Effects of acute doses of ethanol on pre- and postnatal development in the mouse.

Investigators on fetal alcohol syndrome (FAS) both clinical and experimental, have generally concentrated attention on chronic alcoholism. In an attempt to elucidate the effects of binge drinking on pre- and postnatal development, single doses of 0.01, 0.02 and 0.03 ml/g body weight of alcohol solutions (25% v/v of ethanol in saline) were administered intraperitoneally to groups of mice on days 8-12 of gestation. Controls were untreated or starved for 6 h. Fetuses collected on day 18 revealed retardation, fetal mortality and craniofacial and digital malformations. While growth retardation and

Hypoglycemia in the fetal alcohol syndrome in rat.

As a treatable cause of central nervous system dysfunctions in the fetal alcohol syndrome, ethanol-induced hypoglycemia was studied in experimental rat models. Female Wistar rats were divided into ethanol and control groups. Before mating and during pregnancy, the ethanol group received 30% ethanol (E), or E with 20% sucrose (S) or 20% glucose (G), and the control group received water (W), or W with S or G. Pregnancies were terminated on gestational day (gd) 15, 18, and 21 by cesarean section or by spontaneous delivery. Dams and offspring were weighed and examined for several biochemical facto

Inhibition of gamma delta T cell development and early thymocyte maturation in IL-7 -/- mice.

While early thymic T cell precursor populations and their maturational sequence have been recently identified, the signals driving differentiation are unknown. While cytokines may play an integral role in T cell development, various mouse models rendered genetically deficient for specific cytokines do not display abnormalities in T cell development. Recently, we have generated IL-7 -/- mice and reported that IL-7 plays a unique and nonredundant role in lymphopoiesis. These mice displayed a 10- to 20-fold reduction in the total number of T and B cells. Here, we show that IL-7 -/- mice display a

Childhood trauma and insulin-like growth factors in amniotic fluid: An exploratory analysis of 79 women.

BACKGROUND: Perinatal stress has adverse effects on fetal outcome, yet the effect of early maternal trauma on fetal outcome has scarcely been studied. We investigated effects of maternal childhood trauma and current environment on important regulators of prenatal growth, fetal insulin-like growth factor (IGF)-1 and IGF-2 in amniotic fluid and assessed the impact of IGFs on newborn anthropometrics. METHODS: 79 pregnant women in their second trimester who underwent amniocentesis (15.9 ± 0.9 weeks of gestational age) and their newborns at birth were analyzed. Maternal childhood trauma was assess

Machine learning-based method for the detection of dextrocardia in ultrasound video clips.

BACKGROUND AND OBJECTIVE: Dextrocardia is a congenital anomaly arising during fetal development, characterised by the abnormal positioning of the heart on the right side of the chest, instead of its usual anatomical location on the left. This paper describes a machine learning-based method to automatically assess ultrasound (US) transverse videos to detect dextrocardia by analysing the Situs and four-chamber (4CH) views. METHODS: The method processes ultrasound video sweeps that users capture, which include the Situs and 4CH views. The automated analysis method consists of three stages. First…

Rethinking fetal central nervous system anomalies: predicting central nervous system anomalies with corpus callosum to head circumference and occipitofrontal diameter ratios.

BACKGROUND: Prenatal diagnosis of fetal central nervous system (CNS) anomalies remains challenging and is influenced by various factors, including imaging equipment quality, and access to advanced modalities such as fetal magnetic resonance imaging (MRI). In resource-limited settings, these constraints highlight the need for practical and accessible screening methods. This study aims to evaluate the predictive value of the fetal corpus callosum length-to-head circumference (CCL/HC) and corpus callosum length-to-occipitofrontal (CCL/OFD) diameter ratios in the detection of fetal central CNS abn

Characteristics of maternal-placental vascular malperfusion and its correlation with neonatal adverse outcomes.

INTRODUCTION: Fetal growth restriction (FGR) is the failure of fetal growth to attain inherent genetic potential owing to a diverse array of factors. Limited studies have investigated the correlation between varying levels of maternal-placental vascular malperfusion (MVM)-type FGR and adverse neonatal outcomes. Thus, we sought to explore the maternal-fetal characteristics of different degrees of MVM-type FGR and their correlation with adverse neonatal outcomes. METHODS: We collected all cases of FGR from 2018 to 2023 and observed the pregnancy, delivery, and newborn characteristics of FGR cas

The prospective associations of fetal growth-related pregnancy complications with subsequent breastfeeding duration and markers of human milk production.

BACKGROUND: The development of the breast for lactation occurs throughout pregnancy. It is unknown whether pregnancy complications resulting in poor fetal growth can affect breastfeeding (BF) success. OBJECTIVES: We examined whether fetal growth-related pregnancy complications were associated with earlier BF cessation and changes in the concentrations of human milk biomarkers of low milk production. METHODS: We used data from the Growing Up in Singapore Toward healthy Outcomes study (n = 954). Human milk concentrations of protein, lactose, citrate, sodium, potassium, and zinc at 3 wk postpar

Cellular and Molecular Pathophysiology of Gestational Diabetes.

Gestational diabetes (GD) is a metabolic disorder characterized by glucose intolerance during pregnancy, significantly impacting maternal and fetal health. Its global prevalence is approximately 14%, with risk factors including obesity, family history of diabetes, advanced maternal age, and ethnicity, which are linked to cellular and molecular disruptions in glucose regulation and insulin resistance. GD is associated with short- and long-term complications for both the mother and the newborn. For mothers, GD increases the risk of developing type 2 diabetes, cardiovascular diseases, and metabol

Hyaluronidase Impairs Neutrophil Function and Promotes Group B Streptococcus Invasion and Preterm Labor in Nonhuman Primates.

