Suppression of cytokines in mice by protein A-V antigen fusion peptide and restoration of synthesis by active immunization.

It is established that an approximately 70-kb Lcr plasmid enables Yersinia pestis, the causative agent of bubonic plague, to multiply in focal necrotic lesions within visceral organs of mice by preventing net synthesis of the cytokines tumor necrosis factor alpha (TNF-alpha) and gamma interferon (IFN-gamma), thereby minimizing inflammation (Lcr+). Rabbit antiserum raised against cloned staphylococcal protein A-V antigen fusion peptide (PAV) is known to passively immunize mice against 10 minimum lethal doses of intravenously injected Lcr+ cells of Y. pestis. In this study, injected PAV suppress

Interlaboratory performance of disk agar diffusion and dilution antimicrobial susceptibility tests, 1979-1981. A summary of the microbiology portion of the College of American Pathologists (CAP) surveys.

The antimicrobial susceptibility test results of the 1979-1981 College of American Pathologists (CAP) Microbiology Surveys are summarized for the disk diffusion and dilution test methods. Improved interpretive accuracy (97.5%) was detected for the Special Bacteriology participants compared with those reported for 1972 and 1977-1978. The performance accuracy rates for the Comprehensive and Basic Surveys subscribers was 95.8% and 93.7%, respectively (all surveys accuracy rates was 95.2% from 196,775 responses). The dilution methods were graded since 1980 and showed an acceptable or good performa

Kinetic epitope mapping of monoclonal antibodies raised against the Yersinia pestis virulence factor LcrV.

Five monoclonal antibodies, mAb7.3, mAb29.3, mAb46.3, mAb12.3 and mAb36.3, raised to the LcrV virulence factor from Yersinia pestis were characterised for their Fab affinity against the purified protein and their Fc affinity to Protein A/G as a proxy for the FcγR receptor. Kinetic measurements were performed label-free in a localised particle plasmon array reader. The Fc-ProteinA/G complex first-order half-life was determined for each antibody and fell in the range of 0.8-3.8h. The Fab first-order half-lives had ranged from 3.4 to 9.2h although two antibodies, mAb12.3 and mAb36.3, showed low a

Kinetics of innate immune response to Yersinia pestis after intradermal infection in a mouse model.

A hallmark of Yersinia pestis infection is a delayed inflammatory response early in infection. In this study, we use an intradermal model of infection to study early innate immune cell recruitment. Mice were injected intradermally in the ear with wild-type (WT) or attenuated Y. pestis lacking the pYV virulence plasmid (pYV(-)). The inflammatory responses in ear and draining lymph node samples were evaluated by flow cytometry and immunohistochemistry. As measured by flow cytometry, total neutrophil and macrophage recruitment to the ear in WT-infected mice did not differ from phosphate-buffered

Mobility, mortality, and the middle ages: identification of migrant individuals in a 14th century black death cemetery population.

Mobility and migration patterns of groups and individuals have long been a topic of interest to archaeologists, used for broad explanatory models of cultural change as well as illustrations of historical particularism. The 14th century AD was a tumultuous period of history in Britain, with severely erratic weather patterns, the Great Famine of 1315-1322, the Scottish Wars of Independence, and the Hundred Years' War providing additional migration pressures to the ordinary economic issues drawing individuals to their capital under more stable conditions. East Smithfield Black Death Cemetery (Roy

Emended description of Actinomyces naeslundii and descriptions of Actinomyces oris sp. nov. and Actinomyces johnsonii sp. nov., previously identified as Actinomyces naeslundii genospecies 1, 2 and WVA 963.

Actinomyces naeslundii is an important early colonizer in the oral biofilm and consists of three genospecies (1, 2 and WVA 963) which cannot be readily differentiated using conventional phenotypic testing or on the basis of 16S rRNA gene sequencing. We have investigated a representative collection of type and reference strains and clinical and oral isolates (n=115) and determined the partial gene sequences of six housekeeping genes (atpA, rpoB, pgi, metG, gltA and gyrA). These sequences identified the three genospecies and differentiated them from Actinomyces viscosus isolated from rodents. Th

Incidence, molecular characterisation and epidemiology of Oriental rat flea, Xenopsylla cheopis (Rothschild, 1903) in synanthropic rodent hosts.

BACKGROUND OBJECTIVES: Rodents and arthropod vectors transmit zoonotic pathogens like Yersinia pestis and Rickettsia typhi , posing a threat to both veterinary and public health. The primary aim of this study was to record incidence of rat fleas, a vector for plague and murine typhus on synanthropic rodents, their epidemiology and molecular characterization. METHODS: A comprehensive survey of synanthropic rodents viz. Bandicota bengalensis (190), Rattus rattus (201), and Tatera indica (180) across various habitats, including crop fields, poultry farms, fish markets, residential areas, shops,

Markers of tuberculous infection in 17th-century Lucca (central Italy): An investigation through granular impressions on the endocranial surface.

