Sodium p-perfluorous nonenoxybenzenesulfonate directly targets on mitochondrial proteins to disrupt metabolic cascade and promote nephrotoxicity.

Chronic kidney disease is largely driven by environmental exposure such as per- and polyfluoroalkyl substances (PFAS). Mitochondrial proteins, whose homeostasis can be affected by PFAS, largely participate in the progression of renal damage. Despite this connection, the direct mitochondria-xenobiotics interaction and linking mechanisms are still lacking. In this study, integrative thermal proteome profiling and limited proteolysis-mass spectrometry was used to identify the direct protein targets of sodium p-perfluorous nonenoxybenzenesulfonate (OBS), a typical emerging PFAS, to pose nephrotoxi…

Mechanism of crosslinking temperature impacting the structure of calcium ion crosslinked sodium alginate/konjac glucomannan films.

Crosslinking temperature has a significant effect on the progress of the crosslinking reaction. This study systematically investigated the effect of crosslinking temperature (15-55 °C) on the structure of calcium ion crosslinked sodium alginate (SA)/konjac glucomannan (KGM) films as well as the packaging-related properties. The results indicated that crosslinking temperature not only influenced the ionic crosslinking between Ca2+ and SA, but also affected the intermolecular interactions between SA and KGM, leading to significantly different film properties. Specifically, at a relatively low cr…

The SGK1/ENaC axis in renal injury during salt-sensitive hypertension: from sodium sensing to inflammation.

Salt-sensitive hypertension (SSHT) is commonly framed as a disorder of renal sodium transport and vascular dysfunction. Yet increasing evidence indicates that immune cells also detect sodium excess and convert it into inflammatory signaling. In this article, we argue that the serum and glucocorticoid-regulated kinase 1 (SGK1)/epithelial sodium channel (ENaC) immune axis provides a useful framework for linking salt exposure to renal injury. In antigen-presenting cells (APCs), high salt activates SGK1, increases ENaC expression and sodium entry, and promotes oxidative stress, isolevuglandin form…

Sodium Butyrate Attenuates Renal Fibrosis by Regulating Glycolysis and Histone H4K12 Lactylation in Diabetic Kidney Disease.

BACKGROUND: The role of histone lactylation in diabetes associated renal fibrosis remains poorly defined. In this study, we investigated the contribution of histone H4 lysine 12 lactylation (H4K12la) to abnormal glycolysis and renal fibrogenesis in diabetic kidney disease (DKD) and evaluated sodium butyrate (NaB) as a potential therapeutic modulator. METHODS: In this study, db/db mice (Bks. Cg-leprdb/leprdb) were employed as an in vivo model of DKD, and high glucose-treated human tubular epithelial cells (HK-2 cells) were used as an in vitro model to investigate whether NaB exerts protective …

Sivelestat sodium alleviates blood-brain barrier dysfunction after traumatic brain injury by reduction of NETs formation.

BACKGROUND: Brain endothelial cells (bECs) dysfunction plays a key role in blood-brain barrier (BBB) disruption after traumatic brain injury (TBI), contributing to a vicious cycle that worsens disease progression. Although neutrophil extracellular traps (NETs) are known to impair the BBB, their mechanisms in TBI remain unclear. Sivelestat sodium, an inhibitor of neutrophil elastase (NE) required for NETs formation, may offer a therapeutic strategy for TBI. METHODS: A controlled cortical impact (CCI) model was used to induce TBI in mice. NETs involvement was confirmed by immunofluorescence and…

Regulation of sodium/calcium homeostasis by BacNav gene therapy rescues cardiac dysfunction in chronic heart failure.

Despite continued progress, therapies to augment contractile function and prevent arrhythmias in patients with heart failure remain limited. Here, we present a two-pronged gene therapy approach that simultaneously augments peak Na+ current and Ca2+ transient amplitude in cardiomyocytes (CMs) to alleviate heart failure pathogenesis. Using in vitro engineered neonatal rat heart tissues, ex vivo adult mouse CMs, and in silico rabbit CMs, we show that expression of prokaryotic Na+ channels (BacNav) dose-dependently enhances Ca2+ transients and contractility of CMs by modulating Na+/Ca2+ exchanger

Rice husk cellulose/sodium alginate/gelatin hydrogels: Sustainable high-performance trapping and immobilization of iodine vapor.

