A knock-in mutation at cysteine 144 of TRIM72 is cardioprotective and reduces myocardial TRIM72 release.

TRIM72 is a membrane repair protein that protects against ischemia reperfusion (I/R) injury. We previously identified Cys144 (C144) on TRIM72 as a site of S-nitrosylation. To study the importance of C144, we generated a knock-in mouse with C144 mutated to a serine (TRIM72 C144S). We subjected ex vivo perfused mouse hearts to 20 min of ischemia followed by 90 min of reperfusion and observed less injury in TRIM72 C144S compared to WT hearts. Infarct size was smaller (54 vs 27% infarct size) and cardiac functional recovery (37 vs 62% RPP) was higher for the TRIM72 C144S mouse hearts. We also demo

Comparison of two fluid solutions for resuscitation in a rabbit model of crush syndrome.

BACKGROUND: Crush syndrome is a common injury, the main characteristics of which include acute kidney injury. However, there is still lack of reliable animal model of crush syndrome, and it also remains controversial as to which type of fluid should be chosen as a more appropriate treatment option for prevention and treatment of acute kidney injury. METHODS: The rabbits were crushed at the lower limbs for 6 h with 36 times the body weight, which means the pressure of each leg was also 36 times the body weight. Fluid resuscitation was performed from 1 h prior to the end of the crush treatment

Endovascular thrombectomy for isolated posterior cerebral artery occlusion: distinct clinical presentation patterns and neurological outcomes in P1 versus P2 segments.

BACKGROUND: Endovascular thrombectomy (EVT) for isolated posterior cerebral artery (PCA) occlusion remains incompletely characterized, particularly regarding segment-specific presentation and outcomes. We aimed to compare clinical profiles, procedural results, and early neurological and imaging outcomes between P1 and P2 PCA occlusions treated with EVT. METHODS: We retrospectively analyzed EVT-treated patients with acute ischemic stroke at a tertiary center from 2018-2025 (n = 1,166). Thirty-nine patients with isolated PCA occlusion were included in the final analysis (P1 n = 27; P2 n = 12).

A potential therapeutic target at Connexin 43 serine phosphorylation against ischaemia/reperfusion arrhythmia.

BACKGROUND AND PURPOSE: Dephosphorylation of several phosphor residues in connexin 43 (Cx43) is critically involved in ischaemia/reperfusion (I/R) arrhythmias. Here, we sought to identify constitutive Cx43-serine 282 (S282) dephosphorylation as an independent and targetable substrate to delineate reperfusion arrhythmogenesis. EXPERIMENTAL APPROACH: Reperfusion arrhythmias were induced by rat I/R (15/45 min), identified in heterozygous knock-in mice with S282 to alanine (Cx43-S282A/+) and rescued by LB100, a specific protein phosphatase 2A (PP2A) inhibitor, via surface electrocardiogram and ep

Targeting the molecular mechanisms of ischemic damage: Protective effects of alpha-crystallin-B.

AIMS: Molecular chaperones constitute protectors of intracellular protein integrity and seem to confer short-term defence against various cell insults. Myocardial damage is associated to a loss of protective chaperones. Ischemic post-conditioning (IPost-Co) is a procedure that seems to protect against reperfusion injury. However, little is known on alpha-crystallin-B-chain (cryab/HspB5) evolution in IPost-Co. Here we have investigated cryab in myocardial ischemia and IPost-Co. METHODS AND RESULTS: Pigs underwent closed-chest 1.5h mid-left anterior descending (LAD) balloon occlusion and were e…

Longitudinal quantification of radical bursts during pulmonary ischaemia and reperfusion.

OBJECTIVES: Pulmonary ischaemia-reperfusion injury (IRI) is associated with several life-threatening pulmonary disorders, and may severely compromise the outcome of lung transplantation. Highly reactive molecules such as superoxide, nitric oxide (NO) and peroxynitrite (ONOO(-)) are presumed to contribute to IRI pathogenesis, but this assumption is based on indirect measurements. We use electron spin resonance (ESR) to directly quantify free radical formation after pulmonary ischaemia and reperfusion. METHODS: Five groups of 10 Swiss mice were subjected to left pulmonary hilum clamping for 1 h…

NLRP3 Inflammasome Involvement in the Organ Damage and Impaired Spermatogenesis Induced by Testicular Ischemia and Reperfusion in Mice.

We investigated the role of the nucleotide-binding oligomerization domain (NOD)-like receptor family pyrin domain containing 3 (NLRP3) inflammasome during testis ischemia and reperfusion injury (TI/R) in wild-type (WT) and NLRP3 knock-out (KO) mice. WT and KO mice underwent 1 hour testicular ischemia followed by 4 hours and 1 and 7 days of reperfusion or a sham TI/R. Furthermore, two groups of WT mice were treated at the beginning of reperfusion and up to 7 days with two inflammasome inhibitors, BAY 11-7082 (20 mg/kg i.p.) or Brilliant Blue G (45.5 mg/kg i.p.), or vehicle. Neuroprotection of the leaf and stem of Vitis amurensis and their active compounds against ischemic brain damage in rats and excitotoxicity in cultured neurons.

