In August 1997, the World Health Organization (WHO) and the Joint United Nations Programme on HIV/AIDS (UNAIDS) convened an expert working group to discuss current strategies for the development of HIV type 1 vaccines. Based on the recent findings of investigators from Japan's National Institute of Infectious Diseases (NIID) in Tokyo using recombinant bacillus Calmette-Guérin (rBCG) as a potential vectored vaccine for HIV, a recommendation was made that further work in this area is a priority. As a result, the working group reconvened in September 1998 to discuss the progress to date with this

The first scientific attempts to control an infectious disease can be attributed to Edward Jenner, who, in 1796 inoculated an 8-year-old boy with cowpox (vaccinia), giving the boy protection against subsequent challenge with virulent smallpox. Thanks to the successful development of vaccines, many major diseases, such as diphtheria, poliomyelitis and measles, are nowadays kept under control, and in the case of smallpox, the dream of eradication has been fulfilled. Yet, there is a growing need for improvements of existing vaccines in terms of increased efficacy and improved safety, besides the

Recent developments have made it possible to construct non-reverting live bacterial vaccine candidates with defined deletions of two or more genes. Such vaccines have proven safe and immunogenic in human volunteers. Since the virulent parent strains are only pathogenic to man (S. typhi, S. flexneri, and V. cholerae), they pose no threat to the environment. Besides holding promise as efficacious vaccines for protection against typhoid fever, bacillary dysentery and cholera, the attenuated strains are well suited as vectors for delivery of heterologous antigenic epitopes from micro-organisms suc

The structure of the gene coding for iduronate sulphate sulphatase (IDS) has been determined. We have used exon to exon and vectorette PCR to identify 9 exons within the IDS gene and to characterise the surrounding intron sequences. The results of this study will be useful for the complete analysis of many mutations giving rise to Hunter syndrome. IDS is the first member of the group of lysosomal nonarylsulphatase genes for which the gene structure has been determined. It bears no relationship to the exon organisation of steroid sulphatase, despite the homology between these two proteins. This

In an attempt to obtain cell lines from the different components found in primary breast cancers, we used a low-calcium medium to culture epithelial cells of mixed phenotype and a recombinant T antigen containing retrovirus to immortalize cells found in these cultures. In each case, the histology of the sample used for culture was examined in detail and the best growth was obtained from samples associated with a substantial in situ or benign component and from lobular rather than ductal carcinomas. Clonal cell lines were developed from each of 4 tumours: 1 infiltrating ductal (tumour number 2)

We have developed a novel positive cloning vector whose use precludes the cloning of any fragments less than 0.8 kb as well as 3.4-kb EcoRI fragments of DYZ1, the largest repeating-DNA family on the long arm of the human Y chromosome. Using this vector, we subcloned inserts of a Y-chromosome-specific phage library constructed from EcoRI-digested flow-sorted Y-chromosome DNA. Ten novel Y-specific fragments were obtained. Their localization on the Y chromosome was determined by deletion mapping using clinical samples with structurally abnormal Y chromosomes. The long arm of the Y chromosome was

Male tsetse, when infected in the laboratory with trypanosomes of the subgenus Trypanozoon, usually produce greater salivary gland infection rates than females of the same species. We show that a single sex-linked gene model can be fitted to most recently published data for salivary gland infection rates in tsetse. The maturation of Trypanosoma congolense infections is shown to be independent of fly sex. The possible effects of genetic control of maturation of Trypanozoon infections in tsetse populations on the transmission of sleeping sickness are considered.

Human lymphoblasts deficient in the enzyme hypoxanthine-guanine phosphoribosyltransferase (HPRT) were infected with an amphotropic helper-free retroviral vector expressing human HPRT cDNA. The stability and expression of the HPRT provirus in five cell lines with different proviral integration sites were examined by determining HPRT mutation and reversion frequencies and by blot hybridization studies. Mutation to the HPRT-negative phenotype occurred at frequencies of approximately 4 X 10(-5) to 3 X 10(-6) per generation. Most mutations in each of the five cell lines were associated with partial

We utilized a plasmid vector host cell reactivation assay to probe the biological functioning of DNA expression vectors and their encoded genes. We studied the effect of ultraviolet radiation or acid-heat treatment on the transient expression of genes transfected into normal human cells and into DNA repair deficient (xeroderma pigmentosum) cells and modification of gene expression by sodium butyrate. U.v. inactivation of transient expression of the bacterial gpt gene contained in a non-replicating expression vector plasmid, pSV2catSVgpt, was much greater in three xeroderma pigmentosum lines th

BACKGROUND: Aging, noise, infection, and ototoxic drugs are the major causes of human acquired sensorineural hearing loss, but treatment options are limited. CRISPR/Cas9 technology has tremendous potential to become a new therapeutic modality for acquired non-inherited sensorineural hearing loss. Here, we develop CRISPR/Cas9 strategies to prevent aminoglycoside-induced deafness, a common type of acquired non-inherited sensorineural hearing loss, via disrupting the Htra2 gene in the inner ear which is involved in apoptosis but has not been investigated in cochlear hair cell protection.

