Homozygous loss of function variant in LMNB2 gene causes major brain malformation and perinatal death.

Lamins play a major role in the mechanical stability of cell nuclei, the organisation of chromatin and the DNA replication, transcription and repair. The expression profiles of A-type and B-type lamins vary depending on developmental stages, cell types and tissues. Lamin B2 is expressed very early in embryogenesis, especially in the central nervous system, where it is essential for neuronal migration and brain development. Pathogenic missense variants in lamin B2 have been linked to conditions such as lipodystrophy, progressive myoclonic epilepsy and primary microcephaly. Here, we report clini

Early-onset phenotype in a patient with an intermediate allele and a large SCA1 expansion: a case report.

BACKGROUND: Spinocerebellar ataxia type 1, is a rare neurodegenerative disorder with autosomal dominant inheritance belonging to the polyglutamine diseases. The diagnosis of this disease requires genetic testing that may also include the search for CAT interruption of the CAG repeat tract. CASE PRESENTATION: One 23-years-old patient suffers from a severe ataxia, with early-onset and rapid progression of the disease. His father might have been affected, but no molecular confirmation has been performed. The genetic results were negative for the Friedreich's ataxia, spinocerebellar ataxia type 2

Ex Vivo Patient-Derived Explant Model for Neurofibromatosis Type 1-Related Cutaneous Neurofibromas.

Cutaneous neurofibromas (CNFs) are benign tumors that occur in the dermis of individuals with the inherited tumor predisposition disorder, neurofibromatosis type 1. CNFs cause disfigurement, pain, burning, and itching, resulting in substantially reduced QOL in patients with neurofibromatosis type 1. CNFs are benign tumors that exhibit cellular and molecular heterogeneity, making it difficult to develop tractable in vitro or in vivo models. As a result, CNF research and drug discovery efforts have been limited. To address this need, we developed a reproducible patient-derived explant (PDE) ex v

Selective neuroimmune modulation by type I interferon drives neuropathology and neurologic dysfunction following traumatic brain injury.

Accumulating evidence suggests that type I interferon (IFN-I) signaling is a key contributor to immune cell-mediated neuropathology in neurodegenerative diseases. Recently, we demonstrated a robust upregulation of type I interferon-stimulated genes in microglia and astrocytes following experimental traumatic brain injury (TBI). The specific molecular and cellular mechanisms by which IFN-I signaling impacts the neuroimmune response and neuropathology following TBI remains unknown. Using the lateral fluid percussion injury model (FPI) in adult male mice, we demonstrated that IFN α/β receptor (IF

Lewis acidic zinc(II) salen-type Schiff-base complexes: sensing properties and responsive nanostructures.

In this frontier article some peculiar characteristics of Zn(salen)-type Schiff-base complexes are reviewed. The paper is mainly focused on the most recent and relevant achievements on responsive supramolecular nanostructures and sensing properties, both of them related to the Lewis acidic character of the ZnII centre in these molecular species, providing an interpretation of these features. The sensing properties of Zn(salen)-type complexes mainly originate from optical spectroscopic changes associated with the formation of the adducts upon addition of a Lewis base (analyte), either by deaggr

Genotypic Diversity Is Independent of Pathogenicity in Colombian Strains of Cryptococcus neoformans and Cryptococcus gattii in Galleria mellonella.

Cryptococcosis is a potentially fatal opportunistic mycosis that affects the lungs and central nervous system. It has been suggested that certain strains of C. neoformans/C. gattii may have the potential to be more virulent according to the molecular type. This study aims to investigate the association between virulence in the G. mellonella model and genotypic diversity of Colombian clinical and environmental isolates of C. neoformans/C. gattii. A total of 33 clinical and 12 environmental isolates were selected according to their geographical origin and sequence types (STs). Pathogenicity was

Molecular mechanisms underlying cardiac antihypertrophic and antifibrotic effects of natriuretic peptides.

Natriuretic peptides (NPs) exert well-characterized protective effects on the cardiovascular system, such as vasorelaxation, natri- and diuresis, increase of endothelial permeability, and inhibition of renin-angiotensin-aldosterone system. It has been reported that they also possess antihypertrophic and antifibrotic properties and contribute actively to cardiac remodeling. As a consequence, they are involved in several aspects of cardiovascular diseases. Antihypertrophic and antifibrotic actions of NPs appear to be mediated by specific signaling pathways within a more complex cellular network.

Comparative transcriptional and biochemical studies in muscle of myotonic dystrophies (DM1 and DM2).

Myotonic dystrophy type 1 (DM1) and myotonic dystrophy type 2 (proximal muscular myopaty/DM2) are caused by similar dynamic mutations at two distinct genetic loci. The two diseases also lead to similar phenotypes but different clinical severity. Dysregulation of alternative splicing has been suggested as the common pathogenic mechanism. Here, we investigate the molecular differences between DM1 and DM2 using reverse transcriptase-polymerase chain reaction of troponin T (TnT) and the insulin receptor (IR), as well as immunoblotting of TnT in muscle biopsies from DM1 and DM2 patients. We found t

[A case of anaphylaxis caused by the health food chitosan].

Chitosan is a linear polysaccharide with about 10(5)-10(6) molecular weight existing in cell walls of zygomycetes. Chitosan decreases serum neutral fat and cholesterol levels and has dietary fiber action, and many products have recently been sold as health foods. Here, we report a case of immediate-type allergy for chitosan-containing health food with a review of the literature. The patient was a 47-year-old female person who developed systemic urticaria and difficulty in breathing after oral ingestion of chitosan. Since skin tests (prick test and scratch patch test) were positive, the test wa

Human lysyl hydroxylase: purification to homogeneity, partial characterization and comparison of catalytic properties with those of a mutant enzyme from Ehlers-Danlos syndrome type VI fibroblasts.

