Predicting brain tumour growth patterns using a novel MRI-based tumour spread map: application to radiotherapy planning.

BACKGROUND: The treatment of glioblastomas (GBM) with radiation therapy is extremely challenging due to their invasive nature and high recurrence rate within normal brain tissue. PURPOSE: In this work, we present a new metric called the tumour spread (TS) map, which utilizes diffusion tensor imaging (DTI) to predict the probable direction of tumour cells spread along fiber tracts. We hypothesized that the TS map could serve as a predictive tool for identifying patterns of likely recurrence in patients with GBM and, therefore, be used to modify the delivery of radiation treatment to pre-emptiv

Amplicon-based DNA and RNA unified NGS for enhanced fusion variant detection in suboptimal real-world NSCLC FFPE specimens.

BACKGROUND: Timely identification of actionable fusion and exon-skipping events is essential for selecting targeted therapies in non-small cell lung cancer (NSCLC), yet routine RNA-based assays frequently fail in formalin-fixed paraffin-embedded (FFPE) specimens, leading to missed treatment opportunities. Amplicon-based DNA and RNA unified next-generation sequencing (D+R NGS) may enable guideline-recommended profiling from limited or degraded tissue, but its performance in real-world specimens is unclear. METHODS: The D+R NGS assay uses 20 ng of co-extracted RNA and DNA from FFPE tissue. Libr

Regeneration of the pulp-dentin complex in mature teeth using autologous demineralized dentin matrix and dental pulp stem cells: two case reports.

BACKGROUND: Regenerating the true pulp-dentin complex in mature teeth with necrotic pulps or apical lesions remains challenging, and consistent clinical evidence of dentin bridge formation is limited. This report describes two adult cases treated with regenerative endodontic therapy (RET) using autologous dental pulp stem cells (DPSCs) and demineralized dentin matrix (DDM). CASE PRESENTATION: In Case 1, a 33-year-old woman developed pulp necrosis with apical periodontitis in a maxillary left central incisor following traumatic subluxation. After canal disinfection, autologous DPSCs isolated f

Isolated tumor cells in the bone marrow (ITC-BM) of breast cancer patients before and after anthracyclin based therapy: influenced by the HER2- and Topoisomerase IIalpha-status of the primary tumor?

PURPOSE: The presence of isolated tumor cells in the bone marrow (ITC-BM) is an independent prognostic factor in all stages of breast cancer. Both the expression/amplification of human epithelial growth factor receptor 2 (HER2) and Topoisomerase IIalpha (TOP IIa), a key enzyme of DNA replication and main target of anthracyclins, in breast cancer tissue seem to have predictive value regarding the effectiveness of systemic therapies. METHODS: To investigate the correlation between these factors and their influence on clinical outcome, tumor tissue of 54 patients who were screened for ITC-BM bef

Intranasal administration of Cytoglobin modifies human umbilical cord‑derived mesenchymal stem cells and improves hypoxic‑ischemia brain damage in neonatal rats by modulating p38 MAPK signaling‑mediated apoptosis.

Neonatal hypoxic‑ischemic brain damage (HIBD) is a common clinical syndrome in newborns. Hypothermia is the only approved therapy for the clinical treatment; however, the therapeutic window of hypothermia is confined to 6 h after birth and even then, >40% of the infants either die or survive with various impairments, including cerebral palsy, seizure disorder and intellectual disability following hypothermic treatment. The aim of the present study was to determine whether nasal transplantation of Cytoglobin (CYGB) genetically modified human umbilical cord‑derived mesenchymal stem cells (CYGB‑H

The RELA-FTH1 axis in senescent cancer-associated fibroblasts creates a stromal iron sink that drives ferroptosis resistance and therapy failure across solid malignant tumors.

BACKGROUND: Therapy resistance remains the foremost barrier to improving outcomes in solid malignancies. While tumor-cell-intrinsic resistance mechanisms have been extensively studied, the contribution of the tumor microenvironment (TME) to treatment failure is poorly understood. Cellular senescence accumulates across diverse solid tumor types and reshapes the stromal landscape, yet which population dominates this senescent compartment and how senescent cancer-associated fibroblasts (senCAFs) drive resistance remain unresolved. In particular, whether senescent fibroblasts can suppress ferropto

Identification of novel therapeutic targets for chronic kidney disease and kidney function by integrating multi-omics proteome with transcriptome.

BACKGROUND: Chronic kidney disease (CKD) is a progressive disease for which there is no effective cure. We aimed to identify potential drug targets for CKD and kidney function by integrating plasma proteome and transcriptome. METHODS: We designed a comprehensive analysis pipeline involving two-sample Mendelian randomization (MR) (for proteins), summary-based MR (SMR) (for mRNA), and colocalization (for coding genes) to identify potential multi-omics biomarkers for CKD and combined the protein-protein interaction, Gene Ontology (GO), and single-cell annotation to explore the potential biologic

Spatial single-cell multi-omics characterization of the tumor microenvironment heterogeneity by HER-2 expression status in gastric cancer.

BACKGROUND: The success of novel antibody-drug conjugates, such as trastuzumab deruxtecan (T-DXd) and disitamab vedotin (RC48), has been pivotal in rendering “HER-2-low” gastric cancer (GC) a therapeutically targetable entity, expanding the population benefiting from anti-HER2 therapy by 2-3-fold. This advancement underscores the imperative to redefine and subclassify HER-2 status in GC. In this study, we innovatively proposed a four-tiered HER-2 classification standard for GC and, for the first time, conducted comparative analyses at the spatial single-cell multi-omics level across these four

Elucidating the immune infiltration in acne and its comparison with rosacea by integrated bioinformatics analysis.

