NK cell recognition of hematopoietic cells by SLAM-SAP families.

The signaling lymphocyte activation molecule (SLAM) family of receptors (SFRs) are ubiquitously expressed on immune cells, and they regulate multiple immune events by recruiting SH2 (Src homology 2) domain-containing SAP family adapters, including SAP and its homologs, Ewing's sarcoma-associated transcript 2 (EAT-2) and EAT-2 related transducer (ERT). In human patients with X-linked lymphoproliferative (XLP) disease, which is caused by SAP mutations, SFRs alternatively bind other inhibitory SH2 domain-containing molecules to suppress immune cell activation and development. NK cells express mul

CIP2A Promotes T-Cell Activation and Immune Response to Listeria monocytogenes Infection.

The oncoprotein Cancerous Inhibitor of Protein Phosphatase 2A (CIP2A) is overexpressed in most malignancies and is an obvious candidate target protein for future cancer therapies. However, the physiological importance of CIP2A-mediated PP2A inhibition is largely unknown. As PP2A regulates immune responses, we investigated the role of CIP2A in normal immune system development and during immune response in vivo. We show that CIP2A-deficient mice (CIP2AHOZ) present a normal immune system development and function in unchallenged conditions. However when challenged with Listeria monocytogenes, CIP2

Advances in Campylobacter biology and implications for biotechnological applications.

Campylobacter jejuni is a major foodborne pathogen of animal origin and a leading cause of bacterial gastroenteritis in humans. During the past decade, especially since the publication of the first C. jejuni genome sequence, major advances have been made in understanding the pathobiology and physiology of this organism. It is apparent that C. jejuni utilizes sophisticated mechanisms for effective colonization of the intestinal tracts in various animal species. Although Campylobacter is fragile in the environment and requires fastidious growth conditions, it exhibits great flexibility in the ad

Bronchopulmonary afferent nerves.

Vagal afferent nerves are the primary communication pathways between the bronchopulmonary system and the central nervous system. Input from airway afferent nerves to the CNS is integrated in the brainstem and ultimately leads to sensations and various reflex outputs. Afferent nerves innervating the airways can be classified into various distinct phenotypes. However, there is no single classification scheme that takes all features, including conduction velocity, cell body diameter, ganglionic origin, and stimuli to which they respond (modality) into account. At present, bronchopulmonary afferen

Exercise-driven microbiome changes regulate host immunity.

Exercise reshapes host physiology and immunity, yet the mechanisms linking physical activity to disease protection remain incompletely defined. Emerging evidence indicates that exercise remodels the gut microbiome, producing compositional and metabolic adaptations that can regulate immune function. In this Review, we discuss how exercise-responsive microbiota and microbial metabolites, including short-chain fatty acids, secondary bile acids, and the one-carbon metabolite formate, influence systemic inflammation, antitumor CD8 T-cell immunity, autoimmunity, and metabolic disease. Defining causa

Nutrition Recalibration in Transplant Recipients: A Phase-Based Metabolic Strategy for Optimizing Graft and Patient Outcomes.

Metabolic recovery after solid organ transplantation is dynamic. Traditional nutritional strategies often assume rapid normalization of appetite and metabolic function. However, emerging evidence suggests that transplant recipients undergo a complex metabolic transition. This transition is characterized by insulin resistance, steroid exposure, altered protein turnover, and fluctuating renal function. The concept of "Nutrition Recalibration" incorporates the need for progressive, physiology-aligned nutritional adaptation. This narrative review proposes a mechanistic and clinically actionable fr

Physical Exercise as a Model for Investigation of the Inflammation Response in Mice.

Animal models, particularly murine protocols on physical exercise, are widely used to investigate physiological adaptations and to study, prevent, and treat chronic non-communicable diseases. Controlled treadmill running enables precise manipulation of intensity and duration, overcoming limitations of voluntary wheel running for modeling inflammation. An incremental-speed treadmill test to fatigue disrupts homeostasis and acts as an acute inflammatory stimulus, eliciting local skeletal muscle and systemic immune responses. This chapter presents a standardized physical exercise protocol in mice

Unique Tunicate Traits Possibly Encoded by Horizontally Transferred Genes.

While horizontal gene transfer (HGT) is common among prokaryotes, its prevalence and impact among metazoans are debatable. A clear example of HGT in animals is the cellulose synthase gene in tunicates, invertebrate chordates that possess the unique ability to produce a cellulose-containing tunic, despite sharing a conserved larval body plan with vertebrates. To investigate evolutionary roles of HGT in animals, we surveyed the genome of a tunicate and identified eight groups of candidate genes that are unlikely to have been vertically transferred. Notably, these candidates encode proteins poten

The Mononuclear Phagocyte System: A Dispersed Organ; Introduction and Comments.

