Hydrogen/Deuterium Exchange and Protein Oxidative Footprinting with Mass Spectrometry Collectively Discriminate the Binding of Small-Molecule Therapeutics to Bcl-2.

Characterizing protein-ligand interactions is crucial to understanding cellular metabolism and guiding drug discovery and development. Herein, we explore complementing hydrogen/deuterium exchange mass spectrometry (HDX-MS) with a recently developed Fenton chemistry-based approach to protein oxidative footprinting mass spectrometry (OX-MS) to discriminate the binding of small-molecule therapeutics. Using drug-dependent perturbation as the experimental report, this combination of techniques more clearly differentiates the in-solution binding profiles of Venetoclax (ABT-199, GDC-0199-AbbVie and G

Hydrogen Bonding Enabled C-H Polarization for Multi-Site Concerted Electron-Proton Transfer in Electrochemical Dimerization of β-Ketoester.

We present the first report of direct electro-oxidation of the α-CH bond of β-ketoesters employing a multisite concerted electron-proton transfer (MS-CEPT) mechanism to access dimerized products through C(sp3)-C(sp3) bond formation. The sustainable approach utilized a graphite anode and a Pt cathode, employing acetonitrile and methanol solvents, NH4PF6 electrolyte, and NaH as a base at a constant potential of 1.8 V for 3 h. The MS-CEPT mechanism involved H+ transfer to the base (NH3) associated through hydrogen bonding with the α-CH bond and simultaneous transfer of an electron to the electrod

Oxygen Vacancies Promote Formaldehyde Base-Free Reforming into Hydrogen over Cu Doping-Induced Cu-CuxZn1-xO Heterointerfaces.

Element doping is a viable strategy to regulate the metal-support interface for enhancing the catalytic performance of supported metal catalysts. Herein, Cu/ZnO:Cu-TH catalysts are prepared by immobilizing Cu nanoparticles (NPs) on ZnO nanorods featuring an adjustable oxygen vacancy, in which partial Cu atoms at the Cu-ZnO interface are incorporated into the ZnO lattice to form CuxZn1-xO species. Such Cu atom doping induces the creation of distinctive Cu-CuxZn1-xO interface sites and optimizes electron transfer from ZnO to Cu NPs, thereby achieving intermediate activation and ultimately endowi

Preparation of nitrogen-doped biocarbon from sewage sludge and pine sawdust for superior hydrogen sulfide removal: Experimental and DFT studies.

Hydrogen sulfide (H2S) is a major air pollutant posing a serious threat to both the environment and public health. In this study, a novel nitrogen-rich biocarbon that effectively removes H2S was produced from a mixture of sewage sludge and pine sawdust using melamine as nitrogen source. Compared with pristine biocarbons, nitrogen (N)-doped biocarbons possessed an adjustable porosity, e.g., higher specific surface area of 786-1547 m2/g, richer nitrogen content of 9.02-10.22%, and larger pore volume of 0.31-0.88 cm3/g. In particular, when H2S flow rate was 100 mL/min at 25 °C under dry condition

A Near-Infrared Fluorescent Probe with a Large Stokes Shift for Detecting Hydrogen Sulfide in Environmental Waters, Wine Samples, and Living Systems.

Hydrogen sulfide (H2S) is closely related to physiological diseases, environmental pollution, food safety, and plant growth. Herein, a new nopinone-based near-infrared fluorescent probe has been developed to detect H2S with superior performance, including a large Stokes shift, low detection limit, good selectivity, and fast response time. NPD-H2S was used to detect H2S in environmental water and wine samples quantitatively. In addition, NPD-H2S could be used to fabricate nanofiber membranes as a portable tool for sensitively detecting H2S vapor using a smartphone. Moreover, NPD-H2S has been ef

In vivo monitoring of endogenous hydrogen sulfide and evaluation of natural protectants in liver injury mice using a highly selective bioluminescent probe.

Hydrogen sulfide (H2S) is an essential endogenous gasotransmitter that can regulate a wide range of physiological processes. However, overproduction of H2S is toxic to humans and causes liver injury, cardiovascular diseases, central nervous system-related disease, diabetes, even cancer. Hence, designing efficient imaging probes for real-time monitoring of the alterations in endogenous H2S is a viable tactic for accurate diagnosis of these diseases. In this work, a bioluminescence (BL) probe, namely Luc-H2S, has been developed to achieve H2S detection in vitro and in vivo. This sensing probe en

Mitigating hydrogen sulfide emissions from wastewater sludge using ferric chloride-foam spraying: an effective alternative to FC mixing.

