PURPOSE: Programmed death receptor ligand-1 (PD-L1) expression and tumor mutational burden (TMB) are approved screening biomarkers for immune checkpoint inhibition (ICI) in advanced triple negative breast cancer. We examined these biomarkers along with characterization of the tumor microenvironment (TME) between breast tumors (BrTs), axillary metastases (AxMs), liver metastases (LvMs), non-axillary lymph node metastases, and non-liver metastases to determine differences related to site of metastatic disease.
METHODS: 3076 unpaired biopsies from breast cancer patients were analyzed using whole

The Bacillus Calmette-Guérin (BCG) vaccine is the oldest cancer immunotherapeutic agent in use. Despite its effectiveness, its initial mechanisms of action remain largely unknown. Here, we elucidate the earliest cellular mechanisms involved in BCG-induced tumor clearance. We developed a fast preclinical in vivo assay to visualize in real time and at single-cell resolution the initial interactions among bladder cancer cells, BCG and innate immunity using the zebrafish xenograft model. We show that BCG induced the recruitment and polarization of macrophages towards a pro-inflammatory phenotype,

UNLABELLED: Type I interferons (IFN) are immune-stimulatory cytokines involved in antiviral and antitumor immune responses. They enhance the efficacy of immunogenic anticancer therapies such as radiotherapy by activating both innate and adaptive immune cells. Macrophages are one of the most abundant innate immune cells in the immune microenvironment of melanoma brain metastases (MBM) and can exert potent immune-suppressive functions. Here, we investigate the potential of tumoral type I IFNs to repolarize tumor-associated macrophages (TAM) in two murine MBM models and assess the effects of radi

Fumarate hydratase (FH) is an important enzymatic component in the tricarboxylic acid cycle. Studies have reported that FH plays an important role in hereditary leiomyomatosis and renal cell cancer (HLRCC). However, the role of FH in human different cancers remains unknown. This study is aimed at analyzing the prognostic value of FH and demonstrating the correlation between FH expression and tumor immunity. Results showed that FH was mutated or copy number varied in 27 types of cancer. FH mRNA was abnormally upregulated across various cancers. Survival analysis suggested high expression of FH

BACKGROUND: Lung adenocarcinoma is prone to brain metastasis, and the prognosis of patients is extremely poor. The inhibitor of apoptosis-stimulating protein of p53 (iASPP) protein, encoded by the protein phosphatase 1 regulatory subunit 13-like (PPP1R13L) gene, is a key inhibitor of the p53 pathway and promotes carcinogenesis in various tumors, but its role in brain metastasis of lung adenocarcinoma is unknown. This study aims to analyze the tumor microenvironment characteristics of patients with brain metastasis of lung adenocarcinoma and explore the expression of PPP1R13L in brain metastasi

Patients diagnosed with microsatellite stable (MSS) metastatic colorectal cancer (mCRC) typically have an immunosuppressive tumor microenvironment, which leads to a low response rate when treated with immunotherapy. Some studies indicate that chemotherapy and anti-angiogenic therapy could potentially improve the responsiveness of these patients to immunotherapy. Therefore, this study is designed to assess the effectiveness and safety of combining chemotherapy with bevacizumab and anti-PD-1 immunotherapy as a second-line treatment option for MSS mCRC. A retrospective analysis was conducted on p

Eicosanoids, bio-active lipid molecules, evoke a multitude of biological effects that directly affect cancer cells and indirectly affect tumor microenvironment. An emerging role has been shown for eicosanoids in the pathogenesis of gynecological malignancies which include cancers of the vulva, vagina, cervix, uterine, and ovary. Eicosanoid biosynthesis pathways start at the metabolism of phospholipids by phospholipase A2 then proceeding to one of three pathways: the cyclooxygenase (COX), lipoxygenase (LOX), or P450 epoxygenase pathways. The most studied eicosanoid pathways include COX and LOX;

Immunomodulatory drugs and monoclonal antibody-based immunotherapies have significantly improved the prognosis of the patients with multiple myeloma (MM) in the recent years. These new classes of reagents target malignant plasma cells (PCs) and further modulate the immune microenvironment, which prolongs anti-MM responses and may prevent tumor occurrence. Since MM remains an incurable cancer for most patients, there continues to be a need to identify new tumor target molecules and investigate alternative cellular approaches using gene therapeutic strategies and novel treatment mechanisms. BACKGROUND: A remolded microenvironment in hepatocellular carcinoma (HCC) caused by abnormally expressed matricellular proteins could promote HCC progression. The cell-matrix interactions mediated by integrins play an important role in tumor microenvironment. Epidermal Growth Factor-like repeats and Discoidin I-Like Domains 3 (EDIL3), an extracellular matrix (ECM) protein with angiogenic and anti-inflammatory effects, is abnormally highly expressed in HCC. Here we aim to analyze its expression in liver and HCC tissues, investigate the underlined mechanisms accounted for HCC progression.