Lysyl hydroxylase was isolated as an essentially homogeneous protein from human fetal tissues and as a homogeneous protein from placental tissue by a procedure involving ammonium sulfate fractionation, affinity chromatography on concanavalin A-agarose, affinity chromatography on collagen linked to agarose and gel filtration. The specific activity of the best enzyme preparations from human fetal tissues was about 80,000 times, and from human placenta about 63,000 times that in the 15,000 X g supernatant of the corresponding tissue homogenate. Physicochemical heterogeneity of hepatitis B e antigen detected in asymptomatic carriers and carriers in a hemodialysis unit.

Physicochemical studies of hepatitis B e antigen (HBeAg) revealed a clear cut difference between e1 and e2 antigen. The e1 antigen was found to have a MW of Ca 150,000 and a pI of 6.4-7.2, whereas both the MW and pI of the e2 antigen were heterogeneous depending upon the source of serum. Sera obtained from asymptomatic carriers were characterized by low titers of HBs antigen, HBc antigen and DNA polymerase and contained e2 antigen of larger molecular weight (200,000-300,000) with a narrow distribution range and a pI of 4.8 to 5.2 (type 1). On the other hand, the sera from patients in a hemodia

Rational Sequence Modification Upgrades a Food-Derived Peptide into a Stronger Dual-Functional Regulator of α-Glucosidase Activity and Insulin Resistance.

Inhibition of α-glucosidase and improvement of insulin resistance (IR) are key strategies for treating type 2 diabetes mellitus (T2DM). Our previous work obtained a novel α-glucosidase inhibitory peptide, FAPSW. However, its activity was weaker than acarbose. To further enhance its activities, 17 novel peptides were constructed by amino acid substitution using FAPSW as a template. Among them, YAPSW (IC50 = 0.126 ± 0.008 mM) exhibited stronger α-glucosidase inhibition than FAPSW and acarbose. Molecular docking suggested that hydrogen bonds were not the sole determinants of peptide-mediated α-gl

Inhibition of Glucose-Induced Glycation of HSA by Pioglitazone: Multi-Spectroscopic and Bioinformatic Evidence.

Diabetes mellitus is a growing health concern that causes numerous complications. Glycation produces advanced glycation end-products (AGEs), which promote diabetic complications. Targeting glycation is a strategy for combating the progression of diabetic complications. Pioglitazone enhances insulin sensitivity in patients with type 2 diabetes mellitus, but its impact on glycation remains unclear. This study aims to evaluate whether pioglitazone can inhibit glucose-induced glycation of human serum albumin (HSA), using in vitro assays and in silico tools. Pioglitazone inhibited >70% of early gly

Underexplored Ligand-Binding Features of FabI From Staphylococcus aureus and Escherichia coli: A Comparative Pharmacophoric Modeling and Surface Mapping Approach.

The rapid spread of antimicrobial resistance, particularly among pathogens such as Staphylococcus aureus and Escherichia coli, highlights the urgent need for novel antibacterial agents with new mechanisms of action. The bacterial enoyl-acyl carrier protein reductase (FabI), an essential enzyme in fatty acid biosynthesis, represents a promising target for narrow-spectrum antimicrobials. This study aimed to define consensus pharmacophore models and interaction profiles for FabI through an integrated computational approach. ConPhar and FTMap analyses were applied to experimental structures and mo

Targeting Insulin Signaling and TRAF2/JNK Pathway: A Comprehensive In Silico Study of Uncaria tomentosa Compounds.

Type 2 diabetes (T2D) is a metabolic syndrome frequently associated with obesity and endoplasmic reticulum stress-mediated inflammation, which can trigger the unfolded protein response (UPR), impair insulin signaling, and promote apoptosis. To identify potential natural therapeutic candidates, this study investigated the mechanisms of action of 14 compounds from Uncaria tomentosa (UT), a medicinal plant from the Amazon rainforest, using in silico modeling. The study focused on the UPR, TRAF2/JNK pro-inflammatory signaling pathway, and insulin signaling pathways, which play key roles in T2D. So

Discovery of Ligands for the TNFR1 Extracellular Domain Using Fragment-Based Drug Discovery.

Tumor necrosis factor receptor 1 (TNFR1) plays a major role in immunoregulation. It is involved in inflammatory and autoimmune diseases like rheumatoid arthritis, psoriasis, Alzheimer's disease, and multiple sclerosis. However, few small-molecule inhibitors of TNFR1 have been reported, even though they constitute a good alternative to already existing antibody therapies targeting the TNF pathway. Here, we report the discovery of a new class of molecules for the extracellular domain of TNFR1 using a fragment-based approach through primary screening by NMR spectroscopy, followed by orthogonal va

A salophen-type macrocyclic Schiff base ligand and its metal complexes: exploring in vitro anticancer efficacy via in silico topoisomerase IIβ enzyme targeting.

Cancer persists as a principal cause of global mortality. Despite significant progress in cancer therapeutics in recent decades, chemotherapy remains a primary modality for cancer treatment. Based on their mechanism of action, commonly used chemotherapeutic agents can be classified into several categories, including topoisomerase inhibitors. Here, a newly synthesized salophen-type macrocyclic Schiff-base ligand (5E,19E)-3-bromo-12,13-dihydrodibenzo[e,m]pyrido[2,3-i][1,4]dioxa[8,11]diazacyclotetradecine (4) and its metal complexes (5a-5d) have been reported as potent anticancer agents exhibitin
