Oxyphenbutazone suppresses non-canonical inflammasome activation and protects against LPS-induced sepsis.

Sepsis remains a leading cause of mortality worldwide, yet therapies targeting proximal inflammatory receptors (e.g., TLR4) or mediators have shown limited clinical success. Non-canonical inflammasome-mediated pyroptosis represents a critical downstream pathological driver and an attractive therapeutic target for Gram-negative sepsis. Although cyclooxygenase (COX)-inhibiting non-steroidal anti-inflammatory drugs (NSAIDs) have been reported to attenuate sepsis in preclinical studies, whether COX activity regulates non-canonical inflammasome activation and subsequent pyroptosis remains unclear.

Anti-inflammatory Diterpenoids and Sesquiterpenoids from the Rhizomes of Shell Ginger (Alpinia zerumbet).

Shell ginger (Alpinia zerumbet) is a perennial species widely utilized as both an edible spice and a medicinal plant. Phytochemical investigation of its rhizomes yielded 33 terpenoids, of which 12 are previously undescribed compounds, comprising both diterpenes and sesquiterpenes. Notably, compound 1 represents a rare, highly oxygenated isospongiane-type diterpenoid with a novel skeleton. The anti-inflammatory potential of all isolates was assessed using lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages and BV2 microglial cells. Subsequent mechanism-oriented assays demonstrated that co

Pulsed Electromagnetic Field Exposure Attenuates Ultraviolet B-Induced Dermal Collagen Loss in Association with A2A Adenosine Receptor Signaling.

Ultraviolet (UV) exposure accelerates skin aging through wavelength-dependent effects. Ultraviolet A (UVA) penetrates relatively deeply into the dermis and promotes oxidative stress and extracellular-matrix remodeling, whereas ultraviolet B (UVB) is absorbed predominantly in the epidermis and produces direct DNA damage and inflammation; nevertheless, UVB can also alter dermal fibroblast matrix metabolism in experimental models. The present study specifically used UVB irradiation (peak wavelength, 306 nm) to examine inflammatory collagen loss. UVB-induced cellular injury activates the NOD-like

Necroptosis activates NLRP3 inflammasome to drive necroinflammation in T-2 toxin-induced cartilage damage.

As a highly toxic mycotoxin present in contaminated food sources, T-2 toxin disrupts skeletal development by inducing cartilage injury. However, the underlying molecular mechanisms remain unclear. Following four weeks of gavage administration of T-2 toxin (200 ng/g body weight/day) to three-week-old male C57BL/6 mice, chondrocyte death in articular cartilage was accompanied by increased levels of p-RIPK3, p-MLKL, NLRP3, cleaved caspase-1, and IL-1β. In C28/I2 chondrocytes, T-2 toxin decreased cell viability and increased LDH release, accompanied by enhanced NLRP3 inflammasome-related inflammat

A KCa 2.2/2.3 Opener Reverses ET-1-Induced NLRP3 Activation in Hypertensive Mice Corpora Cavernosa.

Hypertension-induced erectile dysfunction is associated with endothelial dysfunction in the corpus cavernosum. Membrane depolarization activates the NLRP3 inflammasome, with downregulation of endothelial Ca2+-activated K+ channels type 2.3 (KCa 2.3) and upregulation of endothelin-1 (ET-1) linked to erectile dysfunction. However, underlying mechanisms remain incompletely understood. We hypothesized that activating KCa 2.2/2.3 channels reverses erectile dysfunction and ET-1-induced NLRP3 activation in hypertensive DOCA/salt mice. Hypertension was induced in mice using a DOCA/salt model, with uni

NLRP6 drives cutaneous wound healing during Staphylococcus aureus infection in mice.

AIMS: Infection is a major complication of wound healing worldwide, with Staphylococcus aureus being the most isolated bacterial species from infected wounds. Pattern recognition receptors, such as the NOD-like receptor family (NLRs), play crucial roles in coordinating subsequent stages of wound-healing via specific cells activation through the production of cytokines, such as IL-1β, whose release is strongly dependent on inflammasome complex assembly and is critical for controlling the bacterial load. This study investigated if the NOD-like receptor family pyrin domain-containing 3 (NLRP3), N

Berberine-associated lipid-inflammatory regulation in atherosclerosis: an integrative transcriptomic, single-cell, and in vivo study.

Atherosclerotic plaques combine disturbed lipid metabolism with myeloid inflammation, yet the human plaque context of berberine (BBR)-related targets remains poorly defined. Paired GSE43292 samples were used to characterize plaque-associated changes, GSE100927 to test reproducibility, and GSE159677 to resolve their cellular distribution. Network pharmacology prioritized BBR-atherosclerosis candidates, with molecular docking and 100-ns molecular dynamics used for structural evaluation. High-fat diet-fed ApoE-/- mice received BBR or nanoliposomal BBR (BNL), with atorvastatin as a comparator. In

7-dehydrocholesterol reductase inhibitors control Leishmania spp. in human macrophages.

