Human Organotypic Airway and Lung Organoid Cells of Bronchiolar and Alveolar Differentiation Are Permissive to Infection by Influenza and SARS-CoV-2 Respiratory Virus.

The ongoing coronavirus disease 2019 (COVID-19) pandemic has led to the initiation of unprecedented research efforts to understand the pathogenesis mediated by Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2). More knowledge is needed regarding the cell type-specific cytopathology and its impact on cellular tropism. Furthermore, the impact of novel SARS-CoV-2 mutations on cellular tropism, alternative routes of entry, the impact of co-infections, and virus replication kinetics along the respiratory tract remains to be explored in improved models. Most applied virology models are no

Development of a multicellular in vitro model of the meningeal blood-CSF barrier to study Neisseria meningitidis infection.

BACKGROUND: Bacterial meningitis is a life-threatening disease that occurs when pathogens such as Neisseria meningitidis cross the meningeal blood cerebrospinal fluid barrier (mBCSFB) and infect the meninges. Due to the human-specific nature of N. meningitidis, previous research investigating this complex host-pathogen interaction has mostly been done in vitro using immortalized brain endothelial cells (BECs) alone, which often do not retain relevant barrier properties in culture. Here, we developed physiologically relevant mBCSFB models using BECs in co-culture with leptomeningeal cells (LMCs

Double remission of chronic lymphocytic leukemia and secondary acute myeloid leukemia after venetoclax monotherapy: A case report.

RATIONALE: The abnormal expression of B-cell lymphoma-2 (Bcl-2) family members is often associated with the progression of the disease. Bcl-2 inhibitors (eg, venetoclax) were first reported to inhibit the proliferation of malignant lymphocytes and have a significant effect on patients with chronic lymphoblastic leukemia, but research on myeloid tumors is relatively delayed. Venetoclax was approved in 2018 for the treatment of acute myeloid leukemia (AML) patients who were not suitable for high-dose chemotherapy. The approval of venetoclax is an advance in the treatment of hematological tumors.

Discovery biology of neuropsychiatric syndromes (DBNS): a center for integrating clinical medicine and basic science.

BACKGROUND: There is emerging evidence that there are shared genetic, environmental and developmental risk factors in psychiatry, that cut across traditional diagnostic boundaries. With this background, the Discovery biology of neuropsychiatric syndromes (DBNS) proposes to recruit patients from five different syndromes (schizophrenia, bipolar disorder, obsessive compulsive disorder, Alzheimer's dementia and substance use disorders), identify those with multiple affected relatives, and invite these families to participate in this study. The families will be assessed: 1) To compare neuro-endophe

A cost-effective cell- and matrix-based minimally invasive single-stage chondroregenerative technique developed with validated vertical translation methodology.

Introduction The morbidity and significant health economic impact associated with the chondral lesion has led to a large number of strategies for therapeutic neochondrogenesis. The challenge has been to develop techniques that are cost effective single-stage procedures with minimal surgical trauma that have undergone rigorous preclinical scrutiny and robust reproducible assessment of effectiveness. A biological repair requires the generation of a cellular and matrix composite with appropriate signalling for chondrogenic differentiation. Methods and results A technique was developed that allowe

Evaluation of neurotoxicity of drugs from morphological and electrophysiological endpoints using human iPSC-derived neural cell models.

INTRODUCTION: Genome-edited human induced pluripotent stem cells (iPSCs) were first generated in 2007, and have been applied in pharmaceutical research, development, and clinical therapy as an in vitro platform for personalised phenotyping and drug discovery. iPSCs can differentiate into multiple types of neural cells, the in vitro hiPSC-derived neural model may also be an effective tool for early screening and evaluating potential neurotoxicants. Neurotoxicity is one of the limiting factors in the clinical application of many drugs. However, there is currently no set of standardized in vitro

Animal models and medical countermeasures development for radiation-induced lung damage: report from an NIAID Workshop.

Since 9/11, there have been concerns that terrorists may detonate a radiological or nuclear device in an American city. Aside from several decorporation and blocking agents for use against internal radionuclide contamination, there are currently no medications within the Strategic National Stockpile that are approved to treat the immediate or delayed complications resulting from accidental exposure to radiation. Although the majority of research attention has focused on developing countermeasures that target the bone marrow and gastrointestinal tract, since they represent the most acutely radi

Neurodegenerative Diseases in Children: A Comprehensive Review.

