Taming autoimmunity: Alpha-1 antitrypsin overexpressing mesenchymal stromal cells promote regulatory T cell crosstalk to reverse diabetes.

Mesenchymal stem/stromal cell (MSC) therapy holds promise as a therapeutic option in diabetes treatment. Anti-inflammatory and immunomodulatory activities are enhanced when MSCs are engineered to overexpress alpha-1 antitrypsin (AAT-MSCs). Because a single infusion of AAT-MSCs reversed new-onset diabetes in over 50% of the female non-obese diabetic mice, we used single-cell RNA sequencing, flow cytometry, and functional analyses to characterize how AAT-MSCs modulate CD4+ and CD8+ T cells in pancreatic lymph nodes (PLNs) and islets. AAT-MSC treatment increased T regulatory cells (Tregs) and mor

Treatment of keratinocytes with 4-phenylbutyrate in epidermolysis bullosa: Lessons for therapies in keratin disorders.

BACKGROUND: Missense mutations in keratin 5 and 14 genes cause the severe skin fragility disorder epidermolysis bullosa simplex (EBS) by collapsing of the keratin cytoskeleton into cytoplasmic protein aggregates. Despite intense efforts, no molecular therapies are available, mostly due to the complex phenotype of EBS, comprising cell fragility, diminished adhesion, skin inflammation and itch. METHODS: We extensively characterized KRT5 and KRT14 mutant keratinocytes from patients with severe generalized EBS following exposure to the chemical chaperone 4-phenylbutyrate (4-PBA). FINDINGS: 4-PBA

A new mode of pancreatic islet innervation revealed by live imaging in zebrafish.

Pancreatic islets are innervated by autonomic and sensory nerves that influence their function. Analyzing the innervation process should provide insight into the nerve-endocrine interactions and their roles in development and disease. Here, using in vivo time-lapse imaging and genetic analyses in zebrafish, we determined the events leading to islet innervation. Comparable neural density in the absence of vasculature indicates that it is dispensable for early pancreatic innervation. Neural crest cells are in close contact with endocrine cells early in development. We find these cells give rise

Estradiol is a critical mediator of macrophage-nerve cross talk in peritoneal endometriosis.

Endometriosis occurs in approximately 10% of women and is associated with persistent pelvic pain. It is defined by the presence of endometrial tissue (lesions) outside the uterus, most commonly on the peritoneum. Peripheral neuroinflammation, a process characterized by the infiltration of nerve fibers and macrophages into lesions, plays a pivotal role in endometriosis-associated pain. Our objective was to determine the role of estradiol (E2) in regulating the interaction between macrophages and nerves in peritoneal endometriosis. By using human tissues and a mouse model of endometriosis, we de

A critical assessment of the methodologies to investigate the role of inhibition of apoptosis in rodent hepatocarcinogenesis.

Non-genotoxic carcinogens act by promoting the clonal expansion of preneoplastic cells by directly or indirectly stimulating cell division or inhibiting cell loss in the target organ. The specific mode-of-action (MoA) by which some non-genotoxic carcinogens ultimately cause cancer is not completely understood. To date, there are several proposed MoAs for non-genotoxic carcinogens, and some of these propose inhibition of apoptosis as one of the key events. In general, inhibition of apoptosis is considered a necessary step for cell survival and in theory can occur in combination or in associatio

Paracrine Factors Secreted by MSCs Promote Astrocyte Survival Associated With GFAP Downregulation After Ischemic Stroke via p38 MAPK and JNK.

Astrocytes are critical for ischemic stroke, and understanding their role in mesenchymal stem cell (MSC)-mediated protection against ischemic injury is important. The paracrine capacity of MSCs has been proposed as the principal mechanism contributing to the protection and repair of brain tissue. In the present study, an in vitro oxygen-glucose deprivation (OGD) model was used to mimic ischemic injury. OGD-induced astrocytes were reperfused with MSC-conditioned medium (MSC-CM) or co-cultured with MSCs for 24 h to create an environment abundant in paracrine factors. The results indicated that b

Suppression of experimental autoimmune myasthenia gravis by autologous T regulatory cells.

