetics.

We discuss the case of a 56-year-old woman who presented with diabetes from the age of 24. The diagnosis of familial partial lipodystrophy was made after the discovery of the lamin A/C gene 20 years later. The diagnosis enabled the detection of a severe cardiac rhythm disorder with the need for an implantable defibrillator. This disease is a rare disorder characterized by an altered body fat repartition, cardiac rhythm anomalies and muscular dystrophy.

Obesity increases risk for diabetes.

The relationship of obesity to the development of diabetes has been reviewed. Older individuals are more obese and have more diabetes. Both the duration and magnitude of the obesity increase risk of diabetes. Family history of diabetes mellitus is also important in whether obesity leads to diabetes. Abdominal body fat increases the risk of diabetes. These factors help in understanding the reasons why obesity poses such a high risk for developing diabetes mellitus.

The Effect of Frailty on Body Composition and Its Impact on the Use of SGLT-2 Inhibitors and GLP-1RA in Older Persons with Diabetes.

The association of frailty with body composition is complex. Frailty can be associated with significant anorexia and weight loss or overweight and obesity. In addition, the development of frailty leads to changes in muscle mass, muscle fibre type, and visceral fat. In older people with diabetes, frailty-induced body composition changes are clinically relevant as they may affect the metabolic profile of the frail person. The determinants of insulin resistance in frail older persons with diabetes include factors such as total body weight and the ratio of lean muscle mass to visceral fat mass. Th

Re-evaluation of recommended insecticides for resistance development, cross-resistance pattern, and underlying mechanisms in Helicoverpa armigera (Lepidoptera: Noctuidae).

The rise of insecticide resistance threatens biodiversity and agricultural sustainability as insects develop mechanisms like enhanced detoxification to withstand toxic exposures. This study assessed resistance levels of Helicoverpa armigera (Lepidoptera: Noctuidae) populations from various regions of India to emamectin benzoate, indoxacarb, spinetoram, and lambda-cyhalothrin, and investigated the role of detoxification enzymes in resistance. Field populations showed variable susceptibility, with negligible resistance to emamectin benzoate, indoxacarb, and spinetoram. Among the tested H. armige

Feasibility Study of Bariatric Surgery in a Rat Model of Spinal Cord Injury to Achieve Beneficial Body Weight Outcome.

Approximately two thirds of spinal cord injury (SCI) persons become overweight or obese. Obesity increases the risk of developing type 2 diabetes and limits self-help techniques. Weight-loss surgery (WLS), including vertical sleeve gastrectomy (VSG), is regarded as highly effective in the long-term treatment of obesity and remission of associated type 2 diabetes. Given the increased risk of obesity post-SCI, WLS offers an attractive intervention strategy. Alterations in the physiology of energy homeostasis after SCI necessitate that SCI persons should not be regarded as similar to able-bodied

The increase in fat-free mass during weight loss does not prevent greater than predicted reductions in resting energy expenditure in recreational athletes.

Energy restriction and resistance training (RT) are commonly used by athletes to induce body weight and fat losses and improve power to mass ratio. Weight loss-induced greater than predicted decrease in resting energy expenditure (GPDR) has repeatedly been reported in individuals with obesity. However, less is known about GPDR in trained athletes who consume adequate protein and who perform RT to maintain fat-free mass (FFM) during weight loss. Thirty-six recreational athletes (14 F; 22 M) from two separate weight loss studies were included for secondary analysis. We examined the effects of sh

Early-Life High-Fat-Diet Exposure Induced Pre-Puberty Obesity-Related MASLD via Autophagy-Mediated Ferroptosis in Male C57BL/6J Mice.

OBJECTIVES: Exposure to high-fat diets in early life plays an important role in metabolic dysfunction-associated steatotic liver disease (MASLD); however, the mechanism remains unclear. In this study, we explore the role of autophagy and ferroptosis in pre-puberty obesity-related MASLD caused by high-fat diets in early life. METHODS: Twenty-four male C57BL/6J mice were fed over a 6-week period, and were divided into three groups: control, lactation HFD, and lactation + post-weaning HFD group. AML12 cells were treated with 0.5 mM free fatty acids (palmitic acid:oleic acid = 1:2) for 24 h to es

ASK1-Induced FGF21 Synthesis in the Liver Prevents Obesity in Mice.

OBJECTIVE: Fibroblast growth factor 21 (FGF21) is a stress-induced hormone secreted by the liver. It has a beneficial role in the regulation of glucose and energy metabolism. Herein, we identify apoptosis signal regulating kinase 1 (ASK1) as an inducer of hepatic FGF21 synthesis. METHODS: Glucose and energy metabolism were assessed in liver-specific ASK1 overexpressing (ASK1+hep) and control littermate (ASK1f/f) mice fed a regular chow or high fat diet (HFD) for 20 weeks. Liver and fat depots were analyzed using RNA sequencing. Hepatic Fgf21 expression was downregulated using adeno-associated

Fucoidan P Alleviates Sarcopenic Obesity by Regulating Muscle Protein and Energy Metabolism.