OBJECTIVE: Granular impressions (GIs) on the endocranial surface have been regarded as evidence of tuberculous meningitis. However, recent studies suggest that these subtle lesions more likely reflect disseminated tuberculosis. This study examines the frequency of GIs in an epidemic skeletal assemblage from Lucca (Italy) dating to the 17th century CE. MATERIALS: Excavations at the convent of St Francesco uncovered a cemetery archaeologically and historically linked to an epidemic outbreak, likely the plague of 1630-31. The assemblage comprises 98 individuals of poor preservation. METHODS: En

Blockchain-enabled strategic optimization: A decision analytics framework for multinational enterprises' e-commerce entry model.

With e-commerce booming, MNEs (Multinational Enterprises) often tap into online markets through E-retailers in multinational e-commerce platforms. Therefore, which e-commerce entry model MNEs choose can be critical, especially when the MNE owning a retail division in the same market. Notably, the MNE can address the plague of low-quality images of e-retailers by introducing blockchain technology, but the effectiveness of blockchain technology for quality verification also varies across different e-commerce entry models. In this paper, we develop a two-tier supply chain model with a manufacturi

Infectious medical waste (IMW) and disease transmission: A critical public health challenge in Africa and the global implications - A systematic review.

BACKGROUND: Infectious Medical Waste (IMW) from both human and animal healthcare sources have the potential to transmit diseases in human populations due to its pathogenic composition. Conversely, a paucity of data regarding cause-and-effect relationships inhibits our understanding of the extent to which disease transmission occurs. In parts of Africa where the management of IMW is not prioritized, the risks to public health, along with its global implications cannot be ignored. METHODOLOGY: A systematic literature search following the PRISMA guidelines was conducted across five electronic da

Rethinking TYK2 P1104A: a flawed evolutionary trade-off in tuberculosis?

The TYK2 P1104A variant has been cast as a textbook example of evolutionary compromise-protecting against autoimmunity whilst increasing susceptibility to tuberculosis (TB). This narrative has been invoked to explain its decline in frequency in European populations since the Bronze Age. However, accumulating evidence suggests that this widely accepted trade-off may be more complex than originally proposed. At the mechanistic level, P1104A reduces TYK2 catalytic activity without abolishing cytokine signalling, owing to compensatory, non-hierarchical JAK activation. Critically, IL-12-dependent i

Protective Effect of Chitosan-saponin-bentonite Nanocomposite against Infertility in Male Rats.

INTRODUCTION: Pollution of the environment and infertility are two current worldwide health problems that plague people everywhere. Hexavalent chromium has been linked to reproductive toxicity, as seen by a decrease in sperm motility, count, and testosterone levels, as well as an increase in sperm abnormalities. The current study assessed the fertility effect of chitosan-saponinbentonite nanocomposite (CSB NC) against potassium dichromate-induced reproductive toxicity in male rats. METHODS: The 49 rats were placed into seven groups of seven rats each. All groups, except for control, were admi

Prevalence and diversity of Aphanomyces astaci in cambarid crayfish of Pennsylvania: where native and introduced hosts meet.

The crayfish plague pathogen Aphanomyces astaci (Oomycota: Saprolegniales) is native to North America but expanded with its crayfish hosts to other regions. In most of its invaded range, A. astaci haplotypes are associated with specific American crayfish, probably due to introduction bottlenecks, but haplotype diversity is higher and clear host-specific associations are lacking in its native range. However, little is known about the infection rate and load of this pathogen in North America. We investigated the distribution, prevalence and genetic variation of A. astaci in Pennsylvania (eastern

Detecting and Salvaging Head Impacts with Decoupling Artifacts from Instrumented Mouthguards.

In response to growing evidence that repetitive head impact exposure and concussions can lead to long-term health consequences, many research studies are attempting to quantify the frequency and severity of head impacts incurred in various sports and occupations. The most popular apparatus for doing so is the instrumented mouthguard (iMG). While these devices hold greater promise of head kinematic accuracy than their helmet-mounted predecessors, data artifacts related to iMG decoupling still plague results. We recreated iMG decoupling artifacts in a laboratory test series using an iMG fit to a

Comparison of Wear Compliance Across Three Accelerometer Protocols in Adolescents.

Introduction. While physical inactivity remains a pervasive public health problem, issues surrounding measuring physical activity plague researchers. Accelerometers are the gold-standard tool to assess physical activity and consumer-grade devices are increasingly used in research. However, accelerometer wear compliance varies by population and is influenced by factors, such as social acceptability, among others. Non-compliance threatens data validity and limits the usefulness regarding health promotion. Therefore, the purpose of this study is to compare the effects of three consumer-grade acce

Machine-learning models for Alzheimer's disease diagnosis using neuroimaging data: survey, reproducibility, and generalizability evaluation.

Clinical diagnosis of Alzheimer's disease (AD) is usually made after symptoms such as short-term memory loss are exhibited, which minimizes the intervention and treatment options. The existing screening techniques cannot distinguish between stable MCI (sMCI) cases (i.e., patients who do not convert to AD for at least three years) and progressive MCI (pMCI) cases (i.e., patients who convert to AD in three years or sooner). Delayed diagnosis of AD also disproportionately affects underrepresented and socioeconomically disadvantaged populations. The significant positive impact of an early diagnosi
