Is there a role for apolipoprotein E in perinatal brain injury?

Perinatal brain injury remains a major cause of neonatal morbidity and lifelong neurological disability. Despite advances in neonatal care, the molecular determinants that modulate vulnerability and recovery in the immature brain remain poorly defined. Genetic variation is increasingly recognized as a contributor to the heterogeneous susceptibility observed among affected infants. Among the genes implicated in neurodevelopmental outcomes, apolipoprotein E (ApoE) has emerged as a key regulator of lipid metabolism, neuroinflammatory signaling, and neuronal repair in the central nervous system.Th…

Connectomic Analysis of Mitochondria in the Central Brain of Drosophila.

Mitochondria are integral to the metabolism and cell biology of a neuron. Electron microscopy images of fly brain volumes, taken for connectomics, can be analyzed for mitochondria as well as the cells and synapses already reported. Here, from the Drosophila Hemibrain connectome dataset, we extract, classify, and measure approximately 6 million mitochondria, with the majority located among more than 20 thousand neurons and over 5500 cell types. Each mitochondrion is annotated with its location, orientation, voxel size, and appearance (dark and dense, light and sparse, or medium), and each synap…

A decade of comparing brains to DNNs: Progress and perspectives.

Over the past decade, comparisons between deep neural networks (DNNs) and the human brain have become central to cognitive neuroscience. Early work focused on vision, driven by the success of convolutional neural networks in object recognition, before such comparisons later gained traction in language with the rise of large-scale language models. These comparisons have validated existing hypotheses and generated new ones, challenging views of information processing, connectivity, and computational goals. Despite progress, debates persist over the interpretability and validity of mapping DNNs t…

Overcoming Perfusion Impairment in the Ischemic Brain: A Review.

Perfusion impairment is one of the major barriers to using machine perfusion to prepare brain tissue for research and clinical applications. As ischemia progresses, multiple mechanisms, including intravascular obstructions, perivascular cellular edema, mural cell contraction, and vessel wall breakdown, progressively limit the uniform delivery of preservative solutions. This problem is particularly important for connectomics, wherein successful preservation requires both widespread distribution of preservative chemicals and the maintenance of the cellular ultrastructure needed for circuit recon…

Skull channels as gateways for immune surveillance of the brain.

The central nervous system (CNS), once considered immune privileged, is, in fact, protected by a dynamic immune compartment located in the meninges. These immune cells are crucial for brain protection, controlling neuroinflammation, and also supporting neural function. Recent studies show that meningeal immune cells, including myeloid and B cells, partly originate from the skull bone marrow, which communicates directly with the meninges via specialized skull channels identified in mice and humans. These channels allow immune cell trafficking and cerebrospinal fluid (CSF) exchange, positioning …

Mild chronic hypoxia and the brain: an ambiguous relationship.

Mild hypoxia is a common condition encountered in various situations, such as high-altitude living, respiratory diseases, anaemia, and certain cardiovascular disorders. At the same time, mild hypoxia often goes unnoticed and can lead to long-term brain impairments. Understanding the subtle effects of mild hypoxia on the brain is essential for early diagnosis and intervention to prevent serious neurological and psychiatric complications. The pathophysiology of mild chronic hypoxia is complex; nevertheless, based on the literature, two main pathways can be distinguished. The first involves the d…

The foundational role of autonomy for brain-computer interfaces.

Invasive brain-computer interfaces (BCIs) pose a challenge for describing human-system interactions because they involve a depth of engagement that is difficult to disentangle. This is particularly problematic when clear authorship of decisions is needed. This presupposes that such devices might also decide autonomously in certain situations. Although the use and implementation of such devices are highly disputed in practical ethics, they seem to offer at least therapeutic benefits for persons with, e.g., Parkinson's disease. That is why it is indispensable to develop a conceptual framework th…

On the Increased and Decreased Connectivity of the Demented Human Brain.

With major advances in cerebral imaging techniques, a large amount of data is available for studying the aging and demented brain. In this contribution, we apply the OASIS-3 dataset to identify small areas of human gray matter with higher or lower structural connectivity in dementia. As was anticipated, we have found that finer structures of the hippocampus and the temporal lobe have decreased connectivity in dementia. More surprisingly, the precuneus, the cuneus, and some finer structures of the insula, the paracentral lobule, and the precentral and paracentral gyri showed higher connectivity…

Impact of meningioma and glioma on whole-brain dynamics.

Brain tumors, particularly meningiomas and gliomas, can profoundly affect neural function, yet their impact on brain dynamics remains incompletely understood. This study investigates alterations in normal brain function among meningioma and glioma patients by assessing dynamical complexity through the Intrinsic Ignition Framework. We analyzed resting-state fMRI data from 34 participants to quantify brain dynamics using intrinsic ignition and metastability metrics. Our results revealed distinct patterns of disruption: glioma patients showed significant reductions in both metrics compared to con…

[Intraoperative radiotherapy for brain metastases: an experience in Argentina].

