Unraveling the pathogenesis of non-alcoholic fatty liver diseases through genome-wide association studies.

Non-alcoholic fatty liver disease (NAFLD) is a significant health burden around the world, affecting approximately 25% of the population. Recent advances in human genetic databases have allowed for the identification of various single nucleotide polymorphisms associated with NAFLD-related traits. Investigating the functions of these genetic factors provides insight into the pathogenesis of NAFLD and potentially identifies novel therapeutic targets for NAFLD. In this review, we summarized current research on genes with NAFLD-associated mutations, highlighting phospholipid remodeling and spatial

Cardiovascular Aging and Longevity: JACC State-of-the-Art Review.

Cardiovascular aging and longevity are interrelated through many pathophysiological mechanisms. Many factors that promote atherosclerotic cardiovascular disease are also implicated in the aging process and vice versa. Indeed, cardiometabolic disorders such as hyperglycemia, insulin resistance, dyslipidemia, and arterial hypertension share common pathophysiological mechanisms with aging and longevity. Moreover, genetic modulators of longevity have a significant impact on cardiovascular aging. The current knowledge of genetic, molecular, and biochemical pathways of aging may serve as a substrate

Molecular clues of a microepidemy among homeless tuberculosis patients in Budapest due to a new and local Mycobacterium tuberculosis clade.

In Budapest, the capital of Hungary, one of the most important tuberculosis related risk factors is homelessness. The aim of this retrospective study was the genetic characterization of Mycobacterium tuberculosis strains isolated from 66 homeless tuberculosis patients by spoligotyping and mycobacterial interspersed repetitive unit (MIRU) typing. The study identified a local microepidemy in the district with the highest tuberculosis incidence of Budapest. Further genetic characterization has shown that the microepidemy was due to a locally emerged Budapest-specific lineage of M. tuberculosis. T

Test of pangamy by genetic analysis of Schistosoma mansoni pairs within its natural murine host in Guadeloupe.

Mating system plays a determinant role in the maintenance and distribution of genetic variations. It can be assessed indirectly by analyzing the distribution of the genetic variability within populations or directly by considering how mating pairs are formed. In the present study, 71 pairs of adult Schistosoma mansoni worms sampled from naturally infected rats were genotyped to investigate how male and female schistosomes paired according to their genetic relatedness. Among all samples, pangamy, the random association between males and females, could not be rejected. Whereas the schistosome ma

Molecular and genetic mechanisms of osteoporosis: implication for treatment.

Osteoporosis is a leading public health problem in our rapidly growing, aging population. It is characterized by reduced bone mass and microarchitectural deterioration of bone tissue, with a consequent increase in bone fragility and susceptibility to fracture risk. Osteoporosis is a complex multifactorial disease, determined by genetic and environmental factors as well as their interactions. A large number of molecular, genetic and environmental factors underlying osteoporosis have been identified in past decades. In this article, we review 1) the molecular mechanisms of several principal syst

Genetic heterogeneity in Alzheimer's disease: a grade of membership analysis.

Grade of membership analysis (GoM) may have particular relevance for genetic epidemiology. The method can flexibly relate genetic markers, clinical features, and environmental exposures to possible subtypes of disease termed pure types even when population allele frequencies and penetrance functions are not known. Hence, GoM may complement existing strategies that sometimes fail in the presence of heterogeneity or when case definition is not well established. To illustrate the method, individuals in the Seattle data set were evaluated with respect to affection status, age at onset, pedigree, s

The genetics of cannabis lifetime use.

Cannabis is one of the most commonly used drugs in the world, and use is trending alarmingly higher. We aimed to examine the genetic basis of cannabis lifetime use (CanLU) and its genetic relationships with a variety of psychiatric- and physical health-related phenotypes. We conducted a multi-ancestral genome-wide association study (GWAS) of CanLU using data from All of Us in five genetic populations. We meta-analyzed the results of EUR participants with previously published CanLU data (total effective sample size: 258,823), and conducted a set of post-GWAS analyses, including genetic correlat

Genome Structural Variants Shape Adaptive Success of an Invasive Urban Malaria Vector Anopheles stephensi.

Global changes are associated with the emergence of several invasive species, although genetic determinants of their adaptive success remain poorly understood. To address this problem, we investigated the role genome structural variants (SVs) play in adaptations of Anopheles stephensi, a primary vector of urban malaria in South Asia and an invasive malaria vector in South Asian islands and Africa. Using whole genome sequencing data, we identified 2,988 duplications and 16,038 deletions of SVs in 115 mosquitoes from invasive island populations and four locations from mainland India, the species

Whole exome sequencing: Unlocking the molecular diagnostic odyssey in Pakhtun ethnic group of Pakistani population.

The Pakhtuns are 40.86 million people, accounting for 16.9 % of the Pakistani population, and primarily reside in the Khyber Pakhtunkhwa province. The Pakistani population in general, and the Pakhtun belt in particular, is divided into small, isolated tribes where inbreeding is very high (>55 %). This leads to high frequency of rare genetic diseases, making "Rare" conditions relatively common in this population. Due to the high frequency and overlapping phenotypes of affected individuals, molecular diagnostics are often difficult and time consuming. We investigated 1374 individuals from 272 fa

Evolutionary and demographic consequences of temperature-induced masculinization under climate warming: the effects of mate choice.

