Hepatitis C virus and ethanol alter antigen presentation in liver cells.

Alcoholic patients have a high incidence of hepatitis C virus (HCV) infection. Alcohol consumption enhances the severity of the HCV disease course and worsens the outcome of chronic hepatitis C. The accumulation of virally infected cells in the liver is related to the HCV-induced inability of the immune system to recognize infected cells and to develop the immune responses. This review covers the effects of HCV proteins and ethanol on major histocompatibility complex (MHC) class I- and class II-restricted antigen presentation. Here, we discuss the liver which functions as an immune privilege o

Immunopathogenesis of Behçet's disease with special emphasize on the possible role of antigen presenting cells.

Behçet's disease (BD) is a systemic vasculitis with unclear etiology and pathogenesis. Although several viral and bacterial causes have been investigated in detail for several years, of late it is widely accepted that microorganisms may play a role as a trigger, or as a cross-reactive antigens that interfere with self-antigens, such as heat-shock proteins. Genes such as HLA-B51, MICA and TNF, considered to play a crucial role in the pathogenesis of BD, are located in the major histocompatibility complex (MHC). However, it has been accepted that, only HLA-B51 is directly related with pathogenes

Experimental autoimmune uveitis in HLA-B27 transgenic mice.

The major histocompatibility complex (MHC) gene, HLA-B27 is strongly associated with auto-immune uveitis and spondyloarthropathies in humans. Experimental mouse models of autoimmune uveitis involve systemic immunization with the retinal autoantigen interphotoreceptor retinoid binding protein (IRBP). To assess possible roles of HLA-B27 in autoimmune uveitis, as well as to investigate a possible new animal model of human uveitis, inbred strains of C57BL/6 and C57BL/6 possessing the human HLA-B27 or HLA-A2 transgene were immunized with IRBP emulsified in complete Freund's adjuvant (CFA). Dilated

Inflammation biomarkers in cancer: mechanistic drivers, diagnostic innovations, and future clinical horizons.

Chronic inflammation is a universally acknowledged characteristic of cancer, playing the role of a vital catalyst in cancer initiation and progression, and a chief predictor of the success or failure of cancer therapies. About 20% of all cancer cases are associated with states of chronic inflammation, and an even greater percentage of cancer cases exploit states of smoldering inflammatory microenvironments for cancer progression, immune escape, angiogenesis, and metastasis. In accordance with the Preferred Reporting Items for Systematic Reviews and Meta-Analyses extension for Scoping Reviews (

Oral anaerobe bacteria-a common risk for cardiovascular disease and mortality and some forms of cancer?

This review explores the results of research on oral health concerning cardiovascular diseases and some forms of cancer and is based on results from published systematic reviews and some studies. The research results will have a strong focus on exploring the relationship between different aspects of oral infections. The relationship between oral health parameters, cardiovascular diseases (CVD), and certain cancers was examined from different angles, including prospective analyses, in a population-based health study in Oslo from the year 2000 (Oslo II study). A major finding was that low levels

scDrugPrio: a framework for the analysis of single-cell transcriptomics to address multiple problems in precision medicine in immune-mediated inflammatory diseases.

BACKGROUND: Ineffective drug treatment is a major problem for many patients with immune-mediated inflammatory diseases (IMIDs). Important reasons are the lack of systematic solutions for drug prioritisation and repurposing based on characterisation of the complex and heterogeneous cellular and molecular changes in IMIDs. METHODS: Here, we propose a computational framework, scDrugPrio, which constructs network models of inflammatory disease based on single-cell RNA sequencing (scRNA-seq) data. scDrugPrio constructs detailed network models of inflammatory diseases that integrate information on

Autoimmune hemolytic anemia: current knowledge and perspectives.

Autoimmune hemolytic anemia (AIHA) is an acquired, heterogeneous group of diseases which includes warm AIHA, cold agglutinin disease (CAD), mixed AIHA, paroxysmal cold hemoglobinuria and atypical AIHA. Currently CAD is defined as a chronic, clonal lymphoproliferative disorder, while the presence of cold agglutinins underlying other diseases is known as cold agglutinin syndrome. AIHA is mediated by autoantibodies directed against red blood cells (RBCs) causing premature erythrocyte destruction. The pathogenesis of AIHA is complex and still not fully understood. Recent studies indicate the invol

Models of mycoplasma respiratory and genital tract infections.

