Durvalumab, carboplatin, and etoposide in patients who are treatment-naive with extensive-stage small-cell lung cancer and poor performance status (NEJ045A): a single-arm phase 2 trial.

BACKGROUND: Treating patients with extensive-stage small-cell lung cancer (SCLC) with poor performance status poses considerable challenges. We aimed to evaluate the combination of an immune checkpoint inhibitor with platinum-based therapy in this population. METHODS: This open-label, single-arm phase 2 NEJ045A trial enrolled untreated patients with extensive-stage SCLC with performance status 2 or 3. Participants received four cycles of durvalumab, carboplatin, and etoposide, followed by durvalumab maintenance. A dose adjustment strategy was used, with initial reductions in carboplatin-etopo

Carfilzomib in combination with R-CHOP for initial treatment of patients with non-germinal center diffuse large B-cell lymphoma: a multicenter, single arm, phase 1/2 study.

We performed a phase I/II trial to explore the safety and efficacy of carfilzomib (K) in combination with R-CHOP (KR-CHOP) in patients with diffuse large B cell lymphoma (DLBCL). A total of 48 patients were enrolled and 47 were treated. The overall response rate (ORR) was 89% (70% complete response). At a median follow-up of 31 months, 3-year Kaplan-Meier estimates of PFS and OS were 79% and 87%, respectively. Treatment with KR-CHOP for non-GC DLBCL was associated with a decreased risk of disease progression and death relative to standard of care treatment with R-CHOP with hazard ratios (HR) o

A multicenter, single-arm study using a modified faricimab treat-and-extend regimen in patients with macular edema due to central retinal vein occlusion: RVOSTAR study design protocol.

INTRODUCTION: Anti-vascular endothelial growth factor (anti-VEGF) agents are generally considered to be the first line of therapy for macular edema due to retinal vein occlusion (RVO-ME). However, current anti-VEGF treatment regimens in Japan are unable to maintain long-term vision improvement, particularly in patients with central RVO-ME (CRVO-ME). Faricimab is a dual angiopoietin-2/VEGF inhibitor approved for the treatment of RVO-ME in Japan. The RVOSTAR study is being conducted to evaluate the long-term maintenance of vision outcomes with faricimab using a modified treat-and-extend (T&E) re

Suzetrigine, a Non-Opioid NaV1.8 Inhibitor With Broad Applicability for Moderate-to-Severe Acute Pain: A Phase 3 Single-Arm Study for Surgical or Non-Surgical Acute Pain.

BACKGROUND: Many patients experience inadequate pain control due to limited options that are both efficacious and safe for treating moderate-to-severe acute pain; therefore, opioids are still frequently prescribed for their effectiveness despite known tolerability issues and safety concerns. Suzetrigine, an oral, non-opioid, offers a promising alternative by selectively inhibiting the voltage-gated sodium channel 1.8 (NaV1.8), a novel therapeutic target for pain management. Given the high selectivity of suzetrigine for NaV1.8 (does not bind to other sodium channels/receptors with CNS activity)

Efficacy of a mechanism-based psychological intervention for persistent gastrointestinal symptoms in ulcerative colitis and irritable bowel syndrome: results of a three-arm randomised controlled trial (SOMA.GUT-RCT).

BACKGROUND: Irritable Bowel Syndrome (IBS) and Ulcerative Colitis (UC) are chronic gastrointestinal conditions with differing pathologies but overlapping symptoms and shared risk factors contributing to symptom persistence. Illness-related anxiety and dysfunctional symptom expectations constitute two empirically proven mechanisms of symptom persistence. Yet, the efficacy of a targeted psychological intervention focussing on these mechanisms remains untested. The SOMA.GUT trial tested whether such a mechanism-based intervention can reduce gastrointestinal symptom severity in individuals with IB

Camp-style lifestyle modification program (CAMP) for diabetes prevention among rural women with prior GDM: study protocol for a three-arm cluster hybrid type 2 randomized controlled trial.

BACKGROUND: Intensive lifestyle interventions were effective to reduce the risk of type 2 diabetes mellitus (T2DM) for women with gestational diabetes mellitus (GDM) history. However, reaching these mothers and maintaining participation in lifestyle interventions is suboptimal in real-world settings. Effective, feasible and sustainable new lifestyle interventions are needed. The objectives of this three-arm trial are to (1) compare diabetes risk outcomes of an evidence-based intensive lifestyle modification (ILSM) intervention, a camp-style lifestyle modification program (CAMP) intervention, a

Efficacy of combining aspirin with hydroxychloroquine in pregnancies at high risk for pre-eclampsia: a prospective, multicentre, open-label, single-arm clinical trial, investigator-initiated study (HUGS study).

INTRODUCTION: The use of hydroxychloroquine (HCQ) during pregnancies complicated by systemic lupus erythematosus or refractory antiphospholipid antibody syndrome has demonstrated a significant ability to prevent pre-eclampsia (PE). As such, the potential for the administration of HCQ to prevent PE in other high-risk pregnancies is an important clinical research agenda among maternal and fetal medicine specialists. Mechanistically, the anti-inflammatory and immunomodulatory effects of HCQ can offer vascular protection and inhibit the placental dysfunction-associated thrombotic changes underlyin

Quantifying Radiation Exposure in Minimally Invasive Spinal Surgery: A Single-Surgeon Study of Minimally Invasive Surgery-Oblique Lateral Lumbar Interbody Fusion (MIS-OLLIF) With Double C-Arm Technique.

In the advancement of spinal surgery, minimally invasive techniques such as the oblique lateral lumbar interbody fusion (OLLIF) have emerged, offering improved clinical outcomes and the use of dual C-Arm intraoperative imaging. Despite the benefits, the radiation exposure to surgeons and staff from this dual-source imaging has not been thoroughly examined. This study aims to quantify the radiation exposure received by surgeons during OLLIF procedures, filling a gap in current research that primarily focuses on single-source imaging. Over 12 months, radiation doses were measured across 121 surg
