We studied the phosphoinositide (PI) turnover in the pineal gland of spontaneously hypertensive rats (SHR) and compared it to that in Wistar-Kyoto and Wistar rats. Both basal and norepinephrine-stimulated PI turnover were significantly higher in the SHR than in the other strains. Prazosin (1 mumol/L) completely blocked the norepinephrine-induced PI hydrolysis in all groups. However, prazosin failed to lower the enhanced basal PI turnover in SHR. The chronic toxicity and possible carcinogenicity of the sugar replacer isomalt was studied in Wistar rats and Swiss mice. Groups of 50 animals of each sex were fed 0, 2.5, 5 or 10% isomalt in the diet for nearly 2.5 yr (rats) or 2 yr (mice). Since nickel is a potent carcinogen the influence of nickel-containing staples on the frequency of gastrointestinal tumour formation was tested in caecum-resected and carcinogen-treated rats. Tumour formation was elevated after stapling compared with simple laparotomy or vicryl sutures. This demonstrates a potential hazard of stapling techniques. In the vicryl group there was only one suture line carcinoma.

Fordyce's granules were observed in the gingiva of the upper incisor and molar teeth in F344 rats. The data were based on 734 males and 722 females that were used as control and treated animals in 26-week, 65-week, and 2-year studies by the National Toxicology Program. In the present paper, effects of scopolia drugs (scopolamine, anisodine, anisodamine) on experimental gastric mucosal lesion models in rats were investigated. It is well recognized that plasma fluctuations resulting from oral levodopa therapy may cause an unstable clinical response in parkinsonian patients. We have therefore developed a slow-release polymer matrix system that can deliver levodopa continuously for extended periods of time (at least 225 days) after subcutaneous implantation in rats. The chronic use of dichloroacetate (DCA) for diabetes mellitus or hyperlipoproteinemias has been compromised by neurologic and other forms of toxicity. DCA is metabolized to glyoxylate, which is converted to oxalate and, in the presence of adequate thiamine levels, to other metabolites. Radiometric assays revealed that thiamine deficiency in rats to whom hydroxythiamine was administered in variable doses, is concomitant with activation of gluconeogenesis from pyruvate in liver tissue. The most probable mechanism of this effect is the cAMP-dependent activation of key enzymes of intracellular glucose synthesis. This process is facilitated by the diminution of the ratio of free forms of NAD+ and NADPH in the cytoplasm.

The extent to which tea, cocoa and carob (foods rich in polyphenols) influence fecal nitrogen (N) excretion was investigated in rats. The studies took into account the methylxanthine contents of tea and cocoa as well as the poor digestibility of cocoa nitrogen.