Tumor blood vascular endothelial cells targeting has emerged as a powerful strategy to enhance the accumulation and penetration of nanomedicines. However, the complex tumor microenvironment, especially the newly discovered breast cancer blood vascular basement membrane, can severely trap accumulated nanomedicines and compromise therapeutic efficacy. In this study, a γ-glutamyl transpeptidase (GGT)-targeting calcium nanomachine is designed and fabricated through decoration of GGT-targeting polysarcosine and encapsulation of nitric oxide (NO) precursors. Upon NIR laser irradiation, these calcium

CD8+ T cells play a crucial role in anti-tumor immunity; however, their functionality is compromised by exhaustion driven by chronic antigen exposure, poor tumor microenvironment, and metabolic dysfunction. Activated CD8+ T cells undergo extensive proteomic remodeling, which includes the induction of the IRE1-XBP1 signaling axis of the unfolded protein response. However, the functional consequences of sustained IRE1-XBP1 activation in exhausted CD8+ T cells remain elusive. Using a model of constitutive XBP1 activation in CD8+ T cells, we demonstrate that sustained XBP1s signaling leads to redu

Intermittent fasting (IF) exerts antitumor activity in colorectal cancer (CRC), but the tumor-intrinsic metabolic mechanism linking nutrient restriction to antitumor immunity remains unclear. In this study, IF reduced taurine availability and suppressed tumor growth in a CD8+ T cell-dependent manner. Taurine supplementation attenuated this effect, whereas pharmacological inhibition of taurine uptake partially recapitulated fasting-associated tumor suppression in preclinical models. Mechanistically, taurine is associated with the endoplasmic reticulum chaperone GRP78 and contributes to the main

Chimeric antigen receptor (CAR) T cell therapy has shown remarkable success in hematologic malignancies and autoimmune diseases but remains limited in solid tumors due to antigen heterogeneity, escape, and an immunosuppressive tumor microenvironment (TME) dominated by tumor-associated macrophages (TAMs). We developed a macrophage-directed CAR T strategy targeting TREM2+ immunosuppressive TAMs, achieving potent in vitro activity and robust antitumor efficacy in vivo. To enhance intratumoral activity, we incorporated synthetic CAR-responsive biosensors containing NFAT, IRF, and AP1 motifs that e

Chimeric antigen receptor-modified T cells (CAR-T) have shown remarkable success in hematologic malignancies, but their efficacy against solid tumors remains limited. While immunosuppressive cells and molecules in the tumor microenvironment (TME) are known to impair CAR-T function, these are not CAR-T-specific barriers. Using multiple mouse models, we found that the impact of CAR-T cells on tumor growth is dose-dependent, capable of promoting, having no effect on, or inhibiting tumor growth. Mechanistically, tumor-infiltrating CAR-T cells play a dual role: they release antitumor effector molec

Successful anti-tumor immune surveillance depends on stem-like CD8+ T cells that are enriched in tumor-draining lymph nodes (LN), but how they are maintained over time remains poorly understood. Here, we identify a continuous lymphatic circuit that sustains stem-like CD8+ T cells. Using photoconversion to fate-map intratumoral T cells we demonstrate that effector cells exit the tumor microenvironment and migrate back to the draining LN. These tumor-specific, migratory effector T cells avoid chronic antigen stimulation, re-express the transcription factor associated with self-renewal, TCF1, and

CD8+ T cell abundance within the tumor microenvironment is a critical determinant of immunotherapy efficacy. Here we show that CD8⁺ T cells lacking STING or TOX display markedly improved antitumor activity, with enhanced tumor infiltration and elevated IFN-γ and granzyme B production. These STING or TOX deficient cells exhibit a stem-like transcriptional state and resist ferroptosis by suppressing lipid peroxidation pathways while promoting mitochondrial biogenesis. Mechanistically, STING and TOX form a positive regulatory loop that represses HO-1 expression, leading to iron accumulation, mito

Sonodynamic therapy (SDT) for deep-seated tumors is limited by tumor microenvironment (TME) barriers. We developed a hyaluronic acid (HA)-modified mesoporous calcium carbonate nanoplatform (HA/CaCO3@Ce6) to synergistically enhance calcium overload and SDT. The CD44-targeted nanoplatform demonstrated pH-responsive degradation in acidic TME, resulting in the release of Ca2+ and chlorin e6 (Ce6). The released Ca2+ induced mitochondrial calcium overload, causing 71% collapse in membrane potential and 1.6-fold increase in reactive oxygen species (ROS) generation, establishing a "Ca2+-ROS positive f