Adoptive transfer of regulatory T (Treg) cells have been employed effectively for suppression of several animal models for autoimmune diseases. In order to employ Treg cell therapy in patients, it is necessary to generate Treg cells from the patient's own cells (autologous) that would be able to suppress effectively the disease in vivo, upon their reintroduction to the patient. Towards this objective, we report in the present study on a protocol for a successful immune-regulation of experimental autoimmune myasthenia gravis (EAMG) by ex vivo--generated autologous Treg cells. For this protocol

Simvastatin reduces leucocyte- and platelet-endothelial cell interaction in murine antigen-induced arthritis in vivo.

OBJECTIVES: The use of statins in the prevention and treatment of cardiovascular diseases is well established. Their use as anti-inflammatory and immunomodulatory agents in the treatment of rheumatoid arthritis (RA) has also been investigated, with several clinical and experimental studies indicating an anti-inflammatory effect of statins for RA, but other studies showing no effect or even the opposite. The current study was designed to examine the effect of simvastatin in an in vivo murine model of arthritis using intravital microscopy. METHOD: We assigned four groups (n = 7, female C57Bl6 m

Integrins in the wounded and unwounded stratified squamous epithelium of the cornea.

PURPOSE: The authors determined the synthesis, cell surface expression, and localization of integrins in the rat corneal epithelium to detect whether any changes in integrins occur during epithelial migration in response to simple debridement wounding. METHODS: Immunoprecipitation analysis of extracts from either metabolically or surface-labeled rat epithelia was done to assess the synthesis and cell surface expression of integrins in the normal cornea. The localization of integrins was determined by indirect immunofluorescence of frozen sections obtained from control corneas and from those a

Interplay between the RBP SYNCRIP and RNA methylation in determining sEV miRNA-cargo and function in cell-to-cell communication.

Small extracellular vesicles (sEVs) are critical mediators of tumor microenvironment communication, largely through the selective transfer of microRNAs (miRNAs) that reprogram recipient cells. Active miRNA sorting into sEVs depends on RNA‑binding proteins (RBPs), sequence determinants, and RNA modifications. Here, a functional interplay between the RBP SYNCRIP and N6‑methyladenosine (m6A) RNA methylation controlling miRNA loading into hepatocellular carcinoma (HCC)‑derived sEVs has been disclosed. It is reported that (i) METTL3 (Methyltransferase-like-3)‑dependent m6A modification is required

Interruption of cross-communication pathways alters the immune cell signature of pancreatic cancer and decreases tumor growth.

BACKGROUND: Pancreatic ductal adenocarcinoma (PDAC) is an aggressive malignancy with limited therapeutic options in part due to a dense stroma and the ability of the tumor cells to defy therapy by creating an immunosuppressive milieu. A histologic feature of PDAC is the abundance of immunosuppressive M2-polarized tumor-associated macrophages (TAMs) that favor cancer growth and metastases. Cancer cells communicate with immune cells of the tumor microenvironment (TME) by several mechanisms including exosomes to induce plasticity of the immune cells favoring pro-cancer instead of anti-cancer phen

IL-1β/CXCL12 signalling orchestrates adipocyte-pancreatic neuroendocrine tumor crosstalk.

BACKGROUND: Pancreatic neuroendocrine tumors (PanNETs) are rare neoplasms belonging to the broader group of neuroendocrine neoplasms. Epidemiological evidence indicates that visceral obesity is associated with both an increased incidence of PanNETs and unfavorable clinicopathological features. However, the mechanisms underlying adipocyte-PanNETs interactions remain poorly understood. METHODS: Here, we investigated the crosstalk between adipocytes and PanNET cells (BON-1 and QGP-1 cell lines) using indirect co-culture systems and conditioned media derived from differentiated 3T3-L1 adipocytes

Lactate attenuates hepatic ischemia-reperfusion injury by reducing intrahepatic inflammation and suppressing hepatocyte-neutrophil interaction via the IL-33/ST2 pathway.