BACKGROUND: One of the dangers of global climate change to wildlife is distorting sex ratios by temperature-induced sex reversals in populations where sex determination is not exclusively genetic, potentially leading to population collapse and/or sex-determination system transformation. Here we introduce a new concept on how these outcomes may be altered by mate choice if sex-chromosome-linked phenotypic traits allow females to choose between normal and sex-reversed (genetically female) males. RESULTS: We developed a theoretical model to investigate if an already existing autosomal allele enc

In the European population HLA-class II genes are not associated with Helicobacter pylori infection.

OBJECTIVE: Genetic variability influences susceptibility to several diseases and depends on the specific ethnic background of individuals. HLA-class II genes have repeatedly been investigated as candidate genes for predisposition to Helicobacter pylori infection. Certain HLA-DQA1 alleles have been reported to be associated with gastric and duodenal ulcer disease in infected patients in the Japanese population. But conflicting results were reported on European and Japanese populations. METHODS: HLA-DRB1 typing of 382 German individuals with well-defined H. pylori status and different clinical

Holistic view of understanding genetic predisposition and perceptions of genomic research in African communities: tradition, trust and transformation.

Genetic research holds immense potential to advance personalised medicine and enhance patient outcomes. However, Africa’s diverse and historically marginalised populations remain significantly underrepresented in these studies, thus limiting the relevance and reach of genomic advancements. By examining African communities through an anthropological lens, this narrative review highlights the importance of their lived realities and holistic perspectives on Genetic and Genomic Research (GGR). Beyond advocating for their inclusion, raising the importance of embedding intentional integrated medical

Alternative Reproductive Tactic Frequencies Vary with Stream Size for an Appalachian Salamander.

AbstractSpatial variation in allele frequencies can provide clues about the mechanisms responsible for the maintenance of genetic polymorphisms. In some populations of two-lined salamanders, males exhibit two alternative reproductive tactics determined by a Y-linked genetic polymorphism: searching males locate and court females in terrestrial habitats, and guarding males defend mates at aquatic nesting sites. Here, we study the fine-scale spatial variation in the frequency of these tactics by genotyping >1,000 total larvae from five pairs of small and large stream reaches in the Appalachian Mo

Multi-generational koala pedigree analysis reveals rapid changes in heritable provirus load associated with life history traits.

Retroviruses that colonize the host germline can be passed on as inherited genetic variants. The koala (Phascolarctos cinereus) is currently experiencing germline colonization by two retroviruses, the koala retrovirus (KoRV) and phaCin-β. We analyze the integration site segregation and diversity of endogenous KoRV, phaCin-β, and the related phaCin-β-like in 111 pedigreed koalas from the San Diego Zoo Wildlife Alliance and seven European Zoos. The use of multigenerational pedigrees and the inclusion of health information for each individual koala reveal elimination of retroviruses from proto-on

Worldwide genetic diversity of Plasmodium vivax Pv47 is consistent with natural selection by anopheline mosquitoes.

Pv47 is the Plasmodium vivax ortholog of Pfs47, a protein that allows the Plasmodium falciparum malaria parasite to evade mosquito immunity and adapt to diverse vectors. We analyzed global genetic diversity of Pv47 and compared it with Pfs47, finding that most common Pv47 polymorphisms are non-synonymous and cluster in regions similar to those in Pfs47. Pv47 domain 2 presents an excess of non-synonymous substitutions, suggesting positive selection. The greatest haplotype diversity is found in Pv47 from East/Southeast Asia and Oceania. Like Pfs47, Pv47 also exhibits a marked geographic populati

Phagosomal RNA sensing through TLR8 controls susceptibility to tuberculosis.

Genetic determinants of susceptibility to Mycobacterium tuberculosis (Mtb) remain poorly understood but could provide insights into critical pathways involved in infection, informing host-directed therapies and enabling risk stratification at individual and population levels. Through a genome-wide forward genetic screen, we identify Toll-like receptor 8 (TLR8) as a key regulator of intracellular killing of Mtb. Pharmacological TLR8 activation enhances the killing of phylogenetically diverse clinical isolates of drug-susceptible and multidrug-resistant Mtb by macrophages and during in vivo infe

Hotspots of genetic change in Yersinia pestis.

The relative contributions of mutation rate variation, selection, and recombination in shaping genomic variation in bacterial populations remain poorly understood. Here we analyze 3318 Yersinia pestis genomes, spanning nearly a century and including 2336 newly sequenced strains, to shed light on the patterns of genetic diversity and variation distribution at the population level. We identify 45 genomic regions ("hot regions", HRs) that, although comprising a minor fraction of the genome, are hotbeds of genetic variation. These HRs are distributed non-randomly across Y. pestis phylogenetic line
