Burkitt Lymphoma and Other High-Grade B-Cell Lymphomas with or without MYC, BCL2, and/or BCL6 Rearrangements.

Burkitt lymphoma (BL) is highly aggressive and requires very intensive chemotherapy approaches for successful treatment. Although "standard" approaches are tolerated in young patients, this is not the case in older adults, and new approaches that lower toxicity but maintain high curability are needed and are currently in development. Recently, high-grade B-cell lymphomas with MYC and BCL2 and/or BCL6 rearrangements have been categorized separately from diffuse large B-cell lymphoma or BL. These have poor outcomes and many exciting novel approaches are being tested in an attempt to augment cura

Expression of c-Ets1 is associated with malignant potential in endometrial carcinoma.

BACKGROUND: The protooncogene c-ets1 is a transcriptional factor that controls the expression of a number of genes involved in extracellular matrix remodeling. It might play a role in the regulation of physiologic processes such as cell proliferation and differentiation and also is associated with angiogenesis, cell migration, and tumor invasion. METHODS: To elucidate the involvement of c-Ets1 in endometrial carcinogenesis, the authors analyzed serial frozen sections for c-Ets1 protein expression in 20 cases of endometrial carcinoma and 20 cases of normal endometria by fluorescent immunohisto

[Genetics of Hirschsprung disease].

Hirschsprung's disease (HD) is one of the commonest gastrointestinal malformations, as it affects one child out of 5,000 births. It classically induces severe neonatal intestinal obstruction requiring surgical treatment which currently ensures a favourable prognosis for most of the affected children. Although the great majority of cases are sporadic, the existence of familial forms (10% of cases) has allowed localization and then identification of an autosomal dominant gene on chromosome 10, the RET proto-oncogene, responsible for 50% of familial forms and 15% of sporadic cases. A second gene

Expression of c-fos and c-myc in Raji Burkitt's lymphoma cells during the progression of DMSO-induced G1 cells into S phase.

Using flow cytometry, we have recently found that dimethyl sulfoxide (DMSO) reversibly induces G1-arrest in the cell cycle of human lymphoid cell lines such as Raji, Akata, and Molt-4. Here we investigated c-fos and c-myc expression in Raji Burkitt's lymphoma cells at DMSO-induced G1 arrest and after release from DMSO. A small but significant accumulation of c-fos mRNA was observed in G1-arrested Raji cells after treatment with 1.5% DMSO for 96 h. When G1-arrested Raji cells were transferred to DMSO-free medium, a transient increase in c-fos transcripts was detected 30-60 min after the release

Molecular pathology of adenoid cystic carcinoma.

Adenoid cystic carcinoma (ACC) is a slow-growing but locally aggressive salivary gland tumor. ACC is composed of ductal/tubular epithelial cells and basal/myoepithelial cells, which form cribriform, tubular, and solid growth patterns in variable combinations and dominance. ACC from different anatomic sites have similar morphological, molecular, and genetic changes. The key molecular alteration in ACC is chromosomal fusion/rearrangement/translocation involving MYB or MYBL1, usually with NFIB as a fusion partner. In this review, we summarize the pathology and molecular alterations in ACC and the

Targeting friend leukemia integration 1: A promising approach for prevention and treatment of solid tumors.

Friend leukemia integration 1 (FLI1) is an ETS transcription factor first identified in erythroleukemia. This protein contributes to various cellular functions such as cell growth and proliferation, apoptosis, angiogenesis, etc. FLI1 is also known to be involved in tumorigenesis. The role of this transcription factor as a proto-oncogene, promoting cancer progression, especially Ewing sarcoma, is well reported. Recent research has found the connection of FLI1 with other solid cancers, including breast cancer, prostate cancer, glioma, and lung cancer. The role of this protein in solid cancers is

Uterine Myxoid Mesenchymal Tumor With a Novel SS18::VEZF1 Gene Fusion, Lacking Worrisome Histological Features.

We report a uterine myxoid mesenchymal tumor with a novel SS18::VEZF1 gene fusion. The current lesion was identified in a 53-year-old woman who presented with symptomatic "fibroids" showing accelerated growth and heterogeneous morphology on radiologic assessment. Microscopic examination revealed a well-demarcated neoplasm, and the tumor exhibited alternating hypocellular/hyalinized and hypercellular areas, composed of a monomorphic proliferation of spindle, ovoid, and epithelioid cells arranged in sheets. These cells were embedded within either a hyalinized collagenous stroma or abundant myxoi

Knockout of c-Cbl/Cbl-b slows c-Met trafficking resulting in enhanced signaling in corneal epithelial cells.

In many cell types, the E3 ubiquitin ligases c-Cbl and Cbl-b induce ligand-dependent ubiquitylation of the hepatocyte growth factor (HGF)-stimulated c-Met receptor and target it for lysosomal degradation. This study determines whether c-Cbl/Cbl-b are negative regulators of c-Met in the corneal epithelium (CE) and if their inhibition can augment c-Met-mediated CE homeostasis. Immortalized human corneal epithelial cells were transfected with Cas9 only (Cas9, control cells) or with Cas9 and c-Cbl/Cbl-b guide RNAs to knockout each gene singularly (-c-Cbl or -Cbl-b cells) or both genes (double KO [

Analysis of V600E BRAF and D816V KIT mutations in systemic mastocytosis.