Mononuclear phagocytes are widely distributed throughout the body and interact with other cellular systems to preserve homeostasis and health. Recent studies have revealed the remarkable plasticity and adaptability of their cellular recognition and trophic responses beyond phagocytosis and immunity. We invited several early-career investigators to review the nature and interactions of selected Mononuclear Phagocyte populations within established organ systems, apart from lympho-haematopoietic tissues. This special issue indicates that the Mononuclear Phagocyte System (MPS) can be considered on

UCI Sports Nutrition Project: Competition and Training Demands, Physical Characteristics, and Nutritional Strategies for Performance in Bicycle Motorcross Race.

Bicycle motorcross racing is a unique sprint cycling discipline that takes place on a 300- to 450-m outdoor track with an 8-m start hill. A single race is 30-40 s in duration, and athletes can compete in up to seven races per day. Athletes require highly specific physiological attributes to meet the training and competition demands, and successful riders can produce high peak power outputs and demonstrate technical and tactical superiority over their opponents. Nutrition has a key role in optimizing athlete health, training adaptations, and overall athlete performance. This review will focus o

The role of IL-1 family cytokines in diabetic cardiomyopathy.

Diabetic cardiomyopathy (DCM) is the primary cause of heart failure in patients with diabetes and is characterised by contractile dysfunction and left ventricular hypertrophy. The complex pathological and physiological mechanisms underlying DCM have contributed to a limited number of available treatment options. A substantial body of evidence has established that DCM is a low-grade inflammatory cardiovascular disorder, with the interleukin-1 (IL-1) family of cytokines playing crucial roles in initiating inflammatory responses and shaping innate and adaptive immunity. In this review, we aim to

microRNAs and Cardiovascular Remodeling.

Heart failure (HF) is associated with significant morbidity and mortality attributable largely to structural changes in the heart and with associated cardiac dysfunction. Remodeling is defined as alteration of the mass, dimensions, or shape of the heart (termed cardiac or ventricular remodeling) and vessels (vascular remodeling) in response to hemodynamic load and/or cardiovascular injury in association with neurohormonal activation. Remodeling may be described as physiologic or pathologic; alternatively, remodeling may be classified as adaptive or maladaptive. The importance of remodeling as

[Perinatal stress in brain programming and pathogenesis of psychoneurological disorders].

We analyzed data from our studies and other author's works concerning the influence of stress stimuli on the development of neuroendocrine and immune systems. The results of our analysis suggest that "immunological stress" induced by viral or bacterial infection, an unhealthy diet, and the mother's behavior during pregnancy and lactation are significant risk factors for the developing offspring. The brain is the key target for these factors. The continuous effect of stress stimuli during critical periods of brain development induces hyperactivity and increased response of hypothalamic-pituitar

Demonstrating cerebral vascular networks: a comparison of methods for the teaching laboratory.

One challenge of neuroscience educators is to make accessible to students as many aspects of brain structure and function as possible. The anatomy and function of the cerebrovasculature is among many topics of neuroscience that are underrepresented in undergraduate neuroscience education. Recognizing this deficit, we evaluated methods to produce archival tissue specimens of the cerebrovasculature and the "neurovascular unit" for instruction and demonstration in the teaching lab. An additional goal of this project was to identify the costs of each method as well as to determine which method(s)

Effects of aerobic training in patients with mitochondrial myopathies.

We studied the physiologic adaptation of patients with mitochondrial myopathies to aerobic training. Ten patients underwent individually supervised, moderate-intensity aerobic training on a treadmill for 8 weeks. Biochemical and functional measures improved with training. Estimated aerobic capacity increased by 30%. Blood lactate concentrations at rest and after exercise decreased by 30%. Muscle phosphorus magnetic resonance spectroscopy measurements of adenosine diphosphate recovery after exercise improved by more than 60%. Fatigue and tolerance to daily activities also improved. Although the

Managing hypernatremia in fluid deficient elderly.

The institutionalized elderly are at risk for developing fluid volume depletion with progression to hypernatremia. This is particularly common in patients transferred to an acute care setting from a nursing home. A marked reduction in intracellular fluid and the increase in body fat associated with normal aging predispose the elderly to water loss with very little environmental prompting. Conditions contributing to the development of fluid volume deficit include febrile illness, utilization of enteral supplements, gastrointestinal bleeding, use of loop diuretics, renal failure, prolonged vomit

[Neurotic inhibition].

Inhibition is a normal neuro-physiologic regulation phenomenon which antagonizes the dangerous aspects of excitation and which facilitates the adaptation of man to his surroundings. Inhibition becomes neurotic: -- when it fails in its normative-connection with excitation. -- when accompanied by anxiety, -- when the subject cannot control it, -- however without blocking all the psychic functions as the psychotic inhibition does. Accompanying symptoms diversify its ways of expression. Contemporary to trauma in neurasthenia, it proceeds from early trauma in psychasthenia and in hystero-phobic neu