Sarcopenic obesity (SO) is a complex condition involving increased fat accumulation along with reduced muscle mass and impaired muscle function; however, there are currently no approved pharmacological interventions targeting these pathological features. Akt and AMPK are key signaling pathways regulating muscle homeostasis and energy metabolism. Natural compounds that modulate these pathways may offer a promising multi-targeted therapeutic strategy. This study investigated the effects of Fucoidan P on SO and its underlying molecular pathways, while assessing translational relevance using clini

Fishing for Solutions: How Pre-Conceptional Fish Oil Supplementation in Obese Fathers Reduces Risk of Non-Alcoholic Fatty Liver Disease in Offspring Mice.

Metabolic dysfunction associated fatty liver disease (MAFLD) is a chronic condition with hepatic fat accumulation. The intergenerational effect of obesity has predominantly focused on mothers, with limited studies on paternal obesity. Nutritional intervention with fish oil (FO) has beneficial effects in reducing markers of obesity. We hypothesized that supplementing obese fathers with FO before conception could enhance the metabolic health of their offspring liver. Male mice were assigned to low-fat (LF), high fat (HF), or HF supplemented with FO for 10 weeks. Subsequently, these males were ma

Dragon Fruit Peel (Hylocereus undatus) Modulates Hepatic Lipid Metabolism and Inflammation in a Rat Model of High-Fat, High-Fructose-Induced Metabolic Dysfunction.

Metabolic dysfunction and hepatic abnormalities, such as those associated with high-fat, high-fructose (HFHFr) diets, are major contributors to obesity-related health issues. The growing interest in sustainable dietary interventions has highlighted the potential of plant-based byproducts. Dragon fruit (Hylocereus undatus) peel waste, rich in bioactive compounds such as dietary fibers, phenolics, and betacyanins, represents a promising functional ingredient for managing these disorders. This study investigated the effects of dragon fruit peel powder (DFP) on metabolic dysfunction and hepatic ab

Small Intestine Neuromuscular Dysfunctions and Neurogliopathy in a Mouse Model of High-Fat Diet-Induced Obesity: Involvement of Toll-Like Receptor 4.

Obesity is associated with enteric dysfunctions, including gut dysmotility and neurodegeneration, which may involve Toll-like receptor 4 (TLR4) signaling. To investigate this relationship, we examined the impact of TLR4 deficiency on the enteric nervous system (ENS) of the small intestine in a mouse model of high-fat diet (HFD)-induced obesity. Male TLR4-/- and wild-type (WT) C57BL/6J mice were fed either a standard diet (SD; 18% kcal fat) or an HFD (60% kcal fat) for 8 weeks. ENS alterations were evaluated using real-time qPCR and confocal immunofluorescence microscopy on longitudinal muscle-

Tirzepatide, a dual GIP/GLP-1 receptor agonist, alleviates metabolic dysfunction-associated steatotic liver disease by reducing the expression of CD36 and OBP2A.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is defined by excessive hepatic lipid accumulation. This study evaluated the therapeutic effects and molecular mechanisms of tirzepatide, a dual GIP and GLP-1 receptor agonist, in treating hepatic steatosis. Eight-week-old C57BL/6J mice were fed either a high-fat diet or a high-fat, high-fructose, and high-cholesterol diet for 12 weeks to induce MASLD. From week 8, some mice received weekly intraperitoneal tirzepatide injections for four weeks. Tirzepatide significantly reduced body and liver weight gain. Histological analysis co

Adipose development is consistent across hunter-gatherers and diverges from western references.

Despite agreement that humans have evolved to be unusually fat primates, adipose patterning among hunter-gatherers has received little empirical consideration. Here we consider the development of adiposity among four contemporary groups of hunter-gatherers, the Aka, Savanna Pumé, Ju'/Hoansi and Agta using multi-level generalized additive mixed modelling to characterize the growth of tricep skinfolds from early childhood through adolescence. In contrast to references, hunter-gatherers show several consistent patterns: (i) children are lean with little fat accumulation; (ii) no adiposity rebound

Lactococcus lactis KF140 Ameliorates Nonalcoholic Fatty Liver Disease Induced by Nε-Carboxymethyl-Lysine and High-Fat Diet.

SCOPE: Long-term consumption of excessive dietary advanced glycation end-products such as Nε-carboxymethyl-lysine (CML), which are produced by the Maillard reaction during food thermal processing, leads to nonalcoholic fatty liver disease (NAFLD) along with high fat consumption. The study previously finds that administration of Lactococcus lactis KF140 (LL-KF140) detoxifies CML by decreasing CML absorption both in a rat model and clinical trial. METHODS AND RESULTS: The present study evaluates the ameliorative effect of LL-KF140 on NAFLD and fatty liver-related biomarkers in a mouse model ind
