Cryptosporidium infections: molecular advances.

Cryptosporidium host cell interaction remains fairly obscure compared with other apicomplexans such as Plasmodium or Toxoplasma. The reason for this is probably the inability of this parasite to complete its life cycle in vitro and the lack of a system to genetically modify Cryptosporidium. However, there is a substantial set of data about the molecules involved in attachment and invasion and about the host cell pathways involved in actin arrangement that are altered by the parasite. Here we summarize the recent advances in research on host cell infection regarding the excystation process, att…

Protein trafficking through the endosomal system prepares intracellular parasites for a home invasion.

Toxoplasma (toxoplasmosis) and Plasmodium (malaria) use unique secretory organelles for migration, cell invasion, manipulation of host cell functions, and cell egress. In particular, the apical secretory micronemes and rhoptries of apicomplexan parasites are essential for successful host infection. New findings reveal that the contents of these organelles, which are transported through the endoplasmic reticulum (ER) and Golgi, also require the parasite endosome-like system to access their respective organelles. In this review, we discuss recent findings that demonstrate that these parasites re…

[Protozoons and arthropods found in eyes].

Protozoons and arthropods can be observed commonly all around the world including our country. These parasites can cause different kind of disorders in human and animals. Some of these can cause eye disorders. The aim of this review was to present information about how the protozoons such as Toxoplasma gondii, Leishmania spp., Trypanosoma spp., Giardia spp., Acanthamoeba spp., Plasmodium spp., the arthropods insects of myiasis, Phthirus pubis, ticks, Demodex folliculorum and Linguatula serrata (under discussion as to which order it beongs) invade the eye of host leading to clinical symptoms, d…

Expansion of CD8+CD57+ T Cells in an Immunocompetent Patient with Acute Toxoplasmosis.

CD57+ T cells increase in several viral infections like cytomegalovirus, herpesvirus, parvovirus, HIV and hepatitis C virus and are associated with several clinical conditions related to immune dysfunction and ageing. We report for the first time an expansion of CD8+ CD57+ T cells in a young patient with an acute infection with Toxoplasma gondii. Our report supports the concept that CD8+ CD57+ T cells could be important in the control of chronic phase of intracellular microorganisms and that the high numbers of these cells may reflect the continuing survey of the immune system, searching for p…

Modeling the molecular basis of atovaquone resistance in parasites and pathogenic fungi.

Atovaquone is a substituted hydroxynaphthoquinone that is used therapeutically for treating Plasmodium falciparum malaria, Pneumocystis jirovecii pneumonia and Toxoplasma gondii toxoplasmosis. It is thought to act on these organisms by inhibiting parasite and fungal respiration by binding to the cytochrome bc1 complex. The recent, growing failure of atovaquone treatment and increased mortality of patients with malaria or Pneumocystis pneumonia has been linked to the appearance of mutations in the cytochrome b gene. To better understand the molecular basis of drug resistance, we have developed …

Chronic ocular lesions in tawny owls (Strix aluco) injured by road traffic.

The chronic ocular lesions suffered by 50 tawny owls (Strix aluco) injured by road traffic were investigated. They included non-healing corneal erosions, cataracts and retinal scarring, and periretinal membrane formation. The intraocular pressure was significantly lower in the eyes with intraocular inflammatory pathology and higher in the eyes with irido- or cyclodialysis than in ophthalmically normal eyes. Cicatritial retinal lesions were not associated with high titres of antibodies to Toxoplasma species. The findings correlate with those observed in human beings with eye injuries due to hig…

Zoonotic protozoa--food for thought.

Outbreaks of water- and foodborne diseases caused by Cryptosporidium, Giardia and Toxoplasma are well documented. Three features of these zoonotic protozoa ensure a high level of environmental contamination and enhance the likelihood of waterborne transmission. Firstly, they are responsible for disease in a broad range of hosts including man, have a low infectious dose enhancing the possibility of zoonotic transmission, secondly, their transmissive stages are small in size and environmentally robust and thirdly are insensitive to the disinfectants commonly used in the water industry. In additi…

[A case of primary toxoplasmosis in an immunocompetent patient].

CASE REPORT: A case of an immunocompetent patient presenting primary systemic toxoplasma infection involving the eye (condition seen in less than 3% of primary infections). The patient showed reactivation of this primary focus two years later. DISCUSSION: Diagnosis of toxoplasm retinitis is based on a typical lesion consisting in an area of active retinitis adjacent to an inactive corioretinal scar. Differential diagnosis must consider other causes of retinal coroiditis in primary infection cases: sarcoidosis, tuberculosis, syphilis as well as viral and fungal infections. Ocular toxoplasmosis…

Ocular toxoplasmosis: a 50th anniversary tribute to the contributions of Helenor Campbell Wilder Foerster.

In 1952, Helenor Campbell Wilder (later Helenor Campbell Wilder Foerster) confirmed the growing suspicion that Toxoplasma gondii was a cause of uveitis in otherwise healthy adults by identifying the presence of parasites in eyes enucleated because of severe intraocular inflammation. Ocular toxoplasmosis was previously known to occur only in newborns with congenital T gondii infection. Her report ushered in a new era in the field of uveitis in which toxoplasmosis, rather than tuberculosis, was confirmed to be the most common cause of retinochoroiditis. Fifty years later, issues raised in her la…

Neuroretinitis: an unusual presentation of ocular toxoplasmosis.