Efficient capture of radioactive iodine vapor during nuclear fuel reprocessing remains a critical challenge in nuclear cycle operations. To address this challenge, a hydrogel adsorbent (designated CSAG) has been synthesized via an aqueous-phase process using cellulose derived from agricultural waste (rice husks), sodium alginate, and gelatin as precursors. The optimized CSAG-50 formulation, with a 5:5 mass ratio, exhibits exceptional iodine adsorption performance, achieving a remarkable capacity of 2485.5 mg/g at 85 °C. Comprehensive characterization reveals that CSAG-50 possesses a distinctiv…

A comparative study of dextran sodium sulfate-induced and spleen-kidney syndrome-based ulcerative colitis models.

OBJECTIVE: Accurate animal models are essential for elucidating the pathogenesis of ulcerative colitis (UC). The widely used dextran sodium sulfate-induced UC (DSS-UC) model inadequately reflects the refractory nature of human UC. This study compared the traditional Chinese medicine (TCM)-based SKYD-UC model with the DSS-UC model to identify a more representative model. METHODS: Network pharmacology was used to analyze the fundamental TCM syndrome of UC and to identify differential targets and pathways between DSS-UC and SKYD-UC. Both UC animal models were subsequently established, and body w…

Postbiotic Lactobacillus reuteri LRE02 Enriched with Sodium Selenite Demonstrates Antidepressant- and Anxiolytic-Like Effects in Mice.

Postbiotics, including inactivated microorganisms and their bioactive components, have emerged as innovative candidates for microbiota-targeted therapies. Among them, Lactobacillus reuteri-derived postbiotics represent a promising and underexplored strategy for modulating host physiology without the risks associated with live bacteria. Selenium enrichment may further enhance these benefits by strengthening antioxidant defenses and regulating inflammation, two central mechanisms to microbiota-gut-brain axis communication. In this study, we investigated the antidepressant- and anxiolytic-like ef

Sodium bicarbonate attenuates myocardial ischemia-reperfusion injury via activation of the splenic cholinergic anti-inflammatory pathway.

OBJECTIVE: The spleen plays a central role in mediating inflammatory responses during myocardial ischemia-reperfusion injury (IRI) via a neuroimmune axis involving vagal signaling and cholinergic modulation. Sodium bicarbonate (NaHCO3) has been implicated in the activation of a splenic cholinergic anti-inflammatory pathway, but its role in myocardial IRI remains incompletely defined. The present study aims to determine whether NaHCO3 administration before reperfusion attenuates myocardial injury by modulating splenic inflammatory signaling. METHODS: C57BL/6 mice underwent 40 minutes of left c…

Vasculoprotective Effects of Sodium-Glucose Co-Transporter Inhibitors in Non-Diabetic Experimental Settings: A Narrative Review.

Sodium-glucose co-transporter (SGLT) inhibitors are a novel class of glucose-lowering drugs with beneficial pleiotropic effects that have been widely investigated in the past decade in several experimental models and patients in the absence of diabetes. There are two types of transporters: the SGLT1 isoform that is distributed across a broad range of tissues, including the cardiovascular system, and the SGLT2 isoform, which is mostly expressed in renal proximal tubular cells. It is known that inflammation and oxidative stress are key contributors to vascular damage and the progression of ather…

Skipjack tuna peptides exert sleep-promoting effects by regulating neurotransmitter metabolism in pentobarbital sodium-induced mice.

It is estimated that sleep disorders affect approximately 35% of the global population, thus becoming the second most prevalent mental health disorder. In this work, the sleep-promoting effect of skipjack tuna peptide GHHAA (S4) was evaluated systematically in pentobarbital sodium (PS)-induced mice, and its mechanism was explored through measuring the content changes of sleep-related neurotransmitters and c-Fos expression levels in mouse brain regions. In comparison with the control group, S4 was able to dose-dependently reduce the sleep latency (6.33 ± 1.15 min) and prolong sleep time (60.33 …

Gut microbiota perturbation and systemic inflammation are associated with salcaprozate sodium (SNAC)-enabled oral semaglutide delivery.

Semaglutide (SEM) is a glucagon-like peptide-1 (GLP-1) receptor agonist formulated for oral delivery with the absorption enhancer salcaprozate sodium (SNAC). Although oral SEM achieves 0.4-1% bioavailability through gastric epithelial uptake, gastrointestinal (GI) adverse events remain a major cause of therapy discontinuation. This study examined the effects of SEM (0.74 mg/kg/day), SNAC (22 mg/kg/day), and combined SEM-SNAC (1:33 w/w) treatments on microbiota and metabolic function, in healthy Sprague Dawley rats over 21 days. Whilst microbial α-diversity remained stable, SNAC significantly a…