Invasive bacterial infections during pregnancy are a major risk factor for preterm birth, stillbirth, and fetal injury. Group B streptococci (GBS) are Gram-positive bacteria that asymptomatically colonize the lower genital tract but infect the amniotic fluid and induce preterm birth or stillbirth. Experimental models that closely emulate human pregnancy are pivotal for the development of successful strategies to prevent these adverse pregnancy outcomes. Using a unique nonhuman primate model that mimics human pregnancy and informs temporal events surrounding amniotic cavity invasion and preterm

Accelerated fetal growth in early pregnancy and risk of preterm birth: a prospective cohort study.

BACKGROUND: Preterm birth (occurring before 37 completed weeks of gestation) affects 15 million infants annually, 7.5% of which die due to related complications. The detection and early diagnosis are therefore paramount in order to prevent the development of prematurity and its consequences. So far, focus has been laid on the association between reduced intrauterine fetal growth during late gestation and prematurity. The aim of the current study was to investigate the association between accelerated fetal growth in early pregnancy and the risk of preterm birth. METHODS: This prospective cohor

Circular RNA expression profiles and features in human tissues: a study using RNA-seq data.

BACKGROUND: Circular RNA (circRNA) is one type of noncoding RNA that forms a covalently closed continuous loop. Similar to long noncoding RNA (lncRNA), circRNA can act as microRNA (miRNA) 'sponges' to regulate gene expression, and its abnormal expression is related to diseases such as atherosclerosis, nervous system disorders and cancer. So far, there have been no systematic studies on circRNA abundance and expression profiles in human adult and fetal tissues. RESULTS: We explored circRNA expression profiles using RNA-seq data for six adult and fetal normal tissues (colon, heart, kidney, live

Maternal prepregnancy obesity and cause-specific stillbirth.

BACKGROUND: In high-income countries, maternal obesity is one of the most important modifiable causes of stillbirth, yet the pathways underpinning this association remain unclear. OBJECTIVE: We estimated the association between maternal prepregnancy body mass index (BMI) and the risk of stillbirth defined by pathophysiologic contributors or causes. DESIGN: Using a case-cohort design, we randomly sampled 1829 singleton deliveries from a cohort of 68,437 eligible deliveries at Magee-Womens Hospital in Pittsburgh, Pennsylvania (2003-2010), and augmented it with all remaining cases of stillbirth

Anti-60-kDa heat shock protein antibodies in fetal serum: a biomarker for unexplained small for gestational age fetuses.

INTRODUCTION: Small for gestational age (SGA) is an important problem affecting 10% of pregnancies and is associated with significant perinatal morbidity. In about 80% of cases, a probable etiology or a major risk factor can be identified. But almost 20% of SGA cases are considered unexplained. The 60-kDa heat shock protein (HSP60) is a highly immunogenic protein whose synthesis is greatly upregulated under nonphysiological conditions. Bacterial and human HSP60 share a high degree of sequence homology, and immunity to conserved epitopes may result in development of autoimmunity following a bac

Tissue-specific ontogenic expression of prostaglandin H synthase 2 in the ovine myometrium, endometrium, and placenta during late gestation and at spontaneous term labor.

OBJECTIVE: The purposes of this study were to determine (1) whether uterine tissues as well as the fetal placenta are involved in the development of prostaglandin-synthesizing capacity associated with impending labor in pregnant sheep and (2) whether the key enzyme of prostaglandin synthesis, prostaglandin H synthase 2, is differentially expressed in the different intrauterine tissues during late gestation and in association with labor. STUDY DESIGN: Myometrium, endometrium, and fetal placenta were removed from ewes at 95 days' gestation, (n = 3), 101 to 110 days' gestation (n = 3), 111 to 12

Macrophage is crucial for tongue development by regulating myogenesis and vascularization.

BACKGROUND: Abnormal tongue development is a craniofacial deformity that affects dental-maxillofacial esthetics and function. Recent evidence has identified macrophages as multi-functional immune cells crucial for heart and brain development. However, it is still unknown whether macrophages affect tongue development. Therefore, this study aims to assess the distribution, phenotype, and roles of macrophages in the developing tongue. METHODS: In this study, immunohistochemical (IHC) and multiplex immunofluorescence (mIF) staining were conducted on C57BL/6 mice at embryonic day (E) 13.5, E14.5,

A de novo SALL4 mutation causes unilateral renal agenesis by misregulating genes involved in kidney development.

BACKGROUND: SALL4 is a transcription factor that plays a crucial role in early embryonic development and organogenesis, particularly in kidney development, although its specific regulatory mechanisms remain unclear. METHODS: We performed whole-exome sequencing (WES) to identify pathogenic variants in a fetus with unilateral renal agenesis and confirmed a variant in SALL4 using Sanger sequencing. The expression of wild-type or mutant SALL4 proteins in cells was used to determine whether the level and localization of the proteins were altered by the SALL4 variant. RNA sequencing was used to ide

Expression of insulin-like growth factor II in human placentas from normal and diabetic pregnancies.

The growth and development of the placenta is critical to fetal growth and development; however, little is known regarding the mechanisms controlling placental growth and development. Human placental membranes are known to possess receptors for insulin-like growth factors I and II (IGF-I and IGF-II) from early gestation, and increasing evidence supports a major role for IGF-I and/or IGF-II in fetal growth and development. Therefore, the IGFs may also play a significant role in regulating placental growth and development. We report here that an adult human liver IGF-II cDNA hybridizes to poly(A