Hydrogen sulfide (H2S) emissions from wastewater sludge generate intense odors and cause corrosion in wastewater treatment facilities. While mixing ferric chloride (FC) with sludge is commonly employed to reduce H2S formation and emissions, its effectiveness diminishes at high concentrations. This study explores the application of FC-foam on sludge as an alternative method to mitigate H2S emissions and compares its efficacy with traditional FC-mixing techniques. In batch tests, FC-foam application achieved an 86.44 % reduction in cumulative H2S emissions, compared to a 49.22 % reduction with F

Iron single atom enzyme-mediated hydrogen sulfide delivery amplifies reactive oxygen species cascade to induce ferroptosis susceptibility.

Lipid peroxidation (LPO) represents one of the most deleterious processes contributing to ferroptosis susceptibility. However, the tumor microenvironment is often characterized by an overproduction of endogenous glutathione (GSH) and reactive oxygen species (ROS)-scavenging enzymes, which limit the ferroptosis susceptibility. Herein, we introduce an iron single-atom enzyme (Fe/SAE) nanoplatform, termed Fe/SAE@A, that acts as hydrogen sulfide (H2S) donor anethole trithione (ADT) delivery system to amplify LPO-mediated ferroptosis vulnerability. Upon internalization by cancer cells, Fe/SAE, feat

Copper-Modified Nanocarbon Composites for Enhanced Hydrogen Sulfide Adsorption: Synergistic Effects of Persistent Free Radicals and Cu-Based Oxides.

In this study, copper-modified nanocarbon composites (OMC) were successfully prepared using two-dimensional carbon nanosheets as the material substrate, the low-temperature hydrothermal method as the main process, and copper nitrate as the modifier. The effects of the modifier dosage ratio, hydrothermal temperature, and residence time on the structure and hydrogen sulfide (H2S) adsorption performance of OMC were investigated. The results show that the OMC with persistent free radicals and copper oxides prepared under the conditions of a mass ratio of copper nitrate to two-dimensional carbon na

Nitrogen sites induce homogeneous dispersion of iron species on biochar for enhanced hydrogen sulfide removal in a low-temperature anaerobic environment.

Hydrogen sulfide (H2S) removal under dry, anaerobic, and low-temperature conditions remains challenging due to limited active-site accessibility and rapid pore blockage in conventional adsorbents. This study addresses these limitations by developing a nitrogen-rich biochar-supported iron composite derived from spent coffee grounds. Nitrogen-rich biochar features a hierarchical pore structure, a high specific surface area (1,919.5 m2 g-1), and abundant nitrogen sites that facilitate Fe-N coordination, enabling uniform dispersion of iron species. The iron-based nitrogen-rich biochar (Fe30/NrB) e

Dual Effect of Defect-Free AlOxFy Layer for Suppressing Hydrogen Influence in Indium-Gallium-Zinc Oxide Thin-Film Transistors.

It has been demonstrated that mixing atomic-layer deposition subcycles of aluminum oxide (Al2O3) and aluminum fluoride (AlF3) at an optimal ratio yields an aluminum oxyfluoride (AlOxFy) compound with virtually no internal defects. By systematically analyzing the defect structures formed in AlOxFy thin films as a function of the Al2O3 to AlF3 subcycle ratio, the optimal composition was identified. Because hydrogen migrates either through intrinsic defects in the film or via repeated bonding and dissociation with oxygen, a defect-free AlOxFy film in which AlF3 sublayers providing no hydrogen-dif

pH-responsive Oral liposomal delivery of hydrogen sulfide donor GYY4137 enables colon-targeted therapy for inflammatory bowel disease.

BACKGROUND: Although therapeutic options for inflammatory bowel disease (IBD) have advanced, many patients still experience suboptimal clinical response, systemic side effects, or difficulty maintaining long-term treatment adherence. Hydrogen sulfide (H₂S), an endogenous gasotransmitter with potent anti-inflammatory and mucosal-protective properties, has shown promise as a therapeutic agent for IBD. However, clinical application has been constrained by its instability in low pH and the need for parenteral administration. RESULTS: Here, we report the first orally administered, pH-responsive li

Augmenting Antioxidant Enzyme-Like Activity and Hydrogen Evolution of MoS2@Bi2S3 Heterojunction via Ultrasound-Evoked Piezoelectricity for Anti-Inflammation.