Neurodegenerative diseases (NDDs) in children represent a heterogeneous group of rare but collectively significant disorders characterized by progressive neurological decline, developmental regression, and substantial morbidity and mortality. Unlike adult-onset neurodegeneration, pediatric conditions are predominantly genetic and frequently arise from defects in fundamental cellular pathways, including lysosomal degradation, mitochondrial oxidative phosphorylation, peroxisomal lipid metabolism, and myelin maintenance. This comprehensive review synthesizes current knowledge regarding the epidem

The Role of Gut Microbiota in Acute Myeloid Leukemia.

Gut microbiota plays a crucial role in regulating immune system function and shaping immunological responses to pathogens capable of causing infections. Alterations in the composition of the intestinal microbiome are associated with immune system dysfunction and increased susceptibility to infections. Patients with acute myeloid leukemia (AML) are highly susceptible to infections due to immune system deregulation caused by the disease itself, as well as chemotherapy-induced bone marrow aplasia. In these patients, gut microbiota dysbiosis and reduced microbial diversity (i.e., imbalances in the

Educational and training needs of regulatory professionals for non-animal methodologies: Insights from a survey.

The increasing application of non-animal new approach methodologies (NAMs) in regulatory decision-making requires regulatory professionals to develop new competencies to confidently interpret data derived from this expanding array of methods. Tailored educational programs could facilitate this, but little research has examined the specific educational needs and preferences of regulatory professionals across sectors. This cross-sectional survey aims to address this gap and generate actionable insights for designing such programs. An online questionnaire was administered to regulatory profession

A Machine Learning-Based Prognostic Model for Sepsis-Associated Liver Injury Using Routine Indicators.

UNLABELLED: <p>Objective: Sepsis-associated liver injury (SALI) occurs in approximately 40% of sepsis cases and is linked to high mortality, a challenge that may stem from the absence of effective prognostic models. We developed a machine learning (ML)-based prognostic model for SALI using conventional biomarkers to guide precise clinical interventions and reduce mortality. METHODS: We retrospectively analyzed 307 SALI patients (2010-2024), stratified into favorable (n = 139) and poor (n = 168) prognosis groups by post-treatment progression. The cohort was randomly split into a training set (

Antiparasitic agents in oncology: Innovative mechanisms, emerging evidence and clinical potential in cancer treatment.

The repurpose of established antiparasitic agents for oncological applications represents a promising and cost-effective strategy in the search for novel anticancer therapeutics. Drugs originally developed to treat parasitic infections, including ivermectin, mebendazole, niclosamide, albendazole, artesunate, flubendazole, and other antiparasitic agents, have demonstrated compelling anticancer properties across a wide range of preclinical models. Their mechanisms of action are multifaceted, encompassing disruption of microtubule dynamics, induction of apoptotic signaling cascades, inhibition of

Inflammation at the Maternal-Fetal Interface: Mechanisms Linking Maternal-Fetal Immunity to Preeclampsia and Fetal Growth Restriction.

Inflammation is a physiological and tightly regulated component of normal pregnancy, contributing to implantation, placental development, and the initiation of parturition. The placenta functions as an active immunological hub, coordinating innate and adaptive immune responses to maintain tolerance while protecting against infection. Preeclampsia and fetal growth restriction (FGR) are major causes of maternal and perinatal morbidity worldwide and represent central manifestations of placental disease. Increasing evidence indicates that these conditions share key pathophysiological mechanisms, i

Maintenance Strategies in High-Risk Myeloma: A Multicenter Comparison of Bortezomib-Lenalidomide Versus Lenalidomide Alone: A USMIRC Multicenter Analysis.

Background: Lenalidomide maintenance after autologous stem cell transplantation (ASCT) improves progression-free survival (PFS) and overall survival (OS) in multiple myeloma; however, these benefits are attenuated in high-risk multiple myeloma (HRMM). No standard post-transplant maintenance strategy is established for HRMM, and some centers employ doublet maintenance with bortezomib plus lenalidomide (VR). We evaluated outcomes with VR versus lenalidomide alone (R) in HRMM. Methods: We conducted a multicenter retrospective study through the US Myeloma Innovations Research Collaborative (USMIRC

Modeling Williams syndrome from a neurodevelopmental perspective: recent advances, model-based translational insights and future directions.

BACKGROUND: Williams syndrome (WS; OMIM #194,050) is a multisystem pediatric genetic disorder caused by a heterozygous microdeletion of a 1.5-1.8 Mb region at chromosome 7q11.23, encompassing 26 to 28 genes. Clinical hallmarks include cardiovascular anomalies, distinctive craniofacial morphology and neurodevelopmental deficits characterized by hypersociability, cognitive impairment and anxiety. Although causative therapies for WS still remain elusive, advances in gene editing and forebrain organoids have already greatly furthered our understanding of the underlying mechanisms.