Vitis amurensis (Vitaceae) has been reported to have anti-oxidant and anti-inflammatory activities. The present study investigated a methanol extract from the leaf and stem of V. amurensis for neuroprotective effects on cerebral ischemic damage in rats and on excitotoxicity induced by glutamate in cultured rat cortical neurons. Transient focal cerebral ischemia was induced by 2h middle cerebral artery occlusion followed by 24h reperfusion (MCAO/reperfusion) in rats. Orally administered V. amurensis (25-100 mg/kg) reduced MCAO/reperfusion-induced infarct and edema formation, neurological defici

Protection of ischemic preconditioning on renal neural function in rats with acute renal failure.

We tested whether tolerance induced by ischemic preconditioning (IPC) in kidneys was related to renal nerves. Experimental acute renal failure (ARF) in a rat model was induced for 45 min of left renal arterial occlusion (RAO), followed by 6 or 24 h of reperfusion (ischemic reperfusion (I/R) group). The episode of IPC was four cycles of 4 min of RAO at 11 min intervals and then the I/R injury was treated as above (IPC-I/R group). After 6 h of reperfusion, polyuria was found in the I/R group associated with an enhancement of afferent renal nerve activity (ARNA) and a reflexive decrease in effere

Association Between Antiarrhythmic, Electrophysiological, and Antioxidative Effects of Melatonin in Ischemia/Reperfusion.

Melatonin is assumed to confer cardioprotective action via antioxidative properties. We evaluated the association between ventricular tachycardia and/or ventricular fibrillation (VT/VF) incidence, oxidative stress, and myocardial electrophysiological parameters in experimental ischemia/reperfusion under melatonin treatment. Melatonin was given to 28 rats (10 mg/kg/day, orally, for 7 days) and 13 animals received placebo. In the anesthetized animals, coronary occlusion was induced for 5 min followed by reperfusion with recording of unipolar electrograms from ventricular epicardium with a 64-lea

Preventing the inflammatory response to open-heart surgery: the role of aprotinin and other protease inhibitors.

This review discusses the role of protease inhibition in reducing or preventing certain of the deleterious effects of major surgery. The ability of agents such as aprotinin to inhibit bleeding and reduce the need for donor blood transfusions is now well established. What is less well recognized is that aprotinin is not simply an antifibrinolytic but is a polyvalent enzyme inhibitor of many of the enzymes which are involved in inflammatory and hemostatic processes. Inappropriate activation of these pathways is considered to be important in the genesis of the inflammatory response to open-heart

ECMO-facilitated rapid and deep hypothermia reduces brain injury on MRI following prolonged cardiac arrest in a translational swine model.

INTRODUCTION: Neuroprotective interventions after cardiac arrest are essential but largely lack evidence of efficacy. Early therapeutic hypothermia (TH) is the only intervention that has shown promise in humans. However, despite a consistent signal for efficacy in animal models, conflicting clinical data hamper clinical acceptance. Two potential causes for the lack of translation from animal studies to humans are the time to achieve target temperature in humans and the inability to cool to deep hypothermic states due to the inherent detrimental cardiac effects accompanying deep hypothermia. Myocardial susceptibility to ischaemia/reperfusion in obesity: a re-evaluation of the effects of age.

BACKGROUND: Reports on the effect of age and obesity on myocardial ischaemia/reperfusion (I/R) injury and ischaemic preconditioning are contradictory. The aim of this study was to re-evaluate the effects of age and diet-induced obesity (DIO) on myocardial I/R injury and preconditioning potential. METHODS: Four groups of Wistar male rats were used: age-matched controls (AMC) receiving standard rat chow for (i) 16 weeks and (ii) 16 months respectively; DIO rats receiving a sucrose-supplemented diet for (iii) 16 weeks and (iv) 16 months respectively. The ages of groups (i) and (iii) were 22 week

Erythropoietin and autoimmune neuroinflammation: lessons from experimental autoimmune encephalomyelitis and experimental autoimmune neuritis.

Erythropoietin (EPO) is known to have numerous biological functions. While its primary function is during haematopoiesis, recent studies have shown that EPO plays important role in cytoprotection, immunomodulation, and antiapoptosis. These secondary functions of EPO are integral to tissue protection following hypoxic injury, ischemia-reperfusion injury, and spinal cord injury in the central nervous system. This review focuses on experimental evidence documenting the neuroprotective effects of EPO in organ-specific autoimmune nervous system disorders such as experimental autoimmune encephalomye…

Kynurenine-mediated redox regulation provides neuroprotection in central retinal artery occlusion.

BACKGROUND: Central retinal artery occlusion (CRAO) causes irreversible vision loss through ischemia-reperfusion (I/R) injury, characterized by oxidative stress and retinal ganglion cell (RGC) death. Current therapies are inadequate. This study investigates kynurenine (Kyn)—a tryptophan metabolite—as a novel therapeutic agent targeting dual pathways to combat retinal I/R injury. METHODS: The unilateral pterygopalatine ophthalmic artery occlusion (UPOAO) model was established by inserting a silicone filament embolus into the pterygopalatine artery.