BACKGROUND: Hepatic ischemia-reperfusion injury (HIRI) is a critical contributor to adverse clinical outcomes and increased mortality after liver surgery or transplantation. Lactate, formerly regarded as a metabolic waste product, has emerged as a critical immunomodulatory molecule. This study investigated how lactate modulates HIRI by regulating intrahepatic inflammation and intercellular communication. METHODS: A mouse model of HIRI was constructed, along with in vitro hypoxia/reoxygenation (H/R) of primary hepatocytes and LPS-stimulated bone marrow-derived neutrophils, to evaluate the prote

Charting spatial ligand-target activity using Renoir.

The advancement of single-cell RNA sequencing and spatial transcriptomics has enabled the inference of cellular interactions in a tissue microenvironment. Despite advances in cell-cell interaction inference, methods capable of mapping the influence of ligands on downstream target genes across spatial niches harboring specific cell type composition, crucial for resolving niche-specific relationship between ligands and their downstream targets are still lacking. Here, we present Renoir for charting the ligand-target activities across a spatial topology, delineating spatial communication niches h

Lactate-Activated GPR132 Signaling Drives a Tumor Microenvironmental Autocrine Metabolic Loop in Kidney Cancer.

UNLABELLED: Despite the presence of oxygen, tumors frequently preferentially perform fermentative glycolysis, producing lactate and acidifying the tumor microenvironment (TME). Although studies have observed that high concentrations of lactate in the TME help tumors gain a proliferative advantage, a detailed understanding of the molecular mechanisms is needed to uncover strategies to overcome lactate-mediated growth. In this study, we investigated how lactate exerts progrowth effects in clear cell renal cell carcinoma (ccRCC), a highly glycolytic tumor primarily caused by alterations in the vo

Deciphering the pulmonary fibrosis niche: A single-cell transcriptomic perspective on cellular crosstalk and microenvironment remodeling.

Idiopathic pulmonary fibrosis (IPF) is a progressive, fatal interstitial lung disease driven by aberrant repair after repetitive alveolar micro-injury. The classical paradigm - that fibrosis is a passive consequence of epithelial injury followed by fibroblast activation - has been substantially reshaped by single-cell RNA sequencing (scRNA-seq) and, more recently, spatial transcriptomics. These technologies have resolved the lung into a constellation of molecularly distinct cell states and exposed a highly structured fibrotic niche in which epithelial, mesenchymal, immune, and endothelial comp

A lymphatic organ-on-a-chip reveals flow-responsive LEC paracrine signals promoting BMSC osteogenesis in jawbone regeneration.

The lymphatic system is essential for fluid homeostasis and immune surveillance, yet its role in skeletal regeneration remains poorly defined. Here, we introduce a biomimetic organ-on-a-chip (OoC) model that recapitulates lymphatic endothelial cell (LEC) behavior under precisely controlled flow-mediated environments and elucidates their regulatory contribution in alveolar bone repair. Under flow-mediated shear stress, LECs exhibited enhanced sprouting, forming lumenized lymphatic structures in vitro. Furthermore, conditioned medium from perfused LECs significantly promoted the proliferation an

Beyond Histamine: Mast Cell-Neuron Communication in Chronic Pruritic and Pain Disorders.

Chronic pruritic and cutaneous pain disorders are increasingly recognized as neuroimmune conditions driven by dynamic interactions between immune and sensory systems. While histamine has historically dominated models of mast cell-mediated itch, accumulating evidence indicates that non-histaminergic pathways are central to chronic disease. In this review, we synthesize current understanding of mast cell-neuron communication and propose a unifying framework in which these cells form functional neuroimmune units that sustain itch and pain through bidirectional signalling and feed-forward amplific