Neuroretinitis is a clinical condition usually seen in young healthy adults that is characterized by rapid profound unilateral loss of vision. Funduscopic findings include swollen disc, splinter hemorrhages, and macular star. Catscratch fever and leptospirosis have been suggested as possible etiologies in this condition. We report a case of a patient with neuroretinitis whose only laboratory abnormality was an elevated enzyme-linked immunosorbent assay immunoglobulin G Toxoplasma titer. The patient responded favorably to treatment with clindamycin, triple sulfa, and systemic corticosteroids. W…

Presence of gamma interferon in human acute and congenital toxoplasmosis.

The production of gamma interferon in acute acquired and congenital toxoplasmosis was studied. Gamma interferon was produced at significant titers (P less than 0.001) in the course of both congenital toxoplasmosis and acquired toxoplasmosis at an early stage of infection, when Toxoplasma gondii was multiplying. Its presence in fetal blood was correlated with the positive inoculation of fetal blood or amniotic fluid into mice (95%). The data suggest that the fetus is able to synthesize gamma interferon as early as week 21 of pregnancy. This test, easily and rapidly performed, could be included …

[Sero-immunologic diagnosis of human African trypanosomiasis: cross reactions of various parasitoses and hyperglobulinemias].

The immunological test: latex agglutination, counter immuno-electrophoresis, ELISA, and immunoperoxydase test, have been applied on the serodiagnosis of african sleeping sickness, and have been compared with each other, with special reference to the cross-reactions in parasitic diseases and hyperglobulinemia. In addition to malaria and leishmaniasis well known interferences, this study gives clear indication of the importance of false positive reactions in active toxoplasma infection. Any how, the serodiagnostic can be worked out from the analysis of responses to various specific antigens, whi…

Infectious mononucleosis and related syndromes.

The "mono syndrome" is an acute febrile disease of older children and young adults which involves the lymphatic system and is characterized hematologically by the presence of 50% or more lymphocytes and monocytes and 10% or more atypical lymphocytes. Epstein-Barr virus (EBV) causes over 90% of the syndrome, cytomegalovirus (CMV) about 5% to 7%, and Toxoplasma gondii less than 1%. Viral hepatitis, adenovirus, rubella, and herpes simplex are rare causes. Only EBV produces classical heterophile-positive infectious mononucleosis. This article reviews the epidemiological and clinical features of th…

Evaluation of the potential therapeutic efficacy of Cerastes cerastes venom in acute experimental toxoplasmosis.

BACKGROUND: The control of toxoplasmosis relies on conventional chemotherapeutics, which have hitherto unresolved concerns. METHODS: Swiss albino mice were intraperitoneally (IP) infected with 5 × 103 tachyzoites of RH HXGPRT( -) strain of Toxoplasma gondii, then IP treated with one-fourth lethal dose 50 (one-fourth LD50) of Cerastes cerastes venom (CCV) for three consecutive days (LD = 0.535 mg/kg). The anti-Toxoplasma activity of CCV was evaluated, for the first time, in immunocompetent (IC) and immunosuppressed (IS) mice via estimation of their mortality and survival time, microscopical co…

Prevalence of toxoplasmosis in natural ungulates as human zoonotic meat-borne pathogens: A systematic review and meta-analysis.

The increasing consumer interest in game meat, along with the potential transmission of Toxoplasma gondii from game meat to humans, poses significant challenges in assessing the related public health risks. The diverse transmission pathways of T. gondii and its ability to infiltrate host organisms offer an excellent opportunity for enhancing our comprehension of this pathogen's transmission dynamics. Toxoplasma has traditionally been a valuable model for studying host-pathogen interactions and investigating population, community, and environmental or ecological aspects. Therefore, a meta-analy…

CRISPR/Cas12a combined with RPA for detection of T. gondii in mouse whole blood.

BACKGROUND: Toxoplasma gondii is an opportunistic protozoan that is ubiquitous in humans and animals. It can invade any human organ and cause severe diseases, including toxoplasma ophthalmopathy, meningoencephalitis, and liver necrosis. Porcine toxoplasmosis is prevalent in China. CRISPR (Clustered Regularly Interspaced Short Palindromic Repeats) and Cas (CRISPR-Associated Protein) systems are widely used for gene editing and pathogen detection. CRISPR-based diagnostics are molecular assays that have been developed to detect parasites with high sensitivity and specificity. METHODS: This study…

Foodborne protozoan parasites in fresh mussels and oysters purchased at retail in Canada.

Studies worldwide have reported the presence of protozoan parasites in a variety of commercial bivalve shellfish. The uptake of these parasites by shellfish occurs during filter feeding in faecally-contaminated waters. The objective of the present study was to determine the prevalence of Giardia, Cryptosporidium and Toxoplasma in fresh, live shellfish purchased in three Canadian provinces as part of the retail surveillance activities led by FoodNet Canada (Public Health Agency of Canada). Packages containing mussels (n = 253) or oysters (n = 130) were purchased at grocery stores in FoodNet Can…