Most patients with systemic mastocytosis (SM) carry a D816 V KIT mutation causing a ligand-independent activation of the receptor. Down-stream of KIT is several components known to be mutated in different malignancies. RAF is among the most frequently mutated kinases, where BRAF V600E mutation occurs in most hairy cell leukemias (HCL) and half of malignant melanomas. We investigated BRAF mutations in 36 subjects with different forms of SM, but could not detect BRAF mutation in any of the cases, not even in the mast cell lineage of a patient with V600E BRAF-positive HCL. Thus, although BRAF is

Sex-specific Risks in Lung Cancer-Associated Thrombosis: Focus on Female Patients.

Sex and lung cancer-associated thrombosis (CAT) matter. Venous thromboembolism (VTE) is a frequent and serious complication in lung cancer, and accumulating evidence suggests that sex-related factors may influence thrombotic risk. In this review, we highlight the epidemiological, clinical, and biological specificities of women in relation to thrombotic risk in lung cancer. We explore sex-specific determinants of CAT, including host-, tumor- and treatment-related factors. Special attention is given to oncogene-addicted adenocarcinoma, particularly enriched in women and their implications for VT

Holding but not folding: How a single charge flip uncouples the DNAJC7-Hsp70 relay in amyotrophic lateral sclerosis.

Genetic mutations impact protein function through various routes: Some catalyze new oncogenic activities, while others trigger complete structural collapse. However, the E425K mutation in DNAJC7, associated with Amyotrophic Lateral Sclerosis (ALS), presents a far more subtle and intriguing case. In their recent study in The FEBS Journal, Elmaleh et al. (2026) FEBS Lett employed high-resolution NMR to demonstrate that this mutation leaves the protein's overall structure intact while selectively paralyzing its ability to communicate with the Hsp70 chaperone machinery. In this commentary, we show

A Patient With Novel PPP1CB -ALK Fusion Advanced NSCLC Achieved Long Survival From Alectinib: A Case Report.

ALK gene fusions are well-established oncogenic drivers that occur in approximately 5% of NSCLCs. The EML4 gene is the most frequent fusion partner. With the advent of next-generation sequencing, numerous novel ALK fusions have been discovered; however, many have not been represented in pivotal clinical trials evaluating ALK tyrosine kinase inhibitors. We present a case of advanced NSCLC with a rare PPP1CB-ALK fusion. The patient achieved a durable response to alectinib, with a progression-free survival of nearly four years, and the patient continues to be progression free at the most recent f

Regulation of TIPIN and replication fork dynamics by USP37.

The deubiquitinating enzyme, USP37, is associated with control of replication and the replication stress response. Cells with oncogene-induced replication stress harbor elevated levels of USP37 and are more sensitive to its loss. These cells experience increased replication stress and delayed progression of the replication fork in the absence of USP37. Consistent with these observations, we find that USP37-deficient cells more frequently fail to restart stalled forks and that these forks are more susceptible to nuclease activity. Underlying these phenomena, we show that USP37 resides at the re

Hypoxia-induced BHLHE41 promotes intrahepatic cholangiocarcinoma progression by enhancing fatty acid synthesis via upregulating ACLY.

Intrahepatic cholangiocarcinoma (ICC) remains one of the most aggressive human malignancies, with its pathogenesis critically influenced by the hypoxic tumor microenvironment. Through RNA sequencing of clinical samples and hypoxia-exposed ICC cells, we identified basic helix-loop-helix transcription factor 41 (BHLHE41) as a significantly upregulated gene. Functional studies demonstrated that, by reprogramming fatty acid synthesis, hypoxia-induced BHLHE41 promotes the proliferation and migration of ICC cells. Combined CUT&Tag and RNA sequencing analyses confirmed that BHLHE41 directly targets a

A middle-aged man with progressive facial swelling and generalized violaceous plaques.

This report describes a patient with AIDS-related angiomatous Kaposi sarcoma (KS) presenting with facial lymphoedema. Angiomatous KS is a rare histological subtype of KS that can mimic benign haemangiomas clinically and histologically. Furthermore, the patient did not have prior history or other clinical clues of HIV infection at the initial presentation, making the diagnosis challenging. Although lymphoedema has not previously been reported in association with angiomatous KS, this case demonstrates that lymphoedema may develop in patients with this histological subtype of KS, reflecting the m

A Noncanonical MET Exon 14 Splice-Site Variant in Pulmonary Sarcomatoid Carcinoma With Response to Capmatinib.

MET exon 14 skipping is an actionable oncogenic driver in non-small cell lung carcinoma (NSCLC); however, noncanonical splice-region variants are frequently classified as variants of unknown significance (VUS), which may result in missed therapeutic opportunities and highlight limitations in current DNA next-generation sequencing (NGS) reporting criteria. We report a patient with pulmonary sarcomatoid carcinoma harboring a noncanonical MET splice donor-proximal indel (c.3022_3028 + 13delinsA), initially interpreted as a VUS, who achieved a rapid and durable response to capmatinib. Case Series: Large B-cell Lymphoma With IRF4 Rearrangement Involving the Lung and Tonsil in Two Different Patients.

Large B-cell lymphoma (LBCL) with interferon regulatory factor 4 (IRF4) rearrangement is characterized by the monotonous proliferation of medium- to large-sized cells with strong expression of IRF4/multiple myeloma oncogene 1 (MUM1). It predominantly affects children and young adults with a median age of 12. The reported disease sites include head and neck lymph nodes, the Waldeyer's ring, and the gastrointestinal tract. Here, we report two cases of LBCL with IRF4 rearrangement in 73-year-old and 71-year-old patients. The first patient presented with a lung nodule, while the second patient pre