Animal models for the study of human diseases must be replaced by in vitro methods, whenever possible. However, when critically evaluated, they remain indispensable for the solution of some specific problems in infectious diseases. These include the pathogenesis, the host response, as well as the study of antimicrobial agents and vaccines. Under these conditions animal models, which closely reflect the situation in man, are especially valuable. Two models may serve as "ideal" examples for the study of human diseases: firstly mouse typhoid for the study of systemic infections with Salmonella ty

Alveolar Macrophages: Development, Respiratory Homeostasis, Signal Pathways, Diseases, and Therapeutic Opportunities.

Alveolar macrophages (AMs), as primary immune sentinels in the respiratory tract, are uniquely adapted to tissue-specific demands shaped by local factors and the physiological environment, maintaining airway homeostasis through the clearance of surfactants and particulates. While AMs exhibit remarkable plasticity shaped by dynamic microenvironmental cues, the traditional M1/M2 paradigm fails to adequately capture this complex heterogeneity, and the specific role of AMs in the pathogenesis of pulmonary diseases, as well as AM-centric therapies, remain to be fully elucidated. This review systema

Pharmacogenetics of Mycophenolic Acid in Patients of African Descent: Underrepresentation of African Data.

Mycophenolic acid (MPA) is a potent antiproliferative immunosuppressive agent used to prevent organ transplant rejection and to treat various immune-mediated diseases. MPA is the active metabolite formed from the biotransformation of the prodrugs mycophenolate mofetil (MMF) or enteric-coated mycophenolate sodium (EC-MPS). MPA exerts its therapeutic effects by inhibiting guanosine nucleotide synthesis in lymphocytes. Through this inhibition, cell and humoral immunity is suppressed, resulting in a reduction of cytotoxicity and inflammation. Systemic exposure to MPA is influenced by a complex pha

Tubulointerstitial inflammation and fibrosis induced by proteinuria: fresh insights.

Proteinuria is a well-established risk factor for chronic kidney disease progression, and its reduction is associated with improved kidney outcomes. During proteinuric chronic kidney diseases, the abnormal passage of plasma-derived proteins through the damaged glomerular filtration barrier can lead to the activation of various pathophysiological processes in the tubular compartment. These mainly comprise complement activation and excessive proximal tubule reuptake of filtered proteins, such as albumin and associated lipids, which trigger cytotoxic effects through the dysregulation of multiple

The multifaceted role of AIF-1 in metabolic dysregulation: bridging inflammation, insulin resistance, and obesity.

Allograft inflammatory factor-1 (AIF-1), a cytokine secreted by activated monocytes, macrophages, and lymphocytes, has emerged as a critical regulator of pathological processes spanning renal diseases, rheumatoid arthritis, cancer, cardiovascular disorders, neurological pathologies, and transplant-related conditions. Population-based studies have associated sequence variants near the AIF-1 locus with obesity, though AIF-1's potential pathophysiological involvement remains uninvestigated. Understanding its molecular characteristics, receptor interactions, and signaling pathways is essential for

The RNA-Binding Protein PARN Remodeled 3' UTR Structure Defines Poly(A)-Loading Sites to Mediate Immunoglobulin Homeostasis.

Class Switch Recombination (CSR) is essential for generating high-affinity antibody isotypes from IgM during adaptive humoral responses. Despite well-established roles for various transcription factors, whether CSR is subject to dedicated post-transcriptional control represents a significant gap in knowledge. By integrating conditional knockout models with SLE disease contexts, this study is the first to identify that the RNA-binding protein, poly(A)-specific ribonuclease (PARN), serves as a key positive regulator of antibody-secreting cell function. Mechanistically, PARN preferentially binds

Roles of peptidylarginine deiminase (PAD) and protein citrullination in viral infections.

Peptidylarginine deiminases (PADs) are calcium-dependent enzymes that catalyze protein citrullination, a post-translational modification implicated in both physiological processes and the pathogenesis of various diseases. This review provides a systematic overview of the PAD family's roles in viral infections, with a focus on their tissue distribution, activation mechanisms, and functional significance in host-pathogen interactions. Accumulating evidence indicates that diverse viruses have evolved to exploit specific host PAD isoforms as a strategy to subvert antiviral immunity and enhance vir