Diamond-Blackfan anemia is a congenital erythroid hypoplasia and is associated with physical malformations and a predisposition to cancer. Twenty-five percent of patients with Diamond-Blackfan anemia have mutations in a gene encoding ribosomal protein S19 (RPS19). Through overexpression of RPS19 using a lentiviral vector with the spleen focus-forming virus promoter, we demonstrated that the Diamond-Blackfan anemia phenotype can be successfully treated in Rps19-deficient mice. In our present study, we assessed the efficacy of a clinically relevant promoter, the human elongation factor 1α short

The main challenge in the "post-GWAS" era is to determine the functional meaning of genetic variants and their contribution to disease pathogenesis. Development of suitable mouse models is critical because disease susceptibility is triggered by complex interactions between genetic, epigenetic, and environmental factors that cannot be modeled by in vitro models. Thyroglobulin (TG) is a key gene for autoimmune thyroid disease (AITD) and several single nucleotide polymorphisms (SNPs) in the TG coding region have been associated with AITD. The classical model of experimental autoimmune thyroiditis

BACKGROUND: Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disorder characterized by the loss of both upper and lower motor neurons, leading to progressive paralysis. Both genetic alterations and epigenetic modifications contribute to neuronal dysfunction in the pathogenesis of ALS. However, the mechanism behind genetic mutations in the non-coding region of genes that affect epigenetic modifications remains unclear.
METHODS: Convolutional neural network was used to identify an ALS-associated SNP located in the intronic region of MEF2C (rs304152), residing in a putative enhan

BACKGROUND: The kinetoplastid protozoan Leishmania tropica mainly causes cutaneous leishmaniasis in humans in the Middle East, and relapse or treatment failure after treatment are common in this area. L. tropica's digenic life cycle includes distinct stages in the vector sandfly and the mammalian host. Sexual reproduction and genetic exchange appear to occur more frequently than in other Leishmania species. Understanding these processes is complicated by chromosome instability during cell division that yields aneuploidy, recombination and heterozygosity. This combination of rare recombination

Factors affecting transmission of D. immitis between dogs are summarized in table 4. Although this list may not be exhaustive, it illustrates the complexity of heartworm epidemiology. There are interesting and provocative similarities in the epidemiology of canine and human filariasis. Ottesen (148) has described the spectrum of disease observed in regions endemic for lymphatic filariasis. It includes patients with 1) microfilariae, but no symptoms; 2) disease, but no microfilariae; 3) tropical pulmonary eosinophilia of filarial etiology; and 4) no disease and no evidence of infection. Bovine Viral Diarrhea Virus (BVDV) is a key contributor to the development of Bovine Respiratory Disease complex, which can cause respiratory disease, congenital defects, severe diarrhea, immunosuppression, abortion, or birth of persistently infected calves. Current commercial vaccines are formulated using strains from BVDV-1a and BVDV-2a, conferring protection against some, but not all, homologous species and low neutralization titers against heterologous and emergent species. This study aimed to develop a live vaccine capable of inducing broad protection against diverse BVDV strains. A conte

INTRODUCTION: Actigraphy-quantified physical activity (PA) allows for continuous measurements of PA that are reflective of real-world day-to-day functioning and morbidity in persons living with cardiomyopathy. This analysis reports the results of actigraphy monitoring and relates these to other clinical outcome assessments in the phase 3, multinational REALM-dilated cardiomyopathy (DCM) (NCT03439514) clinical trial in LMNA-related DCM.
METHODS: Between 2020 and 2022, REALM-DCM randomized 37 patients with actigraphy worn on the nondominant wrist continuously to monitor daily PA. Of those, 35 p

Resource availability can alter infection outcomes through its impact on host immunity and on parasite reproduction. On one hand, enhanced nutrition could favor immunity, limiting the parasite, and on the other hand, it could favor establishment and reproduction of the parasite. Our study aimed to determine the effect of diet on (1) host susceptibility to infection and (2) parasite production in a snail-trematode system. We fed Biomphalaria sudanica, a snail vector of Schistosoma mansoni, either a strict lettuce (low nutrient) or pellet (high nutrient) diet for two generations before exposing

BACKGROUND: Malaria prevention still relies greatly on vector control interventions. However, increasing levels of resistance to pyrethroids across Africa have significantly reduced the efficacy of pyrethroid-based interventions leading to an increase of malaria burden. Consequently, it is imperative to characterize the extent and molecular basis of this resistance.
METHODS: This study was conducted from 2020 to 2021 in a South-North transect across Cameroon. WHO tube assay was used to assess the susceptibility profile of Anopheles funestus to the four main classes of insecticides. The effica

The apicomplexan parasites are globally considered as the major cause of numerous infectious diseases in humans and animals. Apicomplexan parasites include Plasmodium, Toxoplasma gondii, Cryptosporidium, Eimeria and Babesia. The rise in the drug resistance have made the traditional control measures, such as chemotherapy and vector management, inadequate against them. These are the intracellular infectious agent and possess complex life cycles, antigenic variability, and immune evasion abilities. These different abilities hinder the development of vaccines against them. Hence, there is urgent n