Leishmaniasis, caused by Leishmania spp., is a neglected parasitic disease with a broad clinical spectrum, whose treatments are toxic and present failures and resistance. Leishmania braziliensis and Leishmania amazonensis are highly prevalent species in Brazil. Cholesterol metabolism affects immune responses. 7-dehydrocholesterol reductase (DHCR7) converts 7-dehydrocholesterol (7-DHC) to cholesterol and this enzyme regulates macrophage polarization towards the M2 profile. Inhibiting DHCR7 enhances pro-inflammatory responses and may control infections, prompting investigation into its effects o

Activation of GPR35 by kynurenic acid inhibits IL-1β secretion in macrophages during CR-hvKP-induced pneumonia.

Carbapenemase-resistant hypervirulent Klebsiella pneumoniae (CR-hvKP) is a common pathogen that can cause severe pneumonia. The innate immune response, especially the response of macrophages, plays a crucial role in the host's defense against bacterial infections. Glycolysis is implicated in the modulation of immune functions in macrophages. Here, we provide evidence supporting the role of GPR35 in decreasing glycolysis and reducing the secretion of IL-1β in macrophages by inhibiting the transcription of HK2 during K. pneumoniae-induced pneumonia. Mice with GPR35 knock-out exhibit higher morta

Structurally diverse sesquiterpenoid glycosides with anti-inflammatory activity from Cissampelopsis spelaeicola.

Sixteen previously undescribed sesquiterpenoid glycosides (1-16), bearing eudesmane-type (1-4) and cadinane-type skeletons (5-16), were obtained from the stems of Cissampelopsis spelaeicola. The structural elucidation and absolute configuration assignment of compounds 1-16 were accomplished through multiple spectroscopic analyses, computed ECD/NMR, and X-ray crystallography. Furthermore, anti-neuroinflammatory effects of all isolates were assessed by NO inhibition in LPS-stimulated BV-2 microglia. Compounds 12 and 14-16 exhibited potent NO inhibitory effects (IC50 = 5.62-19.29 μM), outperformi

Resolvin D1 suppresses inflammation in human fibroblast-like synoviocytes via the p-38, NF-κB, and AKT signaling pathways.

Synovitis represents the initial pathological change in osteoarthritis and contributes to its progression. Resolvin D1 (RV-D1) is a novel and endogenous docosahexaenoic acid-derived lipid mediator, which regulates the duration and magnitude of inflammation by downregulating pro-inflammatory genes and mediators. However, the effects of RV-D1 on synovitis remain unknown. The aim of the present study was to investigate the anti-inflammatory effects of RV-D1 in human fibroblast-like synoviocytes (HFLSs) and the underlying mechanisms. The expression of the HFLS formyl peptide receptor 2 (ALX/FPR) w

Role of CD34 in calcification of human aortic valve interstitial cells from patients with aortic valve stenosis.

Various osteogenic factors are involved in ectopic human aortic valve calcification; however, the key cell species involved in calcification remains unclear. In a previous study, we reported that mesenchymal stem (CD73, 90, 105) and endothelial (VEGFR2) cell markers are positive in almost all human aortic valve interstitial cells (HAVICs) obtained from a patient with calcified aortic valve stenosis (CAVS). Further, CD34-negative HAVICs are highly sensitive to calcification stimulations. Here, we aimed to pathophysiologically clarify the role of CD34 in HAVICs obtained from individual patients

Neutrophil extracellular traps promote the activation of the NLRP3 inflammasome and PBMCs pyroptosis via the ROS-dependent signaling pathway in Kawasaki disease.

Kawasaki disease (KD) is an acute systemic vasculitis predominantly affecting infants and children under the age of 5. Recent studies have indicated that excessively released neutrophil extracellular traps (NETs) are involved in the progression of vasculitis. The purpose of this study was to investigate the role of NETs, especially their interaction with peripheral blood mononuclear cells (PBMCs), in the pathogenesis of KD. First, we demonstrated that the levels of NETs (cfDNA, MPO, and NE) in the serum of KD patients were significantly higher than those in healthy controls (HCs) and notably d

GSNOR negatively regulates the NLRP3 inflammasome via S-nitrosation of MAPK14.

Hyperactivation of the NLRP3 inflammasome has been implicated in the pathogenesis of numerous diseases. However, the precise molecular mechanisms that modulate the transcriptional regulation of NLRP3 remain largely unknown. In this study, we demonstrated that S-nitrosoglutathione reductase (GSNOR) deficiency in macrophages leads to significant increases in the Nlrp3 and Il-1β expression levels and interleukin-1β (IL-1β) secretion in response to NLRP3 inflammasome stimulation. Furthermore, in vivo experiments utilizing Gsnor-/- mice revealed increased disease severity in both lipopolysaccharide