BACKGROUND: Acne vulgaris and rosacea are common inflammatory complications of the skin, both characterized by abnormal infiltration of immune cells. The two diseases can be differentiated based on characteristic profile of the immune cell infiltrates at the periphery of disease lesions. In addition, dysregulated infiltration of immune cells not only occur in the acne lesions but also in non-lesional areas of patients with the disease, thus characterizing the immune infiltration in these sites can further enhance our understanding on the pathogenesis of acne. METHODS: Five microarray data-set

Lesion-specific features of macrophage polarization contribute to Mycobacterium tuberculosis infection in the lungs of patients with pulmonary tuberculosis.

BACKGROUND: The innate immunity, linked to macrophage polarization, plays a critical role in the progression of tuberculosis (TB) disease caused by M. tuberculosis (Mtb) infection. Within a single patient with active pulmonary TB, Mtb infection leads to the formation of extensive lung pathology with a spectrum of morphologically and physiologically distinct TB lesions at different stages of progression, where macrophages are believed to represent varying functional phenotypes. Understanding the dynamics of macrophage polarization with identification of the cytokine network, molecular pathways,

Ex Vivo Evaluation of the Sepsis Triple Therapy High-Dose Vitamin C in Combination with Vitamin B1 and Hydrocortisone in a Human Peripheral Blood Mononuclear Cells (PBMCs) Model.

Sepsis is an extremely complex clinical syndrome, usually involving an excessive inflammatory response including an overshooting cytokine release that damages tissue and organs of the patient. Due to the severity of this condition, it is estimated that over 11 million people die from sepsis each year. Despite intensive research in the field, there is still no specific therapy for sepsis. Many sepsis patients show a marked deficiency of vitamin C. 9 out of 10 sepsis patients have a hypovitaminosis C, and every third patient even shows a clinical deficiency in the scurvy range. In addition, low

Influence of photobiomodulation therapy on the treatment of pulmonary inflammatory conditions and its impact on COVID-19.

We are currently facing a pandemic that continuously causes high death rates and has negative economic and psychosocial impacts. Therefore, this period requires a quick search for viable procedures that can allow us to use safe and non-invasive clinical tools as prophylactic or even adjuvant methods in the treatment of COVID-19. Some evidence shows that photobiomodulation therapy (PBMT) can attenuate the inflammatory response and reduce respiratory disorders similar to acute lung injury (ALI), complications associated with infections, such as the one caused by the new Coronavirus (SARS-CoV-2).

Role of Direct Oral Anticoagulation Agents as Thromboprophylaxis in Antiphospholipid Syndrome.

Antiphospholipid syndrome (APS) is an autoimmune disorder that causes venous, arterial and small-vessel thrombosis, pregnancy loss, and premature birth. Cardiac valvular disease, renal thrombotic microangiopathy, thrombocytopenia, hemolytic anemia, and cognitive impairment are some of its other clinical symptoms. Antiphospholipid antibodies cause endothelial cells, monocytes, and platelets to become activated, as well as an increase in tissue factor and thromboxane A2. Complement activation might play a key function in pathogenesis. Long-term oral anticoagulation is used to treat thrombosis, a

[Long-term survival of a breast cancer patient with carcinomatous pleuritis and carcinomatous cardiac tamponade successfully treated by multimodality therapy].

A 69-year old woman was admitted to our hospital because of dyspnea and pain in her left breast. Computed tomography revealed a massive quantity of left pleural effusion, a tumor in the left breast(5 cm in diameter), left cervical and supraclavicular lymph node metastasis, and a large left axillary metastatic mass. Based on a core needle biopsy, her breast tumor was diagnosed pathologically as scirrhous carcinoma, which was positive for estrogen receptor/progesterone receptor and negative for HER2 using the FISH assay, and left pleural metastasis was diagnosed cytologically. The carcinomatous

Hypoxia and Microvascular Alterations Are Early Predictors of IDH-Mutated Anaplastic Glioma Recurrence.

Anaplastic gliomas (AG) represents aggressive brain tumors that often affect young adults. Although isocitrate-dehydrogenase (IDH) gene mutation has been identified as a more favorable prognostic factor, most IDH-mutated AG patients are confronted with tumor recurrence. Hence, increased knowledge about pathophysiological precursors of AG recurrence is urgently needed in order to develop precise diagnostic monitoring and tailored therapeutic approaches. In this study, 142 physiological magnetic resonance imaging (phyMRI) follow-up examinations in 60 AG patients after standard therapy were evalu

Biodistribution of the Bombesin Receptor-Targeted Radiopharmaceutical Precursor BBN/C1-C2 in a Prostate Cancer Model.

UNLABELLED: Gastrin-releasing peptide receptor (GRPR) is a G protein-coupled receptor expressed in the central nervous system, the gastrointestinal tract, the pancreas, and the adrenal cortical tissue regulating their physiological functions. In addition to normal tissues, GRPR is overexpressed in many solid cancers. At present, several radiolabeled ligands targeting GRPR have been introduced into clinical practice for cancer diagnostics and radioligand therapy. However, there were found the high uptake of radiopharmaceuticals in normal organs and low bioavailability of the drugs caused by the

Application of a new coaxial bipolar electrode for the treatment of vertebral metastases: a pilot study in an ovine model.

INTRODUCTION: Spinal metastases account for approximately 90% of masses detected through spinal imaging. Therefore, it is imperative to advance therapies. Electroporation modifies the permeability of cancer cell membranes using electric energy, increasing the local uptake of chemotherapeutics and promoting local tumor control. The aim of this study was to evaluate the safety of delivering electrical pulses that induce tissue ablation using novel coaxial bipolar electrodes in healthy bone and clinically relevant structures in an ovine model. METHODS: Electroporation was performed on sheep vert