Postoperative stereotactic radiosurgery or hypofractionated radiotherapy represent the standard local adjuvant treatment modality after surgical resection of single brain metastases. Intraoperative radiotherapy (IORT) offers the advantage of delivering an ablative dose during the same surgical procedure, avoiding delays and allowing early reinitiation of systemic therapies. This work describes our first national experience with IORT, with preliminary results from a case series. The series included three male patients diagnosed with a single supratentorial brain metastasis (two right frontal an…

Optogenetics for therapeutic use in the brain, eye, and ear.

Optogenetics is the process of photosensitizing cells and thereby controlling their activity with light. The discovery of channelrhodopsins … have been envisioned as potential treatments that can target and reverse pathologic pathways in the brain. Thus, in the recent years, there is a great interest in therapeutic optogenetics, centered mainly around restoration of the major senses of vision and hearing. This chapter will first highlight the ongoing evolution of optogenetics from an investig …

Neuroimaging in Hypertension: A Multifaceted Approach to Investigate the Brain.

The brain is the most metabolically demanding organ, whose integrity and homeostasis are highly sensitive to changes in blood perfusion. Cerebrovascular alterations are therefore major drivers in the development of neurodegenerative diseases and cognitive impairment. Among cardiovascular conditions, hypertension is associated with a wide spectrum of structural and functional brain changes, being a major risk factor for dementia. The multitude of cerebrovascular alterations and consequent injuries linked to hypertension covers distinct, yet interdependent, brain aspects, from modifications in v…

Characterization of a distinct form of vimentin in the neurodegenerative brain.

Alzheimer's disease (AD) and Parkinson's disease (PD) are characterized by brain accumulation of aggregated proteins, leading to neuronal and glial dysfunction. Compelling data indicate that changes in vimentin expression, structure, and localization reflect alterations in cellular homeostasis and may correlate with disease progression. Yet, the involvement of abnormal vimentin in AD and PD remains unclear. Here, we have thoroughly characterized the distribution of modified, disease-associated vimentin in the AD and PD brain parenchyma, as well as in cerebrospinal fluid (CSF), hiPSC-derived as…

Translocation of bacteria from the gut to the brain in mice.

Recent advances suggest a correlation between gut dysbiosis and neurological diseases, however, relatively little is known about how gut bacteria impact the brain. Here, we reveal that bacteria can translocate directly from the gut to the brain in small numbers when mice are fed an atherogenic, high-fat diet (Paigen diet) that causes alterations in gut microbiome composition and gut barrier permeability. The bacteria were not found in other systemic sites or the blood, but were detected in the vagus nerve. Right cervical vagotomy reduced bacterial burden in the brain, implicating the vagus ner…

Traumatic Brain Injury and the Road to Alzheimer's Disease.

Alzheimer's disease (AD) is associated with both mild and moderate to severe traumatic brain injury (TBI). This narrative review gives an account of the association of TBI and AD and highlights possible cellular and molecular pathways linking these pathologies. Following TBI, the immune system of the brain is rapidly activated and gives rise to acute neuroinflammation. While neuroinflammation is protective in nature, it may persist chronically in the case of less-controlled prolonged responses, triggering neuroprotective loss and neurotoxicity. Moreover, reduced clearance of amyloid-beta may o…

[Gene and Cell Technologies for Treatment of Brain Malignancies].

Brain malignancies, such as glioblastoma, remain one of the most aggressive and difficult-to-treat cancers because of their heterogeneity, invasive growth, and resistance to conventional therapy. Significant recent progress in the treatment of these diseases has been associated with the development of gene and cell technologies, which open up new possibilities for targeted therapy, immunotherapy, and personalized approaches. The review systematically summarizes the data on current treatment strategies, including gene therapy, cell therapy, and combined approaches. Particular attention is paid …

[The pathology of the brain eating amoeba Naegleria fowleri].

The genus Naegleria is a taxonomic subfamily consisting of 47 free-living amoebae. The genus can be found in warm aqueous or soil habitats worldwide. The species Naegleria fowleri is probably the best-known species of this genus. As a facultative parasite, the protist is not dependent on hosts to complete its life cycle. However, it can infect humans by entering the nose during water contact, such as swimming, and travel along the olfactory nerve to the brain. There it causes a purulent meningitis (primary amoebic meningoencephalitis or PAME). Symptoms are severe and death usually occurs withi…

Transcranial Photobiomodulation for Neuromodulation of Brain Disorders: A Perspective.

INTRODUCTION: Photobiomodulation (PBM) uses light to modulate biological activity. Because red and near-infrared wavelengths penetrate the skin and skull, PBM can be applied transcranially (tPBM) to noninvasively modulate brain activity, offering therapeutic potential for neurologic, psychiatric, neurodevelopmental, neurodegenerative, and neuroimmunologic disorders, and for cognitive enhancement. MATERIALS AND METHODS: A scoping review is performed integrating literature on tPBM in humans, excluding indications for which only preclinical data exist. RESULTS: tPBM can be delivered through the…