Inorganic piezoelectric biomaterials exhibit significant potential for diverse biomedical applications, yet their limited piezoelectric effect has hindered broader utilization. To address this challenge, the study successfully fabricates a heterojunction MoS2@Bi2S3 with exceptional piezoelectric properties through doping engineering. Comprehensive studies reveal that MoS2@Bi2S3 efficiently converts mechanical energy into electrical energy, facilitates hydrogen (H2) evolution, and enhances the antioxidant enzyme-like performances under ultrasonic irradiation. Moreover, a GSH and GSSG switch cyc

Ultra-Low Hysteresis Under Large Deformation Enabled by Fast Chains Relaxation in Highly Competitive Dynamic Hydrogen Bond Networks.

High-quality capture of diverse motion signals in flexible sensors requires soft sensing materials to perform signal conversion and transmission stably and without delay over extended periods. However, low hysteresis achieved through purely elastic mechanisms easily exhibits poor crack propagation resistance. By leveraging the significant gap between the strain rate during large-strain service conditions and the crack propagation rate in the fracture process, this study presents a facile strategy for constructing a highly competitive dynamic hydrogen bonding system to produce near-zero-hystere

Exogenous hydrogen sulfide improves non-alcoholic fatty liver disease by inhibiting endoplasmic reticulum stress/NLRP3 inflammasome pathway.

Non-alcoholic fatty liver disease (NAFLD) is a common chronic liver disease worldwide, and its exact pathogenesis has not been fully studied. Hydrogen sulfide (H2S) is the third gas signaling molecule discovered in mammals, following nitric oxide and carbon monoxide. It has the effects of anti-inflammation, anti-apoptosis, and so on, thereby playing an important role in many diseases. However, the role and mechanism of exogenous H2S in NAFLD are not fully understood. In this study, we constructed in vitro and in vivo NAFLD models by feeding mice a high-fat diet and stimulating hepatocytes with

Blockade of Aryl Hydrocarbon Receptor Ameliorates Functional Insufficiency in 5/6 Nephrectomized Rat Kidneys by Restoring Hydrogen Sulfide Formation.

Aims: We previously demonstrated that aryl hydrocarbon receptor (AhR) activation attenuates the cytoprotective effect of hydrogen sulfide (H2S), leading to indoxyl sulfate (IS)-mediated renal tubular damage. However, it is unclear whether this pathway would be present in an in vivo uremic model. Results: In a rat chronic kidney disease (CKD) model with 5/6 nephrectomized (Nx), we found that poor renal filtration is associated with accumulation of IS and homocysteine (Hcy), an H2S precursor. Compared with controls, the protein and mRNA levels of H2S-producing enzymes, including cystathionine β-

Molecular Hydrogen Ameliorates Anti-Desmoglein 1 Antibody-Induced Pemphigus-Associated Interstitial Lung Disease by Inhibiting Oxidative Stress.

Pemphigus-associated interstitial lung disease (P-ILD) is a severe complication observed in pemphigus patients that is characterized by pulmonary interstitial inflammation and fibrosis. This study investigated the role of anti-desmoglein (Dsg) 1/3 antibodies in P-ILD pathogenesis and evaluated the therapeutic potential of molecular hydrogen (H2). Using a BALB/cJGpt mouse model, we demonstrated that anti-Dsg 1 antibodies, but not anti-Dsg 3 antibodies, induced interstitial inflammation and fibrosis. Immunofluorescence staining confirmed IgG deposition in the alveolar epithelium, suggesting immu

Hydrogen sulfide attenuates sepsis-induced cardiac dysfunction in infant rats by inhibiting the expression of cold-inducible RNA-binding protein.

Sepsis-induced cardiac dysfunction is one of the most common complications of sepsis. It is also a major cause of death in pediatric intensive care units. The underlying mechanism of sepsis-induced cardiac dysfunction remains elusive. Cold-inducible RNA-binding protein (CIRP) is a damage-associated molecular pattern that is up-regulated during sepsis. Hydrogen sulfide (H2S) has been shown to play a protective role in sepsis-induced cardiac dysfunction in adult animals. The present study aimed to determine whether H2S ameliorates the cardiac function in infant rats by inhibiting CIRP-mediated s
